    Red cell function at extreme altitudes on Mount Everest

    As part of the American Medical Research Expedition to Everest in 1981, we measured hemoglobin concentration, red cell 2,3-diphosphoglycerate (2,3-DPG), Po2 at which hemoglobin is 50% saturated (P50), and acid-base status in expedition members at various altitudes. All measurements were made in expedition laboratories and, with the exception of samples from the South Col of Mt. Everest (8,050 m), within 2 h of blood collection. In vivo conditions were estimated from direct measurements of arterial blood gases and pH or inferred from base excess and alveolar PCO2. As expected, increased 2,3-DPG was associated with slightly increased P50, when expressed at pH 7.4. Because of respiratory alkalosis, however, the subjects' in vivo P50 at 6,300 m (27.6 Torr) was slightly less than at sea level (28.1 Torr). The estimated in vivo P50 was progressively lower at 8,050 m (24.9 Torr) and on the summit at 8,848 m (19.4 Torr in one subject). Our data suggest that, at extreme altitude, the blood O2 equilibrium curve shifts progressively leftward because of respiratory alkalosis. This left shift protects arterial O2 saturation at extreme altitude

    Electrophysiological measures of conflict detection and resolution in the Stroop task

    Conflict detection and resolution is crucial in a cognitive task like the Stroop task. Previous studies have identified an early negativity component (Ninc) as a prominent marker of Stroop conflict in event-related potentials (ERPs). However, to what extent this ERP component reflects conflict detection and/or resolution is still unclear. Here, we report a Stroop task in which the stimulus onset asynchrony (SOA) of color and word stimuli presentation was manipulated in order to disentangle the roles of conflict detection and conflict resolution in generating Stroop-related ERP components. Separating the word from the color information gives us precise control over the timing of conflict. If the Ninc is related with conflict resolution it should be absent when the word appears during response preparation, as in a long-latency positive SOA. Our data shows that the Ninc occurs in all SOAs, even after a response has been made, supporting its role in the detection of stimulus conflict rather than conflict resolution. The use of SOA manipulation therefore allows for the examination of a wider temporal spectrum of interference in order to specify the functions of this conflict-related component. These results provide insight into the neural signatures of conflict processes, and have implications for models of cognitive control mechanisms in the brain

    Counting matrices over finite fields with support on skew Young diagrams and complements of Rothe diagrams

    We consider the problem of finding the number of matrices over a finite field with a certain rank and with support that avoids a subset of the entries. These matrices are a q-analogue of permutations with restricted positions (i.e., rook placements). For general sets of entries these numbers of matrices are not polynomials in q (Stembridge 98); however, when the set of entries is a Young diagram, the numbers, up to a power of q-1, are polynomials with nonnegative coefficients (Haglund 98). In this paper, we give a number of conditions under which these numbers are polynomials in q, or even polynomials with nonnegative integer coefficients. We extend Haglund's result to complements of skew Young diagrams, and we apply this result to the case when the set of entries is the Rothe diagram of a permutation. In particular, we give a necessary and sufficient condition on the permutation for its Rothe diagram to be the complement of a skew Young diagram up to rearrangement of rows and columns. We end by giving conjectures connecting invertible matrices whose support avoids a Rothe diagram and Poincar\'e polynomials of the strong Bruhat order.Comment: 24 pages, 9 figures, 1 tabl

    The structural basis for high‐affinity uptake of lignin‐derived aromatic compounds by proteobacterial TRAP transporters

    The organic polymer lignin is a component of plant cell walls, which like (hemi)-cellulose is highly abundant in nature and relatively resistant to degradation. However, extracellular enzymes released by natural microbial consortia can cleave the ÎČ-aryl ether linkages in lignin, releasing monoaromatic phenylpropanoids that can be further catabolised by diverse species of bacteria. Biodegradation of lignin is therefore important in global carbon cycling, and its natural abundance also makes it an attractive biotechnological feedstock for the industrial production of commodity chemicals. Whilst the pathways for degradation of lignin-derived aromatics have been extensively characterised, much less is understood about how they are recognised and taken up from the environment. The purple phototrophic bacterium Rhodopseudomonas palustris can grow on a range of phenylpropanoid monomers and is a model organism for studying their uptake and breakdown. R. palustris encodes a tripartite ATP-independent periplasmic (TRAP) transporter (TarPQM) linked to genes encoding phenylpropanoid-degrading enzymes. The periplasmic solute-binding protein component of this transporter, TarP, has previously been shown to bind aromatic substrates. Here, we determine the high-resolution crystal structure of TarP from R. palustris as well as the structures of homologous proteins from the salt marsh bacterium Sagittula stellata and the halophile Chromohalobacter salexigens, which also grow on lignin-derived aromatics. In combination with tryptophan fluorescence ligand-binding assays, our ligand-bound co-crystal structures reveal the molecular basis for high-affinity recognition of phenylpropanoids by these TRAP transporters, which have potential for improving uptake of these compounds for biotechnological transformations of lignin

    Can we continue research in splenectomized dogs? Mycoplasma haemocanis: Old problem - New insight

    We report the appearance of a Mycoplasma haemocanis infection in laboratory dogs, which has been reported previously, yet, never before in Europe. Outbreak of the disease was triggered by a splenectomy intended to prepare the dogs for a hemorrhagic shock study. The clinical course of the dogs was dramatic including anorexia and hemolytic anemia. Treatment included allogeneic transfusion, prednisone, and oxytetracycline. Systematic follow-up (n=12, blood smears, antibody testing and specific polymerase chain reaction) gives clear evidence that persistent eradication of M. haemocanis is unlikely. We, therefore, had to abandon the intended shock study. In the absence of effective surveillance and screening for M. haemocanis, the question arises whether it is prudent to continue shock research in splenectomized dogs. Copyright (C) 2004 S.     Selectivity and functional diversity in arbuscular mycorrhizas of co-occurring fungi and plants from a temperate deciduous woodland

    1 The arbuscular mycorrhizal (AM) fungi colonizing plants at a woodland site in North Yorkshire (UK) have been characterized from the roots of five plant species (Rubus fruticosus agg. L., Epilobium angustifolium L., Acer pseudoplatanus L., Ajuga reptans L. and Glechoma hederacea L.), and identified using small-subunit rRNA (SSUrRNA) gene amplification and sequencing. 2 Interactions between five plant species from the site and four co-occurring glomalean fungi were investigated in artificial one-to-one AM symbioses. Three of the fungi were isolated from the site; the fourth was a culture genetically similar to a taxon found at the site. Phosphorus uptake and growth responses were compared with non-mycorrhizal controls. 3 Individual fungi colonized each plant with different spatial distribution and intensity. Some did not colonize at all, indicating incompatibility under the conditions used in the experiments. 4 Glomus hoi consistently occupied a large proportion of root systems and outperformed the other fungi, improving P uptake and enhancing the growth of four out of the five plant species. Only G. hoi colonized and increased P uptake in Acer pseudoplatanus, the host plant with which it associates almost exclusively under field conditions. Colonization of all plant species by Scutellospora dipurpurescens was sparse, and beneficial to only one of the host plants (Teucrium scorodonia). Archaeospora trappei and Glomus sp. UY1225 had variable effects on the host plants, conferring a range of P uptake and growth benefits on Lysimachia nummularia and T. scorodonia, increasing P uptake whilst not affecting biomass in Ajuga reptans and Glechoma hederacea, and failing to form mycorrhizas with A. pseudoplatanus. 5 These experimental mycorrhizas show that root colonization, symbiont compatibility and plant performance vary with each fungus-plant combination, even when the plants and fungi naturally co-exist. 6 We provide evidence of physical and functional selectivity in AM. The small number of described AM fungal species (154) has been ascribed to their supposed lack of host specificity, but if the selectivity we have observed is the general rule, then we may predict that many more, probably hard-to-culture glomalean species await discovery, or that members of species as currently perceived may be physiologically or functionally distinct

    Improved Effective Potential in Curved Spacetime and Quantum Matter - Higher Derivative Gravity Theory

    \noindent{\large\bf Abstract.} We develop a general formalism to study the renormalization group (RG) improved effective potential for renormalizable gauge theories ---including matter-R2R^2-gravity--- in curved spacetime. The result is given up to quadratic terms in curvature, and one-loop effective potentials may be easiliy obtained from it. As an example, we consider scalar QED, where dimensional transmutation in curved space and the phase structure of the potential (in particular, curvature-induced phase trnasitions), are discussed. For scalar QED with higher-derivative quantum gravity (QG), we examine the influence of QG on dimensional transmutation and calculate QG corrections to the scalar-to-vector mass ratio. The phase structure of the RG-improved effective potential is also studied in this case, and the values of the induced Newton and cosmological coupling constants at the critical point are estimated. Stability of the running scalar coupling in the Yukawa theory with conformally invariant higher-derivative QG, and in the Standard Model with the same addition, is numerically analyzed.     Growing up in Bradford:Protocol for the age 7-11 follow up of the Born in Bradford birth cohort

    Background: Born in Bradford (BiB) is a prospective multi-ethnic pregnancy and birth cohort study that was established to examine determinants of health and development during childhood and, subsequently, adult life in a deprived multi-ethnic population in the north of England. Between 2007 and 2010, the BiB cohort recruited 12,453 women who experienced 13,776 pregnancies and 13,858 births, along with 3353 of their partners. Forty five percent of the cohort are of Pakistani origin. Now that children are at primary school, the first full follow-up of the cohort is taking place. The aims of the follow-up are to investigate the determinants of children's pre-pubertal health and development, including through understanding parents' health and wellbeing, and to obtain data on exposures in childhood that might influence future health. Methods: We are employing a multi-method approach across three data collection arms (community-based family visits, school based physical assessment, and whole classroom cognitive, motor function and wellbeing measures) to follow-up over 9000 BiB children aged 7-11 years and their families between 2017 and 2021. We are collecting detailed parent and child questionnaires, cognitive and sensorimotor assessments, blood pressure, anthropometry and blood samples from parents and children. Dual x-ray absorptiometry body scans, accelerometry and urine samples are collected on subsamples. Informed consent is collected for continued routine data linkage to health, social care and education records. A range of engagement activities are being used to raise the profile of BiB and to disseminate findings. Discussion: Our multi-method approach to recruitment and assessment provides an efficient method of collecting rich data on all family members. Data collected will enhance BiB as a resource for the international research community to study the interplay between ethnicity, socioeconomic circumstances and biology in relation to cardiometabolic health, mental health, education, cognitive and sensorimotor development and wellbeing.</p
