    Building a Digital Platform for Behavioral Intervention Technology Research and Deployment

    While there is demonstrated potential for behavioral intervention technologies (BITs) to improve access to beneficial mental health interventions, there is a continuing need for research to evaluate effectiveness of new and existing applications for specific population groups. This paper reports experience in developing a digital platform to support delivery of an ecosystem of online mental health screening and behavioral intervention technologies to promote the wellbeing of New Zealand youth. Key platform requirements center on identity management, usage and assessment tracking, and implementation of research protocol workflow. A reusable and scalable solution based on Web services is presented in the context of ongoing iterative development and end user studies. The platform is providing consistent research and service delivery support for multiple concurrent threads of the overall youth mental health program

    Anonymization of DICOM electronic medical records for radiation therapy

    Electronic medical records (EMR) and treatment plans are used in research on patient outcomes and radiation effects. In many situations researchers must remove protected health information (PHI) from EMRs. The literature contains several studies describing the anonymization of generic Digital Imaging and Communication in Medicine (DICOM) files and DICOM image sets but no publications were found that discuss the anonymization of DICOM radiation therapy plans, a key component of an EMR in a cancer clinic. In addition to this we were unable to find a commercial software tool that met the minimum requirements for anonymization and preservation of data integrity for radiation therapy research. The purpose of this study was to develop a prototype software code to meet the requirements for the anonymization of radiation therapy treatment plans and to develop a way to validate that code and demonstrate that it properly anonymized treatment plans and preserved data integrity. We extended an open-source code to process all relevant PHI and to allow for the automatic anonymization of multiple EMRs. The prototype code successfully anonymized multiple treatment plans in less than 1. min/patient. We also tested commercial optical character recognition (OCR) algorithms for the detection of burned-in text on the images, but they were unable to reliably recognize text. In addition, we developed and tested an image filtering algorithm that allowed us to isolate and redact alpha-numeric text from a test radiograph. Validation tests verified that PHI was anonymized and data integrity, such as the relationship between DICOM unique identifiers (UID) was preserved. © 2014 Elsevier Ltd

    Pasture Improvement Needs and Options for New Zealand Sheep and Beef Farms

    In recent years in New Zealand, sheep and beef farming has been outcompeted for prime land. This means that the government and industry targets to increase sheep and beef production have to be achieved on farms with significant constraints on pasture production. They are increasingly restricted to hilly and other locations with variable climates and soils, and landscape constraints on farming practices. Thus there are limits on the ability to improve pasture production – whether by pasture renewal or through means like grazing management, fertiliser use, or weed and pest control

    Recreational fisheries (RF) are complex social-ecological systems that play an important role in aquatic environments while generating significant social and economic benefits around the world. The nature of RF is diverse and rapidly evolving, including the participants, their priorities and behaviors, and the related ecological impacts and social and economic benefits. RF can lead to negative ecological impacts, particularly through overexploitation of fish populations and spread of non-native species and genotypes through stocking. Hence, careful management and monitoring of RF is essential to sustain these ecologically and socioeconomically important resources. This special issue on recreational fisheries contains diverse research, syntheses, and perspectives that highlight the advances being made in RF research, monitoring, management, and practice, which we summarize here. Co-management actions are rising, often involving diverse interest groups including government and non-government organizations; applying collaborative management practices can help balance social and economic benefits with conservation targets. Technological and methodological advances are improving the ability to monitor biological, social, and economic dynamics of RF, which underpin the ability to maximize RF benefits through management actions. To ensure RF sustainability, much research focuses on the ecological aspects of RF, as well as the development of management and angling practices that reduce negative impacts on fish populations. For example, angler behavior can be influenced to conform to conservation-minded angling practices through regulations, but is often best accomplished through growing bottom-up social change movements. Anglers can also play an important role in fisheries monitoring and conservation, including providing data on fish abundance and assemblages (i.e., citizen science). The increasing impacts that growing human populations are having on the global environment are threatening many of the natural resources and ecosystem services they provide, including valuable RF. However, with careful development of research initiatives, monitoring and management, sustainable RF can generate positive outcomes for both society and natural ecosystems and help solve allocation conflicts with commercial fisheries and conservation

    Epigenetic and oncogenic regulation of SLC16A7 (MCT2) results in protein over-expression, impacting on signalling and cellular phenotypes in prostate cancer

Anne Y Warren research time funded by: Cambridge Biomedical Research Centre.Monocarboxylate Transporter 2 (MCT2) is a major pyruvate transporter encoded by the SLC16A7 gene. Recent studies pointed to a consistent overexpression of MCT2 in prostate cancer (PCa) suggesting MCT2 as a putative biomarker and molecular target. Despite the importance of this observation the mechanisms involved in MCT2 regulation are unknown. Through an integrative analysis we have discovered that selective demethylation of an internal SLC16A7/MCT2 promoter is a recurrent event in independent PCa cohorts. This demethylation is associated with expression of isoforms differing only in 5'-UTR translational control motifs, providing one contributing mechanism for MCT2 protein overexpression in PCa. Genes co-expressed with SLC16A7/MCT2 also clustered in oncogenic-related pathways and effectors of these signalling pathways were found to bind at the SLC16A7/MCT2 gene locus. Finally, MCT2 knock-down attenuated the growth of PCa cells. The present study unveils an unexpected epigenetic regulation of SLC16A7/MCT2 isoforms and identifies a link between SLC16A7/MCT2, Androgen Receptor (AR), ETS-related gene (ERG) and other oncogenic pathways in PCa. These results underscore the importance of combining data from epigenetic, transcriptomic and protein level changes to allow more comprehensive insights into the mechanisms underlying protein expression, that in our case provide additional weight to MCT2 as a candidate biomarker and molecular target in PCa.Publisher PDFPeer reviewe
