    Mexican Trypanosoma cruzi T. cruzi I Strains with Different Degrees of Virulence Induce Diverse Humoral and Cellular Immune Responses in a Murine Experimental Infection Model

    It is has been shown that the majority of T. cruzi strains isolated from Mexico belong to the T. cruzi I (TCI). The immune response produced in response to Mexican T. cruzi I strains has not been well characterized. In this study, two Mexican T. cruzi I strains were used to infect Balb/c mice. The Queretaro (TBAR/MX/0000/Queretaro)(Qro) strain resulted in 100% mortality. In contrast, no mortality was observed in mice infected with the Ninoa (MHOM/MX/1994/Ninoa) strain. Both strains produced extended lymphocyte infiltrates in cardiac tissue. Ninoa infection induced a diverse humoral response with a higher variety of immunoglobulin isotypes than were found in Qro-infected mice. Also, a stronger inflammatory TH1 response, represented by IL-12p40, IFNγ, RANTES, MIG, MIP-1β, and MCP-1 production was observed in Qro-infected mice when compared with Ninoa-infected mice. We propose that an exacerbated TH1 immune response is a likely cause of pathological damage observed in cardiac tissue and the primary cause of death in Qro-infected mice

    Elastic electron scattering from 3-hydroxytetrahydrofuran: experimental and theoretical studies

    We report the results of measurements and calculations for elastic electron scattering from 3-hydroxytetrahydrofuran (C4H8O2). The measurements are performed with a crossed electron-target beam apparatus and the absolute cross-sections are determined using the relative flow technique. The calculations are carried out using the Schwinger multichannel method in the static-exchange plus polarization (SEP) approximation. A set of angular differential cross-sections (DCS) is provided at five incident energies (6.5, 8, 10, 15 and 20 eV) over an angular range of 20–130°, and the energy dependence of the elastic DCS at a scattering angle of 120° is also presented. Integral elastic and elastic momentum transfer cross-sections have also been derived and calculated. The results are compared with those of recent measurements and calculations for the structurally similar molecule tetrahydrofuran (C4H8O)

    Sociocultural values, attitudes and risk factors associated with adolescent cyberbullying in East Asia: a systematic review

    Cyberbullying amongst adolescents is a rapidly growing and alarming global phenomenon that can significantly harm their well-being. Studying cyberbullying in East Asia is especially important, where peer pressure based on collectivistic ideals and rigid cultural scripts for social interactions remain strong. Furthermore, the countries represented in this review are amongst the top globally for internet usage, suggesting that adolescents in East Asia are likely to be excessive users of social media communication and be more exposed to various forms of cyberbullying. This systematic review summarizes findings from peer-reviewed journal articles and book chapters on cyberbullying amongst adolescents between the ages of 10 and 19 in East Asian countries (N = 21). SCOPUS, Google Scholar, and PsycINFO databases were searched for relevant work published between 2008 and 2020. Search strategies involved using keywords related to cyberbullying, adolescents, East Asia, and the name of each country represented in the region (China, South Korea, Japan, Hong Kong and Taiwan). Key factors associated with cyberbullying specific to adolescents in this region are identified and discussed in this review, such as gender socialization patterns and literacy with digital media communication, emphasis on academic achievement and school factors, urban-rural digital divide, relationship with parents and teachers, and collectivistic values. The present review highlights the need to pay further attention to the sociocultural context in future cyberbullying research and calls for more context-specific cyberbullying prevention programs and awareness initiatives

    Private Financing or Not, That is the Question: Lessons from the Light Rail Systems in Spain

    The objective of this paper is to analyze if there is any difference between the light rail systems in Spain according to whether they have been carried out through public financing or private financing (totally or partially). The importance of this study lies in the fact that, for decades, the public–private partnership has been proposed as an alternative to public financing of public transport projects in order to obtain additional financial resources, reduce the public deficit, and increase efficiency. However, there are hardly any detailed studies describing how these initiatives have turned out. Therefore, the present study analyzes if there is any difference in the main variables explaining the performance of light rail projects in Spain depending on their source of funding can be found. For this, the relationship between variables related to design, operation and costs of the projects, and the percentage of private financing were statistically analyzed. As the most relevant conclusion, we underline the fact that the investment per passenger increases when financing is completely private. This would indicate that the most cost-effective lines, from a social standpoint, were financed totally or partially by the public administrations, whereas the least beneficial ones for society were assigned to private enterprises.     Macrocyclic colibactin induces DNA double-strand breaks via copper-mediated oxidative cleavage.

    Colibactin is an assumed human gut bacterial genotoxin, whose biosynthesis is linked to the clb genomic island that has a widespread distribution in pathogenic and commensal human enterobacteria. Colibactin-producing gut microbes promote colon tumour formation and enhance the progression of colorectal cancer via cellular senescence and death induced by DNA double-strand breaks (DSBs); however, the chemical basis that contributes to the pathogenesis at the molecular level has not been fully characterized. Here, we report the discovery of colibactin-645, a macrocyclic colibactin metabolite that recapitulates the previously assumed genotoxicity and cytotoxicity. Colibactin-645 shows strong DNA DSB activity in vitro and in human cell cultures via a unique copper-mediated oxidative mechanism. We also delineate a complete biosynthetic model for colibactin-645, which highlights a unique fate of the aminomalonate-building monomer in forming the C-terminal 5-hydroxy-4-oxazolecarboxylic acid moiety through the activities of both the polyketide synthase ClbO and the amidase ClbL. This work thus provides a molecular basis for colibactin's DNA DSB activity and facilitates further mechanistic study of colibactin-related colorectal cancer incidence and prevention

    The PRIDE database and related tools and resources in 2019: improving support for quantification data

    The PRoteomics IDEntifications (PRIDE) database (https://www.ebi.ac.uk/pride/) is the world's largest data repository of mass spectrometry-based proteomics data, and is one of the founding members of the global ProteomeXchange (PX) consortium. In this manuscript, we summarize the developments in PRIDE resources and related tools since the previous update manuscript was published in Nucleic Acids Research in 2016. In the last 3years, public data sharing through PRIDE (as part of PX) has definitely become the norm in the field. In parallel, data re-use of public proteomics data has increased enormously, with multiple applications. We first describe the new architecture of PRIDE Archive, the archival component of PRIDE. PRIDE Archive and the related data submission framework have been further developed to support the increase in submitted data volumes and additional data types. A new scalable and fault tolerant storage backend, Application Programming Interface and web interface have been implemented, as a part of an ongoing process. Additionally, we emphasize the improved support for quantitative proteomics data through the mzTab format. At last, we outline key statistics on the current data contents and volume of downloads, and how PRIDE data are starting to be disseminated to added-value resources including Ensembl, UniProt and Expression Atlas
