    The influence of working memory load on response inhibition in children with attention-deficit/hyperactivity disorder or reading disorder

    The aim of the present study was to examine the relationship between response inhibition and working memory in 8-12-year-old children with attention-deficit/hyperactivity disorder (ADHD; n = 19), reading disorder (RD; n = 17), ADHD + RD (n = 21), and control children (n = 19). For the first time a within-task methodology was used to study the combined effect of both executive functions on a common measure of task performance in two often comorbid childhood disorders, ADHD and RD. We found evidence of an interaction between both domains, suggesting that they rely on a common pool of resources. In addition, we found that children with ADHD or RD were not more seriously affected by the combined load of both executive functions than children without ADHD or RD

    The 10-word learning task in the differential diagnosis of early Alzheimer's disease and elderly depression: a cross-sectional pilot study

    Objectives: Identification of early Alzheimer's disease (AD) has become very important. Episodic memory tasks appear to have predictive power for indicating early AD. Deficits in encoding and storage processes that are characteristic of AD, however, must be distinguished from non-AD deficits that can also affect memory, including difficulties that may be present in depression. This pilot study was set up to ascertain whether a 10-word-list-learning task (delayed recognition and rate of forgetting) may be useful in making the differentiation between mild AD and depression. Method: A Dutch version of Rey's auditory verbal learning test was administered to 36 mild AD patients, 41 depressed patients, and 47 healthy controls. Data were analyzed in a cross-sectional manner. Results: Receiver operating characteristic analyses showed that for differentiating mild AD and depression, both delayed recognition and percentage of forgetting have sufficient diagnostic accuracy. Conclusion: Percentage of forgetting had the highest diagnostic accuracy for differentiating mild AD and depressed patients and may be useful in the early detection of AD

    ADHD subtypes: do they differ in their executive functioning profile?

    The present study was designed to investigate the hypothesis that children with Attention Deficit Hyperactivity Disorder combined subtype (ADHD-C) have a generalized executive functioning (EF) [Barkley, R. A. (1997). Behavioural inhibition, sustained attention, and executive functions: Constructing a unifying theory of AD/HD. Psychological Bulletin, 121, 65-94; Barkley, R. A. (1997). ADHD and the nature of self-control. New York: The Guilford Press]. We tested whether ADHD-C and ADHD inattentive subtype (ADHD-I) can be differentiated from each other on EF measures. We compared 16 normally developing boys with 16 boys with ADHD-C and 16 with ADHD-I on five EF domains. The boys were all matched on age, IQ, and the presence of oppositional defiant disorder (ODD)/conduct disorder (CD). Despite carefully diagnosed groups and methodological controls, the results do not support the EF-hypothesis of ADHD-C. Children with ADHD-C differed from normal controls (NC) on tasks related to inhibition; they did not exhibit EF deficits on all EF tasks. Children with ADHD-C also exhibited deficits on non-EF tasks. Furthermore, the ADHD-C and ADHD-I subtypes did not differ from one another. Neuropsychological findings on the domains under study did not yield evidence for the distinctiveness of ADHD-C and ADHD-I subtypes. Ā© 2004 National Academy of Neuropsychology.     A comprehensive investigation of memory impairment in attention deficit hyperactivity disorder and oppositional defiant disorder

    We conducted a comprehensive and systematic assessment of memory functioning indrug-naĆÆve boys with attention deficit hyperactivity disorder (ADHD) and oppositional defiant disorder (ODD).Ā Boys performed verbal and spatial working memory (WM) component (storage and central executive) and verbal and spatial storage load tasks, and the spatial span, spatial executive WM, spatial recognition memory and verbal recognition memory tasks from the Cambridge Neuropsychological Test Automated Battery. Groups comprised: (a) ADHD only (N = 21); (b) ADHD+ODD (N = 27); (c) ODD only (N = 21); and (d) typically developing (TYP) boys (N = 26). Groups were matched for age (M = 9.7 years) and sex (all boys).Ā Confirmatory factor analyses confirmed the presence of five factors: verbal functioning, spatial functioning, WM storage, WM central executive and long-termĀ memory (LTM). All three clinical groups demonstrated impaired memory performance. Boys with ODD and ODD+ADHD but not ADHD alone performed poorly on verbal memory tasks, whilst all three clinical groups showed impaired performance on spatial memory tasks. All three clinical groups performed poorly on the storage and central executive WM factors and the LTM factor.Ā ADHD and ODD are characterised by impaired performance storage and central executive WM tasks and LTM tasks. This is, we believe, the first report of impaired WM and LTM performance in ODD. This study suggests that verbal memory difficulties are more closely associated with ODD than ADHD symptoms and that combined ADHD+ODD represents a true comorbidity. The data also support a small but growing number of suggestions in the literature of impaired LTM in ADHD

    The Prevalence and Psychopathological Correlates of Sibling Bullying in Children with and without Autism Spectrum Disorder

    Using data from a prospective population based study, the prevalence and psychopathological correlates of sibling bullying in children with and without autism spectrum disorder (ASD) were estimated. There were 475 children with ASD and 13,702 children without ASD aged 11Ā years. Children with ASD were more likely to be bullied by their siblings compared to those without ASD. They were also more likely than those without ASD to both bully and be bullied by their siblings, which was associated with lower prosocial skills as well as more internalizing and externalizing problems compared to those not involved in any sibling bullying. Interventions to improve social and emotional outcomes in children with ASD should focus on both the affected and the unaffected sibling

    Elderly with Autism: Executive Functions and Memory

    Cognitive autism research is mainly focusing on children and young adults even though we know that autism is a life-long disorder and that healthy aging already has a strong impact on cognitive functioning. We compared the neuropsychological profile of 23 individuals with autism and 23 healthy controls (age range 51ā€“83Ā years). Deficits were observed in attention, working memory, and fluency. Aging had a smaller impact on fluency in the high functioning autism (HFA) group than in the control group, while aging had a more profound effect on visual memory performance in the HFA group. Hence, we provide novel evidence that elderly with HFA have subtle neuropsychological deficits and that the developmental trajectories differ between elderly with and without HFA in particular cognitive domains

    Cytochrome c(551) from Starkeya novella - Characterization, spectroscopic properties, and phylogeny of a diheme protein of the SoxAX family

    Cytochromes from the SoxAX family have a major role in thiosulfate oxidation via the thiosulfate-oxidizing multi-enzyme system (TOMES). Previously characterized SoxAX proteins from Rhodovulum sulficlophilum and Paracoccus pantotrophus contain three heme c groups, two of which are located on the SoxA subunit. In contrast, the SoxAX protein purified from Starkeya novella was found to contain only two heme groups. Mass spectrometry showed that a disulfide bond replaced the second heme group found in the diheme SoxA subunits. Apparent molecular masses of 27,229 +/- 10.3 Da and 20,258.6 +/- 1 Da were determined for SoxA and SoxX with an overall mass of 49.7 kDa, indicating a heterodimeric structure. Optical redox potentiometry found that the two heme cofactors are reduced at similar potentials (versus NHE) that are as follows: + 133 mV (pH 6.0); + 104 mV (pH 7.0); +49 (pH 7.9) and +10 mV (pH 8.7). EPR spectroscopy revealed that both ferric heme groups are in the low spin state, and the spectra were consistent with one heme having a His/Cys axial ligation and the other having a His/Met axial ligation. The His/Cys ligated heme is present in different conformational states and gives rise to three distinct signals. Amino acid sequencing was used to unambiguously assign the protein to the encoding genes, soxAX, which are part of a complete sox gene cluster found in S. novella. Phylogenetic analysis of soxA- and soxX-related gene sequences indicates a parallel development of SoxA and SoxY, with the diheme and monoheme SoxA sequences located on clearly separated branches of a phylogenetic tree
