    Soslovie and the “Foreign” Clergies of Imperial Russia

    AbstractThis essay addresses a significant imperial dimension of the soslovie question by analyzing the estate status of the religious servitors of Russia’s non-Orthodox faiths (the “foreign confessions”). Its goal is to ascertain the extent to which one may discern a clerical estate (dukhovnoe sostoianie) for the foreign confessions, the standards by which individuals were recognized as belonging to non-Orthodox clergies, and the rights and privileges to which these religious servitors were entitled. Drawing on a distinction offered by Vasilii Kliuchevskii between “estate rights” (soslovnye prava) and “service rights” (dolzhnostnye prava), the author argues that Christian servitors, beginning with the Orthodox clergy, gradually acquired estate rights, while non-Christian servitors were generally able to acquire only service rights. The reasons for this outcome should be sought in a combination of practical and ideological concerns having to do with the state’s limited knowledge about non-Orthodox servitors, its commitments to the privileging of (Orthodox) Christianity in Russia’s social order, and broader shifts in the state’s soslovie policies.RĂ©sumĂ©Par son analyse du statut social des serviteurs religieux des confessions non-orthodoxes de Russie (« les confessions Ă©trangĂšres »), cet essai aborde une dimension impĂ©riale significative de la question des ordres (sosloviia).     Investigating the effect of target of rapamycin kinase inhibition on the Chlamydomonas reinhardtii phosphoproteome: from known homologs to new targets

    Recuperado de: https://www.biorxiv.org/content/10.1101/310102v1Target of rapamycin (TOR) kinase is a conserved regulator of cell growth whose activity is modulated in response to nutrients, energy and stress. Key proteins involved in the pathway are conserved in the model photosynthetic microalga Chlamydomonas reinhardtii, but the substrates of TOR kinase and downstream signaling network have not been elucidated. Our study provides a new resource for investigating the phosphorylation networks governed by the TOR kinase pathway in Chlamydomonas. We used quantitative phosphoproteomics to investigate the effects of inhibiting Chlamydomonas TOR kinase on dynamic protein phosphorylation. Wild-type and AZD-insensitive Chlamydomonas strains were treated with TOR-specific chemical inhibitors (rapamycin, AZD8055 and Torin1), after which differentially affected phosphosites were identified. Our quantitative phosphoproteomic dataset comprised 2547 unique phosphosites from 1432 different proteins. Inhibition of TOR kinase caused significant quantitative changes in phosphorylation at 258 phosphosites, from 219 unique phosphopeptides. Our results include Chlamydomonas homologs of TOR signaling-related proteins, including a site on RPS6 with a decrease in phosphorylation. Additionally, phosphosites on proteins involved in translation and carotenoid biosynthesis were identified. Follow-up experiments guided by these phosphoproteomic findings in lycopene beta/epsilon cyclase showed that carotenoid levels are affected by TORC1 inhibition and carotenoid production is under TOR control in algae.National Science Foundation CAREER MCB-155252

    Terahertz frequency standard based on three-photon coherent population trapping

    A scheme for a THz frequency standard based on three-photon coherent population trapping in stored ions is proposed. Assuming the propagation directions of the three lasers obey the phase matching condition, we show that stability of few 10−14^{-14} at one second can be reached with a precision limited by power broadening to 10−1110^{-11} in the less favorable case.     Evaluation of Ceftaroline Activity against Heteroresistant Vancomycin-Intermediate Staphylococcus aureus and Vancomycin-Intermediate Methicillin-Resistant S. aureus Strains in an In Vitro Pharmacokinetic/Pharmacodynamic Model: Exploring the “Seesaw Effect”

    A “seesaw effect” in methicillin-resistant Staphylococcus aureus (MRSA) has been demonstrated, whereby susceptibility to ÎČ-lactam antimicrobials increases as glyco- and lipopeptide susceptibility decreases. We investigated this effect by evaluating the activity of the anti-MRSA cephalosporin ceftaroline against isogenic pairs of MRSA strains with various susceptibilities to vancomycin in an in vitro pharmacokinetic/pharmacodynamic (PK/PD) model. The activities of ceftaroline at 600 mg every 12 h (q12h) (targeted free maximum concentration of drug in serum [fCmax], 15.2 ÎŒg/ml; half-life [t1/2], 2.3 h) and vancomycin at 1 g q12h (targeted fCmax, 18 ÎŒg/ml; t1/2, 6 h) were evaluated against 3 pairs of isogenic clinical strains of MRSA that developed increased MICs to vancomycin in patients while on therapy using a two-compartment hollow-fiber PK/PD model with a starting inoculum of ∌107 CFU/ml over a 96-h period. Bacterial killing and development of resistance were evaluated. Expression of penicillin-binding proteins (PBPs) 2 and 4 was evaluated by reverse transcription (RT)-PCR. The achieved pharmacokinetic parameters were 98 to 119% of the targeted values. Ceftaroline and vancomycin were bactericidal against 5/6 and 1/6 strains, respectively, at 96 h. Ceftaroline was more active against the mutant strains than the parent strains, with this difference being statistically significant for 2/3 strain pairs at 96 h. The level of PBP2 expression was 4.4× higher in the vancomycin-intermediate S. aureus (VISA) strain in 1/3 pairs. The levels of PBP2 and PBP4 expression were otherwise similar between the parent and mutant strains. These data support the seesaw hypothesis that ceftaroline, like traditional ÎČ-lactams, is more active against strains that are less susceptible to vancomycin even when the ceftaroline MICs are identical.     HITRAP: A facility at GSI for highly charged ions

    An overview and status report of the new trapping facility for highly charged ions at the Gesellschaft fuer Schwerionenforschung is presented. The construction of this facility started in 2005 and is expected to be completed in 2008. Once operational, highly charged ions will be loaded from the experimental storage ring ESR into the HITRAP facility, where they are decelerated and cooled. The kinetic energy of the initially fast ions is reduced by more than fourteen orders of magnitude and their thermal energy is cooled to cryogenic temperatures. The cold ions are then delivered to a broad range of atomic physics experiments.Comment: 8 pages, 11 figure
