    Contributos para a adaptação da entrevista clínica geracional à população portuguesa

    O presente estudo teve como objetivo contribuir para a adaptação e validação da Intervista Clinica Generazionale (ICG) de Cigoli e Tamanza (2009), anteriormente submetida a um processo de adaptação transcultural para a língua portuguesa por Raguso, Facchin, Molgora e Gonçalves (2010) e intitulada como Entrevista Clínica Geracional (ECG). A ECG é um instrumento clínico para a avaliação e investigação das relações familiares centrado em particular no conceito de Generatividade das famílias. A ECG é composta por diferentes dimensões a saber pelo Eixo I - As Origens, pelo Eixo II - O Casal e o pelo Eixo III - Passagem Generativa. Foi no presente estudo aplicada a 10 casais portugueses visando o teste das propriedades de medida, especificamente a validade de constructo, através do uso do teste da validade convergente (Freire & Almeida, 2001). Para estudar a validade convergente utilizou-se a análise de comparação usando o teste de Fisher para testar a associação entre os Eixos da ECG (Eixo I - As Origens, Eixo II - O Casal e o Eixo III - Passagem Generativa) com outros instrumentos: o Family Environment Scale (FES) e o Family Adaptability and Cohesion Evaluation Scales (FACES IV); a Escala de Avaliação da Satisfação em Áreas da Vida Conjugal (EASAVIC); e, por último, a Loyola Generativity Scale (LGS), respetivamente; que avaliam constructos teoricamente relacionados (e.g., coesão familiar, expressividade, comunicação, etc) e calculou-se, subsequentemente, as suas correlações (coeficiente de correlação não-paramétrica de Spearman). As análises de correlação entre Eixos I (Q1, Q6, Q7), II (Q5,6,8.1/8.2), III (Q3.1/3.2) da ECG com os scores das escalas/subescalas da FES (‘Ambiente familiar’, ‘Expressividade’) e FACES IV (‘Coesão equilibrada’, ‘Flexibilidade’), EASAVIC (‘Próprio’, ‘outro’, total’) e LGS mostraram, respetivamente, no mesmo sentido da análise de Fisher, que existem coeficientes de correlação positiva estatisticamente significativos, indicando validade convergente para as respetivas dimensões. De salientar que a validade convergente entre Eixo I (Q1) e a subescalas da FES (‘Ambiente familiar’) apenas foram verificadas para os cônjuges do género feminino, assim como entre o Eixos I (Q7) e a subescalas da FES (‘Expressividade’), assim como, entre Eixo I (Q6,7) e a subescalas da FACES IV (‘Coesão equilibrada’, ‘Flexibilidade’) apenas ocorreram para os cônjuges do género masculino. Em suma, os presentes resultados mostram que a ECG, na sua versão portuguesa, possui boa qualidade psicométrica e validade, e aponta para a sua utilidade na avaliação da dinâmica familiar para as respetivas dimensões onde foram encontradas validade convergente.This research project aims to contribute to the adaptation and validation of the Interview Clinic Generazionale (ICG) of Cigoli and Tamanza (2009), previously subjected to a process of cultural adaptation to Portuguese by Raguso, Facchin, Molgora and Gonçalves (2010), entitled ‘The Clinical Generational Interview’ (CGI). The CGI is a clinical tool for the evaluation and research of family relationships focused in particular on the concept of Family Generativity. The CGI is composed by different dimensions namely the Axis I - The family of origin, the Axis II - The couple and the Axis III - The generational passage. In this study the CGI was applied to 10 Portuguese couples in order to analyze their measurement properties , in particular the construct validity through the use of convergent validity procedures (Freire & Almeida,2001). To study the convergent validity was used the comparison analysis using the Fisher's exact test and the coefficient of non-parametric Spearman correlation to explore the association/correlation between the axes of the CGI (Axis I - The family of origin, Axis II - The couple and Axis III - The generational passage) with other instruments, such as: the Family Environment Scale (FES) and the Family Adaptability and Cohesion Evaluation Scales (FACES IV); the Scale of Assessment of Satisfaction in Areas of Conjugal Life (EASAVIC) and, lastly, the Generativity Loyola Scale (LGS), respectively; since these instruments assess theoretical constructs related to those covered by the CGI (e.g., family cohesion, expressiveness, communication, etc.). The correlation analysis between Axis I (Q1, Q6, Q7), II (Q5,6,8.1 / 8.2), III (Q3.1 / 3.2) of the CGI with the scores of the subscales of FES ( 'Family environment’ and 'Expressive') and of FACES IV ( 'Balanced Cohesion' and 'Flexibility'), EASAVIC ( 'Self', 'Other' and total scores ') and LGS showed, respectively, in the same sense of Fisher's analysis, statistically significant positive relationships, indicating convergent validity for the respective dimensions. To point out that the convergent validity between Axis I (Q1) and the subscales of the FES ('Family environment’') were only founded for the female gender spouses group, as well as the association between the Axis I (Q7) and the subscales of the FES ( 'expressiveness ') and the Axis I (Q6,7) and subscales faces IR (' Balanced cohesion, '' Flexibility ') only was observed for the males spouses group. In short, the present results show that the CGI in its Portuguese version, has good psychometric quality and validity, and points to its usefulness in the assessment of family dynamics to the respective dimensions which were found convergent validity

São encarados como um serviço promissor que possibilita a variedade de programações e, por isso, uma captação e fidelização de públicos mais vasta. É com a percepção do fenómeno dos serviços educativos e da importância que lhes deve ser prestada no sentido de se projectar e influenciar as práticas culturais que surge o objecto de estudo deste relatório. O capítulo I é dedicado à teorização do conceito de serviço educativo, no que diz respeito aos seus objectivos, missões e conceitos que o sustentam. O capítulo II é apresentado como um relatório de estágio, desenvolvido no CCB Fábrica das Artes com a duração de seis meses, de Abril a Outubro de 2010, sendo descritas determinadas actividades programadas, as funções e as aquisições.ABSTRACT - Educational services in Portugal are beginning to gain notoriety within the action of cultural entities. They are seen as a promising service that enables the variety of programming and therefore, a wider capture and retention of public. It is with the perception of the phenomenon of educational services and the importance that they should be given, in order to project and induce cultural practices, that arises the object of study in this report. Chapter I is devoted to theorizing the concept of educational services, concerning its objectives, tasks and concepts that support it.     The defensive functions of plant inhibitors are not restricted to insect enzyme inhibition

    Three plant proteinase inhibitors BbKI (kallikrein inhibitor) and BbCI (cruzipain inhibitor) from Bauhinia bouhinioides, and a BrTI (trypsin inhibitor) from B. rufa, were examined for other effects in Callosobruchus maculatus development; of these only BrTI affected bruchid emergence. BrTI and BbKI share 81% identities in their primary sequences and the major differences between them are the regions comprising the RGD and RGE motifs in BrTI. These sequences were shown to be essential for BrTI insecticidal activity, since a modified BbKI [that is a recombinant form (BbKIm) with some amino acid residues replaced by those found in BrTI sequence] also strongly inhibited insect development. By using synthetic peptides related to the BrTI sequence, YLEAPVARGDGGLA-NH(2) (RGE) and IVYYPDRGETGL-NH(2) (RGE), it was found that the peptide with an RGE sequence was able to block normal development of C. maculatus larvae (ED(50) 0.16% and LD(50) 0.09%), this being even more effective than the native protein.     Exuberant Hand-Foot-Mouth Disease: An Immunocompetent Adult with Atypical Findings

    Non-polio enteroviruses are ubiquitous viruses responsible for a wide spectrum of disease in people of all ages, although infection and illness disproportionately affect infants and young children. Hand-foot-mouth disease (HFMD) is an enteroviral clinical syndrome most frequently caused by coxsackievirus-A16 and enterovirus-A71. Since 2008, a novel coxsackievirus-A6 genotype has been associated with more severe HFMD in both children and adults, presenting with a unique constellation of findings, and whose prevalence has been increasing over the last few years. In this case report, an atypical clinical picture of confirmed enterovirus HFMD is described in an immunocompetent adult, with exuberant clinical findings, clinically consistent with coxsackievirus-A6 infection.     The aim of this study was to evaluate the effect of essential oils (EOs) of plants naturally occurring in northern Portugal on the spoilage of fresh Maronesa beef burgers stored at 2 and 8 C under different packaging conditions. EOs were obtained from dried leaves of laurel(Laurus Nobilis L.) and rosemary (Rosmarinus officinallis L.) by hydro-distillation using a Clevenger-type apparatus. Analysis of volatile composition of essential oils of rosemary and laurel was achieved by Gas Chromatography–Mass Spectrometry (GC–MS) and Gas Chromatography-Thermal Conductivity Detection (GC-TCD) resulting in the detection of 95.8% and 89.4% of its compounds, respectively. Fresh beef (semitendinosus and semimembranosus) of DOP-Maronesa breed (males; n = 4) were obtained from local market and transported to the laboratory. Samples were stored at 2 and 8 C in two different conditions: aerobiosis (A) and vacuum (V) and analyzed at 0, 1, 2, 3, 5, 7, 10, 14, 21 and 28 days for Lactic acid bacteria (LAB), Enterobacteriaceae,Pseudomonas spp., Fungi, Total mesophilic (TM) and psychrotrophic (TP), color (L*a*b*) and pH. Laurel was the most effective EO keeping pH from increasing. Coordinates L* and a* were higher on samples containing laurel EO for both A and V packaging. Laurel also showed better effect in reducing microbiologic counts in samples packed in A at both 2 and 8 C and packed in V at 8 C. Rosemary was effective in reducing microbial counts on all V samples stored at 2 C. This study allows to conclude that Laurel EO has significant effect in shelf-life, maintaining fresh beef colo

    Purification and characterization of a new trypsin inhibitor from Dimorphandra mollis seeds

    A second trypsin inhibitor (DMTI-II) was purified from the seed of Dimorphandra mollis (Leguminosae-Mimosoideae) by ammonium sulfate precipitation (30-60%), gel filtration, and ion-exchange and affinity chromatography.     Tumor cell invasion is vital for cancer progression and metastasis. Adhesion, migration, and degradation of the extracellular matrix are important events involved in the establishment of cancer cells at a new site, and therefore molecular targets are sought to inhibit such processes. the effect of a plant proteinase inhibitor, Enterolobium contortisiliquum trypsin inhibitor (EcTI), on the adhesion, migration, and invasion of gastric cancer cells was the focus of this study. EcTI showed no effect on the proliferation of gastric cancer cells or fibroblasts but inhibited the adhesion, migration, and cell invasion of gastric cancer cells; however, EcTI had no effect upon the adhesion of fibroblasts. EcTI was shown to decrease the expression and disrupt the cellular organization of molecules involved in the formation and maturation of invadopodia, such as integrin beta 1, cortactin, neuronal Wiskott-Aldrich syndrome protein, membrane type 1 metalloprotease, and metalloproteinase-2. Moreover, gastric cancer cells treated with EcTI presented a significant decrease in intracellular phosphorylated Src and focal adhesion kinase, integrin-dependent cell signaling components.
