    Cumulative stress restricts niche filling potential of habitat-forming kelps in a future climate

    1. Climate change is driving range contractions and local population extinctions across the globe. When this affects ecosystem engineers the vacant niches left behind are likely to alter the wider ecosystem unless a similar species can fulfil them. 2. Here, we explore the stress physiology of two coexisting kelps undergoing opposing range shifts in the Northeast Atlantic and discuss what differences in stress physiology may mean for future niche filling. 3. We used chlorophyll florescence (Fv/Fm) and differentiation of the heat shock response (HSR) to determine the capacity of the expanding kelp, Laminaria ochroleuca, to move into the higher shore position of the retreating kelp, Laminaria digitata. We applied both single and consecutive exposures to immersed and emersed high and low temperature treatments, replicating low tide exposures experienced in summer and winter. 4. No interspecific differences in HSR were observed which was surprising given the species? different biogeographic distributions. However, chlorophyll florescence revealed clear differences between species with L. ochroleuca better equipped to tolerate high immersed temperatures but showed little capacity to tolerate frosts or high emersion temperatures. 5. Many patterns observed were only apparent after consecutive exposures. Such cumulative effects have largely been overlooked in tolerance experiments on intertidal organisms despite being more representative of the stress experienced in natural habitats. We therefore suggest future experiments incorporate consecutive stress into their design. 6. Climate change is predicted to result in fewer ground frosts and increased summer temperatures. Therefore, L. ochroleuca may be released from its summer cold limit in winter but still be prevented from moving up the shore due to desiccation in the summer. Laminaria ochroleuca will, however, likely be able to move into tidal pools.     Poverty, food insufficiency and HIV infection and sexual behaviour among young rural Zimbabwean women.

    BACKGROUND: Despite a recent decline, Zimbabwe still has the fifth highest adult HIV prevalence in the world at 14.7%; 56% of the population are currently living in extreme poverty. DESIGN: Cross-sectional population-based survey of 18-22 year olds, conducted in 30 communities in south-eastern Zimbabwe in 2007. OBJECTIVE: To examine whether the risk of HIV infection among young rural Zimbabwean women is associated with socio-economic position and whether different socio-economic domains, including food sufficiency, might be associated with HIV risk in different ways. METHODS: Eligible participants completed a structured questionnaire and provided a finger-prick blood sample tested for antibodies to HIV and HSV-2. The relationship between poverty and HIV was explored for three socio-economic domains: ability to afford essential items; asset wealth; food sufficiency. Analyses were performed to examine whether these domains were associated with HIV infection or risk factors for infection among young women, and to explore which factors might mediate the relationship between poverty and HIV. RESULTS: 2593 eligible females participated in the survey and were included in the analyses. Overall HIV prevalence among these young females was 7.7% (95% CI: 6.7-8.7); HSV-2 prevalence was 11.2% (95% CI: 9.9-12.4). Lower socio-economic position was associated with lower educational attainment, earlier marriage, increased risk of depression and anxiety disorders and increased reporting of higher risk sexual behaviours such as earlier sexual debut, more and older sexual partners and transactional sex. Young women reporting insufficient food were at increased risk of HIV infection and HSV-2. CONCLUSIONS: This study provides evidence from Zimbabwe that among young poor women, economic need and food insufficiency are associated with the adoption of unsafe behaviours. Targeted structural interventions that aim to tackle social and economic constraints including insufficient food should be developed and evaluated alongside behaviour and biomedical interventions, as a component of HIV prevention programming and policy

    Integrated Implementation of Scotland's New Human Rights Framework : Workshop Series Findings

    In March 2021, the Scottish Government committed to an ambitious legal framework that will give rise to a new range of duties and a new focus on groups whose rights are the least realised. We know that when the framework comes into effect, its success will depend upon the capacity of public authorities to translate its ambition into lived realities. Human rights and related equality protections have a constitutional quality. They are constitutional in the sense that they aim to regulate and guide the way that state actors (from central to local government and organisations tasked with the delivery of public services) treat people and prioritise their rights in relevant law, policy, and operational decision-making. Being constitutional in this sense means that legal human rights and related equality protections should provide a baseline – a foundation – for the strategic and operational delivery of public services. If these legal protections were introduced to a clean legislative and policy slate, they would act as a foundation stone, and domestic legislation and policy across the spectrum of services would be systematically built upwards from this foundation. This is not, of course, the context in which any law, including human rights and related equality law, is introduced. The new legal framework will be introduced into developed, and in some areas crowded, legal and policy landscapes. This is not negative – there are undoubtedly already many areas of good practice, including approaches which go beyond what human rights and related equality law demands, and the process of integrating human rights and related equality law provides standards against which such good practice can be identified and acknowledged. Further, it is positive that the process of integrating human rights and related equality law will at the same time provide standards against which gaps can be identified, where Scotland has not yet reached the baseline set in international human rights law. The core aim in Scotland is to meet, and actively plan to meet, this baseline as a way of securing better human rights and equality outcomes in people’s daily lives

    Short-range hunters: exploring the function and constraints of water shooting in dwarf gouramis

    Ballistic predation is a rare foraging adaptation: in fishes, most attention has focused on a single genus, the archerfish, known to manipulate water to shoot down prey above the water surface. However, several gourami species also exhibit apparently similar ‘shooting’ behaviour, spitting water up to 5 cm above the surface. In a series of experiments, we explored the shooting behaviour and aspects of its significance as a foraging ability in the dwarf gourami (Trichogaster lalius). We investigated sex differences in shooting abilities to determine whether gourami shooting is related to the sex-specific bubble nest manufacture where males mix air and water at the surface to form bubbles. We found that, actually, both sexes were equally able to shoot and could learn to shoot a novel target. In a second experiment, we presented untrained gouramis with opportunities to shoot at live prey and found they successfully shot down both fruit flies and crickets. Finally, we explored the effect of target height on shooting performance to establish potential constraints of shooting as a foraging ability. The frequency of attempted shots and success of hitting targets decreased with height, whereas latency to shoot increased. We also observed that repeatable individual differences account for variation in these measures of shooting performance. Together, our results provide evidence that gourami shooting has a foraging function analogous to that of archerfish. Gourami shooting may serve as an example of convergent evolution and provide opportunities for comparative studies into the, as yet unexplored, ecology and evolution of shooting in fishes.Peer reviewe

    Habitat Choice and Speciation

    The role of habitat choice in reproductive isolation and ecological speciation has often been overlooked, despite acknowledgement of its ability to facilitate local adaptation. It can form part of the speciation process through various evolutionary mechanisms, yet where habitat choice has been included in models of ecological speciation little thought has been given to these underlying mechanisms. Here, we propose and describe three independent criteria underlying ten different evolutionary scenarios in which habitat choice may promote or maintain local adaptation. The scenarios are the result of all possible combinations of the independent criteria, providing a conceptual framework in which to discuss examples which illustrate each scenario. These examples show that the different roles of habitat choice in ecological speciation have rarely been effectively distinguished. Making such distinctions is an important challenge for the future, allowing better experimental design, stronger inferences and more meaningful comparisons among systems. We show some of the practical difficulties involved by reviewing the current evidence for the role of habitat choice in local adaptation and reproductive isolation in the intertidal gastropod Littorina saxatilis, a model system for the study of ecological speciation, assessing whether any of the proposed scenarios can be reliably distinguished, given current research

    Bovine Mammary Gland Biopsy Techniques

    Bovine mammary gland biopsies allow researchers to collect tissue samples to study cell biology including gene expression, histological analysis, signaling pathways, and protein translation. This article describes two techniques for biopsy of the bovine mammary gland (MG). Three healthy Holstein dairy cows were the subjects. Before biopsies, cows were milked and subsequently restrained in a cattle chute. An analgesic (flunixin meglumine, 1.1 to 2.2 mg/kg of body weight) was administered via jugular intravenous [IV] injection 15-20 min prior to biopsy. For standing sedation, xylazine hydrochloride (0.01-0.05 mg/kg of body weight) was injected via the coccygeal vessels 5-10 min before the procedure. Once adequately sedated, the biopsy site was aseptically prepared and locally anaesthetized with 6 mL of 2% lidocaine hydrochloride via subcutaneous injection. Using aseptic technique, a 2 to 3 cm vertical incision was made using a number 10 scalpel. Core and needle biopsy tools were used. The core biopsy tool was attached to a cordless drill and inserted into the MG tissue through the incision using a clock-wise drill action. The needle biopsy tool was manually inserted into the incision site. Immediately after the procedure, an assistant applied pressure on the incision site for 20 to 25 min using a sterile towel to achieve hemostasis. Stainless steel surgical staples were used to oppose the skin incision. The staples were removed 10 days post-procedure. The main advantages of core and needle biopsies is that both approaches are minimally invasive procedures that can be safely performed in healthy cows. Milk yield following the biopsy was unaffected. These procedures require a short recovery time and result in fewer risks of complications. Specific limitations may include bleeding after the biopsy and infection on the biopsy site. Applications of these techniques include tissue collection for clinical diagnosis and research purposes, such as primary cell culture

    A detailed phenotypic assessment of individuals affected by MFRP-related oculopathy

    Purpose: To determine the spectrum of mutations and phenotypic variability within patients with mutations in membrane-type frizzled related protein gene (MFRP).Methods: Individuals were initially ascertained based on a phenotype similar to that previously published in association with MFRP mutations. Affected patients underwent a full ophthalmic examination (best-corrected visual acuity, slit-lamp examination, applanation tonometry, and fundoscopy), color fundus photography, optical coherence tomography, autofluorescence imaging, and electrophysiology. MFRP was identified by a genome-wide scan in the fourth-largest autozygous region in one consanguineous family. Sanger sequencing of all the exons and intron-exon boundaries of MFRP was undertaken in the affected individuals.Results: Seven affected individuals from four families were identified as having mutations in MFRP. Patients from two families were homozygous for mutations already previously described (c. 1143_1144 insC and c. 492 delC), while those from the other two were compound heterozygous for mutations (c. 201G>A and c. 491_492 insT, and c. 492 delC, and c. 1622_1625 delTCTG), three of which were novel. There was considerable phenotypic variability within and among families. Autofluorescence imaging revealed the central macula to be relatively well preserved. Foveal cysts and optic nerve head drusen were present in two of the four families. Electrophysiology results showed rod-cone dystrophy with mild to moderate reduction in macular function in all affected members.Conclusions: We report three novel MFRP mutations and expand the phenotypic data available on patients with MFRP mutations

    MHC class I chain-related protein A shedding in chronic HIV-1 infection is associated with profound NK cell dysfunction

    Natural killer (NK) cells play a critical role in host defense against viral infections. However chronic HIV-1 infection is associated with an accumulation of dysfunctional NK cells, that poorly control viral replication. The underlying mechanisms for this NK cell mediated dysfunction are not understood. Certain tumors evade NK cell mediated detection by dampening NK cell activity through the downregulation of NKG2D, via the release of soluble NKG2D-ligands, resulting in a potent suppression of NK cell function. Here we show that chronic HIV-1 infection is associated with a specific defect in NKG2D-mediated NK cell activation, due to reduced expression and transcription of NKG2D. Reduced NKG2D expression was associated with elevated levels of the soluble form of the NKG2D-ligand, MICA, in patient sera, likely released by HIV+CD4+ T cells. Thus, like tumors, HIV-1 may indirectly suppress NK cell recognition of HIV-1-infected CD4+ T cells by enhancing NKG2D-ligand secretion into the serum resulting in a profound impairment of NK cell function
