    Characterisation of the Cell Line HC-AFW1 Derived from a Pediatric Hepatocellular Carcinoma

    Current treatment of paediatric hepatocellular carcinoma (HCC) is often inefficient due to advanced disease at diagnosis and resistance to common drugs. The aim of this study was to generate a cell line derived from a paediatric HCC in order to expand research in this field. We established the HC-AFW1 cell line from a liver neoplasm of a 4-year-old boy through culturing of primary tumor specimens. The cell line has been stable for over one year of culturing and has a doubling time of 40 h. The tumour cells have an epithelial histology and express HCC-associated proteins such as Alpha-fetoprotein (AFP), Glypican 3, E-cadherin, CD10, CD326, HepPar1 and Vimentin. Forty-nine amino acids in exon 3 of β-Catenin that involve the phosphorylation sites of GSK3 were absent and β-Catenin is detectable in the cell nuclei. Cytogenetic analysis revealed large anomalies in the chromosomal map. Several alterations of gene copy numbers were detected by genome-wide SNP array. Among the different drugs tested, cisplatin and irinotecan showed effective inhibition of tumour cell growth in a proliferation assay at concentrations below 5 µg/ml. Subcutaneous xenotransplantation of HC-AFW1 cells into NOD/SCID mice resulted in fast growing dedifferentiated tumours with high levels of serum AFP. Histological analyses of the primary tumour and xenografts included national and international expert pathological review. Consensus reading characterised the primary tumour and the HC-AFW1-derived tumours as HCC. HC-AFW1 is the first cell line derived from a paediatric HCC without a background of viral hepatitis or cirrhosis and represents a valuable tool for investigating the biology of and therapeutic strategies for childhood HCC

    Primary pancreatic lymphoma – pancreatic tumours that are potentially curable without resection, a retrospective review of four cases

    BACKGROUND: Primary pancreatic lymphomas (PPL) are rare tumours of the pancreas. Symptoms, imaging and tumour markers can mimic pancreatic adenocarcinoma, but they are much more amenable to treatment. Treatment for PPL remains controversial, particularly the role of surgical resection. METHODS: Four cases of primary pancreatic lymphoma were identified at Prince of Wales Hospital, Sydney, Australia. A literature review of cases of PPL reported between 1985 and 2005 was conducted, and outcomes were contrasted. RESULTS: All four patients presented with upper abdominal symptoms associated with weight loss. One case was diagnosed without surgery. No patients underwent pancreatectomy. All patients were treated with chemotherapy and radiotherapy, and two of four patients received rituximab. One patient died at 32 months. Three patients are disease free at 15, 25 and 64 months, one after successful retreatment. Literature review identified a further 103 patients in 11 case series. Outcomes in our series and other series of chemotherapy and radiotherapy compared favourably to surgical series. CONCLUSION: Biopsy of all pancreatic masses is essential, to exclude potentially curable conditions such as PPL, and can be performed without laparotomy. Combined multimodality treatment, utilising chemotherapy and radiotherapy, without surgical resection is advocated but a cooperative prospective study would lead to further improvement in treatment outcomes

    Methodology of calculation of construction and hydrodynamic parameters of a foam layer apparatus for mass-transfer processes

    Промислова реалізація методу стабілізації газорідинного шару дозволяє значно розширити галузь застосування пінних апаратів і відкриває нові можливості інтенсифікації технологічних процесів з одночасним створенням маловідходних технологій. У статті встановлені основні параметри, що впливають на гідродинаміку пінних апаратів, розглянуті основні конструкції та режими роботи пінних апаратів. Виявлено зв'язок гідродинамічних параметрів. Розглянуто гідродинамічні закономірності пінного шару. Вказані фактори, що впливають на процес масообміну, як в газовій, так і в рідкій фазах. Проведений аналіз ряду досліджень показав, що перспективним напрямком інтенсифікації процесу масообміну є розробка апаратів з трифазним псевдозрідженим шаром зрошуваної насадки складних форм із сітчастих матеріалів. Отже, необхідне проведення спеціальних досліджень гідродинамічних режимів роботи апарату з сітчастою насадкою і визначенням параметрів, що впливають на швидкість переходу насадки з одного режиму в інший.Industrial implementation of the stabilization method of the gas-liquid layer can significantly expand the field of use of foaming apparatus and opens up new opportunities for intensifying technological processes with the simultaneous creation of low-waste technologies. The article establishes the basic parameters influencing the hydrodynamics of foam apparatus, considers the basic constructions and operating modes of foam apparatus. The connection of hydrodynamic parameters is revealed. The hydrodynamic laws of the foam layer are considered. The indicated factors affecting the process of mass transfer, both in the gas and in the liquid phases. The conducted analysis of a number of studies showed that the perspective direction of intensification of the mass transfer process is the development of apparatuses with a three-phase fluidized bed of an irrigated nozzle of complex forms with mesh materials

    Objective: The evaluation of medical research performance is a key prerequisite for the systematic advancement of medical faculties, research foci, academic departments, and individual scientists' careers. However, it is often based on vaguely defined aims and questionable methods and can thereby lead to unwanted regulatory effects. The current paper aims at defining the position of German academic medicine toward the aims, methods, and consequences of its evaluation. Methods: During the Berlin Forum of the Association of the Scientific Medical Societies in Germany (AWMF) held on 18 October 2013, international experts presented data on methods for evaluating medical research performance. Subsequent discussions among representatives of relevant scientific organizations and within three ad-hoc writing groups led to a first draft of this article. Further discussions within the AWMF Committee for Evaluation of Performance in Research and Teaching and the AWMF Executive Board resulted in the final consented version presented here.Results: The AWMF recommends modifications to the current system of evaluating medical research performance. Evaluations should follow clearly defined and communicated aims and consist of both summative and formative components. Informed peer reviews are valuable but feasible in longer time intervals only. They can be complemented by objective indicators. However, the Journal Impact Factor is not an appropriate measure for evaluating individual publications or their authors. The scientific "impact" rather requires multidimensional evaluation. Indicators of potential relevance in this context may include, e.g., normalized citation rates of scientific publications, other forms of reception by the scientific community and the public, and activities in scientific organizations, research synthesis and science communication. In addition, differentiated recommendations are made for evaluating the acquisition of third-party funds and the promotion of junior scientists.Conclusions: With the explicit recommendations presented in the current position paper, the AWMF suggests enhancements to the practice of evaluating medical research performance by faculties, ministries and research funding organizations.Ziele: Die Evaluation medizinischer Forschungsleistung ist eine zentrale Voraussetzung für die gezielte Weiterentwicklung von Fakultäten, Forschungsschwerpunkten, Instituten bzw. Kliniken sowie Wissenschaftlerkarrieren. Sie basiert aber vielfach auf unklaren Zielvorstellungen und fragwürdigen Methoden und kann zu Fehlsteuerungen führen. Ziel der vorliegenden Arbeit ist eine Positionsbestimmung der wissenschaftlichen Medizin gegenüber den Zielen, Methoden und Konsequenzen ihrer Evaluation. Methoden: Im Rahmen des Berliner Forums der AWMF vom 18.10.2013 wurden von internationalen Experten Befunde zur Methodik der Evaluation medizinischer Forschungsleistung referiert, von Vertretern relevanter Wissenschaftsorganisationen diskutiert und in drei Adhoc-Schreibgruppen in einen Textentwurf gefasst. Dieser wurde mehrfach in der AWMF-Kommission für Leistungsevaluation in Forschung und Lehre sowie im AWMF-Präsidium diskutiert und in der vorliegenden finalen Fassung konsentiert.Ergebnisse: Die AWMF empfiehlt eine Veränderung des Systems der Bewertung medizinischer Forschungsleistungen. Evaluationen sollten eine a priori klar definierte und kommunizierte Zielsetzung besitzen und neben summativen auch formative Elemente enthalten. Neben sinnvollen aber nur in längeren Intervallen praktikablen informierten peer reviews können auch objektive Indikatoren zur Evaluation eingesetzt werden. Die Verwendung des Journal Impact Factors zur Bewertung einzelner Publikationen oder ihrer Autoren wird dabei abgelehnt. Der wissenschaftliche "Impact" erfordert vielmehr eine multidimensionale Beurteilung, in der z.B. normierte Zitationsraten wissenschaftlicher Publikationen ebenso Berücksichtigung finden wie andere Formen der Rezeption in Fachwelt und Öffentlichkeit sowie Tätigkeiten im Bereich der Wissenschaftsorganisation, -synthese und -kommunikation. Daneben werden Vorschläge zu einer differenzierten Bewertung von Drittmitteleinwerbung und Nachwuchsförderung gemacht.Folgerungen: Mit den konkreten Empfehlungen des vorliegenden Positionspapiers legt die AWMF den Fakultäten, Ministerien und Organisationen der Forschungsförderung eine Weiterentwicklung der Evaluationspraxis medizinischer Forschungsleistungen nahe

    Microsatellite markers reveal multiple paternity and sperm storage in the Mediterranean spur-thighed tortoise, Testudo graeca

    The spur-thighed tortoise (Testudo graeca) is a terrestrial species in which multiple mating is frequently observed. We assessed the probability of multiple paternity in clutches (n = 15) laid by eight females, including successive clutches. Paternity was determined by microsatellite analyses at three loci. A large number of alleles per locus (n = 15-22) provided high probabilities of detecting multiple paternity, particularly at all loci combined (P = 0.989). Multiple paternity was found in 20% of the clutches in which offspring displayed more than two paternal alleles. However, this frequency may have been underestimated, given the small clutch sizes and the few loci used. Also, T. graeca is able to store sperm from single or multiple matings and can use it to fertilize subsequent clutches of eggs, as indicated by the fact that the second clutch of a captive female was sired by a different male and that clutches of another female were multiply sired by the same males. These results confirm that multiple paternity exists in T. graeca and that sperm storage in this species may be an important reproductive strategy to fertilize multiple clutches per year.Peer Reviewe
