    Enabling Responsible Living Springer A Multi-Level Framework and Values-Based Indicators to Enable Responsible Living

    Abstract Efforts to enable responsible living require supporting frameworks and tools to bridge science and values at all levels from local to global. At the local level, community action is most effective in a village or neighbourhood where people will invest for the common betterment of their families and neighbours. Educational activities in and outside formal education for children, preadolescents, youth and adults should encourage action for responsible living based on the community's own values and vision of human purpose and well-being. Values-based indicators can help to measure the impact of sustainability education on behaviours in communities and organizations, as demonstrated in a recent pilot project. At the national level, in addition to formal curricular change, the media and diverse organizations of civil society from businesses to faith-based organizations can lead discussions of various dimensions of responsible living. Internationally, the debate on the future of sustainability around the Rio+20 conference has stimulated a re-examination of preconceptions and certitudes about individual and collective purposes and underlining the importance of values and ethical principles to sustainability. Linking local efforts to these international debates and implementing values-based indicators of education for sustainability will help to move from words to action for responsible living

    Galactic Kinematics Towards the South Galactic Pole. First Results from the Yale-San Juan Southern Proper-Motion Program

    The predictions from a Galactic Structure and Kinematic model code are compared to the color counts and absolute proper-motions derived from the Southern Proper-Motion survey covering more than 700 deg⁥2\deg^2 toward the South Galactic Pole in the range 9<BJ≀199 < B_{\rm J} \le 19. The theoretical assumptions and associated computational procedures, the geometry for the kinematic model, and the adopted parameters are presented in detail and compared to other Galactic Kinematic models of its kind. The data to which the model is compared consists of more than 30,000 randomly selected stars, and it is best fit by models with a solar peculiar motion of +5 km s−1^{-1} in the V-component (pointing in the direction of Galactic rotation), a large LSR speed of 270 km s−1^{-1}, and a (disk) velocity ellipsoid that always points towards the Galactic center. The absolute proper-motions in the U-component indicate a solar peculiar motion of 11.0±1.511.0 \pm 1.5 km s−1^{-1}, with no need for a local expansion or contraction term. The fainter absolute motions show an indication that the thick-disk must exhibit a rather steep velocity gradient of about -36 km s−1^{-1} kpc−1^{-1} with respect to the LSR. We are not able to set constraints on the overall rotation for the halo, nor on the thick-disk or halo velocity dispersions. Some substructure in the U & V proper-motions could be present in the brighter bins 10<BJ<1310 < B_{\rm J} < 13, and it might be indicative of (disk) moving groups.Comment: 24 double-column pages, 12 tables, AAS Latex macros v4.0, 19 B&W figures, 1 color figure.     Case report: Life threatening hyponatremia in infants with urinary tract infections: two cases of type III pseudohypoaldosteronism and review of the literature

    We describe two female infants at the age of five and six months with urinary tract infections presenting with vomiting and reduced drinking behavior. On laboratory analysis, severe hyponatremia (106 mmol/L and 109 mmol/L) was seen with hyperkalemia and compensated metabolic acidosis. Endocrinological analyses revealed massively increased levels of aldosterone and renin, leading to the diagnosis of type III pseudohypoaldosteronism (PHA). A review of the current literature 2013–2023 revealed 26 type III PHA cases aged up to ten months with reduced drinking behavior, weight loss and/or failure to thrive being the most common clinical presentations. Given the severe presentation of PHA electrolyte measurements in infants with urinary tract infections and/or in infants with congenital anomalies of the kidney and urinary tract (CAKUT) are strongly recommended

    From its inception holography has proven an extremely productive and attractive area of research. While specific technical applications give rise to 'hot topics', and three-dimensional (3D) visualisation comes in and out of fashion, the core principals involved continue to lead to exciting innovations in a wide range of areas. We humbly submit that it is impossible, in any journal document of this type, to fully reflect current and potential activity; however, our valiant contributors have produced a series of documents that go no small way to neatly capture progress across a wide range of core activities. As editors we have attempted to spread our net wide in order to illustrate the breadth of international activity.     Circulating microRNA Profiles during the Bovine Oestrous Cycle

    Up to 50% of ovulations go undetected in modern dairy herds due to attenuated oestrus behavior and a lack of high-accuracy methods for detection of fertile oestrus. This significantly reduces overall herd productivity and constitutes a high economic burden to the dairy industry. MicroRNAs (miRNAs) are ubiquitous regulators of gene expression during both health and disease and they have been shown to regulate different reproductive processes. Extracellular miRNAs are stable and can provide useful biomarkers of tissue function; changes in circulating miRNA profiles have been reported during menstrual cycles. This study sought to establish the potential of circulating miRNAs as biomarkers of oestrus in cattle. We collected plasma samples from 8 Holstein-Friesian heifers on days Days 0, 8 and 16 of an oestrous cycle and analysed small RNA populations on each Day using two independent high-throughput approaches, namely, Illumina sequencing (n = 24 samples) and Qiagen PCR arrays (n = 9 sample pools, 3-4 samples / pool). Subsequently, we used RT-qPCR (n = 24 samples) to validate the results of high-throughput analyses, as well as to establish the expression profiles of additional miRNAs previously reported to be differentially expressed during reproductive cycles. Overall, we identified four miRNAs (let-7f, miR-125b, miR-145 and miR-99a-5p), the plasma levels of which distinctly increased (up to 2.2-fold, P < 0.05) during oestrus (Day 0) relative to other stages of the cycle (Days 8 and 16). Moreover, we identified several hundred different isomiRs and established their relative abundance in bovine plasma. In summary, our results reveal the dynamic nature of plasma miRNAs during the oestrous cycle and provide evidence of the feasibility of using circulating miRNAs as biomarkers of reproductive function in livestock in the future

    Structural insights into TRPV2 activation by small molecules

    Transient receptor potential vanilloid 2 (TRPV2) is involved in many critical physiological and pathophysiological processes, making it a promising drug target. Here we present cryo-electron microscopy (cryo-EM) structures of rat TRPV2 in lipid nanodiscs activated by 2-aminoethoxydiphenyl borate (2-APB) and propose a TRPV2-specific 2-ABP binding site at the interface of S5 of one monomer and the S4-S5 linker of the adjacent monomer. In silico docking and electrophysiological studies confirm the key role of His521 and Arg539 in 2-APB activation of TRPV2. Additionally, electrophysiological experiments show that the combination of 2-APB and cannabidiol has a synergetic effect on TRPV2 activation, and cryo-EM structures demonstrate that both drugs were able to bind simultaneously. Together, our cryo-EM structures represent multiple functional states of the channel, providing a native picture of TRPV2 activation by small molecules and a structural framework for the development of TRPV2-specific activators

    Assessing ecosystem services for informing land-use decisions: a problem-oriented approach

    Assessments of ecosystem services (ES), that aim at informing decisions on land management, are increasing in number around the globe. Despite selected success stories, evidence for ES information being used in decision making is weak, partly because ES assessments are found to fall short in targeting information needs by decision makers. To improve their applicability in practice, we compared existing concepts of ES assessments with focus on informing land use decisions and identified opportunities for enhancing the relevance of ES assessments for decision making. In a process of codesign, building on experience of four projects in Brazil, China, Madagascar, and Vietnam, we developed a step-wise approach for better targeting ES assessments toward information needs in land use decisions. Our problem-oriented approach aims at (1) structuring ES information according to land use problems identified by stakeholders, (2) targeting context-specific ES information needs by decision makers, and (3) assessing relevant management options. We demonstrate how our approach contributes to making ES assessments more policy relevant and enhances the application of ES assessments as a tool for decision support
