    Improving the precision of dynamic forest parameter estimates using Landsat

    The use of satellite-derived classification maps to improve post-stratified forest parameter estimates is well established.When reducing the variance of post-stratification estimates for forest change parameters such as forest growth, it is logical to use a change-related strata map. At the stand level, a time series of Landsat images is ideally suited for producing such a map. In this study, we generate strata maps based on trajectories of Landsat Thematic Mapper-based normalized difference vegetation index values, with a focus on post-disturbance recovery and recent measurements. These trajectories, from1985 to 2010, are converted to harmonic regression coefficient estimates and classified according to a hierarchical clustering algorithm from a training sample. The resulting strata maps are then used in conjunction with measured plots to estimate forest status and change parameters in an Alabama, USA study area. These estimates and the variance of the estimates are then used to calculate the estimated relative efficiencies of the post-stratified estimates. Estimated relative efficiencies around or above 1.2 were observed for total growth, total mortality, and total removals, with different strata maps being more effective for each. Possible avenues for improvement of the approach include the following: (1) enlarging the study area and (2) using the Landsat images closest to the time of measurement for each plot.     Stimulation of shrimp (Penaeus monodon) hemocytes by lipopolysaccharide-like molecules derived from Novacq™

    Immune stimulation through feed additives is a promising strategy that can help to combat disease in shrimp farming and reduce the use of antibiotics and other chemotherapeutics. The present study investigated the in vitro immunostimulatory effects of lipopolysaccharide (LPS)-like molecules isolated from the microbial based feed additive Novacq™ (N-LPS). The presence of LPS-like molecules was confirmed and quantified Novacq™ using a HEK-TLR4 reporter cell line. Primary hemocytes isolated from adult Penaeus monodon were used to measure the immunostimulatory of N-LPS compared with the control group that were treated with E. coli derived LPS (E-LPS). The N-LPS stimulated a rapid and significant induction of the phenoloxidase (PO) response in the hemocytes. The PO response increased with exposure time and LPS concentration and was significantly higher compared with an E. coli LPS (E-LPS) control. In addition, using gene expression data, we quantified the transcriptome response of the hemocytes at 15, 30 and 60 mins post stimulation. Compared with the controls, the N-LPS treated hemocytes had a significant up-regulation of genes involved in the immune system modulation and control at all time-points. Most noteworthy was the significant induction of transcripts that function as serine protease inhibitors (namely SERPINs), that regulate the overexpression of the PO system. Transcription factors from the Notch family which directly regulate the expression of many immune genes were also induced within the hemocytes. Additionally, we also saw a strong up-regulation of crustacean hyperglycemic hormone (CHH) transcripts, an important neuropeptide involved in immune function. Overall, the transcriptome profile of the hemocytes suggests that the LPS component of Novacq™ is highly immunostimulatory and generates a strong PO response in vitro. The subsequent transcriptional response appears to be directed towards preventing further activation of the PO system most likely in an attempt to limit cytoxicity to the host. Our study highlights the immunostimulatory ability of Novacq™ and provides further evidence of the positive health benefits this microbial based feed additive can have in shrimp.</p

    The ROTSE-III Robotic Telescope System

    The observation of a prompt optical flash from GRB990123 convincingly demonstrated the value of autonomous robotic telescope systems. Pursuing a program of rapid follow-up observations of gamma-ray bursts, the Robotic Optical Transient Search Experiment (ROTSE) has developed a next-generation instrument, ROTSE-III, that will continue the search for fast optical transients. The entire system was designed as an economical robotic facility to be installed at remote sites throughout the world. There are seven major system components: optics, optical tube assembly, CCD camera, telescope mount, enclosure, environmental sensing & protection and data acquisition.     Deep-water macroalgae from the Canary Islands: new records and biogeographical relationships

    Due to the geographical location and paleobiogeography of the Canary Islands, the seaweed flora contains macroalgae with different distributional patterns. In this contribution, the biogeographical relations of several new records of deep-water macroalgae recently collected around the Canarian archipelago are discussed. These are Bryopsidella neglecta (Berthotd) Rietema,Discosporangium mesarthrocarpum (Meneghini) Hauck, Hincksia onslowensis (Amsler et Kapraun)P.C. Silva, Syringoderma floridana Henry, Peyssonnelia harveyana J. Agardh, Cryptonemia seminervis(C. Agardh) J. Agardh, Botryodadia wynnei Ballantine, Gloiocladia blomquistii (Searles) R. E.Norris, PIahchrysis peltata (W. R. Taylor) P. Huv4 et H. Huv4, Leptofauchea brasiliensis Joly, and Sarcodiotheca divaricata W. R. Taylor. These new records, especially those in the Florideophyceae,support the strong affinity of the Canary Islands seaweed flora with the warm-temperate Mediterranean-Atlantic region. Some species are recorded for the first time from the east coast of the Atlantic Ocean, enhancing the biogeographic relations of the Canarian marine flora with that of the western Atlantic regions

    Vertical distribution of buoyant Microcystis blooms in a Lagrangian particle tracking model for short‐term forecasts in Lake Erie

    Cyanobacterial harmful algal blooms (CHABs) are a problem in western Lake Erie, and in eutrophic fresh waters worldwide. Western Lake Erie is a large (3000 km2), shallow (8 m mean depth), freshwater system. CHABs occur from July to October, when stratification is intermittent in response to wind and surface heating or cooling (polymictic). Existing forecast models give the present location and extent of CHABs from satellite imagery, then predict two‐dimensional (surface) CHAB movement in response to meteorology. In this study, we simulated vertical distribution of buoyant Microcystis colonies, and 3‐D advection, using a Lagrangian particle model forced by currents and turbulent diffusivity from the Finite Volume Community Ocean Model (FVCOM). We estimated the frequency distribution of Microcystis colony buoyant velocity from measured size distributions and buoyant velocities. We evaluated several random‐walk numerical schemes to efficiently minimize particle accumulation artifacts. We selected the Milstein scheme, with linear interpolation of the diffusivity profile in place of cubic splines, and varied the time step at each particle and step based on the curvature of the local diffusivity profile to ensure that the Visser time step criterion was satisfied. Inclusion of vertical mixing with buoyancy significantly improved model skill statistics compared to an advection‐only model, and showed greater skill than a persistence forecast through simulation day 6, in a series of 26 hindcast simulations from 2011.     Relationship between quantum decoherence times and solvation dynamics in condensed phase chemical systems

    A relationship between the time scales of quantum coherence loss and short-time solvent response for a solute/bath system is derived for a Gaussian wave packet approximation for the bath. Decoherence and solvent response times are shown to be directly proportional to each other, with the proportionality coefficient given by the ratio of the thermal energy fluctuations to the fluctuations in the system-bath coupling. The relationship allows the prediction of decoherence times for condensed phase chemical systems from well developed experimental methods.Comment: 10 pages, no figures, late

    Trust in Nanotechnology? On Trust as Analytical Tool in Social Research on Emerging Technologies

    Trust has become an important aspect of evaluating the relationship between lay public and technology implementation. Experiences have shown that a focus on trust provides a richer understanding of reasons for backlashes of technology in society than a mere focus of public understanding of risks and science communication. Therefore, trust is also widely used as a key concept for understanding and predicting trust or distrust in emerging technologies. But whereas trust broadens the scope for understanding established technologies with well-defined questions and controversies, it easily fails to do so with emerging technologies, where there are no shared questions, a lack of public familiarity with the technology in question, and a restricted understanding amongst social researchers as to where distrust is likely to arise and how and under which form the technology will actually be implemented. Rather contrary, ‘trust’ might sometimes even direct social research into fixed structures that makes it even more difficult for social research to provide socially robust knowledge. This article therefore suggests that if trust is to maintain its important role in evaluating emerging technologies, the approach has to be widened and initially focus not on people’s motivations for trust, but rather the object of trust it self, as to predicting how and where distrust might appear, how the object is established as an object of trust, and how it is established in relation with the public

    Development of a new real-time PCR for the detection of pilchard orthomyxovirus (POMV) in apparently healthy fish

    Pilchard orthomyxovirus (POMV) is a virus of concern to the Atlantic salmon aquaculture industry in Tasmania. First isolated from wild pilchards in southern Australia in 1998, the virus is now a recognised pathogen of farmed Atlantic salmon (Salmo salar) in Tasmania. While the current real-time PCR for POMV targets segment 5 of the viral genome, recent viral gene expression data suggests that other segments of the POMV genome presented higher transcription levels and thus may be better candidates for the early detection of the virus. This study aimed to design and begin validating a more sensitive reverse transcriptase real-time PCR (RT-qPCR) assay to detect POMV. Primers and probes were developed targeting two independent viral genes derived from segments 7 and 8, which presented higher transcription levels than segment 5 in both cell culture and infected fish. These were compared with the current POMV RT-qPCR. The POMV segment 8 assay had a higher analytical sensitivity than segment 7, detecting at least 1 plasmid copy μl−1, and was 10-fold more sensitive than both POMV segment 7 and 5 assays when analysing nucleic acid from a positive field sample. Both new assays also had high analytical specificity, detecting the 11 POMV isolates tested (inclusivity testing) and not amplifying nucleic acids from other viruses, including ISAV, a related orthomyxovirus. In the latent class model analysis, the diagnostic sensitivity of the segment 8 and 7 assays were higher than segment 5 in 93% and 92% of simulations, respectively. Seven samples (18.4%), all from subclinical fish infected with POMV, returned a positive result only with the segment 8 assay. Both new assays showed reproducible results when applied to aliquots of the same samples tested in three different laboratories. The new POMV segment 8 assay shows promising results as a surveillance tool for detecting POMV in fish without any symptoms.publishedVersio

    Analysis of chloroplast genomes and a supermatrix inform reclassification of the Rhodomelaceae (Rhodophyta).

    With over a thousand species, the Rhodomelaceae is the most species-rich family of red algae. While its genera have been assigned to 14 tribes, the high-level classification of the family has never been evaluated with a molecular phylogeny. Here, we reassess its classification by integrating genome-scale phylogenetic analysis with observations of the morphological characters of clades. In order to resolve relationships among the main lineages of the family we constructed a phylogeny with 55 chloroplast genomes (52 newly determined). The majority of branches were resolved with full bootstrap support. We then added 266 rbcL, 125 18S rRNA gene and 143 cox1 sequences to construct a comprehensive phylogeny containing nearly half of all known species in the family (407 species in 89 genera). These analyses suggest the same subdivision into higher-level lineages, but included many branches with moderate or poor support. The circumscription for nine of the 13 previously described tribes was supported, but the Lophothalieae, Polysiphonieae, Pterosiphonieae and Herposiphonieae required revision, and five new tribes and one resurrected tribe were segregated from them. Rhizoid anatomy is highlighted as a key diagnostic character for the morphological delineation of several lineages. This work provides the most extensive phylogenetic analysis of the Rhodomelaceae to date and successfully resolves the relationships among major clades of the family. Our data show that organellar genomes obtained through high-throughput sequencing produce well-resolved phylogenies of difficult groups, and their more general application in algal systematics will likely permit deciphering questions about classification at many taxonomic levels
