    Long-term effects of caloric restriction or exercise on DNA and RNA oxidation levels in white blood cells and urine in humans

    Excessive adiposity is associated with increased oxidative stress and accelerated aging. Weight loss induced by negative energy balance reduces markers of oxidation in experimental animals and humans. The long-term effects of weight loss induced by calorie restriction or increased energy expenditure induced by exercise on measures of oxidative stress and damage have not been studied in humans. The objective of the present study was to compare the effects of 20% caloric restriction or 20% exercise alone over 1 year on oxidative damage to DNA and RNA, as assessed through white blood cell and urine analyses. Eighteen men and women aged 50 to 60 years with a body mass index (BMI) between 23.5 to 29.9 kg/m(2) were assigned to one of two conditions — 20% CR (n = 9) or 20% EX (n = 9) — which was designed to produce an identical energy deficit through increased energy expenditure. Compared to baseline, both interventions significantly reduced oxidative damage to both DNA (48.5% and 49.6% reduction for the CR and EX groups, respectively) and RNA (35.7% and 52.1% reduction for the CR and EX groups, respectively) measured in white blood cells. However, urinary levels of DNA and RNA oxidation products did not differ from baseline values following either 12-month intervention program. Data from the present study provide evidence that negative energy balances induced through either CR or EX result in substantial and similar improvements in markers of DNA and RNA damage to white blood cells, potentially by reducing systemic oxidative stress

    Characteristics and monetary compensation of caregivers for patients with rheumatic conditions

    Musculoskeletal disorders (MD) are highly prevalent conditions that affect quality of life. MD cause physical and psychological dependence. Usually, the care of a patient with MD is assumed by a caregiver. The aim of this study was to describe the sociodemographic characteristics and the monetary remuneration associated to the care of a patient with MD. A cross sectional study was carried out in order to estimate the monetary remuneration related to the health care of patients with MD. A survey was applied to caregivers of patients with MD. Demographic data was collected. We asked about the relationship with the patient, the time as a caregiver and all data related to the monetary compensation. Descriptive epidemiology was done. We reported monetary data in American Dollars (USD) using the average exchange rate for 2018. We surveyed 132 caregivers. Mean age was 52 years [standard deviation 19], 72% were women, 78% were taking care of a patient with rheumatoid arthritis, 12% osteoarthrosis 2% lupus, and 2% osteoporosis. The remaining 6% were caregivers of patients with ankylosing spondylitis, fibromyalgia and Sjogren syndrome. Regarding the time as a caregiver, 48% had less than a year, 16% between two and three years, 18% more than three years, 13% more than four years, and 5% were temporarily caregivers. In our study, 85% of caregivers were a family member, while 15% a nurse or a non-related person. Regarding the compensation, 97% did not receive any salary or payment for being caregiver, the remaining 3% received between 265 and 530 USD per month. Our study demonstrated that the care for patients with MD is mainly assumed by family members. Our results agree with other studies in chronic conditions where only a small proportion of caregivers is paid. Caregivers should be considered for the health system

    Long-term moderate calorie restriction inhibits inflammation without impairing cell-mediated immunity: A randomized controlled trial in non-obese humans

    Calorie restriction (CR) inhibits inflammation and slows aging in many animal species, but in rodents housed in pathogen-free facilities, CR impairs immunity against certain pathogens. However, little is known about the effects of long-term moderate CR on immune function in humans. In this multi-center, randomized clinical trial to determine CR's effect on inflammation and cell-mediated immunity, 218 healthy non-obese adults (20-50 y), were assigned 25% CR (n=143) or an ad-libitum (AL) diet (n=75), and outcomes tested at baseline, 12, and 24 months of CR. CR induced a 10.4% weight loss over the 2-y period. Relative to AL group, CR reduced circulating inflammatory markers, including total WBC and lymphocyte counts, ICAM-1 and leptin. Serum CRP and TNF-α concentrations were about 40% and 50% lower in CR group, respectively. CR had no effect on the delayed-type hypersensitivity skin response or antibody response to vaccines, nor did it cause difference in clinically significant infections. In conclusion, long-term moderate CR without malnutrition induces a significant and persistent inhibition of inflammation without impairing key in vivo indicators of cell-mediated immunity. Given the established role of these pro-inflammatory molecules in the pathogenesis of multiple chronic diseases, these CR-induced adaptations suggest a shift toward a healthy phenotype

    Calorie restriction in humans inhibits the PI3K/AKT pathway and induces a younger transcription profile

    Caloric restriction (CR) and down-regulation of the insulin/IGF pathway are the most robust interventions known to increase longevity in lower organisms. However, little is known about the molecular adaptations induced by CR in humans. Here, we report that long-term CR in humans inhibits the IGF-1/insulin pathway in skeletal muscle, a key metabolic tissue. We also demonstrate that CR induces dramatic changes of the skeletal muscle transcriptional profile that resemble those of younger individuals. Finally, in both rats and humans, CR evoked similar responses in the transcriptional profiles of skeletal muscle. This common signature consisted of three key pathways typically associated with longevity: IGF-1/insulin signaling, mitochondrial biogenesis, and inflammation.     Nitrogen fixation and transfer in open ocean diatom–cyanobacterial symbioses

    Many diatoms that inhabit low-nutrient waters of the open ocean live in close association with cyanobacteria. Some of these associations are believed to be mutualistic, where N2-fixing cyanobacterial symbionts provide N for the diatoms. Rates of N2 fixation by symbiotic cyanobacteria and the N transfer to their diatom partners were measured using a high-resolution nanometer scale secondary ion mass spectrometry approach in natural populations. Cell-specific rates of N2 fixation (1.15–71.5 fmol N per cell h−1) were similar amongst the symbioses and rapid transfer (within 30 min) of fixed N was also measured. Similar growth rates for the diatoms and their symbionts were determined and the symbiotic growth rates were higher than those estimated for free-living cells. The N2 fixation rates estimated for Richelia and Calothrix symbionts were 171–420 times higher when the cells were symbiotic compared with the rates estimated for the cells living freely. When combined, the latter two results suggest that the diatom partners influence the growth and metabolism of their cyanobacterial symbionts. We estimated that Richelia fix 81–744% more N than needed for their own growth and up to 97.3% of the fixed N is transferred to the diatom partners. This study provides new information on the mechanisms controlling N input into the open ocean by symbiotic microorganisms, which are widespread and important for oceanic primary production. Further, this is the first demonstration of N transfer from an N2 fixer to a unicellular partner. These symbioses are important models for molecular regulation and nutrient exchange in symbiotic systems

    Association of Motivational Climate With Addictive Behaviors Depending on Type of Sport in University Students: Structural Equation Analysis

    This research study aims at contrasting a structural model of the associations between the alcohol consumption, tobacco dependence, and the problematic use of video games with motivational climate toward sport depending on the category of sports practiced in a sample of Physical Education university students. The sample consisted of 775 university students from the Autonomous Community of Andalusia (Spain), aged between 21 and 35 (22.22 ± 3.76) years. The instruments used were the Perceived Motivational Climate in Sport Questionnaire (PMCSQ-2), the Alcohol Use Disorders Identification Test (AUDIT), the Fagerström Test for Nicotine Dependence (FTND), and the Questionnaire of Experiences Related to Video Games (QERV) questionnaires. A path model that fitted properly in the multigroup analysis for both categories of sports was used, χ2 = 19.843; gl = 8; p = .011; comparative fit index (CFI) = 0.911; normed fit index (NFI) = 0.903; incremental fit index (IFI) = 0.912; root mean square error approximation (RMSEA) = 0.085. An inverse association was shown between task climate and tobacco consumption in individual sports, being weaker in collective sports. This association is not significant for collective sports, but it is for individual sports for ego climate. However, a positive association was found between ego climate and the use of video games in individual sports, not being significant in the case of collective sports.     Análisis del impacto en el uso del TIC en la modalidad de clases en línea en el instituto superior tecnológico Tsáchila

    Information and communication technologies in recent years have taken on great importance in the educational field since it has a wide range of applications and tools that can be used to make education dynamic. This research aims to carry out an analysis of the impact on the use of ICT in the modality of online classes at the Tsáchila Higher Technological Institute in times of the COVID-19 pandemic in Ecuador, due to the pandemic the institution had to carry out a hasty change from the face-to-face modality to an online modality to avoid contagion in the educational community, but this implied new challenges for both students and teachers who did not have the knowledge or technological resources necessary to carry out this transition. An exploratory and correlational study is presented below, where 150 teachers and 1500 students from the different careers of the institution were included, to whom an online survey was applied where different types of questions were integrated in a questionnaire in order to collect information. necessary and subsequently carry out the analysis of the investigation. As a result, it was obtained that ICTs are of great contribution to the teaching-learning process in higher education, but to make a correct use of ICT, digital skills must be acquired.Las tecnologías de la información y comunicación a lo largo de los últimos años han tomado gran importancia en el ámbito educativo puesto que cuenta con una gran gama de aplicaciones y herramientas que se pueden usar para hacer dinámica la educación. La presente investigación pretende realizar un análisis del impacto en el uso de las TIC en la modalidad de clases en línea en el Instituto Superior Tecnológico Tsáchila en tiempo de pandemia de la COVID-19 en Ecuador, por causa de la pandemia la institución  tuvo que realizar un cambio apresurado de la modalidad presencial por una modalidad en línea para evitar el contagio en la comunidad educativa, pero esto implico nuevos retos tanto para estudiantes como docentes que no contaban con los conocimientos o recursos tecnológicos necesarios para realizar esta transición. Se presenta a continuación un estudio exploratorio y correlacional, donde se incluyeron 150 docentes y 1500estudiantes de las diferentes carreras de la institución a los cuales se les aplicó una encuesta en línea donde se integraron diferentes tipos de preguntas en un cuestionario para de esa forma recolectar información necesaria y posteriormente realizar el análisis de la investigación. Como resultado se obtuvo que las TIC son de gran aporte para el proceso de enseñanza-aprendizaje en la educación superior, pero para hacer un correcto uso de las TIC se debe adquirir competencias digitales

    Integrated genetic map and genetic analysis of a region associated with root traits on the short arm of rye chromosome 1 in bread wheat

    A rye–wheat centric chromosome translocation 1RS.1BL has been widely used in wheat breeding programs around the world. Increased yield of translocation lines was probably a consequence of increased root biomass. In an effort to map loci-controlling root characteristics, homoeologous recombinants of 1RS with 1BS were used to generate a consensus genetic map comprised of 20 phenotypic and molecular markers, with an average spacing of 2.5 cM. Physically, all recombination events were located in the distal 40% of the arms. A total of 68 recombinants was used and recombination breakpoints were aligned and ordered over map intervals with all the markers, integrated together in a genetic map. This approach enabled dissection of genetic components of quantitative traits, such as root traits, present on 1S. To validate our hypothesis, phenotyping of 45-day-old wheat roots was performed in five lines including three recombinants representative of the entire short arm along with bread wheat parents ‘Pavon 76’ and Pavon 1RS.1BL. Individual root characteristics were ranked and the genotypic rank sums were subjected to Quade analysis to compare the overall rooting ability of the genotypes. It appears that the terminal 15% of the rye 1RS arm carries gene(s) for greater rooting ability in wheat
