    The differential effects of stressors on aversive, novel and appetitive stimuli in rats

    Stressors produce an array of effects on both aversively- and appetitively-motivated behaviors. Because aversive stimuli motivate behaviors in aversively-motivated tasks, assessing stressors effects in these tasks requires re-exposure to aversive stimuli. This is not the case in appetitively-motivated tasks. Based on research that stressors can potentiate responding to subsequent aversive stimuli, the current studies assessed the hypothesis that aversively-motivated behaviors are more sensitive to the effects of stressors than appetitively-motivated behaviors. The studies reported here compared the effects of footshock and restraint in several behavioral procedures that measure responses to aversive and appetitive stimuli. Because of evidence that novel stimuli may also be aversive, the impact of stressors on responding to novel stimuli were also evaluated. CTA studies indicated that footshock enhanced amphetamine CTA (aversive response) and saccharin neophobia (novelty) but not saccharin consumption in saline-injected rats (appetitive response). Runway studies showed that footshock enhanced the response to reward reduction (aversive response) but had not impact on runway responding when reward magnitude was unchanged or increased (appetitive response). Two studies examined the effect of restraint on drug self-administration. Restraint did not influence the acquisition or maintenance of cocaine self-administration, although restraint potentiated cocaine-induced locomotion. Restraint increased ethanol intake on the first post-restraint test only in animals deprived of ethanol on restraint days. Footshock decreased novelty-induced locomotion but only in the initial 5 min of the open-field test. Results of these studies indicate that stressors can alter response to aversive and novel stimuli without altering response to appetitive stimuli. The light/dark emergence test confirmed that Sprague-Dawley rats are more "anxious" than Long-Evans rats. The more reactive Sprague-Dawley rats exhibited a behavioral profile similar to that seen in stressed Long-Evans rats in the earlier studies. These studies support the hypothesis that exposure to stressors preferentially enhance the response to both aversive and novel stimuli without altering responses to appetitive stimuli

    How dark the sky: the JWST backgrounds

    We describe the sources of stray light and thermal background that affect JWST observations; report actual backgrounds as measured from commissioning and early science observations; compare those background levels to pre-launch predictions; estimate the impact of the backgrounds on science performance; and explore how the backgrounds probe the achieved configuration of the deployed observatory. We find the observatory is limited by the irreducible astrophysical backgrounds, rather than scattered stray light and thermal self-emission, for all wavelengths λ<12.5\lambda < 12.5 micron, thus meeting the level 1 requirement. This result was not assured given the open architecture and thermal challenges of JWST, and is the result of meticulous attention to stray light and thermal issues in the design, construction, integration, and test phases. From background considerations alone, JWST will require less integration time in the near-infrared compared to a system that just met the stray light requirements; as such, JWST will be even more powerful than expected for deep imaging at 1--5 micron. In the mid-infrared, the measured thermal backgrounds closely match pre-launch predictions. The background near 10 micron is slightly higher than predicted before launch, but the impact on observations is mitigated by the excellent throughput of MIRI, such that instrument sensitivity will be as good as expected pre-launch. These measured background levels are fully compatible with JWST's science goals and the Cycle 1 science program currently underway.Comment: Submitted to the "JWST Overview" special issue of PAS

    Mining and validating grape (Vitis L.) ESTs to develop EST-SSR markers for genotyping and mapping

    Grape expressed sequence tags (ESTs) are a new resource for developing simple sequence repeat (SSR) functional markers for genotyping and genetic mapping. An integrated pipeline including several computational tools for SSR identification and functional annotation was developed to identify 6,447 EST-SSR sequences from a total collection of 215,609 grape ESTs retrieved from NCBI. The 6,447 EST-SSRs were further reduced to 1,701 non-redundant sequences via clustering analysis, and 1,037 of them were successfully designed with primer pairs flanking the SSR motifs. From them, 150 pairs of primers were randomly selected for PCR amplification, polymorphism and heterozygosity analysis in V. vinifera cvs. Riesling and Cabernet Sauvignon, and V. rotundifolia (muscadine grape) cvs. Summit and Noble, and 145 pairs of these primers yielded PCR products. Pairwise comparisons of loci between the parents Riesling and Cabernet Sauvignon showed that 72 were homozygous in both cultivars, while 70 loci were heterozygous in at least one cultivar of the two. Muscadine parents Noble and Summit had 90 homozygous SSR loci in both parents and contained 50 heterozygous loci in at least one of the two.     The Bolocam Galactic Plane Survey V: HCO+ and N2H+ Spectroscopy of 1.1 mm Dust Continuum Sources

    We present the results of observations of 1882 sources in the Bolocam Galactic Plane Survey (BGPS) at 1.1 mm with the 10m Heinrich Hertz Telescope simultaneously in HCO+ J=3-2 and N2H+ J=3-2. We detect 77% of these sources in HCO^+ and 51% in N2H+ at greater than 3σ\sigma. We find a strong correlation between the integrated intensity of both dense gas tracers and the 1.1 mm dust emission of BGPS sources. We determine kinematic distances for 529 sources (440 in the first quadrant breaking the distance ambiguity and 89 in the second quadrant) We derive the size, mass, and average density for this subset of clumps. The median size of BGPS clumps is 0.75 pc with a median mass of 330 M_{\odot} (assuming T_{Dust}=20 K). The median HCO+ linewidth is 2.9 km s1^{-1} indicating that BGPS clumps are dominated by supersonic turbulence or unresolved kinematic motions. We find no evidence for a size-linewidth relationship for BGPS clumps. We analyze the effects of the assumed dust temperature on the derived clump properties with a Monte Carlo simulation and we find that changing the temperature distribution will change the median source properties (mass, volume-averaged number density, surface density) by factors of a few. The observed differential mass distribution has a power-law slope that is intermediate between that observed for diffuse CO clouds and the stellar IMF. BGPS clumps represent a wide range of objects (from dense cores to more diffuse clumps) and are typically characterized by larger sizes and lower densities than previously published surveys of high-mass star-forming regions. This collection of objects is a less-biased sample of star-forming regions in the Milky Way that likely span a wide range of evolutionary states.Comment: 48 pages, 25 figures, Accepted for publicatio

    The effect of climate change on avian offspring production: A global meta-analysis

    Climate change affects timing of reproduction in many bird species, but few studies have investigated its influence on annual reproductive output. Here, we assess changes in the annual production of young by female breeders in 201 populations of 104 bird species (N = 745,962 clutches) covering all continents between 1970 and 2019. Overall, average offspring production has declined in recent decades, but considerable differences were found among species and populations. A total of 56.7% of populations showed a declining trend in offspring production (significant in 17.4%), whereas 43.3% exhibited an increase (significant in 10.4%). The results show that climatic changes affect offspring production through compounded effects on ecological and life history traits of species. Migratory and larger-bodied species experienced reduced offspring production with increasing temperatures during the chick-rearing period, whereas smaller-bodied, sedentary species tended to produce more offspring. Likewise, multi-brooded species showed increased breeding success with increasing temperatures, whereas rising temperatures were unrelated to reproductive success in single-brooded species.
