    Global secular changes in different tidal high water, low water and range levels

    Tides exert a major control on the coastal zone by influencing high sea levels and coastal flooding, navigation, sediment dynamics, and ecology. Therefore, any changes to tides have wide ranging and important implications. In this paper, we uniquely assess secular changes in 15 regularly used tidal levels (five high water, five low water and five tidal ranges), which have direct practical applications. Using sea level data from 220 tide gauge sites, we found changes have occurred in all analyzed tidal levels in many parts of the world. For the tidal levels assessed, between 36% and 63% of sites had trends significantly different (at 95% confidence level) from zero. At certain locations, the magnitude of the trends in tidal levels were similar to trends in mean sea level over the last century, with observed changes in tidal range and high water levels of over 5 mm/yr and 2 mm/yr, respectively. More positive than negative trends were observed in tidal ranges and high water levels, and vice versa for low water levels. However we found no significant correlation between trends in mean sea level (MSL) and any tidal levels. Spatially coherent trends were observed in some regions, including the north-east Pacific, German Bight and Australasia, and we also found that differences in trends occur between different tidal levels. This implies that analyzing different tidal levels is important. Because changes in the tide are widespread and of similar magnitude to MSL rise at a number sites, changes in tides should be considered in coastal risk assessments

    Urinary catheters: history, current status, adverse events and research agenda

    For more than 3500 years, urinary catheters have been used to drain the bladder when it fails to empty. For people with impaired bladder function and for whom the method is feasible, clean intermittent self-catheterization is the optimal procedure. For those who require an indwelling catheter, whether short- or long-term, the self-retaining Foley catheter is invariably used, as it has been since its introduction nearly 80 years ago, despite the fact that this catheter can cause bacterial colonization, recurrent and chronic infections, bladder stones and septicaemia, damage to the kidneys, the bladder and the urethra, and contribute to the development of antibiotic resistance. In terms of medical, social and economic resources, the burden of urinary retention and incontinence, aggravated by the use of the Foley catheter, is huge. In the UK, the harm resulting from the use of the Foley catheter costs the National Health Service between ÂŁ1.0–2.5 billion and accounts for ∌2100 deaths per year. Therefore, there is an urgent need for the development of an alternative indwelling catheter system. The research agenda is for the new catheter to be easy and safe to insert, either urethrally or suprapubically, to be retained reliably in the bladder and to be withdrawn easily and safely when necessary, to mimic natural physiology by filling at low pressure and emptying completely without damage to the bladder, and to have control mechanisms appropriate for all users

    The Swift X-ray flaring afterglow of GRB 050607

    The unique capability of the Swift satellite to perform a prompt and autonomous slew to a newly detected Gamma-Ray Burst (GRB) has yielded the discovery of interesting new properties of GRB X-ray afterglows, such as the steep early lightcurve decay and the frequent presence of flares detected up to a few hours after the GRB trigger. We present observations of GRB 050607, the fourth case of a GRB discovered by Swift with flares superimposed on the overall fading X-ray afterglow. The flares of GRB 050607 were not symmetric as in previously reported cases, showing a very steep rise and a shallower decay, similar to the Fast Rise, Exponential Decay that are frequently observed in the gamma-ray prompt emission. The brighter flare had a flux increase by a factor of approximately 25,peaking for 30 seconds at a count rate of approximately 30 counts s-1, and it presented hints of addition short time scale activity during the decay phase. There is evidence of spectral evolution during the flares. In particular, at the onset of the flares the observed emission was harder, with a gradual softening as each flare decayed. The very short time scale and the spectral variability during the flaring activity are indicators of possible extended periods of energy emission by the GRB central engine. The flares were followed by a phase of shallow decay, during which the forward shock was being refreshed by a long-lived central engine or by shells of lower Lorentz factors, and by a steepening after approximately 12 ks to a decay slope considered typical of X-ray afterglows.Comment: 23 pages, 5 figures, Accepted by the Astrophysical Journa

    Biochemical studies on Francisella tularensis RelA in (p)ppGpp Biosynthesis

    2 ABSTRACT The bacterial stringent response is induced by nutrient deprivation and is mediated by enzymes of the RSH superfamily that control concentrations of the "alarmones" (p)ppGpp. This regulatory pathway is present in the vast majority of pathogens and has been proposed as a potential antibacterial target. Current understanding of RelA mediated responses are based on biochemical studies using Escherichia coli as a model. In comparison, the Francisella tularensis RelA sequence contains a truncated regulatory C-terminal region and an unusual synthetase motif (EXSD). Biochemical analysis of Francisella tularensis RelA showed the similarities and differences of this enzyme compared to the model RelA from Escherichia coli. Purification of the enzyme yielded a stable dimer capable of reaching concentrations of 10 mg/mL. In contrast to other enzymes from the RelA/SpoT homologue superfamily, activity assays with F. tularensis RelA demonstrate a high degree of specificity for GTP as a pyrophosphate acceptor, with no measurable turnover for GDP. Steady state kinetic analysis of F. tularensis RelA gave saturation activity curves that best fitted a sigmoidal function. This kinetic profile can result from allosteric regulation and further measurements with potential allosteric regulators demonstrated activation by ppGpp with an EC 50 of 60 ± 1.9 ΌM. Activation of F. tularensis RelA by stalled ribosomal complexes formed with ribosomes purified from Escherichia coli MRE600 was observed, but interestingly, significantly weaker activation with ribosomes isolated from Francisella philomiragia

    ENU Mutagenesis Reveals a Novel Phenotype of Reduced Limb Strength in Mice Lacking Fibrillin 2

    Background: Fibrillins 1 (FBN1) and 2 (FBN2) are components of microfibrils, microfilaments that are present in many connective tissues, either alone or in association with elastin. Marfan's syndrome and congenital contractural arachnodactyly (CCA) result from dominant mutations in the genes FBN1 and FBN2 respectively. Patients with both conditions often present with specific muscle atrophy or weakness, yet this has not been reported in the mouse models. In the case of Fbn1, this is due to perinatal lethality of the homozygous null mice making measurements of strength difficult. In the case of Fbn2, four different mutant alleles have been described in the mouse and in all cases syndactyly was reported as the defining phenotypic feature of homozygotes.Methodology/Principal Findings: As part of a large-scale N-ethyl-N-nitrosourea (ENU) mutagenesis screen, we identified a mouse mutant, Mariusz, which exhibited muscle weakness along with hindlimb syndactyly. We identified an amber nonsense mutation in Fbn2 in this mouse mutant. Examination of a previously characterised Fbn2-null mutant, Fbn2(fp), identified a similar muscle weakness phenotype. The two Fbn2 mutant alleles complement each other confirming that the weakness is the result of a lack of Fbn2 activity. Skeletal muscle from mutants proved to be abnormal with higher than average numbers of fibres with centrally placed nuclei, an indicator that there are some regenerating muscle fibres. Physiological tests indicated that the mutant muscle produces significantly less maximal force, possibly as a result of the muscles being relatively smaller in Mariusz mice.Conclusions: These findings indicate that Fbn2 is involved in integrity of structures required for strength in limb movement. As human patients with mutations in the fibrillin genes FBN1 and FBN2 often present with muscle weakness and atrophy as a symptom, Fbn2-null mice will be a useful model for examining this aspect of the disease process further

    Twist-bend nematics, liquid crystal dimers, structure–property relations

    <p>One of the current challenges in liquid crystal science is to understand the molecular factors leading to the formation of the intriguing twist-bend nematic phase (N<sub>TB</sub>) and determine its properties. During our earlier hunt for the N<sub>TB</sub> phase created on cooling directly from the isotropic phase and not the nematic phase, we had prepared 30 symmetric liquid crystal dimers. These had odd spacers and methylene links to the two mesogenic groups; desirable but clearly not essential features for the formation of the N<sub>TB</sub>. Here, we report the phases that the dimers exhibit and their transition temperatures as functions of both the lengths of the spacer and the terminal chains. In addition we describe the transitional entropies, their optical textures, the X-ray scattering patterns and the <sup>2</sup>H NMR spectra employed in characterising the phases. All of which may lead to important properties of the twist-bend nematic phase.</p

    Towards a consensus-based classification of childhood arterial ischemic stroke.

    Background and purposeThe implementation of uniform nomenclature and classification in adult arterial ischemic stroke (AIS) has been critical for defining outcomes and recurrence risks according to etiology and in developing risk-stratified treatments. In contrast, current classification and nomenclature in childhood AIS are often overlapping or contradictory. Our purpose was to develop a comprehensive consensus-based classification system for childhood AIS.MethodsUsing a modified-Delphi method, members of the International Pediatric Stroke Study (IPSS) developed the Childhood AIS Standardized Classification And Diagnostic Evaluation (CASCADE) criteria. Two groups of pediatric stroke specialists from the IPSS classified 7 test cases using 2 methods each: (1) classification typical of the individual clinician's current clinical practice; and (2) classification based on the CASCADE criteria. Group 1 underwent in-person training in the utilization of the CASCADE criteria. Group 2 classified the same cases via an online survey, including definitions but without training. Inter-rater reliability (IRR) was assessed via multi-rater unweighted Îș-statistic.ResultsIn Group 1 (with training), IRR was improved using CASCADE criteria (Îș=0.78, 95% CI=[0.49, 0.94]), compared with typical clinical practice (Îș=0.40, 95% CI=[0.11, 0.60]). In Group 2 (without training), IRR was lower than among trained raters (Îș=0.61, 95% CI=[0.29, 0.77]), but higher than current practice (Îș=0.23, 95% CI=[0.03, 0.36]).ConclusionsA new, consensus-based classification system for childhood AIS, the CASCADE criteria, can be used to classify cases with good IRR. These preliminary findings suggest that the CASCADE criteria may be particularity useful in the setting of prospective multicenter studies in childhood-onset AIS, where standardized training of investigators is feasible

    Inter-Rater Reliability of the CASCADE Criteria: Challenges in Classifying Arteriopathies.

    Background and purposeThere are limited data about the reliability of subtype classification in childhood arterial ischemic stroke, an issue that prompted the IPSS (International Pediatric Stroke Study) to develop the CASCADE criteria (Childhood AIS Standardized Classification and Diagnostic Evaluation). Our purpose was to determine the CASCADE criteria's reliability in a population of children with stroke.MethodsEight raters from the IPSS reviewed neuroimaging and clinical records of 64 cases (16 cases each) randomly selected from a prospectively collected cohort of 113 children with arterial ischemic stroke and classified them using the CASCADE criteria. Clinical data abstracted included history of present illness, risk factors, and acute imaging. Agreement among raters was measured by unweighted Îș statistic.ResultsThe CASCADE criteria demonstrated a moderate inter-rater reliability, with an overall Îș statistic of 0.53 (95% confidence interval [CI]=0.39-0.67). Cardioembolic and bilateral cerebral arteriopathy subtypes had much higher agreement (Îș=0.84; 95% CI=0.70-0.99; and Îș=0.90; 95% CI=0.71-1.00, respectively) than cases of aortic/cervical arteriopathy (Îș=0.36; 95% CI=0.01-0.71), unilateral focal cerebral arteriopathy of childhood (FCA; Îș=0.49; 95% CI=0.23-0.76), and small vessel arteriopathy of childhood (Îș=-0.012; 95% CI=-0.04 to 0.01).ConclusionsThe CASCADE criteria have moderate reliability when used by trained and experienced raters, which suggests that it can be used for classification in multicenter pediatric stroke studies. However, the moderate reliability of the arteriopathic subtypes suggests that further refinement is needed for defining subtypes. Such revisions may reduce the variability in the literature describing risk factors, recurrence, and outcomes associated with childhood arteriopathy
