    Completeness of the cubic and quartic H\'enon-Heiles Hamiltonians

    The quartic H\'enon-Heiles Hamiltonian H=(P12+P22)/2+(Ω1Q12+Ω2Q22)/2+CQ14+BQ12Q22+AQ24+(1/2)(α/Q12+β/Q22)−γQ1H = (P_1^2+P_2^2)/2+(\Omega_1 Q_1^2+\Omega_2 Q_2^2)/2 +C Q_1^4+ B Q_1^2 Q_2^2 + A Q_2^4 +(1/2)(\alpha/Q_1^2+\beta/Q_2^2) - \gamma Q_1 passes the Painlev\'e test for only four sets of values of the constants. Only one of these, identical to the traveling wave reduction of the Manakov system, has been explicitly integrated (Wojciechowski, 1985), while the three others are not yet integrated in the generic case (α,β,γ)≠(0,0,0)(\alpha,\beta,\gamma)\not=(0,0,0). We integrate them by building a birational transformation to two fourth order first degree equations in the classification (Cosgrove, 2000) of such polynomial equations which possess the Painlev\'e property. This transformation involves the stationary reduction of various partial differential equations (PDEs). The result is the same as for the three cubic H\'enon-Heiles Hamiltonians, namely, in all four quartic cases, a general solution which is meromorphic and hyperelliptic with genus two. As a consequence, no additional autonomous term can be added to either the cubic or the quartic Hamiltonians without destroying the Painlev\'e integrability (completeness property).Comment: 10 pages, To appear, Theor.Math.Phys.     Critical role of metals in biochemical properties of xylose isomerase

    Improving the activity of xylose isomerase (XI) is highly desired for achieving efficient fermentation of xylose in lignocellulosic biomass using XI-expressing S. cerevisiae. XI is a metalloenzyme which requires two bivalent metals for its catalytic activity. The enzyme from Piromyces sp. E2 (PirXI)[1],[2] is activated with various metal ions including Mg2+, Mn2+, Ca2+, Co2+, Zn2+ and Fe2+. The biochemical properties of PirXI are dependent on the types of its metal cofactors. Moreover, the enzyme shows different affinities towards these metals. Characterization of these properties is critical for understanding the enzyme behavior in vivo and to further adapt the enzyme to the cytosolic metal environment. Recently, we have shown that altered intracellular metal composition can improve anaerobic growth of a xylose-fermenting strain by enhancing the activity of PirXI[3]. Furthermore, our current study on PirXI and other studies on different XIs have shown that it is also possible to change the metal preferences of the enzyme[4]. A PirXI variant with a single amino acid substitution in the proximity of the metal binding residues showed significant changes in metal preference compared to the wild-type PirXI. Further exploration on metal specificity of PirXI is necessary to optimize the in vivo enzyme activity.     A carbohydrate-binding protein, B-GRANULE CONTENT 1, influences starch granule size distribution in a dose-dependent manner in polyploid wheat

    In Triticeae endosperm (e.g. wheat and barley), starch granules have a bimodal size distribution (with A- and B-type granules) whereas in other grasses the endosperm contains starch granules with a unimodal size distribution. Here, we identify the gene, BGC1 (B-GRANULE CONTENT 1), responsible for B-type starch granule content in Aegilops and wheat. Orthologues of this gene are known to influence starch synthesis in diploids such as rice, Arabidopsis, and barley. However, using polyploid Triticeae species, we uncovered a more complex biological role for BGC1 in starch granule initiation: BGC1 represses the initiation of A-granules in early grain development but promotes the initiation of B-granules in mid grain development. We provide evidence that the influence of BGC1 on starch synthesis is dose dependent and show that three very different starch phenotypes are conditioned by the gene dose of BGC1 in polyploid wheat: normal bimodal starch granule morphology; A-granules with few or no B-granules; or polymorphous starch with few normal A- or B-granules. We conclude from this work that BGC1 participates in controlling B-type starch granule initiation in Triticeae endosperm and that its precise effect on granule size and number varies with gene dose and stage of development

    Microvessel quantification in primary colorectal carcinoma: an immunohistochemical study.

    The vascularisation of human primary colorectal carcinomas was studied immunohistochemically using the endothelial cell markers CD31 and factor VIII-related antigen. Tumour sections were systematically scanned at a magnification of x 100 to find areas of intense neovascularisation. Microvessel counts within these vascular 'hotspots' were performed at magnification x 250. Regions in which tumour cords were surrounded by a collagen IV-positive basement membrane were compared with those in which this was absent and with normal mucosa. CD31 appeared to be a more sensitive marker for endothelial cells than factor VIII-related antigen (mean 185 +/- 59 and 120 +/- 38 microvessels mm-2). Within individual tumour sections microvessel counts in vascular hotspots with highest vessel density correlated significantly with microvessel counts in vascular hotspots with second highest vessel density (P < 0.01). Microvessel counts in tumour areas where collagen IV-positive basement membrane were absent exceeded those in areas where it was present (factor of 1.7) and those in normal mucosa (factor of 1.6). The differences in vessel density between individual tumours and the low variability in vessel density within individual tumours using this quantification technique allow us to investigate the prognostic value of vessel density in areas of intense neovascularisation in human primary colorectal carcinomas

    Mapping of Submerged Aquatic Vegetation in Rivers From Very High Resolution Image Data, Using Object Based Image Analysis Combined with Expert Knowledge

    The use of remote sensing for monitoring of submerged aquatic vegetation (SAV) in fluvial environments has been limited by the spatial and spectral resolution of available image data. The absorption of light in water also complicates the use of common image analysis methods. This paper presents the results of a study that uses very high resolution (VHR) image data, collected with a Near Infrared sensitive DSLR camera, to map the distribution of SAV species for three sites along the Desselse Nete, a lowland river in Flanders, Belgium. Plant species, including Ranunculus aquatilis L., Callitriche obtusangula Le Gall, Potamogeton natans L., Sparganium emersum L. and Potamogeton crispus L., were classified from the data using Object-Based Image Analysis (OBIA) and expert knowledge. A classification rule set based on a combination of both spectral and structural image variation (e.g. texture and shape) was developed for images from two sites. A comparison of the classifications with manually delineated ground truth maps resulted for both sites in 61% overall accuracy. Application of the rule set to a third validation image, resulted in 53% overall accuracy. These consistent results show promise for species level mapping in such biodiverse environments, but also prompt a discussion on assessment of classification accuracy

    Water from abandoned mines as a heat source: practical experiences of open- and closed-loop strategies, United Kingdom

    Pilot heat pump systems have been installed at two former collieries in Yorkshire/Derbyshire, England, to extract heat from mine water. The installations represent three fundamental configurations of heat exchanger. At Caphouse Colliery, mine water is pumped through a heat exchanger coupled to a heat pump and then discharged to waste (an open-loop heat exchange system). The system performs with high thermal efficiency, but the drawbacks are: (1) it can only be operated when mine water is being actively pumped from the colliery shaft for the purposes of regional water-level management, and (2) the fact that the water is partially oxygenated means that iron oxyhydroxide precipitation occurs, necessitating regular removal of filters for cleaning. At Markham Colliery, near Bolsover, a small amount of mine water is pumped from depth in a flooded shaft, circulated through a heat exchanger coupled to a heat pump and then returned to the same mine shaft at a slightly different depth (a standing column arrangement). This system’s fundamental thermal efficiency is negatively impacted by the electrical power required to run the shaft submersible pump, but clogging issues are not significant. In the third system, at Caphouse, a heat exchanger is submerged in a mine water treatment pond (a closed-loop system). This can be run at any time, irrespective of mine pumping regime, and being a closed-loop system, is not susceptible to clogging issues
