    Understanding Nanopore Window Distortions in the Reversible Molecular Valve Zeolite RHO

    Molecular valves are becoming popular for potential biomedical applications. However, little is known concerning their performance in energy and environmental areas. Zeolite RHO shows unique pore deformations upon changes in hydration, cation siting, cation type, or temperature-pressure conditions. By varying the level of distortion of double eight-rings, it is possible to control the adsorption properties, which confer a molecular valve behavior to this material. We have employed interatomic potentials-based simulations to obtain a detailed atomistic view of the structural distortion mechanisms of zeolite RHO, in contrast with the averaged and space group restricted information provided by diffraction studies. We have modeled four aluminosilicate structures, containing Li+^+, Na+^+, K+^+, Ca2+^{2+}, and Sr2+^{2+} cations. The distortions of the three different zeolite rings are coupled, and the six- and eight-membered rings are largely flexible. A large dependence on the polarizing power of the extra-framework cations and with the loading of water has been found for the minimum aperture of the eight-membered rings that control the nanovalve effect. The calculated energy barriers for moving the cations across the eight-membered rings are very high, which explains the experimentally observed slow kinetics of the phase transition as well as the appearance of metastable phases

    Trifluoroethanol Modulates Amyloid Formation by the All α-Helical URN1 FF Domain

    Amyloid fibril formation is implicated in different human diseases. The transition between native α-helices and nonnative intermolecular β-sheets has been suggested to be a trigger of fibrillation in different conformational diseases. The FF domain of the URN1 splicing factor (URN1-FF) is a small all-α protein that populates a molten globule (MG) at low pH. Despite the fact that this conformation maintains most of the domain native secondary structure, it progressively converts into β-sheet enriched and highly ordered amyloid fibrils. In this study, we investigated if 2,2,2-trifluoroethanol (TFE) induced conformational changes that affect URN1-FF amyloid formation. Despite TFE having been shown to induce or increase the aggregation of both globular and disordered proteins at moderate concentrations, we demonstrate here that in the case of URN1-FF it reinforces its intrinsic α-helical structure, which competes the formation of aggregated assemblies. In addition, we show that TFE induces conformational diversity in URN1-FF fibrils, in such a way that the fibrils formed in the presence and absence of the cosolvent represent different polymorphs.     Proliferative glioblastoma cancer cells exhibit persisting temporal control of metabolism and display differential temporal drug susceptibility in chemotherapy

    Even in immortalized cell lines, circadian clocks regulate physiological processes in a time-dependent manner, driving transcriptional and metabolic rhythms, the latter being able to persist without transcription. Circadian rhythm disruptions in modern life (shiftwork, jetlag, etc.) may lead to higher cancer risk. Here, we investigated whether the human glioblastoma T98G cells maintained quiescent or under proliferation keep a functional clock and whether cells display differential time responses to bortezomib chemotherapy. In arrested cultures, mRNAs for clock (Per1, Rev-erbα) and glycerophospholipid (GPL)-synthesizing enzyme genes, 32 P-GPL labeling, and enzyme activities exhibited circadian rhythmicity; oscillations were also found in the redox state/peroxiredoxin oxidation. In proliferating cells, rhythms of gene expression were lost or their periodicity shortened whereas the redox and GPL metabolisms continued to fluctuate with a similar periodicity as under arrest. Cell viability significantly changed over time after bortezomib treatment; however, this rhythmicity and the redox cycles were altered after Bmal1 knock-down, indicating cross-talk between the transcriptional and the metabolic oscillators. An intrinsic metabolic clock continues to function in proliferating cells, controlling diverse metabolisms and highlighting differential states of tumor suitability for more efficient, time-dependent chemotherapy when the redox state is high and GPL metabolism low.Fil: Wagner, Paula Micaela.     "Dos de todo": El cuento chino de los problemas de comparación multiplicativa en la Educación Infantil

    Narración del desarrollo de dos sesiones de un taller de resolución de problemas con niños y niñas de 5 y 6 años, pertenecientes a dos grupos de Educación Infantil. Partiendo de la lectura del cuento chino “Doble de todo”, los pequeños abordan la resolución de dos problemas de comparación multiplicativa. Los niños, a través del uso de diversas estrategias informales de modelización directa, y empleando distintas representaciones manipulativas y gráficas de cantidades, llegan a las soluciones de los problemas y las comparten con sus compañeros. Esta experiencia forma parte de una investigación dirigida al desarrollo del currículo de la Educación Infantil

    Near-Term Effects of Perennial Grasses on Soil Carbon and Nitrogen in Eastern Nebraska

    Incorporating native perennial grasses adjacent to annual row crop systems managed on marginal lands can increase system resiliency by diversifying food and energy production. This study evaluated (1) soil organic C (SOC) and total N stocks (TN) under warm-season grass (WSG) monocultures and a low diversity mixture compared to an adjacent no-till continuous-corn system, and (2) WSG total above-ground biomass (AGB) in response to two levels of N fertilization from 2012 to 2017 in eastern Nebraska, USA. The WSG treatments consisted of (1) switchgrass (SWG), (2) big bluestem (BGB), and (3) low-diversity grass mixture (LDM; big bluestem, Indiangrass, and sideoat grama). Soils were sampled at fixed depth increments (0–120 cm) in the WSG plots and in the adjacent corn experiment in 2012 and 2017. Soil stocks (Mg ha-1) of SOC and TN were calculated on an equivalent soil mass (ESM) basis and compared within the three WSG treatments as well as between experiments (corn compared to the mean of all WSGs). Soil organic C and TN stocks within soil layers and cumulative stocks responded to the main effect of WSG (PWSG \u3c 0.05) but were no different when comparing the WSGs to corn (Pexpt = NS). Both SOC/TN stocks and cumulative stocks were generally greater in the LDM compared to the BGB. Neither SOC nor TN changed over time under either the WSGs or corn. Warm-season grass AGB responded to a three-way interaction of year, N rate, and WSG (p = 0.0007). Decreases in AGB over time were significant across WSGs and N levels except for SWG at 56 kg N ha-1 and LDM at 112 kg N ha-1. Above-ground biomass was generally greater in the LDM after the first harvest year (2013). Results suggest that incorporating WSGs into marginal cropland can maintain SOC and TN stocks while providing a significant source of biomass to be used in energy production or in integrated livestock systems

    Spawning induction of first-generation (F1) greater amberjack Seriola dumerili in the Canary Islands, Spain using GnRHa delivery systems

    The development of a sustainable aquaculture industry requires the closing of the life-cycle of a potential species in captivity, and the establishment of breeding selection programs using hatchery-produced breeding stocks. The greater amberjack Seriola dumerili is a cosmopolitan pelagic species that has been considered as a good candidate for the species diversification of aquaculture production in the Mediterranean region. However, commercial production is still very limited due to bottlenecks in reproduction, larval rearing and management control during grow out. The aim of the present study was to examine the reproductive development of hatchery-produced greater amberjack and to develop a spawning induction protocol based on the use of gonadotropin releasing hormone agonist (GnRHa) in a controlled-release delivery system. The results showed that hatchery-produced greater amberjack undergo normal gametogenesis and can be induced to undergo maturation, ovulation and spawning after multiple administrations of GnRHa implants, over an extended spawning period lasting from May to September in the Canary Islands, Spain. The use of GnRHa-delivery systems resulted in multiple spawns of fertilized and viable eggs. Egg production was high and egg quality adequate for the implementation of larval rearing for commercial purposes. The handling required to administer the GnRHa implants during the prolonged spawning season did not result in any negative effect on the welfare and reproductive performance of the fish based on evaluation of several biochemical parameters. The developed reproduction control method shows great potential to advance the commercial production of greater amberjack, by enabling the use of hatchery-produced broodstocks for further breeding selection.Postprin
