    This thesis describes the development of sea aquarium controller. The technologies that are combined here are Arduino, Android and electronic circuit. In the core of this system lies microconrtoller Arduino. It is attached to electronic circuit, that has in this version attached temperature sensor, clock that runs correctly even after power outage, Bluetooth interface for communication with control station (e.g. Android device), SD memory card for logging of current state and events. Program on microcontroller Arduino is written with C/C++ programming language. At first controller has default settings built in si it would know how to control sensors and components. These settings can be altered with Android application that runs on some device that has Bluetooth connection to controller. With this application user can look back into how the controller was performing so it can be determined if controller reacted correctly to all sensors states and if everything is efficent enough

    Studying Large Multi-Protein Complexes Using Single Molecule Localization Microscopy

    Biology would not be where it is today without fluorescence microscopy. It is arguably one of the most commonly used tools in the biologists toolbox and it has helped scientists study the localization of cellular proteins and other small things for decades, but it is not without its limitations. Due to the diffraction limit, conventional fluorescence microscopy is limited to micrometer-range structures. Science has long relied upon electron microscopy and X-ray crystallography to study phenomena that occur below this limit. However, many of lifes processes occur between these two spatial domains. Super-resolution microscopy, the next stage of evolution of fluorescence microscopy, has the potential to bridge this gap between micro and nano. It combines superior resolutions of down to a few nanometers with the ability to view objects in their natural environments. It is the ideal tool for studying the large, multi-protein complexes that carry out most of lifes functions, but are too complex and fragile to put on an electron microscope or into a synchrotron. A form of super-resolution microscopy called SMLM Microscopy shows especially high promise in this regard. With its ability to detect individual molecules, it combines the high resolution needed for structural studies with the quantitative readout required for obtaining data on the stoichiometry of multi-protein complexes. This thesis describes new tools which expand the toolbox of SMLM with the specific aim of studying multi-protein complexes. First, the development of a novel fluorescent tagging system that is a mix of genetic tagging and immuno-staining. The system, termed BC2, consists of a short, genetically encodable peptide that is targeted by a nanobody (BC2 nanobody). The system brings several advantages. The small tag is not disruptive to the protein it is attached to and the small nanobody can get into tight spaces, making it an excellent tag for dense multi-protein structures. Next, several new variants of some commonly used green-to-red fluorescent proteins. The novel variants, which can be converted with a combination of blue and infrared light are especially useful for live-cell imaging. The developed fluorescent proteins can also be combined with photo-activatable fluorescent proteins to enable imaging of several targets with the same color protein. Finally, an application of the latter technique to study the multi-protein kinetochore complex and gain first glimpses into its spatial organization and the stoichiometry of its subunits

    Usmerjenost k uporabniku je eno od načel, ki jih zasleduje reforma javne uprave v Sloveniji. V tem okviru se javna uprava trudi povečati zadovoljstvo strank z njenimi storitvami. Meje usmerjenosti k uporabniku določa javni interes. Pri izboljševanju kakovosti upravnih storitev je potrebno na strateški ravni odločanja kombinirati pristop od zgoraj navzdol (uvajanje novih idej in projektov, ki jih domača praksa še ne pozna) s pristopom od spodaj navzgor (vznik iniciativ v posameznih institucijah, razvoj in promocija dobrih praks). Za zadovoljstvo uporabnika so pomembni naslednji elementi upravne storitve: informacija o storitvi, prostorska in časovna dostopnost, enostavnost naročila, postopka in plačila, kakovost osebnega stika, strokovnost in urejenost uslužbencev, urejenost poslovnih prostorov, odzivnost na kritike, pripombe, predloge in pohvale ter zanesljivost. Na zadovoljstvo uporabnika bistveno vpliva tudi spoštovanje »klasičnih« načel delovanja uprave, to je načel zakonitosti, pravne varnosti in predvidljivosti, politične nevtralnosti in odgovornosti javne uprave. V slovenski javni upravi so bili uvedeni številni ukrepi za izboljševanje navedenih elementov. Problem, ki ga je treba preseči v naslednjem obdobju, je razkorak med normativnim in stvarnim, med predpisi in realnostjo. Bistvo nadaljevanja reforme javne uprave je torej v doslednem izvajanju sprejetih zakonov in podzakonskih predpisov na vseh področjih. To pa bo mogoče doseči z učinkovitim nadzorom, usposabljanjem menedžerskih struktur in drugih uslužbencev v javni upravi in s spreminjanjem organizacijske kulture v javni upravi

    Energy and sustainability aspects of wood construction

    Nowadays sustainable and energy-efficient construction is one of the top issues, therefore in\ud my graduation thesis we calculated the energy balance of a small family house and checked\ud the suitability of wooden structural complexes of exterior walls according to selected criteria\ud of sustainability.\ud In this thesis we chose wooden structural complexes of exterior walls, which can be found in\ud the Slovenian market. We also specified two conventional structural complexes of brick and\ud reinforced concrete. The theoretical part defines wood composites, which are found in\ud selected structural complexes and concepts of sustainability, sustainable development,\ud sustainable architecture, sustainable building and sustainable engineering.\ud Energy efficency of selected structural complexes is shown in the case of a single-family lowenergy\ud house type called Prima, which is the product of company Marles. In addition, we\ud used the Dovjak and Krainer method [12] to define sustainability aspects and specific criteria,\ud which were analyzed on the basis of the composition of selected wooden structural\ud complexes. We discussed environmental, social, economic and health criteria of\ud sustainability.\ud We found out that the energy efficency of a building is determined by thermal transmittance\ud of frame construction. For this reason it is not necessary that the wood-frame construction is\ud more energy efficient than the conventional one. In the analysis of the sustainability it is\ud proven that wood-frame construction are more sustainable than conventional, though not\ud to meet demands of sustainability (environmental, economic, social, health). In order to fully\ud meet all the criteria should be avoided imports of wood and wood composites, and use wood\ud without adhesives and resins, which are bad for health and the environment

    Nach dem großen Erfolg des Romans Léon und Louise (2011) veröffentlichte Alex Capus das nur 77 Seiten umfassende Buch mit dem Titel Skidoo und dem Untertitel „Meine Reise durch die Geisterstädte des Wilden Westens“. Das Buch umfasst eine Sammlung kleiner, skurriler Geschichten aus der Vergangenheit dieser Städte. Der ironische Ton, der zwar alle Texte Capusʼ prägt, in diesem aber noch besonders bedeutungstragend ist, wird schon auf dem Buchumschlag angekündigt, auf dem die Bemerkung des Autors zitiert wird: „Wenn ein Mann einen Liebesroman geschrieben hat, muss er hernach zum Ausgleich etwas Ordentliches tun. Einen Western schreiben zum Beispiel. Man ist sich das einfach schuldig, nicht wahr?

    Cet article veut dégager une tendance du théâtre actuel, caractérisée par la polysémie et la dimension fictionnelle, dont la complexité se détache des formes d’écriture dramatique les plus accessibles, sans pour autant paraître hermétique. Les principes dramaturgiques sont ainsi considérés dans leurs liens avec le contexte historique, auquel ils peuvent emprunter certaines fonctions. Des formes variables du théâtre postdramatique ont pu, dans la période qui a précédé la fin de la Guerre froide, contribuer à diversifier et à rendre plurielles les représentations établies du monde dans les sociétés de stagnation, tandis que le néo-réalisme des années 1990 contribuait, lui, à forger une image du monde neuve et consensuelle, qui renvoyait dans un premier temps à des représentations ponctuelles et limitées de la vie. Ce n’est que depuis la relative stabilisation de la société que des rapports historico-politiques et sociaux plus complexes peuvent être traités dans des textes de structure complexe, perçus comme tels dans leurs diverses interprétations. Des œuvres de Roland Schimmelpfennig, Ulrike Syha et Theresia Walser illustreront à différents niveaux cette complexification qui contribue à leur polysémie et à leur poéticité.The article tries to define a tendency in contemporary drama which is marked by ambiguity, fictionality and complexity, but it is neither populist nor hermetic. Drama is considered to be bound to a historical context, in which it has certain functions: before the end of the Cold War different forms of the postdramatic theatre could introduce diversity and plurality into stagnating societies with firmly established world views. The new realism of the 1990s helped to establish a new, consensual world view that in the start referred to single events and limited areas of life. Now, in a relatively stable situation, texts with a complex structure, dealing with complex historical, political and social themes, can be perceived and interpreted as such. Examples from texts by Roland Schimmelpfennig, Ulrike Syha and Theresia Walser show on different levels how complexity produces ambiguity and by this the poetical character of the texts.Der vorliegende Artikel versucht, eine Tendenz in der Gegenwartsdramatik zu bestimmen, die durch Mehrdeutigkeit und Fiktionalität gekennzeichnet wird und sich durch erhöhte Komplexität von der leicht zugänglichen dramatischen Schreibart abhebt, ohne deshalb hermetisch zu wirken. Die Dramatik wird dabei in ihrer Abhängigkeit vom historischen Kontext gesehen, in dem sie bestimmte Funktionen übernehmen kann. Unterschiedliche Formen der Postdramatik konnten in der Zeit vor dem Ende des Kalten Kriegs zur Diversifizierung und Pluralisierung der verfestigten Weltbilder in stagnierenden Gesellschaften beitragen, der neue Realismus in den 1990er Jahren hingegen zur Bildung eines neuen, konsensuellen Weltbilds, das sich zunächst auf einzelne Ereignisse und begrenzte Lebensbereiche bezog. Erst nach der relativen Stabilisierung der Gesellschaft können nun komplexere historisch-politische und soziale Zusammenhänge in komplexer strukturierten Texten thematisiert und in unterschiedlichen Interpretationen als solche wahrgenommen werden. An Texten von Roland Schimmelpfennig, Ulrike Syha und Theresia Walser werden Beispiele solcher Komplexitätssteigerung, die zur Mehrdeutigkeit und so zur Poetizität dieser Texte beiträgt, auf unterschiedlichen Ebenen aufgezeigt

    In the novel Stiller and in the play Don Juan, both published in the early 1950s, Frisch already focuses on the problem of identity as one of his major themes, but it is not yet situated in a political context as, for example, in the play Andorra. In these works, the identity-problem is situated in the domain of intimate relationships and artistic creativity, offering the possibility to discuss Frisch’s concepts of art and love, which unfold through Frisch’s treatment of the traditional literary figure of Don Juan and his hubris. Frisch’s Stiller and Don Juan do not believe in God; but their frame of mind stays the same, while the position of God and his attributes are assigned to love, art and geometry. Although both literary figures in the course of events realize that this substitution is a mistake, they are unable to alter their concepts of love and art. Instead of altering the concepts, they rather resign in silence, while Max Frisch continues to search for alternatives in reflecting on the process of fictionalization and in developing innovative narrative and dramatic structures

    Application of innovation management in construction

    Challenged by increasing competition and unprecedented pace of change, companies can no longer concentrate only on today's needs of the customers, but on the anticipation of those of tomorrow. This can only be achieved by innovations that must be managed properly by innovation management. Innovations are accompanied with high level of uncertainty, which is the main reason for their careful incorporation into the companies’ daily operation. On the other hand, today, there are many successful methods of innovation implementation, which help to decrease the possibility of the failure. These methods can be transferred into the new environment only after appropriate modifications. Implementation of innovation is accompanied by high costs and uncertainty. One has to be aware that innovations can only succeed when the environment is supportive. Therefore, the thesis shows how to build an innovative organization, implementation process of innovation, and alliances as a powerful tool for decreasing uncertainty and acquiring the proper sources. The last part of my thesis is dedicated to innovation management in construction with emphasis on non-technological innovation, such as partnering and supply chain management. Specific features of construction sector, such as project based work, high number of small and medium enterprises and uniqueness of construction projects, are discussed along with the influence of these features on innovation management in construction
