    Microbial Quality and Pathogen Decontamination Strategies for Locally-Grown, Fresh Produce from West Virginia and Kentucky

    This study aimed to evaluate the microbiological quality/safety of fresh produce from farmers\u27 markets (FM) and assess the post-harvest washing practice with antimicrobials to inactivate Salmonella and Listeria monocytogenes on fresh produce. In study I, 212 produce samples were tested for the presence of Salmonella and Listeria spp. using modified FDA-BAM methods. Aerobic plate counts (APCs), total coliforms (TCCs), and yeast/molds were analyzed on petri-films. Among the 212 samples, the APCs, TCCs, and yeast/molds were 3.72-5.63, 3.67-5.47, and 3.07-4.13 log CFU/g, respectively, with spinach containing the highest (P\u3c0.05) populations. Among all tested samples, Salmonella enterica spp. enterica was detected on 18.6% of spinach, 10.9% of tomatoes, 18.5% of peppers, and 56.3% of cantaloupes, which is much higher than previous reported. Only 3.78% of the samples were confirmed for Listeria spp., and 50% of them were identified as L. monocytogenes, based on multiplex PCR results. Due to the high percentage of pathogens detected on the farmers\u27 marker produce, an evaluation of post-harvest produce washing with various antimicrobials was conducted in study II. Specifically, spinach, tomatoes, green peppers and cucumbers were inoculated with S. Typhimurium and Tennessee or L. monocytogenes and washed in tap water, vinegar water (10%), lactic acid (5%), a lactic and citric acid blend (2.5%), and sodium hypochlorite (200 ppm) for 30 sec or unwashed. Vinegar water (10%) showed better (P\u3c0.05) reduction of S. Typhimurium and Tennessee on tomatoes and cucumbers, and L. monocytogenes on tomatoes and peppers than tap water. The three antimicrobials reached an additional reduction level of 0.9 to 2.7 (S. Typhimurium and Tennessee) and 0.2 to 1.4 log CFU/g ( L. monocytogenes) compared to tap water. Lactic acid caused the greatest (P\u3c0.05) reduction of S. Typhimurium and Tennessee on spinach and green peppers, and sodium hypochlorite showed the great (P\u3c0.05) reduction of L. monocytogenes on cucumbers. The results supplied important information for FM vendors to develop post-harvest protocols to control foodborne pathogens

    Large-angle Polarization of the Cosmic Microwave Background Radiation and Reionization

    We discuss the effect of matter reionization on the large-angular-scale anisotropy and polarization of the cosmic microwave background radiation (CMBR) in the standard CDM model. We separate three cases in which the anisotropy is induced by pure scalar, pure tensor, and mixed metric perturbations respectively. It is found that, if reionization occurs early enough, the polarization can reach a detectable level of sequentially 6%6\%, 9%9\%, and 6.5%6.5\% of the anisotropy. In general, a higher degree of polarization implies a dominant contribution from the tensor mode or reionization at high redshift. Since early reionization will suppress small-scale CMBR anisotropies and polarizations significantly, measuring the polarization on few degree scales can be a direct probe of the reionization history of the early universe.Comment: Changes in the revised version: 1.     Does precipitation fall preferentially over dry (or wet) soils?

    Previous work suggests precipitation is more likely to fall over dry soils than wet soils. The limited knowledge of the association between surface moisture content and convective activity raises many questions about whether a positive (or negative) feedback exists. A positive feedback implies higher soil moisture values correspond with enhanced precipitation. While soil moisture does not directly influence precipitation, it does moderate latent and sensible heat fluxes, which may lead to influencing convective initiation. In this study we analyze synoptically benign (SB) environments that do not feature a low-level jet (noLLJ). Because these atmospheric conditions are expected to be much less conducive to convection and precipitation, we can better understand the possible impacts of soil moisture. This case study approach will focus on convective events that are characterized as localized convective precipitation as opposed to stratiform precipitation caused by large-scale frontal systems.     Locally self-consistent embedding approach for disordered electronic systems

    We present a new embedding scheme for the locally self-consistent method to study disordered electron systems. We test this method in a tight-binding basis and apply it to the single band Anderson model. The local interaction zone is used to efficiently compute the local Green's function of a supercell embeded into a local typical medium. We find a quick convergence as the size of the local interaction zone which reduces the computational costs as expected. This method captures the Anderson localization transition and accurately predicts the critical disorder strength. The present work opens the path towards the development of a typical medium embedding scheme for the O(N)O(N) multiple scattering methods.Comment: 7 pages, 5 figure

    LYVE1 Marks the Divergence of Yolk Sac Definitive Hemogenic Endothelium from the Primitive Erythroid Lineage.

    The contribution of the different waves and sites of developmental hematopoiesis to fetal and adult blood production remains unclear. Here, we identify lymphatic vessel endothelial hyaluronan receptor-1 (LYVE1) as a marker of yolk sac (YS) endothelium and definitive hematopoietic stem and progenitor cells (HSPCs). Endothelium in mid-gestation YS and vitelline vessels, but not the dorsal aorta and placenta, were labeled by Lyve1-Cre. Most YS HSPCs and erythro-myeloid progenitors were Lyve1-Cre lineage traced, but primitive erythroid cells were not, suggesting that they represent distinct lineages. Fetal liver (FL) and adult HSPCs showed 35%-40% Lyve1-Cre marking. Analysis of circulation-deficient Ncx1-/- concepti identified the YS as a major source of Lyve1-Cre labeled HSPCs. FL proerythroblast marking was extensive at embryonic day (E) 11.5-13.5, but decreased to hematopoietic stem cell (HSC) levels by E16.5, suggesting that HSCs from multiple sources became responsible for erythropoiesis. Lyve1-Cre thus marks the divergence between YS primitive and definitive hematopoiesis and provides a tool for targeting YS definitive hematopoiesis and FL colonization
