    Biophysical and Functional Characterization of Rhesus Macaque IgG Subclasses

    Antibodies raised in Indian rhesus macaques [Macaca mulatta (MM)] in many preclinical vaccine studies are often evaluated in vitro for titer, antigen-recognition breadth, neutralization potency, and/or effector function, and in vivo for potential associations with protection. However, despite reliance on this key animal model in translation of promising candidate vaccines for evaluation in first in man studies, little is known about the properties of MM immunoglobulin G (IgG) subclasses and how they may compare to human IgG subclasses. Here, we evaluate the binding of MM IgG1, IgG2, IgG3, and IgG4 to human Fc gamma receptors (FcgammaR) and their ability to elicit the effector functions of human FcgammaR-bearing cells, and unlike in humans, find a notable absence of subclasses with dramatically silent Fc regions. Biophysical, in vitro, and in vivo characterization revealed MM IgG1 exhibited the greatest effector function activity followed by IgG2 and then IgG3/4. These findings in rhesus are in contrast with the canonical understanding that IgG1 and IgG3 dominate effector function in humans, indicating that subclass-switching profiles observed in rhesus studies may not strictly recapitulate those observed in human vaccine studies

    Methane observations from the Greenhouse Gases Observing SATellite: Comparison to ground‐based TCCON data and model calculations

    We report new short-wave infrared (SWIR) column retrievals of atmospheric methane (X_(CH4)) from the Japanese Greenhouse Gases Observing SATellite (GOSAT) and compare observed spatial and temporal variations with correlative ground-based measurements from the Total Carbon Column Observing Network (TCCON) and with the global 3-D GEOS-Chem chemistry transport model. GOSAT X_(CH4) retrievals are compared with daily TCCON observations at six sites between April 2009 and July 2010 (Bialystok, Park Falls, Lamont, Orleans, Darwin and Wollongong). GOSAT reproduces the site-dependent seasonal cycles as observed by TCCON with correlations typically between 0.5 and 0.7 with an estimated single-sounding precision between 0.4–0.8%. We find a latitudinal-dependent difference between the X_(CH4) retrievals from GOSAT and TCCON which ranges from 17.9 ppb at the most northerly site (Bialystok) to −14.6 ppb at the site with the lowest latitude (Darwin). We estimate that the mean smoothing error difference included in the GOSAT to TCCON comparisons can account for 15.7 to 17.4 ppb for the northerly sites and for 1.1 ppb at the lowest latitude site. The GOSAT X_(CH4) retrievals agree well with the GEOS-Chem model on annual (August 2009 – July 2010) and monthly timescales, capturing over 80% of the zonal variability. Differences between model and observed X_(CH4) are found over key source regions such as Southeast Asia and central Africa which will be further investigated using a formal inverse model analysis

    Sex differences in age-related decline of urinary insulin-like growth factor-binding protein-3 levels in adult bonobos and chimpanzees

    There is increasing interest in the characterization of normative senescence in humans. To assess to what extent aging patterns in humans are unique, comparative data from closely-related species, such as non-human primates, can be very useful. Here we use data from bonobos and chimpanzees, two closely-related species that share a common ancestor with humans, to explore physiological markers that are indicative of aging processes. Many studies on aging in humans focus on the somatotropic axis, consisting of growth hormone (GH), insulin-like growth factors (IGFs), and IGF binding proteins (IGFBPs). In humans, IGFBP-3 levels decline steadily with increasing age. We used urinary IGFBP-3 levels as an alternative endocrine marker for IGF-I, to identify the temporal pattern known to be related with age-related changes in cell proliferation, growth, and apoptosis. We measured urinary IGFBP-3 levels in samples from 71 bonobos and 102 chimpanzees. Focusing on samples from individuals aged ten years or older we found that urinary IGFBP-3 levels decline in both ape species with increasing age. However, in both species, females start with higher urinary IGFBP-3 levels than males, experience a steeper decline with increasing age, and converge with male levels around the age of 30 to 35 years. Our measurements of urinary IGFBP-3 levels indicate that bonobos and chimpanzees mirror human patterns of age-related decline in IGFBP-3 in older individuals (< 10 years) of both sexes. Moreover, like humans, both ape species show sex-specific differences in IGFBP-3 levels with females having higher levels than males, a result that correlates with sex-differences in life expectancy. Using changes in urinary IGFBP-3 levels as a proxy for changes in GH and IGF-I levels that mark age-related changes in cell proliferation, this approach provides an opportunity to investigate trade-offs in life history strategies in cross-sectional and in longitudinal studies, both in captivity and in the wild

    Starch Digestion Enhances Bioaccessibility of Anti-Inflammatory Polyphenols from Borlotti Beans (Phaseolus vulgaris)

    The consumption of beans has been associated with chronic disease prevention which may be attributed to the polyphenols present in the seed coat and endosperm. However, their bioaccessibility is likely to be limited by interactions with bean matrix components, including starch, protein and fibre. The aim of this project was to evaluate the effect of domestic processing and enzymatic digestion on the bioaccessibility of polyphenols from Borlotti beans (Phaseolus vulgaris) and to test their anti-inflammatory properties in a macrophage cell model. In vitro digestion of cooked beans released twenty times more polyphenols (40.4 ± 2.5 mg gallic acid equivalents (GAE)/g) than domestic processing (2.22 ± 0.1 mg GAE/g), with starch digestion contributing to the highest release (30.9 ± 0.75 mg GAE/g). Fluorescence microscopy visualization of isolated bean starch suggests that polyphenols are embedded within the granule structure. LC-MS analysis showed that cooked Borlotti bean contain flavonoids, flavones and hydroxycinnamic acids, and cooked bean extracts exerted moderate anti-inflammatory effects by decreasing mRNA levels of IL1β and iNOS by 25% and 40%, respectively. In conclusion, the bioaccessibility of bean polyphenols is strongly enhanced by starch digestion. These polyphenols may contribute to the health benefits associated with bean consumption

    Design and Multiphysics Analysis of Direct and Cross-Coupled SIW Combline Filters Using Electric and Magnetic Couplings

    In this paper, combline substrate integrated waveguide (SIW) filters using electric and magnetic couplings are thoroughly studied. Thus, a negative coupling scheme consisting on an open-ended coplanar probe is proposed and analyzed in detail. Several in-line 3-pole filters at C-band are designed, manufactured and measured showing how the presented approach can be used for implementing direct couplings while enabling an important size reduction and improved spurious-free band compared to conventional magnetic irises. A fully-packaged quasi-elliptic 4-pole filter is also designed at 5.75 GHz showing how the negative coupling structure can be used for introducing transmission zeros by means of cross-couplings between non-adjacent resonators. Finally, average and peak power handling capabilities of these filters have been also analyzed from a multiphysics point of view. Measured results validate the theoretical predictions confirming that combline SIW filters can handle significant levels of continuous and peak power, providing at the same time easy integration, compact size and advanced filtering responses.This work was supported in part by MINECO (Spanish Government) under projects TEC2013-47037-C5-1-R and TEC2013-48036-C3-3-R.     Enhancing the therapeutic activity of hyperimmune IgG against chikungunya virus using FcγRIIIa affinity chromatography

    INTRODUCTION: Chikungunya virus (CHIKV) is a re-emerging mosquito transmitted alphavirus of global concern. Neutralizing antibodies and antibody Fc-effector functions have been shown to reduce CHIKV disease and infection in animals. However, the ability to improve the therapeutic activity of CHIKV-specific polyclonal IgG by enhancing Fc-effector functions through modulation of IgG subclass and glycoforms remains unknown. Here, we evaluated the protective efficacy of CHIKV-immune IgG enriched for binding to Fc-gamma receptor IIIa (FcγRIIIa) to select for IgG with enhanced Fc effector functions. METHODS: Total IgG was isolated from CHIKV-immune convalescent donors with and without additional purification by FcγRIIIa affinity chromatography. The enriched IgG was characterized in biophysical and biological assays and assessed for therapeutic efficacy during CHIKV infection in mice. RESULTS: FcγRIIIa-column purification enriched for afucosylated IgG glycoforms. In vitro characterization showed the enriched CHIKV-immune IgG had enhanced human FcγRIIIa and mouse FcγRIV affinity and FcγR-mediated effector function without reducing virus neutralization in cellular assays. When administered as post-exposure therapy in mice, CHIKV-immune IgG enriched in afucosylated glycoforms promoted reduction in viral load. DISCUSSION: Our study provides evidence that, in mice, increasing Fc engagement of FcγRs on effector cells, by leveraging FcγRIIIa-affinity chromatography, enhanced the antiviral activity of CHIKV-immune IgG and reveals a path to produce more effective therapeutics against these and potentially other emerging viruses

    Modelling the potential impacts of climate change on the distribution of ichthyoplankton in the Yangtze Estuary, China

    Aim Species distribution models (SDMs) are an effective tool to explore the potential distribution of terrestrial, freshwater and marine organisms; however, SDMs have been seldom used to model ichthyoplankton distributions, and thus, our understanding of how larval stages of fishes will respond to climate change is still limited. Here, we developed SDMs to explore potential impacts of climate change on habitat suitability of ichthyoplankton. Location Yangtze Estuary, China. Methods Using long-term ichthyoplankton survey data and a large set of marine predictor variables, we developed ensemble SDMs for five abundant ichthyoplankton species in the Yangtze Estuary (Coilia mystus, Hypoatherina valenciennei, Larimichthys polyactis, Salanx ariakensis and Chelidonichthys spinosus). Then, we projected their habitat suitability under present and future climate conditions. Results The ensemble SDMs had good predictive performance and were successful in estimating the known distributions of the five species. Model projections highlighted two contrasting patterns of response to future climates: while C. mystus will likely expand its range, the ranges of the other four species will likely contract and shift northward. Main conclusions According to our SDM projections, the five ichthyoplankton species that we tested in the Yangtze Estuary are likely to respond differently to future climate changes. These projected different responses seemingly reflect the differential functional attributes and life-history strategies of these species. To the extent that climate change emerges as a critical driver of the future distribution of these species, our findings provide an important roadmap for designing future conservation strategies for ichthyoplankton in this region.Peer reviewe

    Diagnosing air quality changes in the UK during the COVID-19 lockdown using TROPOMI and GEOS-Chem

    The dramatic and sudden reduction in anthropogenic activity due to lockdown measures in the UK in response to the COVID-19 outbreak has resulted in a concerted effort to estimate local and regional changes in air quality, though changes in underlying emissions remain uncertain. Here we combine satellite observations of tropospheric NO_{2} from TROPOspheric Monitoring Instrument and the Goddard Earth Observing System (GEOS)-Chem 3D chemical transport model to estimate that NO_{x} emissions declined nationwide by ~20% during the lockdown (23 March to 31 May 2020). Regionally, these range from 22% to 23% in the western portion of the country to 29% in the southeast and Manchester, and >40% in London. We apply a uniform 20% lockdown period emission reduction to GEOS-Chem anthropogenic emissions over the UK to determine that decline in lockdown emissions led to a national decline in PM_{2.5} of 1.1 μg m^{−3}, ranging from 0.6 μg m^{−3} in Scotland to 2 μg m^{−3} in the southwest. The decline in emissions in cities (>40%) is greater than the national average and causes an increase in ozone of ~2 ppbv in London and Manchester. The change in ozone and PM_{2.5} concentrations due to emission reductions alone is about half the total change from 2019 to 2020. This emphasizes the need to account for emissions and other factors, in particular meteorology, in future air pollution abatement strategies and regulatory action
