    Vectorcardiographic changes during extended space flight

    To assess the effects of space flight on cardiac electrical properties, vectorcardiograms were taken on the 9 Skylab astronauts during the flights of 28, 59, and 84 days. The Frank lead system was used and observations were made at rest; during 25%, 50% and 75% of maximum exercise; during a short pulse of exercise (150 watts, 2 minutes); and after exercise. Data from 131 in-flight tests were analyzed by computer and compared to preflight and postflight values. Statistically significant increase in QRS vector magnitude (six of nine crewmen); T vector magnitude (five of nine crewmen); and resting PR interval duration (six of nine crewmen) occurred. During exercise the PR interval did not differ from preflight. Exercise heart rates inflight were the same as preflight, but increased in the immediate postflight period. With the exception of the arrhythmias, no deleterious vectorcardiographic changes were observed during the Skylab missions

    The Gut Microbiota Composition in Dichorionic Triplet Sets Suggests a Role for Host Genetic Factors

    peer-reviewedMonozygotic and dizygotic twin studies investigating the relative roles of host genetics and environmental factors in shaping gut microbiota composition have produced conflicting results. In this study, we investigated the gut microbiota composition of a healthy dichorionic triplet set. The dichorionic triplet set contained a pair of monozygotic twins and a fraternal sibling, with similar pre- and post-natal environmental conditions including feeding regime. V4 16S rRNA and rpoB amplicon pyrosequencing was employed to investigate microbiota composition, and the species and strain diversity of the culturable bifidobacterial population was also examined. At month 1, the monozygotic pair shared a similar microbiota distinct to the fraternal sibling. By month 12 however, the profile was more uniform between the three infants. Principal coordinate analysis (PCoA) of the microbiota composition revealed strong clustering of the monozygotic pair at month 1 and a separation of the fraternal infant. At months 2 and 3 the phylogenetic distance between the monozygotic pair and the fraternal sibling has greatly reduced and by month 12 the monozygotic pair no longer clustered separately from the fraternal infant. Pulse field gel electrophoresis (PFGE) analysis of the bifidobacterial population revealed a lack of strain diversity, with identical strains identified in all three infants at month 1 and 12. The microbiota of two antibiotic-treated dichorionic triplet sets was also investigated. Not surprisingly, in both triplet sets early life antibiotic administration appeared to be a major determinant of microbiota composition at month 1, irrespective of zygosity. By month 12, early antibiotic administration appeared to no longer exert such a strong influence on gut microbiota composition. We hypothesize that initially host genetics play a significant role in the composition of an individual’s gut microbiota, unless an antibiotic intervention is given, but by month 12 environmental factors are the major determinant.This study was performed as part of the INFANTMET project (10/RD/Infantmet/MFRC/705) and was funded by the Government of Ireland's Department of Agriculture Fisheries and in part by Alimentary Pharmabiotic Centre.     Liquid Phase Electrochemistry at Ultralow Temperatures

    Fluid electrolyte solutions based on mixtures of butyronitrile (PrCN) and ethyl chloride (EtCl) with or as electrolyte freeze below −180°C and provide excellent media for cryogenic electrochemical experiments. A 1:2 mixture of PrCN and EtCl exhibits the best combination of freezing point and ionic conductivity for ultralow temperature electrochemistry. Diffusion coefficients for bis(pentamethylcyclopentadienyl) iron are measurable by potential step chronoamperometry down to −160°C using a conventionally sized electrode, but the resistivity of the solvent mixture is such that potential sweep voltammetry benefits from the use of microdisk (10 and 25 μm diam Pt) or microband (0.2 μm wide Au) electrodes.     The Composition of Human Milk and Infant Faecal Microbiota Over the First Three Months of Life: A Pilot Study

    peer-reviewedHuman milk contains a diverse array of bioactives and is also a source of bacteria for the developing infant gut. The aim of this study was to characterize the bacterial communities in human milk and infant faeces over the first 3 months of life, in 10 mother-infant pairs. The presence of viable Bifidobacterium and Lactobacillus in human milk was also evaluated. MiSeq sequencing revealed a large diversity of the human milk microbiota, identifying over 207 bacterial genera in milk samples. The phyla Proteobacteria and Firmicutes and the genera Pseudomonas, Staphylococcus and Streptococcus were the predominant bacterial groups. A core of 12 genera represented 81% of the microbiota relative abundance in milk samples at week 1, 3 and 6, decreasing to 73% at week 12. Genera shared between infant faeces and human milk samples accounted for 70–88% of the total relative abundance in infant faecal samples, supporting the hypothesis of vertical transfer of bacteria from milk to the infant gut. In addition, identical strains of Bifidobacterium breve and Lactobacillus plantarum were isolated from the milk and faeces of one mother-infant pair. Vertical transfer of bacteria via breastfeeding may contribute to the initial establishment of the microbiota in the developing infant intestine

    The previous chapter established that nearly all of what we mapped at Chunchucmil was occupied at the end of the Early Classic period. This allows us to combine mapping and excavation to discuss the structure of the city at this critical time. As we explain below, settlement in and around Chunchucmil was not homogeneous. We lump this settlement into five zones each with different characteristics: (1) site center, (2) residential core, (3) residential periphery, (4) settlement fingers, and (5) hinterland. This chapter presents the characteristics (size, settlement density, kinds of occupation) of these zones and then supplies a population estimate

    We prove that if a finite group HH has a generalized involution model, as defined by Bump and Ginzburg, then the wreath product H≀SnH \wr S_n also has a generalized involution model. This extends the work of Baddeley concerning involution models for wreath products. As an application, we construct a Gelfand model for wreath products of the form A≀SnA \wr S_n with AA abelian, and give an alternate proof of a recent result due to Adin, Postnikov, and Roichman describing a particularly elegant Gelfand model for the wreath product \ZZ_r \wr S_n. We conclude by discussing some notable properties of this representation and its decomposition into irreducible constituents, proving a conjecture of Adin, Roichman, and Postnikov's.Comment: 29 page

    STATISTICAL METHODS FOR AFFYMETRIX TILING ARRAY DATA

    Tiling arrays are a microarray technology currently being used for a variety of genomic and epigenomic applications, such as the mapping of transcription, DNA methylation, and histone modifications. Tiling arrays provide high-density coverage of a genome, or a genomic region, through the systematic and sequential placement of probes without regard to genome annotation. In this paper we compare the Affymetrix tiling array to the Affymetrix GeneChip® 3’ expression array and propose methods that address statistical and bioinformatic issues that accompany gene expression data that are generated from Affymetrix tiling arrays.     Bifidobacterium breve with α-Linolenic Acid and Linoleic Acid Alters Fatty Acid Metabolism in the Maternal Separation Model of Irritable Bowel Syndrome

    peer-reviewedThe aim of this study was to compare the impact of dietary supplementation with a Bifidobacterium breve strain together with linoleic acid & α-linolenic acid, for 7 weeks, on colonic sensitivity and fatty acid metabolism in rats. Maternally separated and non-maternally separated Sprague Dawley rats (n = 15) were orally gavaged with either B. breve DPC6330 (109 microorganisms/day) alone or in combination with 0.5% (w/w) linoleic acid & 0.5% (w/w) α-linolenic acid, daily for 7 weeks and compared with trehalose and bovine serum albumin. Tissue fatty acid composition was assessed by gas-liquid chromatography and visceral hypersensitivity was assessed by colorectal distension. Significant differences in the fatty acid profiles of the non-separated controls and maternally separated controls were observed for α-linolenic acid and arachidonic acid in the liver, oleic acid and eicosenoic acid (c11) in adipose tissue, and for palmitoleic acid and docosahexaenoic acid in serum (p<0.05). Administration of B. breve DPC6330 to MS rats significantly increased palmitoleic acid, arachidonic acid and docosahexaenoic acid in the liver, eicosenoic acid (c11) in adipose tissue and palmitoleic acid in the prefrontal cortex (p<0.05), whereas feeding B. breve DPC6330 to non separated rats significantly increased eicosapentaenoic acid and docosapentaenoic acid in serum (p<0.05) compared with the NS un-supplemented controls. Administration of B. breve DPC6330 in combination with linoleic acid and α-linolenic acid to maternally separated rats significantly increased docosapentaenoic acid in the serum (p<0.01) and α-linolenic acid in adipose tissue (p<0.001), whereas feeding B. breve DPC6330 with fatty acid supplementation to non-separated rats significantly increased liver and serum docosapentaenoic acid (p<0.05), and α-linolenic acid in adipose tissue (p<0.001). B. breve DPC6330 influenced host fatty acid metabolism. Administration of B. breve DPC6330 to maternally separated rats significantly modified the palmitoleic acid, arachidonic acid and docosahexaenoic acid contents in tissues.
