    Longitudinal evaluation of aflatoxin exposure in two cohorts in south-western Uganda

    Aflatoxins (AF) are a group of mycotoxins. AF exposure causes acute and chronic adverse health effects such as aflatoxicosis and hepatocellular carcinoma in human populations, especially in the developing world. In this study, AF exposure was evaluated using archived serum samples from human immunodeficiency virus (HIV)-seronegative participants from two cohort studies in south-western Uganda. AFB1-lysine (AFB-Lys) adduct levels were determined via HPLC fluorescence in a total of 713 serum samples from the General Population Cohort (GPC), covering eight time periods between 1989 and 2010. Overall, 90% (642/713) of the samples were positive for AFB-Lys and the median level was 1.58 pg mg(-1) albumin (range = 0.40-168 pg mg(-1) albumin). AFB-Lys adduct levels were also measured in a total of 374 serum samples from the Rakai Community Cohort Study (RCCS), across four time periods between 1999 and 2003. The averaged detection rate was 92.5% (346/374) and the median level was 1.18 pg mg(-1) albumin (range = 0.40-122.5 pg mg(-1) albumin). In the GPC study there were no statistically significant differences between demographic parameters, such as age, sex and level of education, and levels of serum AFB-Lys adduct. In the RCCS study, longitudinal analysis using generalised estimating equations revealed significant differences between the adduct levels and residential areas (p = 0.05) and occupations (p = 0.02). This study indicates that AF exposure in people in two populations in south-western Uganda is persistent and has not significantly changed over time. Data from one study, but not the other, indicated that agriculture workers and rural area residents had more AF exposure than those non-agricultural workers and non-rural area residents. These results suggest the need for further study of AF-induced human adverse health effects, especially the predominant diseases in the region

    Synthesis of Tapered CdS Nanobelts and CdSe Nanowires with Good Optical Property by Hydrogen-Assisted Thermal Evaporation

    The tapered CdS nanobelts and CdSe nanowires were prepared by hydrogen-assisted thermal evaporation method. Different supersaturation leads to two different kinds of 1D nanostructures. The PL measurements recorded from the as-prepared tapered CdS nanobelts and CdSe nanowires show only a bandgap emission with relatively narrow full-width half maximum, which means that they possess good optical property. The as-synthesized high-quality tapered CdS nanobelts and CdSe nanowires may be excellent building blocks for photonic devices

    Antimicrobial Peptide Evolution in the Asiatic Honey Bee Apis cerana

    The Asiatic honeybee, Apis cerana Fabricius, is an important honeybee species in Asian countries. It is still found in the wild, but is also one of the few bee species that can be domesticated. It has acquired some genetic advantages and significantly different biological characteristics compared with other Apis species. However, it has been less studied, and over the past two decades, has become a threatened species in China. We designed primers for the sequences of the four antimicrobial peptide cDNA gene families (abaecin, defensin, apidaecin, and hymenoptaecin) of the Western honeybee, Apis mellifera L. and identified all the antimicrobial peptide cDNA genes in the Asiatic honeybee for the first time. All the sequences were amplified by reverse transcriptase-polymerase chain reaction (RT-PCR). In all, 29 different defensin cDNA genes coding 7 different defensin peptides, 11 different abaecin cDNA genes coding 2 different abaecin peptides, 13 different apidaecin cDNA genes coding 4 apidaecin peptides and 34 different hymenoptaecin cDNA genes coding 13 different hymenoptaecin peptides were cloned and identified from the Asiatic honeybee adult workers. Detailed comparison of these four antimicrobial peptide gene families with those of the Western honeybee revealed that there are many similarities in the quantity and amino acid components of peptides in the abaecin, defensin and apidaecin families, while many more hymenoptaecin peptides are found in the Asiatic honeybee than those in the Western honeybee (13 versus 1). The results indicated that the Asiatic honeybee adult generated more variable antimicrobial peptides, especially hymenoptaecin peptides than the Western honeybee when stimulated by pathogens or injury. This suggests that, compared to the Western honeybee that has a longer history of domestication, selection on the Asiatic honeybee has favored the generation of more variable antimicrobial peptides as protection against pathogens

    Ao longo do processo de transporte do gΓ‘s natural em gasodutos submarinos atΓ© as Unidades de Processamento, a indΓΊstria petrolΓ­fera pode enfrentar diversos problemas, dentre eles, a formação de hidratos. O ambiente submarino possui condiçáes propicias de pressΓ£o (alta) e temperatura (baixa) para que estas estruturas sΓ³lidas se formem. Os hidratos sΓ£o problemas crΓ­ticos para a garantia de escoamento, pois podem causar entupimento das tubulaçáes, prisΓ£o e problemas de acionamento em equipamentos, podem comprometer a produtividade das operaçáes, aumentar custos de produção e consumo de energia, alΓ©m de gerar danos aos equipamentos e as estruturas, comprometendo a seguranΓ§a das atividades e em casos mais extremos, causando a paralisação completa da produção. Por isso, faz-se necessΓ‘rio adotar medidas de controle, planejamento e monitoramento das condiçáes termodinΓ’micas as quais o gΓ‘s natural estΓ‘ sendo transportado. Neste trabalho sΓ£o simuladas as condiçáes prΓ³ximas das reais enfrentadas durante o transporte do gΓ‘s natural produzido na Unidade de Produção de PetrΓ³leo e GΓ‘s Natural FPSO Cidade de VitΓ³ria atΓ© a Unidade de Tratamento de GΓ‘s de Cacimbas. Para verificar as propriedades de fluxo do sistema, a temperatura e a pressΓ£o de formação dos hidratos, e dimensionar de forma otimizada o uso de mΓ©todos quΓ­micos (inibidores) e mΓ©todos tΓ©rmicos (aquecedores) de prevenção serΓ‘ utilizado o software HYSYS. Os resultados obtidos comprovam a eficiΓͺncia do inibidor termodinΓ’mico metanol na prevenção da formação do hidrato e as vazΓ΅es necessΓ‘rias para realizar este controle. AlΓ©m disso, indica-se a quantidade de energia necessΓ‘ria para que um aquecedor elΓ©trico forneΓ§a calor suficiente Γ  tubulação para prevenir a formação de hidratos, mostrando que este mΓ©todo tΓ©rmico pode substituir a injeção do metanol, porΓ©m Γ© necessΓ‘rio avaliar o requisito energΓ©tico, que pode ser elevado.     AFM-Detected Apoptotic Changes in Morphology and Biophysical Property Caused by Paclitaxel in Ishikawa and HeLa Cells

    The apoptosis of cancer cells is associated with changes in the important cell properties including morphology, surface roughness and stiffness. Therefore, the changes in morphology and biophysical properties can be a good way of evaluating the anticancer activity of a drug. This study examined the effect of paclitaxel on the properties of Ishikawa and HeLa cells using atomic force microscopy (AFM), and the relationship between the changes in morphology and the biophysical properties and apoptosis was discussed. The viability and proliferation of the cells were analyzed using the methylthiazol tetrazolium (MTT) method and a TUNEL assay to confirm cellular apoptosis due to a paclitaxel treatment. AFM observations clearly showed the apoptotic morphological and biophysical changes in Ishikawa and HeLa cells. After the paclitaxel treatment, the cell membrane was torn and holed, the surface roughness was increased, and the stiffness was decreased. These changes were observed more apparently after a 24 h treatment and in Ishikawa cells compared to HeLa cells. The MTT and TUNEL assays results revealed the Ishikawa cells to be more sensitive to paclitaxel than HeLa cells and definite apoptosis occurred after a 24 h treatment. These results showed good agreement with the AFM results. Therefore, research on the morphological and biophysical changes by AFM in cancer cells will help to evaluate the anticancer activities of the drugs

    Consistent dust and gas models for protoplanetary disks IV. A panchromatic view of protoplanetary disks

    Context. Consistent modeling of protoplanetary disks requires the simultaneous solution of both continuum and line radiative transfer, heating and cooling balance between dust and gas and, of course, chemistry. Such models depend on panchromatic observations that can provide a complete description of the physical and chemical properties and energy balance of protoplanetary systems. Along these lines, we present a homogeneous, panchromatic collection of data on a sample of 85 T Tauri and Herbig Ae objects for which data cover a range from X-rays to centimeter wavelengths. Datasets consist of photometric measurements, spectra, along with results from the data analysis such as line fluxes from atomic and molecular transitions. Additional properties resulting from modeling of the sources such as disk mass and shape parameters, dust size, and polycyclic aromatic hydrocarbon (PAH) properties are also provided for completeness. Aim. The purpose of this data collection is to provide a solid base that can enable consistent modeling of the properties of protoplanetary disks. To this end, we performed an unbiased collection of publicly available data that were combined to homogeneous datasets adopting consistent criteria. Targets were selected based on both their properties and the availability of data. Methods. Data from more than 50 different telescopes and facilities were retrieved and combined in homogeneous datasets directly from public data archives or after being extracted from more than 100 published articles. X-ray data for a subset of 56 sources represent an exception as they were reduced from scratch and are presented here for the first time. Results. Compiled datasets, along with a subset of continuum and emission-line models are stored in a dedicated database and distributed through a publicly accessible online system. All datasets contain metadata descriptors that allow us to track them back to their original resources. The graphical user interface of the online system allows the user to visually inspect individual objects but also compare between datasets and models. It also offers to the user the possibility to download any of the stored data and metadata for further processing

    Genome-Scale Modeling of Light-Driven Reductant Partitioning and Carbon Fluxes in Diazotrophic Unicellular Cyanobacterium Cyanothece sp. ATCC 51142

    Genome-scale metabolic models have proven useful for answering fundamental questions about metabolic capabilities of a variety of microorganisms, as well as informing their metabolic engineering. However, only a few models are available for oxygenic photosynthetic microorganisms, particularly in cyanobacteria in which photosynthetic and respiratory electron transport chains (ETC) share components. We addressed the complexity of cyanobacterial ETC by developing a genome-scale model for the diazotrophic cyanobacterium, Cyanothece sp. ATCC 51142. The resulting metabolic reconstruction, iCce806, consists of 806 genes associated with 667 metabolic reactions and includes a detailed representation of the ETC and a biomass equation based on experimental measurements. Both computational and experimental approaches were used to investigate light-driven metabolism in Cyanothece sp. ATCC 51142, with a particular focus on reductant production and partitioning within the ETC. The simulation results suggest that growth and metabolic flux distributions are substantially impacted by the relative amounts of light going into the individual photosystems. When growth is limited by the flux through photosystem I, terminal respiratory oxidases are predicted to be an important mechanism for removing excess reductant. Similarly, under photosystem II flux limitation, excess electron carriers must be removed via cyclic electron transport. Furthermore, in silico calculations were in good quantitative agreement with the measured growth rates whereas predictions of reaction usage were qualitatively consistent with protein and mRNA expression data, which we used to further improve the resolution of intracellular flux values
