    Cellular delivery of antibodies: effective targeted subcellular imaging and new therapeutic tool

    It is already more than a century since the pioneering work of the Nobel Laureate Ehrlich gave birth to the side chain theory1, which helped to define antibodies and their ability to target specific biological sites. However, the use of antibodies is still restricted to the extracellular space due to the lack of a suitable delivery vehicle for the efficient transport of antibodies into live cells without inducing toxicity. In this work, we report the efficient encapsulation and delivery of antibodies into live cells with no significant loss of cell viability or any deleterious affect on the cell metabolic activity. This delivery system is based on poly(2-(methacryloyloxy)ethyl phosphorylcholine)-block-(2-(diisopropylamino)ethyl methacrylate), (PMPC-PDPA), a pH sensitive diblock copolymer that self-assembles to form nanometer-sized vesicles, also known as polymersomes, at physiological pH. These polymersomes can successfully deliver relatively high antibody payloads within live cells. Once inside the cells, we demonstrate that these antibodies can target their epitope by immune-labelling of cytoskeleton, Golgi, and transcription factor proteins in live cells. We also demonstrate that this effective antibody delivery mechanism can be used to control specific subcellular events, as well as modulate cell activity and pro-inflammatory process

    A flow-through balance is provided which includes a non-metric portion and a metric portion which form a fluid-conducting passage in fluid communication with an internal bore in the sting. The non-metric and metric portions of the balance are integrally connected together by a plurality of flexure beams such that the non-metric portion, the metric portion and the flexure beams form a one-piece construction which eliminates mechanical hysteresis between the non-metric and the metric portion. The system includes structures for preventing the effects of temperature, pressure and pressurized fluid from producing asymmetric loads on the flexure beams. A temperature sensor and a pressure sensor are located within the fluid-conducting passage of the balance. The system includes a longitudinal bellows member connected at two ends to one of the non-metric portion and the metric portion and at an intermediate portion thereof to the other of (1) and (2). A plurality of strain gages are mounted on the flexure beams to measure strain forces on the flexure beams. The flexure beams are disposed so as to enable symmetric forces on the flexure beams to cancel out so that only asymmetric forces are measured as deviations by the strain gages

    Close Pairs as Proxies for Galaxy Cluster Mergers

    Galaxy cluster merger statistics are an important component in understanding the formation of large-scale structure. Unfortunately, it is difficult to study merger properties and evolution directly because the identification of cluster mergers in observations is problematic. We use large N-body simulations to study the statistical properties of massive halo mergers, specifically investigating the utility of close halo pairs as proxies for mergers. We examine the relationship between pairs and mergers for a wide range of merger timescales, halo masses, and redshifts (0<z<1). We also quantify the utility of pairs in measuring merger bias. While pairs at very small separations will reliably merge, these constitute a small fraction of the total merger population. Thus, pairs do not provide a reliable direct proxy to the total merger population. We do find an intriguing universality in the relation between close pairs and mergers, which in principle could allow for an estimate of the statistical merger rate from the pair fraction within a scaled separation, but including the effects of redshift space distortions strongly degrades this relation. We find similar behavior for galaxy-mass halos, making our results applicable to field galaxy mergers at high redshift. We investigate how the halo merger rate can be statistically described by the halo mass function via the merger kernel (coagulation), finding an interesting environmental dependence of merging: halos within the mass resolution of our simulations merge less efficiently in overdense environments. Specifically, halo pairs with separations less than a few Mpc/h are more likely to merge in underdense environments; at larger separations, pairs are more likely to merge in overdense environments.Comment: 12 pages, 9 figures; Accepted for publication in ApJ.     Prevalence of and factors associated with herpes zoster in England: a cross-sectional analysis of the Health Survey for England

    BACKGROUND: Herpes zoster (commonly called shingles) is caused by the reactivation of varicella zoster virus, and results in substantial morbidity. While the risk of zoster increases significantly with age and immunosuppression, relatively little is known about other risk factors for zoster. Moreover, much evidence to date stems from electronic healthcare or administrative data. Hence, the aim of this study was to explore potential risk factors for herpes zoster using survey data from a nationally-representative sample of the general community-dwelling population in England. METHODS: Data were extracted from the 2015 Health Survey for England, an annual cross-sectional representative survey of households in England. The lifetime prevalence of self-reported herpes zoster was described by age, gender and other socio-demographic factors, health behaviours (physical activity levels, body mass index, smoking status and alcohol consumption) and clinical conditions, including; diabetes, respiratory, digestive and genito-urinary system and mental health disorders. Logistic regression models were then used to identify possible factors associated with shingles, and results were presented as odds ratios with 95% confidence intervals. RESULTS: The lifetime prevalence of shingles among the sample was 11.5% (12.6% among women, 10.3% among men), which increased with age. After adjusting for a range of covariates, increased age, female gender (odds ratio: 1.21; 95%CI: 1.03, 1.43), White ethnic backgrounds (odds ratio: 2.00; 95%CI: 1.40, 2.88), moderate physical activity 7 days per week (odds ratio: 1.29; 95%CI: 1.01, 1.66) and digestive disorders (odds ratio: 1.51; 95%CI: 1.13, 1.51) were each associated with increased odds of having had herpes zoster. CONCLUSIONS: Age, gender, ethnicity and digestive disorders may be risk factors for herpes zoster among a nationally representative sample of adults in England. These potential risk factors and possible mechanisms should be further explored using longitudinal studies

The definitive version was published in Bulletin of Marine Science 93 (2017): 611-639, doi:10.5343/bms.2016.1016.Understanding the promotion and regulation of circadian rhythms in marine fishes is important for studies spanning conservation, evolutionary biology, and physiology. Given numerous challenges inherent to quantifying behavioral activity across the full spectrum of marine environments and fish biodiversity, case studies offer a tractable means of gaining insights or forecasting broad patterns of diel activity. As these studies continue to accumulate, assessing whether, and to what extent, the cumulatively collected data are biased in terms of geography, habitat, or taxa represents a fundamentally important step in the development of a broad overview of circadian rhythms in marine fish. As such investigations require a phylogenetic framework, general trends in the phylogenetic sampling of marine fishes should be simultaneously assessed for biases in the sampling of taxa and trait data. Here, we compile diel activity data for more than 800 marine species from more than five decades of scientific studies to assess general patterns of bias. We found significant geographic biases that largely reflect a preference toward sampling warm tropical waters. Additionally, taxonomic biases likewise reflect a tendency toward conspicuous reef associated clades. Placing these data into a phylogenetic framework that includes all known marine fishes revealed significant under-dispersion of behavioral data and taxon sampling across the whole tree, with a few subclades exhibiting significant over-dispersion. In total, our study illuminates substantial gaps in our understanding of diel activity patterns and highlights significant sampling biases that have the potential to mislead evolutionary or ecological analyses.Partial funding was provided by the North Carolina Museum of Natural Sciences
