    El examen serológico con muestras de sangre obtenidas en papel de filtro

    Se ha puesto a punto una técnica de obtención de sangre total en papel filtro para el muestreo serológico de enfermedades de los conejos tales como: Enfermedad Hemorrágica Viral (RHVD); Encephalitozoonosis; Chlamydia psittaci y Mixomatosis. Se propone como alternativa de muestreo para la determinación de anticuerpos, por ser un método sencillo que no requiere muchos cuidados en el envío al laboratorio. Se evaluaron 94 muestras de suero de conejos llegados al laboratorio para el diagnóstico de las entidades antes citadas. Los resultados serológicos de la muestras de sangre total obtenida por venipuntura y en papel filtro, fueron comparados. Los métodos empleados incluyeron: Inmunofluorescencia Indirecta (IFI) para detectar IgG, Carbón inmunoensayo (CIA) e Inhibición de la Hemoaglutinación (IHA) para la evaluación de anticuerpos totales. Los resultados de sensibilidad, especificidad, índice de concordancia y valores predictivos positivos y negativos obtenidos en este trabajo fueron satisfactorios y nos permitieron decir que la toma y el transporte de muestras de sangre en papel de filtro es una técnica útil con sensibilidad y especificidad adecuada para realizar estudios seroepidemiológicos en conejos

    The Internet as a Reliable Source of Sexual and Reproductive Health Information among Rural School-Aged Children in Oaxaca, Mexico

    Background: Correct and culturally relevant sexual and reproductive health knowledge among children and adolescents is a key component to a healthy life. In Mexico, a country plagued with a teen pregnancy epidemic, sexuality education in the public-school system begins in 4th grade. Our study aims were to characterize the sexual and reproductive health knowledge of middle school students from Oaxaca, and its association with belonging to an indigenous group, gender, sources of sexual and reproductive health information, and parents2019; level of schooling. Methods: Cross-sectional study. Students responded to a multiple-choice paper and pencil selfadministered survey on sexual and reproductive health knowledge.     The IUPAC–IUGS joint Task Group “Isotopes in Geosciences” (TGIG) has evaluated the published literature on the half-lives of six long-lived, geologically relevant radioactive nuclides. Where conflicting literature estimates are present, it is necessary to first identify any systematic bias in accordance with metrological traceability and to exclude the biased estimates from further consideration. The TGIG recommends three robust half-life estimates: 49.61±0.16 Ga for 87Rb, corresponding to a decay constant λ(87Rb) = (1.3972±0.0045)×10–11 a–1; 106.25±0.38 Ga for 147Sm, and a corresponding decay constant λ(147Sm) = (6.524±0.024)×10–12 a–1; 4.4683±0.0096 Ga for 238U, i.e. a decay constant λ(238U) = (1.55125±0.00333)×10–10 a–1. All cited uncertainties have a coverage factor k = 2. For other radionuclides of Sm and U no unambiguous consensus value can be endorsed at present by TGIG, which limits its evaluation to a status report highlighting unaccounted-for potential sources of bias. The improved repeatability of mass spectrometric measurements has revealed systematic bias effects that had been dismissed as subordinate in the past. These issues can only be resolved by future dedicated investigations

    Protective effects of epoxypukalide on pancreatic b-cells and glucose metabolism in STZ-induced diabetic mice

    Producción CientíficaDiabetes is a consequence of a decrease on functional β-cell mass. We have recently demonstrated that epoxypukalide (Epoxy) is a natural compound with beneficial effects on primary cultures of rat islets. In this study, we extend our previous investigations to test the hypothesis that Epoxy protects β-cells and improves glucose metabolism in STZ-induced diabetic mice. We used 3-months old male mice that were treated with Epoxy at 200 μg/kg body weight. Glucose intolerance was induced by multiple intraperitoneal low-doses of streptozotocin (STZ) on 5 consecutive days. Glucose homeostasis was evaluated measuring plasma insulin levels and glucose tolerance. Histomorphometry was used to quantify the number of pancreatic β-cells per islet. β-cell proliferation was assessed by BrdU incorporation, and apoptosis by TUNEL staining. Epoxy treatment significantly improved glucose tolerance and plasma insulin levels. These metabolic changes were associated with increased β-cell numbers, as a result of a two-fold increase in β-cell proliferation and a 50% decrease in β-cell death.     Sexual steroid hormone receptors profiles of ovarian carcinoma in Mexican women

    The significance of the presence of androgen receptor (AR), estrogen receptor alpha (ER) and progesterone receptor (PR) in ovarian cancer patient survival has been a matter of numerous studies. This study was aimed to describe the expression profile of the three sexual steroid receptors in high-grade serous, endometrioid, mucinous and low-grade serous ovarian carcinoma and its association to the proliferation index in patients with primary ovarian carcinoma diagnosis, before any treatment. Eighty-one samples were obtained from the National Institute of Cancerology in Mexico City and were evaluated for the presence of AR, ER, PR and Ki67 by immunohistochemistry. The four subtypes of ovarian carcinoma displays a specific profile of the eight possible combinations of the steroid receptors with significant differences within the profile and the histological subtypes. High-grade serous carcinoma was characterized by a high frequency of both, triple-negative and AR+ ER− PR+ profiles. Endometrioid carcinoma presented a higher frequency of triple-positive profile. The presence of only AR+ profile was not observed in the endometrioid tumors. The relationship of the receptor profile with the proliferation index in the tumor epithelium shows that the expression of only ER is associated to a reduced proliferation index in endometrioid carcinoma. Steroid hormone receptor expression and co-expression could help characterize ovarian carcinoma

    The structural assembly switch of cell division protein FtsZ probed with fluorescent allosteric inhibitors

    FtsZ is a widely conserved tubulin-like GTPase that directs bacterial cell division and a new target for antibiotic discovery. This protein assembly machine cooperatively polymerizes forming single-stranded filaments, by means of self-switching between inactive and actively associating monomer conformations. The structural switch mechanism was proposed to involve a movement of the C-terminal and N-terminal FtsZ domains, opening a cleft between them, allosterically coupled to the formation of a tight association interface between consecutive subunits along the filament. The effective antibacterial benzamide PC190723 binds into the open interdomain cleft and stabilizes FtsZ filaments, thus impairing correct formation of the FtsZ ring for cell division. We have designed fluorescent analogs of PC190723 to probe the FtsZ structural assembly switch. Among them, nitrobenzoxadiazole probes specifically bind to assembled FtsZ rather than to monomers. Probes with several spacer lengths between the fluorophore and benzamide moieties suggest a binding site extension along the interdomain cleft. These probes label FtsZ rings of live Bacillus subtilis and Staphylococcus aureus, without apparently modifying normal cell morphology and growth, but at high concentrations they induce impaired bacterial division phenotypes typical of benzamide antibacterials. During the FtsZ assembly-disassembly process, the fluorescence anisotropy of the probes changes upon binding and dissociating from FtsZ, thus reporting open and closed FtsZ interdomain clefts. Our results demonstrate the structural mechanism of the FtsZ assembly switch, and suggest that the probes bind into the open clefts in cellular FtsZ polymers preferably to unassembled FtsZ in the bacterial cytosol

    Validation and psychometric analysis of the Internet Addiction Test in Spanish among college students

    BACKGROUND: The wide use of the Internet in the workplace, academic or social field, can have an impact on daily life. One of the most used questionnaires worldwide to analyse these problems is the Internet Addiction Test (IAT). Our aim was to validate a Spanish version of the IAT and analyse its psychometric properties. METHODS: Population of study were college students participating in the uniHcos project (Universities of Granada, Huelva, Jaén, León, Salamanca, and Vigo). The questionnaire was translated and back-translated by two native English speakers. Reliability of scores was analysed using Pearson's correlation coefficient and agreement was analysed using the Bland-Altman and Kappa techniques. Test dimensions were analysed by exploratory and confirmatory factor analysis. RESULTS: The reliability of scores was good (r = 0.899, Kappa = 0.650 and mean difference using Bland-Altman = -3.5). The psychometric assessment identified two factors (Emotional Investment; Performance and Time Management) which explained 55 % of the variance (total internal consistency of 0.91) and only 19 items. The confirmatory analysis showed an acceptable goodness of fit, especially when items 6 and 8 were related (RMSEA = 0.07 90%IC = 0.06 - 0.08; WRMR = 1.01, CFI = 0.96; TLI = 0.95). The two dimensions were negatively correlated with age and positively correlated with time spent online, especially for the purposes of leisure and entertainment. DISCUSSION: The results show good reliability and psychometric properties of the Spanish version of IAT with a two-dimensional solution. This result is partially in concordance with previous validations of the IAT in other languages that have found uni- and multi-dimensional solutions using different methodologies. Moreover, we want to highlight the possibility that some item of this questionnaire is outdated due to the technological and lifestyles changes and should be not taken into account. CONCLUSION: The reliability and psychometric properties obtained in this study support the conclusion that this Spanish short version of the IAT represents a useful tool for the analysis of problems arising from misuse of the Internet
