    PMI: A Delta Psi(m) Independent Pharmacological Regulator of Mitophagy

    Mitophagy is central to mitochondrial and cellular homeostasis and operates via the PINK1/Parkin pathway targeting mitochondria devoid of membrane potential (Διm) to autophagosomes. Although mitophagy is recognized as a fundamental cellular process, selective pharmacologic modulators of mitophagy are almost nonexistent. We developed a compound that increases the expression and signaling of the autophagic adaptor molecule P62/SQSTM1 and forces mitochondria into autophagy. The compound, P62-mediated mitophagy inducer (PMI), activates mitophagy without recruiting Parkin or collapsing Διm and retains activity in cells devoid of a fully functional PINK1/Parkin pathway. PMI drives mitochondria to a process of quality control without compromising the bio-energetic competence of the whole network while exposing just those organelles to be recycled. Thus, PMI circumvents the toxicity and some of the nonspecific effects associated with the abrupt dissipation of Διm by ionophores routinely used to induce mitophagy and represents a prototype pharmacological tool to investigate the molecular mechanisms of mitophagy

    Soil survey of Vasse Research Station, Western Australia

    This soil survey of the Vasse Research Station properties was carried out at the request of the Division of Plant Production of the Agriculture Western Australia. The survey followed a preliminary examination and mapping of soil series in the area by Mr T. Stoneman in 1982, after which it was concluded that more detailed survey work was required to assist farm management and research experimentation. The research station consists of two blocks, referred to here as the main block (Sussex Locations 1475, 1439, 2128, 2138, 2137, 2136, 2129, 2130, 2119, 2118, 2126 and 2125) and the hill block (Sussex Locations 3024, 3025 and part of 1864) approximately 12 km due south, and 15 km south-south-east respectively from Busselton. (Figure 1). The main (lowland) portion of the research station is approximately 610 ha in area and the hill block further south covers approximately 160 ha. The country is considered to be representative of much of the inland Swan Coastal Plain and its margin with the Whicher Range between Boyanup and Dunsborough. The work was conducted using a free survey technique where map unit boundaries and field observation sites were selected on the basis of air photo interpretation. Aerial photos enlarged to scales of 1:10,000 and 1:2000 respectively were used for the main and hill blocks. The final map presented in this report is at a uniform scale of 1:10,000.     Local and remote climate impacts of future African aerosol emissions

    The potential future trend in African aerosol emissions is uncertain, with a large range found in future scenarios used to drive climate projections. The future climate impact of these emissions is therefore uncertain. Using the Shared Socioeconomic Pathway (SSP) scenarios, transient future experiments were performed with the UK Earth System Model (UKESM1) to investigate the effect of African emissions following the high emission SSP370 scenario as the rest of the world follows the more sustainable SSP119, relative to a global SSP119 control. This isolates the effect of Africa following a relatively more polluted future emissions pathway. Compared to SSP119, SSP370 projects higher non-biomass-burning (non-BB) aerosol emissions, but lower biomass burning emissions, over Africa. Increased shortwave (SW) absorption by black carbon aerosol leads to a global warming, but the reduction in the local incident surface radiation close to the emissions is larger, causing a local cooling effect. The local cooling persists even when including the higher African CO2 emissions under SSP370 than SSP119. The global warming is significantly higher by 0.07 K when including the non-BB aerosol increases and higher still (0.22 K) when including all aerosols and CO2. Precipitation also exhibits complex changes. Northward shifts in the Inter-tropical Convergence Zone (ITCZ) occur under relatively warm Northern Hemisphere land, and local rainfall is enhanced due to mid-tropospheric instability from black carbon absorption. These results highlight the importance of future African aerosol emissions for regional and global climate and the spatial complexity of this climate influence

    The importance of tau leptons for supersymmetry searches at the Tevatron

    Supersymmetry is perhaps most effectively probed at the Tevatron through production and decay of weak gauginos. Most of the analyses of weak gaugino observables require electrons or muons in the final state. However, it is possible that the gauginos will decay primarily to tau leptons, thus complicating the search for supersymmetry. The motivating reasons for high tau multiplicity final states are discussed in three approaches to supersymmetry model building: minimal supergravity, gauge mediated supersymmetry breaking, and more minimal supersymmetry. The concept of ``e/mu/tau candidate'' is introduced, and an observable with three e/mu/tau candidates is defined in analog to the trilepton observable. The maximum mass reach for supersymmetry is then estimated when gaugino decays to tau leptons have full branching fraction.Comment: 9 pages, latex, 2 figures.     Are adults just big kids? Can the newer paediatric weight estimation systems be used in adults?

    Background. The weight of a patient is an important variable that impacts on their medical care. Although some drugs are prescribed on a so-called ‘adult dose’ basis, we know that adults come in all shapes and sizes – a ‘one-dose-fits-all’ approach is not necessarily appropriate. As a measured weight may not always be available, an alternative method of accurately estimating weight is required.Objectives. To assess and compare the accuracy of weight estimations in adults by patient self-estimation, the Mercy method, Buckley method, Broca index and PAWPER XL-MAC (paediatric advanced weight prediction in the emergency room eXtra length/eXtra large mid-arm circumference) method.Methods. This was a prospective, cross-sectional study conducted at a tertiary academic hospital in a metropolitan area of Johannesburg, South Africa. Anthropometric variables of height, abdominal circumference, thigh circumference, mid-arm circumference and humeral length were measured. These variables were then applied to the various weight estimation methods and compared with the patient’s actual weight.Results. There were 188 adult patients included in the study. None of the methodologies evaluated in this study achieved the recommended >70% of weight estimations within 10% of the patient’s actual weight (PW10). The Mercy method was the closest to achieving greater than the recommended 95% for weight estimation falling within 20% of the patient’s actual weight (PW20). The PW20 for the Mercy method was 91.5%. The PAWPER XL-MAC and patient self-estimate methods achieved a PW20 of 85.1% and 86.1%, respectively. The Broca and Buckley methods performed poorly overall.Conclusions. None of the evaluated weight estimation methodologies was accurate enough for use in adult weight estimation. The Mercy and PAWPER XL-MAC methodologies both showed significant promise for use in adult weight estimation, but need further refinement. Although patient self-estimates were similarly accurate to those found in previous studies, they were not an accurate option; self-estimations would remain the first choice if the patient was able to provide such an estimation. The Broca index and Buckley method cannot be recommended owing to their poor performance

    Movement correction in DCE-MRI through windowed and reconstruction dynamic mode decomposition

    Images of the kidneys using dynamic contrast enhanced magnetic resonance renography (DCE-MRR) contains unwanted complex organ motion due to respiration. This gives rise to motion artefacts that hinder the clinical assessment of kidney function. However, due to the rapid change in contrast agent within the DCE-MR image sequence, commonly used intensity-based image registration techniques are likely to fail. While semi-automated approaches involving human experts are a possible alternative, they pose significant drawbacks including inter-observer variability, and the bottleneck introduced through manual inspection of the multiplicity of images produced during a DCE-MRR study. To address this issue, we present a novel automated, registration-free movement correction approach based on windowed and reconstruction variants of dynamic mode decomposition (WR-DMD). Our proposed method is validated on ten different healthy volunteers’ kidney DCEMRI data sets. The results, using block-matching-block evaluation on the image sequence produced by WR-DMD, show the elimination of 99% of mean motion magnitude when compared to the original data sets, thereby demonstrating the viability of automatic movement correction using WR-DMD

    Intention of preserving forest remnants among landowners in the Atlantic Forest: The role of the ecological context via ecosystem services

    Unravelling the psychological processes determining landowners' support towards forest conservation is crucial, particularly in rural areas of the tropics, where most forest remnants are within private lands. As human–nature connections are known to shape pro‐environmental behaviours, the intention of preserving forest remnants should ultimately be determined by the ecological context people live in. Here, we investigate the pathways through which the ecological context (forest cover), via direct contact with forests and ecosystem services and disservices, influence the psychological antecedents of conservation behaviour (beliefs, attitude and intention of preserving forest remnants). We conceptualized a model based on the Reasoned Action Approach, using the ecological context and these three forest experiences as background factors, and tested the model using Piecewise Structural Equation Modelling. Data were collected through an interview‐based protocol applied to 106 landowners across 13 landscapes varying in forest cover in a consolidated rural region in the Brazilian Atlantic Forest. Our results indicate that: (a) ecosystem services are more important than disservices for shaping intention of preserving forests, particularly non‐provisioning services; (b) contact with forest has an indirect effect on intention, by positively influencing the frequency of receiving ecosystem services; (c) people living in more forested ecological contexts have more contact with forests, receive ecosystem services more frequently and, ultimately, have stronger intention of preserving forests. Hence, our study suggests a dangerous positive feedback loop between deforestation, the extinction of forest experiences and impairment of human–nature connections. Local demands across the full range of ecosystem services, the balance between services and disservices and the ecological context people live in should be considered when developing conservation initiatives in tropical rural areas

    Measurement of the charged pion mass using X-ray spectroscopy of exotic atoms

    The 5g−4f5g-4f transitions in pionic nitrogen and muonic oxygen were measured simultaneously by using a gaseous nitrogen-oxygen mixture at 1.4\,bar. Due to the precise knowledge of the muon mass the muonic line provides the energy calibration for the pionic transition. A value of (139.57077\,±\pm\,0.00018)\,MeV/c2^{2} (±\pm\,1.3ppm) is derived for the mass of the negatively charged pion, which is 4.2ppm larger than the present world average
