    Cannabis sativa extracts protect LDL from Cu2+-mediated oxidation

    Background: Multiple therapeutic properties have been attributed to Cannabis sativa. However, further research is required to unveil the medicinal potential of Cannabis and the relationship between biological activity and chemical profile. Objectives: The primary objective of this study was to characterize the chemical profile and antioxidant properties of three varieties of Cannabis sativa available in Uruguay during progressive stages of maturation. Methods: Fresh samples of female inflorescences from three stable Cannabis sativa phenotypes, collected at different time points during the end of the flowering period were analyzed. Chemical characterization of chloroform extracts was performed by 1 H-NMR. The antioxidant properties of the Cannabis sativa extracts, and pure cannabinoids, were measured in a Cu2+-induced LDL oxidation assay. Results: The main cannabinoids in the youngest inflorescences were tetrahydrocannabinolic acid (THC-A, 242 ± 62 mg/g) and tetrahydrocannabinol (THC, 7.3 ± 6.5 mg/g). Cannabinoid levels increased more than twice in two of the mature samples. A third sample showed a lower and constant concentration of THC-A and THC (177 ± 25 and 1 ± 1, respectively). The THC-A/THC rich cannabis extracts increased the latency phase of LDL oxidation by a factor of 1.2–3.5 per μg, and slowed down the propagation phase of lipoperoxidation (IC50 1.7–4.6 μg/mL). Hemp, a cannabidiol (CBD, 198 mg/g) and cannabidiolic acid (CBD-A, 92 mg/g) rich variety, also prevented the formation of conjugated dienes during LDL oxidation. In fact, 1 μg of extract was able to stretch the latency phase 3.7 times and also to significantly reduce the steepness of the propagation phase (IC50 of 8 μg/mL). Synthetic THC lengthened the duration of the lag phase by a factor of 21 per μg, while for the propagation phase showed an IC50 ≤ 1 μg/mL. Conversely, THC-A was unable to improve any parameter. Meanwhile, the presence of 1 μg of pure CBD and CBD-A increased the initial latency phase 4.8 and 9.4 times, respectively, but did not have an effect on the propagation phase. Conclusion: Cannabis whole extracts acted on both phases of lipid oxidation in copper challenged LDL. Those effects were just partially related with the content of cannabinoids and partially recapitulated by isolated pure cannabinoids. Our results support the potentially beneficial effects of Cannabis sativa whole extracts on the initial phase of atherosclerosis

    Updated revision of the marine mammals collection housed at the Museo Argentino de Ciencias Naturales "Bernardino Rivadavia" (Buenos Aires, Argentina)

    Se realizó una revisión de los ejemplares de la Colección de Mamíferos Marinos del Museo Argentino de Ciencias Naturales "Bernardino Rivadavia". La misma indicó que del total de ejemplares catalogados (n=378), el 56.9% corresponde a Cetacea mientras que el 43.1% restante corresponde a Pinnipedia. Dentro de Cetacea la familia más representada es Delphinidae y el género con más ejemplares, Pontoporia (Pontoporidae); se reportan además, en forma complementaria, tres ejemplares tipo que no fueron listados en un trabajo previo. Dentro de los Pinnipedia, la familia mejor representada es Otariidae y el género con mayor número de ejemplares, Otaria.A thorough revision of the specimens housed at the Marine Mammals collection of the Museo Argentino de Ciencias Naturales "Bernardino Rivadavia" indicated that 56.9% (n=378), of the total number of specimens catalogued, belongs to Cetacea and the remainder (43.1%) to Pinnipedia. Among Cetacea, Delphinidae is the best represented family, whereas Pontoporia (Pontoporidae) is the genus which comprises the major number of specimens. Furthermore, we report three type specimens which were not cited in a previous work.     Ivermectin reduces in vivo coronavirus infection in a mouse experimental model

    SARS-CoV2 is a single strand RNA virus member of the type 2 coronavirus family, responsible for causing COVID-19 disease in humans. The objective of this study was to test the ivermectin drug in a murine model of coronavirus infection using a type 2 family RNA coronavirus similar to SARS-CoV2, the mouse hepatitis virus (MHV). BALB/cJ female mice were infected with 6,000 PFU of MHV-A59 (Group Infected; n=20) and immediately treated with one single dose of 500 μg/kg of ivermectin (Group Infected + IVM; n=20), or were not infected and treated with PBS (Control group; n=16). Five days after infection/treatment, mice were euthanized to obtain different tissues to check general health status and infection levels. Overall results demonstrated that viral infection induces the typical MHV disease in infected animals, with livers showing severe hepatocellular necrosis surrounded by a severe lymphoplasmacytic inflammatory infiltration associated with a high hepatic viral load (52,158 AU), while ivermectin administration showed a better health status with lower viral load (23,192 AU; p<0.05) and few livers with histopathological damage (p<0.05), not showing statistical differences with control mice (P=NS). Furthermore, serum transaminase levels (aspartate aminotransferase and alanine aminotransferase) were significantly lower in treated mice compared to infected animals. In conclusion, ivermectin seems to be effective to diminish MHV viral load and disease in mice, being a useful model for further understanding new therapies against coronavirus diseases.Fil: Arevalo, A.     La pandemia de la covid-19 irrumpió como un evento sin precedentes con especial impacto en las y los trabajadores de salud. El objetivo de este trabajo consistió en analizar en las narrativas de las y los trabajadores de salud pública de Mar del Plata y zonas rurales circundantes, las principales problemáticas y estrategias locales en la transición hacia la postpandemia. Con un diseño cualitativo, se realizaron entrevistas en profundidad entre marzo y diciembre de 2021. Los resultados revelaron el potencial de las y los trabajadores de salud pública como sujeto colectivo para la producción del cuidado en el marco de las tensiones que atraviesan el sistema sanitario. Se discute su capacidad instituyente para la salud colectiva desde la articulación del pensamiento crítico latinoamericano, la psicología social y el marxismo

    Effect of Different Technological Factors on the Gelation of a Low-Lectin Bean Protein Isolate

    Gelling ability of a bean protein isolate (BPI) obtained from a naturally low-lectin variety (Phaseolus vulgaris var. Almonga) was analysed. For that purpose differences on gels processing: concentration (14% and 17%), salt addition (0 and 2%), and pH (6.5 –lot A- and 7 –lot B), were studied to obtain suitable colour, mechanical and viscoelastic properties for making appropriate meat and seafood analogues. Gelation at pH 7 at both 14 and 17% BPI concentrations, produced less rigid, more flexible, time-stable and cohesive gel networks. Colour of the resulting gels was white enough to be considered as an adequate base for making plant-based analogues. The content of total galactoside, inositol phosphates and trypsin inhibitors (bioactive compounds) present in one serving (100 g) of these BPI gels were up to 0.80 mg/g, 8.06 mg/g and 239 TIUs, respectively

    Development, validation and application of a GC-MS method for the simultaneous detection and quantification of neutral lipid species in Trypanosoma cruzi

    The development and validation of an analytical method for the simultaneous analysis of five neutral lipids in Trypanosoma cruzi epimastigotes by GC-MS is presented in this study. The validated method meets all validation parameters for all components and the chromatographic conditions have been optimized during its development. This analytical method has demonstrated good selectivity, accuracy, within-day precision, recovery and linearity in each of the established ranges. In addition, detection and quantification limits for squalene, cholesterol, ergosterol and lanosterol have been improved and it is worth highlighting the fact that this is the first time that squalene-2,3-epoxide validation data have been reported. The new validated method has been applied to epimastigotes treated with compounds with in vitro anti- T.cruzi activity. This new methodology is straightforward and constitutes a tool for screening possible sterol biosynthesis pathway inhibitors in Trypanosoma cruzi, one of the most studied targets in Chagas disease treatment. Therefore, it is an interesting and useful contribution to medicinal chemistry research

    MYC is an oncogenic transcription factor dysregulated in about half of total human tumors. While transcriptomic studies reveal more than 1000 genes regulated by MYC, a much smaller fraction of genes is directly transactivated by MYC. Virtually all Burkitt lymphoma (BL) carry chromosomal translocations involving MYC oncogene. Most endemic BL and a fraction of sporadic BL are associated with Epstein-Barr virus (EBV) infection. The currently accepted mechanism is that EBV is the BL-causing agent inducing MYC translocation. Herein we show that the EBV receptor, CR2 (also called CD21), is a direct MYC target gene. This is based on several pieces of evidence: MYC induces CR2 expression in both proliferating and arrested cells and in the absence of protein synthesis, binds the CR2 promoter and transactivates CR2 in an E-box-dependent manner. Moreover, using mice with conditional MYC ablation we show that MYC induces CR2 in primary B cells. Importantly, modulation of MYC levels directly correlates with EBV's ability of infection in BL cells. Altogether, in contrast to the widely accepted hypothesis for the correlation between EBV and BL, we propose an alternative hypothesis in which MYC dysregulation could be the first event leading to the subsequent EBV infection.S

    La presente comunicación se basa en un proyecto de colaboración entre Ciencias del Mar y Didáctica de las Ciencias, encaminado a formar al profesorado de Educación Primaria, en formación inicial, en Educación Científica para la Sustentabilidad, con perspectiva de género, a través del conocimiento de modelos explicativos del Cambio Global, en la Historia de la Ciencia reciente, con especial atención al fenómeno de la acidificación oceánica. Todo ello se inserta en un modelo de formación del profesorado basado en la adquisición de competencias docentes, que necesita formación científica, didáctica y recursos. Se ha elaborado un instrumento de evaluación de recursos TIC y se concluye la necesidad de elaborar intervenciones didácticas, una vez que se hayan analizado éstos
