    Effective Dose and Persistence of Rhodamine-B in Wild Pig Vibrissae

    As a result of substantial ecological and economic damage attributed to wild pigs (Sus scrofa), there is international interest in using pharmaceutical baits to control populations. To assess the efficacy and specificity of baiting programs, chemical biomarkers can be used to evaluate uptake of pharmaceutical baits. Rhodamine B (RB) is known to be an effective biomarker in wild pigs. However, significant data gaps exist regarding the minimum effective dosage and persistence of RB in wild pigs. We used a controlled doubleblind study experiment conducted in spring of 2014 on the Savannah River Site, Aiken, South Carolina, USA, wherein we administered a one-time dose of RB at 3 treatment levels (5 mg/kg, 15mg/kg, or 30 mg/kg) to 15 captive pigs, with 5 pigs/treatment group to investigate persistence of RB. Facial vibrissae were collected pre-RB ingestion as a control and every 2 weeks post-RB ingestion for 12 weeks. We examined samples for RB presence and used a generalized linear mixed model (GLMM) to determine the influence of treatment dose on persistence of RB. Additionally, we measured distance moved by the RB mark away from the vibrissae root and used a GLMM to assess movement rates of RB bands along growing vibrissae. We found consistently greater persistence of RB in the 15- and 30-mg/kg treatments across the sampling period. A significant, positive movement trend in RB bands was observed within the 15mg/kg and 30 mg/kg groups. Based on our results, a 15 mg/kg dosage can be considered a minimum effective dose for wild pigs and will reliably produce a detectable RB mark up to and likely beyond 12 weeks after ingestion

    Spatial Modeling of Common Raven Density and Occurrence Helps Guide Landscape Management Within Great Basin Sagebrush Ecosystems

    Common ravens (Corvus corax; ravens) are a behaviorally flexible nest predator of several avian species, including species of conservation concern. Movement patterns based on life history phases, particularly territoriality of breeding birds and transiency of nonbreeding birds, are thought to influence the frequency and efficacy of nest predation. As such, predicting where on the landscape territorial resident and non-territorial transient birds may be found in relation to the distribution of sensitive prey is of increasing importance to managers and conservationists. From 2007 to 2019, we conducted raven point count surveys between mid-March and mid-September across 43 different field sites representing typical sagebrush (Artemisia spp.) ecosystems of the Great Basin, USA. The surveys conducted during 2007–2016 were used in previously published maps of raven occurrence and density. Here, we examined the relationship between occurrence and density of ravens using spatially explicit predictions from 2 previously published studies and differentiate areas occupied by higher concentrations of resident ravens as opposed to transients. Surveys conducted during 2017–2019 were subsequently used to evaluate the predicted trends from our analytical approach. Specifically, we used residuals from a generalized linear regression to establish the relationship between occurrence and density, which ultimately resulted in a spatially explicit categorical map that identifies areas of resident versus transient ravens. We evaluated mapped categories using independently collected observed raven group sizes from the 2017–2019 survey data, as well as an independent dataset of global positioning system locations of resident and transient individuals monitored during 2019–2020. We observed moderate agreement between the mapped categories and independent datasets for both evaluation approaches. Our map provides broad inference about spatial variation in potential predation risk from ravens for species such as greater sage-grouse (Centrocercus urophasianus) and can be used as a valuable spatial layer for decision support tools aimed at guiding raven management decisions and, ultimately, improving survival and reproduction of sensitive prey within the Great Basin

    Measuring the impact and costs of a universal group based parenting programme : protocol and implementation of a trial

    Background Sub-optimal parenting is a common risk factor for a wide range of negative health, social and educational outcomes. Most parenting programmes have been developed in the USA in the context of delinquency prevention for targeted or indicated groups and the main theoretical underpinning for these programmes is behaviour management. The Family Links Nurturing Programme (FLNP) focuses on family relationships as well as behaviour management and is offered on a universal basis. As a result it may be better placed to improve health and educational outcomes. Developed in the UK voluntary sector, FLNP is popular with practitioners, has impressed policy makers throughout the UK, has been found to be effective in before/after and qualitative studies, but lacks a randomised controlled trial (RCT) evidence base. Methods/Design A multi-centre, investigator blind, randomised controlled trial of the FLNP with a target sample of 288 south Wales families who have a child aged 2-4 yrs living in or near to Flying Start/Sure Start areas. Changes in parenting, parent child relations and parent and child wellbeing are assessed with validated measures immediately and at 6 months post intervention. Economic components include cost consequences and cost utility analyses based on parental ranking of states of quality of life. Attendance and completion rates and fidelity to the FLNP course delivery are assessed. A nested qualitative study will assess reasons for participation and non-participation and the perceived value of the programme to families. By the end of May 2010, 287 families have been recruited into the trial across four areas of south Wales. Recruitment has not met the planned timescales with barriers including professional anxiety about families entering the control arm of the trial, family concern about video and audio recording, programme facilitator concern about the recording of FLNP sessions for fidelity purposes and delays due to the new UK research governance procedures. Discussion Whilst there are strong theoretical arguments to support universal provision of parenting programmes, few universal programmes have been subjected to randomised controlled trials. In this paper we describe a RCT protocol with quantitative and qualitative outcome measures and an economic evaluation designed to provide clear evidence with regard to effectiveness and costs.     The composition of InxGa1 − xN nanorods grown by molecular beam epitaxy with nominal x = 0.5 has been mapped by electron microscopy using Z-contrast imaging and x-ray microanalysis. This shows a coherent and highly strained core-shell structure with a near atomically sharp boundary between a Ga-rich shell (x∼0.3) and an In-rich core (x∼ 0.7), which itself has In- and Ga-rich platelets alternating along the growth axis. It is proposed that the shell and core regions are lateral and vertical growth sectors, with the core structure determined by spinodal decomposition

    SMaRT: A Science-based Tiered Framework for Common Ravens

    Large-scale increases and expansion of common raven (Corvus corax; raven) populations are occurring across much of North America, leading to increased negative consequences for livestock and agriculture, human health and safety, and sensitive species conservation. We describe a science-based adaptive management framework that incorporates recent quantitative analyses and mapping products for addressing areas with elevated raven numbers and minimizing potential adverse impacts to sensitive species, agricultural damage, and human safety. The framework comprises 5 steps: (1) desktop analysis; (2) field assessments; (3) comparison of raven density estimates to an ecological threshold (in terms of either density or density plus distance to nearest active or previous nest); (4) prescribing management options using a 3-tiered process (i.e., habitat improvements, subsidy reductions, and direct actions using StallPOPd.V4 software); and (5) post-management monitoring. The framework is integrated within the Science-based Management of Ravens Tool (SMaRT), a web-based application outfitted with a user-friendly interface that guides managers through each step to develop a fully customized adaptive plan for raven management. In the SMaRT interface, users can: (1) interact with pre-loaded maps of raven occurrence and density and define their own areas of interest within the Great Basin to delineate proposed survey or treatment sites; (2) enter site-level density estimates from distance sampling methods or perform estimation of raven densities using the rapid assessment protocol that we provide; (3) compare site-level density estimates to an identified ecological threshold; and (4) produce a list of potential management options for their consideration. The SMaRT supports decision-making by operationalizing scientific products for raven management and facilitates realization of diverse management goals including sensitive species conservation, protection of livestock and agriculture, safeguarding human health, and addressing raven overabundance and expansion. We illustrate the use of the framework through SMaRT using an example of greater sage-grouse (Centrocercus urophasianus) conservation efforts within the Great Basin, USA

    In Vitro Praziquantel Test Capable of Detecting Reduced In Vivo Efficacy in Schistosoma mansoni Human Infections

    Although great reductions in human schistosomiasis have been observed after praziquantel (PZQ) mass drug administration (MDA), some individuals remain infected after multiple treatments. Many MDA programs now require monitoring for drug efficacy as a key component. No molecular tools for PZQ resistance currently exist and investigations into the dose of PZQ required to kill 50% of adult worms in vivo (ED50) present ethical, logistical, and temporal restraints. We, therefore, assessed the feasibility and accuracy of a rapid, inexpensive in vitro PZQ test in the laboratory and directly in the field in Uganda under MDA in conjunction with highly detailed infection intensity, clearance, and reinfection data. This test strongly differentiated between subsequently cleared and uncleared infections as well as differences between parasite populations pre- and post-PZQ treatments, advocating its use for on-the-spot monitoring of PZQ efficacy in natural foci. After only a few treatments, uncleared parasites were identified to be phenotypically different from drug-sensitive parasites, emphasizing the urgent need for monitoring of these repeatedly PZQ-treated populations

    Synchronous seasonality in the gut microbiota of wild mouse populations

    The gut microbiome performs many important functions in mammalian hosts, with community composition shaping its functional role. However, the factors that drive individual microbiota variation in wild animals and to what extent these are predictable or idiosyncratic across populations remains poorly understood. Here, we use a multi-population dataset from a common rodent species (the wood mouse, Apodemus sylvaticus), to test whether a consistent “core” gut microbiota is identifiable in this species, and to what extent the predictors of microbiota variation are consistent across populations. Between 2014 and 2018 we used capture-mark-recapture and 16S rRNA profiling to intensively monitor two wild wood mouse populations and their gut microbiota, as well as characterising the microbiota from a laboratory-housed colony of the same species. Although the microbiota was broadly similar at high taxonomic levels, the two wild populations did not share a single bacterial amplicon sequence variant (ASV), despite being only 50km apart. Meanwhile, the laboratory-housed colony shared many ASVs with one of the wild populations from which it is thought to have been founded decades ago. Despite not sharing any ASVs, the two wild populations shared a phylogenetically more similar microbiota than either did with the colony, and the factors predicting compositional variation in each wild population were remarkably similar. We identified a strong and consistent pattern of seasonal microbiota restructuring that occurred at both sites, in all years, and within individual mice. While the microbiota was highly individualised, some seasonal convergence occurred in late winter/early spring. These findings reveal highly repeatable seasonal gut microbiota dynamics in multiple populations of this species, despite different taxa being involved. This provides a platform for future work to understand the drivers and functional implications of such predictable seasonal microbiome restructuring, including whether it might provide the host with adaptive seasonal phenotypic plasticity

    Salt intake and iodine status of women in Samoa

    The objective of this study was to determine iodine nutrition status and whether iodine status differs across salt intake levels among a sample of women aged 18-45 years living in Samoa. A cross-sectional survey was completed and 24-hr urine samples were collected and assessed for iodine (n=152) and salt excretion (n=119). The median urinary iodine concentration (UIC) among the women was 88 μg/L (Interquartile range (IQR)=54-121 μg/L). 62% of the women had a UIC <100 μg/L. The crude estimated mean 24-hr urinary salt excretion was 6.6 (standard deviation 3.2) g/day. More than two-thirds (66%) of the women exceeded the World Health Organization recommended maximum level of 5 g/day. No association was found between median UIC and salt excretion (81 μg/L iodine where urinary salt excretion >=5 g/day versus 76 μg/L where urinary salt excretion <5 g/day; p=0.4). Iodine nutrition appears to be insufficient in this population and may be indicative of iodine deficiency disorders in Samoan women. A collaborative approach in monitoring iodine status and salt intake will strengthen both programs and greatly inform the level of iodine fortification required to ensure optimal iodine intake as population salt reduction programs take effect
