the role of genetic polymorphisms in the ABCB1-gene
P-Glykoprotein (P-Gp) ist ein membranstÀndiges Protein, das als Efflux-
Transporter die intrazellulÀre Konzentration seines Substrats vermindert.
Seine physiologische Funktion liegt im Schutz der Zelle vor schÀdigenden
Substanzen sowie der Elimination endogener Stoffwechselprodukte. P-Gp spielt
eine wichtige Rolle in der zellvermittelten ImmunitÀt sowie bei der Multiplen
Wirkstoffresistenz und ist von zentraler Bedeutung fĂŒr die Pharmakokinetik
zahlreicher therapeutisch wichtiger Wirkstoffe. Beim Menschen wird P-Gp durch
das ABCB1-Gen kodiert. FĂŒr dieses Gen lieĂen sich genetische Polymorphismen
identifizieren, denen eine Auswirkung auf die Expression von ABCB1 und somit
auf die AktivitÀt von P-Gp zugeschrieben wird. Untersuchungen zu dieser Frage
kommen zu teilweise kontrÀren Ergebnissen. Im Rahmen einiger Studien findet
ein kleinerer Medikamententransporter Mini-P-Gp ErwĂ€hnung, der mutmaĂlich
durch einen alternativen SpleiĂvorgang an der ABCB1-RNA entsteht. Ziel dieser
Arbeit war herauszufinden, ob sich die Expression von P-Gp, abgebildet in
ABCB1-mRNA-Mengen, in vier Zelllinien der peripheren lymphatischen Blutreihe
(CD4+-, CD8+-, CD56+- und CD 19+-Lymphozyten) unterscheidet. AuĂerdem sollte
geklÀrt werden, ob die Höhe der P-Gp-Expression mit dem Vorhandensein
bestimmter Genotypen zweier hÀufiger genetischer Polymorphismen im ABCB1-Gen
(G2677T/A und C3435T) assoziiert ist. Durch die Auswahl spezifischer Primer
zur VervielfÀltigung der N- bzw. C-terminalen HÀlfte des ABCB1-Gens erfolgte
die ĂberprĂŒfung der Existenz von Mini-P-Gp. Die Trennung der von uns
untersuchten Zelllinien geschah durch FACS-Zellsortierung nach FĂ€rbung mit
immunfluoreszierenden Antikörpern. Mit Hilfe eines quantitativen RT-PCR-
Versuchsaufbaus wurde die ABCB1-mRNA gemessen. Zur Normalisierung der
gemessenen Konzentrationen diente die gleichzeitige Quantifizierung eines
Housekeeping-Gens. Die Ergebnisse unserer Studie wiesen die höchste ABCB1
-mRNA-Menge in CD 56+-, gefolgt von CD8+-, CD4+- und CD 19+-Lymphozyten nach.
Dies steht in Einklang mit frĂŒheren Studien zur AktivitĂ€t des
Medikamententransporters und unterstreicht die physiologische Rolle von P-Gp
in der zellvermittelten zytotoxischen ImmunitĂ€t. Bei der ĂberprĂŒfung der
genetischen Polymorphismen hinsichtlich der Expression von P-Gp konnte nach
Normalisierung der Daten keine statistisch signifikante Auswirkung auf die
Anzahl der ABCB1-mRNA-Kopien in allen Zellpopulationen gefunden werden. Die
Frage nach der Existenz eines Mini-P-Gp konnte nicht geklÀrt werden. Zwar
wurde in allen Zellen eine etwa halb so hohe Transkriptzahl fĂŒr die
N-terminale HĂ€lfte wie fĂŒr die C-terminale HĂ€lfte gemessen, was als Indiz fĂŒr
die Existenz des kleineren Transporterproteins gewertet werden kann. Doch darf
dabei nicht auĂer Acht gelassen werden, dass die Effizienz der PCR fĂŒr die
N-terminale HĂ€lfte schlechter als fĂŒr die C-terminale HĂ€lfte ist. AuĂerdem
gibt es bisher keine molekularen Analysen, die das PhÀnomen Mini-P-Gp erklÀren
könnten.P-Glycoprotein (P-Gp) is a membrane-bound protein, which acts as an efflux-
pump and reduces the intracellular concentration of its substrates. Its
physiological function is the protection of the cell from toxic substances as
well as the elimination of endogenous metabolic wastes. P-Gp plays an
important role in cell-mediated immunity, in multi-drug resistance and in the
phamacokinetics of numerous therapeutic agents. In humans P-Gp is encoded by
the ABCB1-gene. A number of genetic polymorphisms for this gene have been
identified, which may have implications on the expression of ABCB1 and,
consequently, on the activity of P-Gp. Investigations in this field have
resulted in contradictory findings. In some studies a small drug transporter,
Mini-P-Gp, is mentioned, which may be the result of alternative splicing of
the ABCB1-RNA. The objective of this study was to find out, whether the
expression of P-Gp, as measured in ABCB1-mRNA, differs in 4 lines of the
peripheral blood lymphoid cell pool (CD4+-, CD8+-, CD 56+- and
CD19+-lymphocytes) and whether the amount of P-Gp expression is associated
with two frequent genetic polymorphisms in the ABCB1-gene (G2677T/A and
C3435T). By designing specific primers for the amplification of the N- and
C-terminal half of the ABCB1-gene, respectively, we tried to verify the
existence of Mini-P-Gp. Cell separation was carried out using FACS cell-
sorting after staining with fluorescent antibodies. Quantification was
performed with a quantitative RT-PCR assay. The results were normalised by
simultaneous quantification of a housekeeping-gene. We found the highest
amount of ABCB1-mRNA in CD56+, followed by CD8+-, CD4+,- and CD19+-
lymphocytes. This is in accordance with previous studies on the activity of
the drug transporter and underlines its physiological role in cell-mediated,
cytotoxic immunity. However, no statistically significant effect on the ABCB1
-mRNA-copy number as a result of the different genetic polymorphisms could be
seen after normalising the data. The question of the existence of Mini-P-Gp
could not be clarified. We found approximately half as many transcript copies
of the N-terminal half as of the C-terminal half in all cell lines, which
could be an indicator of the existence of the smaller transport protein.
However, this result may be confounded by a lower PCR efficiency for the
N-terminal half than for the C-terminal half. Moreover, there is to this date
no molecular analysis explaining the phenomenon Mini-P-Gp
Multimodality in Epistemological Foreign Language Research? Two Case Studies in the Field of German as a Foreign and Second Language
This article aims to elaborate on the epistemological interest of cultural studies for German as a foreign and second language and, simultaneously, to point out the limitations of the current focus on certain modes. We intend to broaden this focus by highlighting the connectivity of the epistemological interest to multimodal research using two relevant case studies. The ïŹrst case considers music videos and their associated comments on YouTube, examining how patterns of freedom are negotiated. The second considers textbooks for the so-called cultural orientation of immigrants to Germany and examines one of the ïŹrst textbooks developed for this purpose through the lens of meaning-making of the Holocaust. Both case studies were selected on the basis of existing doctoral projects of the authors. The analyses follow Bateman, Wildfeuer, and Hiippala, but take into account the extensive history of multimodal studies.
