    Polarized growth of pollen tubes is a critical step for successful reproduction in angiosperms and is controlled by ROP GTPases. Spatiotemporal activation of ROP (Rho GTPases of plants) necessitates a complex and sophisticated regulatory system, in which guanine nucleotide exchange factors (RopGEFs) are key components. It was previously shown that a leucine-rich repeat receptor-like kinase, Arabidopsis pollen receptor kinase 2 (AtPRK2), interacted with RopGEF12 for its membrane recruitment. However, the mechanisms underlying AtPRK2-mediated ROP activation in vivo are yet to be defined. It is reported here that over-expression of AtPRK2 induced tube bulging that was accompanied by the ectopic localization of ROP-GTP and the ectopic distribution of actin microfilaments. Tube depolarization was also induced by a potentially kinase-dead mutant, AtPRK2K366R, suggesting that the over-expression effect of AtPRK2 did not require its kinase activity. By contrast, deletions of non-catalytic domains in AtPRK2, i.e. the juxtamembrane (JM) and carboxy-terminal (CT) domains, abolished its ability to affect tube polarization. Notably, AtPRK2K366R retained the ability to interact with RopGEF12, whereas AtPRK2 truncations of these non-catalytic domains did not. Lastly, it has been shown that the JM and CT domains of AtPRK2 were not only critical for its interaction with RopGEF12 but also critical for its distribution at the plasma membrane. These results thus provide further insight into pollen receptor kinase-mediated ROP activation during pollen tube growth

    Overexpression of the Tomato Pollen Receptor Kinase LePRK1 Rewires Pollen Tube Growth to a Blebbing Mode

    The tubular growth of a pollen tube cell is crucial for the sexual reproduction of flowering plants. LePRK1 is a pollen-specific and plasma membrane–localized receptor-like kinase from tomato (Solanum lycopersicum). LePRK1 interacts with another receptor, LePRK2, and with KINASE PARTNER PROTEIN (KPP), a Rop guanine nucleotide exchange factor. Here, we show that pollen tubes overexpressing LePRK1 or a truncated LePRK1 lacking its extracellular domain (LePRK1ΔECD) have enlarged tips but also extend their leading edges by producing “blebs.” Coexpression of LePRK1 and tomato PLIM2a, an actin bundling protein that interacts with KPP in a Ca2+-responsive manner, suppressed these LePRK1 overexpression phenotypes, whereas pollen tubes coexpressing KPP, LePRK1, and PLIM2a resumed the blebbing growth mode. We conclude that overexpression of LePRK1 or LePRK1ΔECD rewires pollen tube growth to a blebbing mode, through KPP- and PLIM2a-mediated bundling of actin filaments from tip plasma membranes. Arabidopsis thaliana pollen tubes expressing LePRK1ΔECD also grew by blebbing. Our results exposed a hidden capability of the pollen tube cell: upon overexpression of a single membrane-localized molecule, LePRK1 or LePRK1ΔECD, it can switch to an alternative mechanism for extension of the leading edge that is analogous to the blebbing growth mode reported for Dictyostelium and for Drosophila melanogaster stem cells.Fil: Gui, Cai Ping.     Thermal Pretreatment of Wood for Co-gasification/co-firing of Biomass and Coal

    Utilization of biomass as a co-feed in coal and biomass co-firing and co-gasification requires size reduction of the biomass. Reducing biomass to below 0.2 mm without pretreatment is difficult and costly because biomass is fibrous and compressible. Torrefaction is a promising thermal pretreatment process and has the advantages of increasing energy density, improving grindability, producing fuels with more homogenous compositions and hydrophobic behavior. Temperature is the most important factor for the torrefaction process. Biomass grindability is related to cell wall structure, thickness and composition. Thermal treatment such as torrefaction can cause chemical changes that significantly affect the strength of biomass. The objectives of this study are to understand the mechanism by which torrefaction improves the grindability of biomass and discuss suitable temperatures for thermal pretreatment for co-gasification/cofiring of biomass and coal. Wild cherry wood was selected as the model for this study. Samples were prepared by sawing a single tangential section from the heartwood and cutting it into eleven pieces. The samples were consecutively heated at 220, 260, 300, 350, 450 and 550oC for 0.5 hr under flowing nitrogen in a tube furnace. Untreated and treated samples were characterized for physical properties (color, dimensions and weight), microstructural changes by SEM, and cell wall composition changes and thermal behaviors by TGA and DSC. The morphology of the wood remained intact through the treatment range but the cell walls were thinner. Thermal treatments were observed to decompose the cell wall components. Hemicellulose decomposed over the range of ~200 to 300oC and resulted in weakening of the cell walls and subsequently improved grindability. Furthermore, wood samples treated above 300oC lost more than 39% in mass. Therefore, thermal pretreatment above the hemicelluloses decomposition temperature but below 300oC is probably sufficient to improve grindability and retain energy value

    Developing and Validating Service Innovation Readiness

    Services have emerged as major economic activities in Taiwan in recent years, since more than 70% of GDP in Taiwan is generated by service sectors. Nevertheless, knowledge of service innovation remained under-explored. To address this research gap, this study proposed the concept of “Service Innovation Readiness” based on expert interviews of five service industries (Department Stores and Retail, Financial, Biotechnology and Medicine, Tourism, and Information Services) and a review of existing literature. Given this, we propose a multi-dimensional construct of Service Innovation Readiness (SIR) consisting of five factors: Strategic Investment, Risk Tolerance, Innovative Champion, IT Experience, and Inter-Organizational Collaboration. To validate the framework of SIR, a survey was conducted in the five service sectors and the final sample consisted of 312 valid cases. The results grant support to the framework, showing that SIR is a multi-dimensional construct. The positive relationship between SIR and service performance provides further evidence to support the predictive validity of the construct measurement. Conclusion and implications are included

    Dying at home: a qualitative study of family carers' views of support provided by GPs community staff

    Background: Dying at home is the preference of many patients with life-limiting illness. This is often not achieved and a key factor is the availability of willing and able family carers. Aim: To elicit family carers’ views about the community support that made death at home possible. Design and setting: Qualitative study in East Devon, North Lancashire, and Cumbria. Method: Participants were bereaved family carers who had provided care at the end of life for patients dying at home. Semi-structured interviews were conducted 6–24 months after the death. Results: Fifty-nine bereaved family carers were interviewed (54% response rate; 69% female). Two-thirds of the patients died from cancer with median time of home care being 5 months and for non-cancer patients the median time for home care was 30 months. An overarching theme was of continuity of care that divided into personal, organisational, and informational continuity. Large numbers and changes in care staff diluted personal continuity and failure of the GPs to visit was viewed negatively. Family carers had low expectations of informational continuity, finding information often did not transfer between secondary and primary care and other care agencies. Organisational continuity when present provided comfort and reassurance, and a sense of control. Conclusion: The requirement for continuity in delivering complex end-of-life care has long been acknowledged. Family carers in this study suggested that minimising the number of carers involved in care, increasing or ensuring personal continuity, and maximising the informational and organisational aspects of care could lead to a more positive experience

    Enterovirus D68 subclade B3 Strain Circulating and Causing an Outbreak in the United States in 2016

    In 2014 the United States experienced a nationwide outbreak of Enterovirus D68 (EV-D68) infection. There were no confirmed cases of EV-D68 in 2015 and CDC was only aware of limited sporadic EV-D68 detection in the US in 2016. In this report, we analyzed 749 nasopharyngeal (NP) specimens collected in 2015 and 2016 from patients in the Lower Hudson Valley, New York using a previously validated EV-D68-specific rRT-PCR assay. EV-D68 was detected in none of 199 NP specimens collected in 2015, and in one of 108 (0.9%) samples from January to May and 159 of 442 (36.0%) samples from July to October 2016. Complete EV-D68 genome sequences from 22 patients in 2016 were obtained using a metagenomic next-generation sequencing assay. Comparative genome analysis confirmed that a new EV-D68 strain belonging to subclade B3, with 3.2-4.8% divergence in nucleotide from subclade B1 strains identified during the 2014 US outbreak, was circulating in the US in 2016 and caused an outbreak in the Lower Hudson Valley, New York with 160 laboratory-confirmed cases. Our data highlight the genetic variability and capacity in causing outbreak by diverse EV-D68 strains, and the necessity of awareness and more surveillance on their active circulation worldwide

    Cultural novelty and international students’ experience: a five-country study

    Research has linked cultural differences between a sojourner’s home and host country with their cultural transformation. Nonetheless, the results of empirical studies are inconclusive due to different operationalizations of cultural differences and testing among different groups of sojourners. We extend previous investigations by examining the effects of cultural novelty (i.e., the subjective perception of cultural differences) on the experience of international students (N = 1114) in Denmark, Germany, Italy, Poland, and the USA. Drawing on acculturation and social learning theories, we conceptualized a model of students’ adjustment and satisfaction taking into account cultural novelty. We tested the model through multi-group structural equation modeling (SEM) and examined the various relationships across subsamples from all five countries. We determined the significant effects of cultural novelty and a range of factors impacting students’ intercultural experience, such as their cultural intelligence, cultural background, second-language skills, time in the host country, and socialization with domestic students, and how the effects may vary by the host country. We discuss implications for future research and practice

    Parametric analysis and optimization for exergoeconomic performance of a combined system based on solid oxide fuel cell-gas turbine and supercritical carbon dioxide Brayton cycle

    Fuel cell-gas turbine hybrid system is a potential field of investigation. This study establishes a modeling and optimization framework for a novel hybrid system consisting of a solid oxide fuel cell, a gas turbine and a supercritical carbon dioxide Brayton cycle. Based on the proposed thermodynamical model, a parametric analysis is investigated to determine the impacts of several key parameters on the system exergoeconomic performance. Meanwhile, bi-objective optimization is conducted for maximizing the exergy efficiency and minimizing the levelized cost of electricity via the Epsilon-constraint approach. The Linear Programming Techniques for Multidimensional Analysis of Preference decision-making approach is further employed to select the Pareto optimum solution from Pareto frontiers. The results show that several extreme values for the exergy efficiency and the levelized cost of electricity exist in a series of sensitivity curves, respectively. The Pareto frontiers indicates that with the increase of the exergy efficiency, the levelized cost of electricity shows a moderately increasing trend at first and increases rapidly afterward. Overall, at the Pareto optimum solution, the combined system can achieve an optimal exergy efficiency and levelized cost of electricity by 68% and 0.0575 $ kWh −1 , respectively

    The Role of Eye Movement Driven Attention in Functional Strabismic Amblyopia

    Strabismic amblyopia “blunt vision” is a developmental anomaly that affects binocular vision and results in lowered visual acuity. Strabismus is a term for a misalignment of the visual axes and is usually characterized by impaired ability of the strabismic eye to take up fixation. Such impaired fixation is usually a function of the temporally and spatially impaired binocular eye movements that normally underlie binocular shifts in visual attention. In this review, we discuss how abnormal eye movement function in children with misaligned eyes influences the development of normal binocular visual attention and results in deficits in visual function such as depth perception. We also discuss how eye movement function deficits in adult amblyopia patients can also lead to other abnormalities in visual perception. Finally, we examine how the nonamblyopic eye of an amblyope is also affected in strabismic amblyopia
