Data Deluge in Astrophysics: Photometric Redshifts as a Template Use Case
Astronomy has entered the big data era and Machine Learning based methods
have found widespread use in a large variety of astronomical applications. This
is demonstrated by the recent huge increase in the number of publications
making use of this new approach. The usage of machine learning methods, however
is still far from trivial and many problems still need to be solved. Using the
evaluation of photometric redshifts as a case study, we outline the main

Law of Large Numbers: The Theory, Applications and Technology-Based Education
Modern approaches for technology-based blended education utilize a variety of recently developed novel pedagogical, computational and network resources. Such attempts employ technology to deliver integrated, dynamically-linked, interactive-content and heterogeneous learning environments, which may improve student comprehension and information retention. In this paper, we describe one such innovative effort of using technological tools to expose students in probability and statistics courses to the theory, practice and usability of the Law of Large Numbers (LLN). We base our approach on integrating pedagogical instruments with the computational libraries developed by the Statistics Online Computational Resource (www.SOCR.ucla.edu). To achieve this merger we designed a new interactive Java applet and a corresponding demonstration activity that illustrate the concept and the applications of the LLN. The LLN applet and activity have common goals – to provide graphical representation of the LLN principle, build lasting student intuition and present the common misconceptions about the law of large numbers. CLASH-VLT: Substructure in the galaxy cluster MACS J1206.2-0847 from kinematics of galaxy populations
In the effort to understand the link between the structure of galaxy clusters
and their galaxy populations, we focus on MACSJ1206.2-0847 at z~0.44 and probe
its substructure in the projected phase space through the spectrophotometric
properties of a large number of galaxies from the CLASH-VLT survey. Our
analysis is mainly based on an extensive spectroscopic dataset of 445 member
galaxies, mostly acquired with VIMOS@VLT as part of our ESO Large Programme,
sampling the cluster out to a radius ~2R200 (4 Mpc). We classify 412 galaxies
as passive, with strong Hdelta absorption (red and blue galaxies, and with
emission lines from weak to very strong. A number of tests for substructure
detection are applied to analyze the galaxy distribution in the velocity space,
in 2D space, and in 3D projected phase-space. Studied in its entirety, the
cluster appears as a large-scale relaxed system with a few secondary, minor
overdensities in 2D distribution. We detect no velocity gradients or evidence
of deviations in local mean velocities. The main feature is the WNW-ESE
elongation. The analysis of galaxy populations per spectral class highlights a
more complex scenario. The passive galaxies and red strong Hdelta galaxies
trace the cluster center and the WNW-ESE elongated structure. The red strong
Hdelta galaxies also mark a secondary, dense peak ~2 Mpc at ESE. The emission
line galaxies cluster in several loose structures, mostly outside R200. The
observational scenario agrees with MACS J1206.2-0847 having WNW-ESE as the
direction of the main cluster accretion, traced by passive galaxies and red
strong Hdelta galaxies. The red strong Hdelta galaxies, interpreted as
poststarburst galaxies, date a likely important event 1-2 Gyr before the epoch
of observation. The emission line galaxies trace a secondary, ongoing infall

CLASH-VLT: The mass, velocity-anisotropy, and pseudo-phase-space density profiles of the z=0.44 galaxy cluster MACS 1206.2-0847
We use an unprecedented data-set of about 600 redshifts for cluster members,
obtained as part of a VLT/VIMOS large programme, to constrain the mass profile
of the z=0.44 cluster MACS J1206.2-0847 over the radial range 0-5 Mpc (0-2.5
virial radii) using the MAMPOSSt and Caustic methods. We then add external
constraints from our previous gravitational lensing analysis. We invert the
Jeans equation to obtain the velocity-anisotropy profiles of cluster members.
With the mass-density and velocity-anisotropy profiles we then obtain the first
determination of a cluster pseudo-phase-space density profile. The kinematics
and lensing determinations of the cluster mass profile are in excellent
agreement. This is very well fitted by a NFW model with mass M200=(1.4 +- 0.2)
10^15 Msun and concentration c200=6 +- 1, only slightly higher than theoretical
expectations. Other mass profile models also provide acceptable fits to our
data, of (slightly) lower (Burkert, Hernquist, and Softened Isothermal Sphere)
or comparable (Einasto) quality than NFW. The velocity anisotropy profiles of
the passive and star-forming cluster members are similar, close to isotropic
near the center and increasingly radial outside. Passive cluster members follow
extremely well the theoretical expectations for the pseudo-phase-space density
profile and the relation between the slope of the mass-density profile and the
velocity anisotropy. Star-forming cluster members show marginal deviations from
theoretical expectations. This is the most accurate determination of a cluster
mass profile out to a radius of 5 Mpc, and the only determination of the
velocity-anisotropy and pseudo-phase-space density profiles of both passive and

Comparison of Evaluations for Heart Transplant Before Durable Left Ventricular Assist Device and Subsequent Receipt of Transplant at Transplant vs Nontransplant Centers
IMPORTANCE: In 2020, the Centers for Medicare & Medicaid Services revised its national coverage determination, removing the requirement to obtain review from a Medicare-approved heart transplant center to implant a durable left ventricular assist device (LVAD) for bridge-to-transplant (BTT) intent at an LVAD-only center. The association between center-level transplant availability and access to heart transplant, the gold-standard therapy for advanced heart failure (HF), is unknown.
OBJECTIVE: To investigate the association of center transplant availability with LVAD implant strategies and subsequent heart transplant following LVAD implant before the Centers for Medicare & Medicaid Services policy change.
DESIGN, SETTING, AND PARTICIPANTS: A retrospective cohort study of the Society of Thoracic Surgeons Intermacs multicenter US registry database was conducted from April 1, 2012, to June 30, 2020. The population included patients with HF receiving a primary durable LVAD.
EXPOSURES: LVAD center transplant availability (LVAD/transplant vs LVAD only).
MAIN OUTCOMES AND MEASURES: The primary outcomes were implant strategy as BTT and subsequent transplant by 2 years. Covariates that might affect listing strategy and outcomes were included (eg, patient demographic characteristics, comorbidities) in multivariable models. Parameters for BTT listing were estimated using logistic regression with center-level random effects and for receipt of a transplant using a Cox proportional hazards regression model with death as a competing event.
RESULTS: The sample included 22 221 LVAD recipients with a median age of 59.0 (IQR, 50.0-67.0) years, of whom 17 420 (78.4%) were male and 3156 (14.2%) received implants at LVAD-only centers. Receiving an LVAD at an LVAD/transplant center was associated with a 79% increased adjusted odds of BTT LVAD designation (odds ratio, 1.79; 95% CI, 1.35-2.38; P \u3c .001). The 2-year transplant rate following LVAD implant was 25.6% at LVAD/transplant centers and 11.9% at LVAD-only centers. There was an associated 33% increased rate of transplant at LVAD/transplant centers compared with LVAD-only centers (adjusted hazard ratio, 1.33; 95% CI, 1.17-1.51) with a similar hazard for death at 2 years (adjusted hazard ratio, 0.99; 95% CI, 0.90-1.08).
CONCLUSIONS AND RELEVANCE: Receiving an LVAD at an LVAD-transplant center was associated with increased odds of BTT intent at implant and subsequent transplant receipt for patients at 2 years. The findings of this study suggest that Centers for Medicare & Medicaid Services policy change may have the unintended consequence of further increasing inequities in access to transplant among patients at LVAD-only centers
The attitudes of brain cancer patients and their caregivers towards death and dying: a qualitative study
<p>Abstract</p> <p>Background</p> <p>Much money and energy has been spent on the study of the molecular biology of malignant brain tumours. However, little attention has been paid to the wishes of patients afflicted with these incurable tumours, and how this might influence treatment considerations.</p> <p>Methods</p> <p>We interviewed 29 individuals – 7 patients dying of a malignant brain tumor and 22 loved ones. One-on-one interviews were conducted according to a pre-designed interview guide. A combination of open-ended questions, as well as clinical scenarios was presented to participants in order to understand what is meaningful and valuable to them when determining treatment options and management approaches. The results were analyzed, coded, and interpreted using qualitative analytic techniques in order to arrive at several common overarching themes.</p> <p>Results</p> <p>Seven major themes were identified. In general, respondents were united in viewing brain cancer as unique amongst malignancies, due in large part to the premium placed on mental competence and cognitive functioning. Importantly, participants found their experiences, however difficult, led to the discovery of inner strength and resilience. Responses were usually framed within an interpersonal context, and participants were generally grateful for the opportunity to speak about their experiences. Attitudes towards religion, spirituality, and euthanasia were also probed.</p> <p>Conclusion</p> <p>Several important themes underlie the experiences of brain cancer patients and their caregivers. It is important to consider these when managing these patients and to respect not only their autonomy but also the complex interpersonal toll that a malignant diagnosis can have.</p
Bioisosteric discovery of NPA101.3, a second generation RET/VEGFR2 inhibitor optimized for single-agent polypharmacology
RET receptor tyrosine kinase is a driver oncogene in human cancer. We recently identified the clinical drug candidate Pz-1, which targets RET and VEGFR2. A key in vivo metabolite of Pz-1 is its less active demethylated pyrazole analogue. Using bioisosteric substitution methods, here, we report the identification of NPA101.3, lacking the structural liability for demethylation. NPA101.3 showed selective inhibitory profile and an inhibitory concentration 50 (IC50) of <0.001 ÎĽM for both RET and VEGFR2. NPA101.3 inhibited phosphorylation of all tested RET oncoproteins as well as VEGFR2 and proliferation of cells transformed by RET. Oral administration of NPA101.3 (10 mg/kg/day) completely prevented formation of tumours induced by RET/C634Y-transformed cells, while it decreased, but did not abrogate, formation of tumours induced by a control oncogene (HRAS/G12V). The balanced synchronous inhibition of both RET and VEGFR2, as well the resistance to demethylation, renders NPA101.3 a potential clinical candidate for RET-driven cancers
Water-repellent cellulose fiber networks with multifunctional properties.
We demonstrate a simple but highly efficient technique to introduce multifunctional properties to cellulose fiber networks by wetting them with ethyl-cyanoacrylate monomer solutions containing various suspended organic submicrometer particles or inorganic nanoparticles. Solutions can be applied on cellulosic surfaces by simple solution casting techniques or by dip coating, both being suitable for large area applications. Immediately after solvent evaporation, ethyl-cyanoacrylate starts cross-linking around cellulose fibers under ambient conditions because of naturally occurring surface hydroxyl groups and adsorbed moisture, encapsulating them with a hydrophobic polymer shell. Furthermore, by dispersing various functional particles in the monomer solutions, hydrophobic ethyl-cyanoacrylate nanocomposites with desired functionalities can be formed around the cellulose fibers.
