    Baculovirus PTP2 Functions as a Pro-Apoptotic Protein.

Available from MDPI via the DOI in this record.The family Baculoviridae encompasses a large number of invertebrate viruses, mainly infecting caterpillars of the order Lepidoptera. The baculovirus Spodoptera exigua multiple nucleopolyhedrovirus (SeMNPV) induces physiological and behavioral changes in its host Spodoptera exigua, as well as immunological responses, which may affect virus transmission. Here we show that the SeMNPV-encoded protein tyrosine phosphatase 2 (PTP2) induces mild apoptosis in Spodoptera frugiperda (Sf) 21 cells upon transient expression. Transient expression of a catalytic-site mutant of ptp2 did not lead to apoptosis, indicating that the phosphatase activity of PTP2 is needed to induce apoptosis. We also found that the caspase level (indicator of apoptosis) was higher in cells transfected with the ptp2 gene than in cells transfected with the catalytic mutant. Adding a caspase inhibitor reduced the level of ptp2-induced apoptosis. Moreover, deletion of the ptp2 gene from the viral genome prevented the induction of apoptosis in S. exigua hemocytes. The virus titer and virulence indices (the viral infectivity and the time to death) were not affected by deletion of the ptp2 gene. However, the viral occlusion body yield from S. exigua larvae infected with the mutant virus lacking the ptp2 gene was much lower than the yield from larvae infected with the wild-type (WT) virus. We hypothesize that the observed pro-apoptotic effects of PTP2 are the result of PTP2-mediated immune suppression in larvae, which consequently leads to higher viral occlusion body yields

    The Twin--Jet System in NGC 1052: VLBI-Scrutiny of the Obscuring Torus

    NGC 1052 offers the possibility to study the obscuring torus around a supermassive black hole, predicted by the standard model of active galactic nuclei, over a wide range of wavelengths from the radio to the X-ray regime. We present a detailed VLBI study of the parsec-scale structure of the ``twin-jet'' system in NGC 1052 in both total and polarized intensity and at multiple frequencies. We report the detection of linearly polarized emission from the base of the eastern jet at 5 GHz. While the radio spectrum in this region might be still consistent with synchrotron self absorption, the highly inverted spectrum of the western jet base represents a clear sign of pronounced free-free absorption in a circumnuclear torus. We observe an abrupt change of the brightness temperature gradient at a distance of 0.2\sim 0.2 pc to 0.3 pc from the central engine. This might provide an observational signature of the edge of the central torus, where the transition from an external pressure-dominated jet regime to a more or less freely expanding jet takes place. We determine the absorbing column density towards the western jet core to be 2.2×1022\sim 2.2 \times 10^{22} cm2^{-2} in good agreement with the values derived from various X-ray observations.     Environmental assay on the effect of poultry manure application on soil organisms in agroecosystems

    This paper reports the effects produced on the organisms of the soil (plants, invertebrates and microorganisms), after the application of two types of poultry manure (sawdust and straw bed) on an agricultural land. The test was made using a terrestrial microcosm, Multi-Species Soil System (MS3) developed in INIA. There was no difference in the germination for any of the three species of plants considered in the study. The biomass was increased in the wheat (Triticum aestivum) coming from ground treated with both kinds of poultry manure. Oilseed rape (Brasica rapa) was not affected and regarding vetch (Vicia sativa) only straw poultry manure showed significant difference. For length only Vicia sativa was affected showing a reduction when straw was exposed to poultry manure. When the effect on invertebrates was studied, we observed a reduction in the number of worms during the test, especially from the ground control (13.7%), higher than in the ground with sawdust poultry manure (6.7%), whereas in the ground with straw poultry manure, there was no reduction. The biomass was affected and at the end of the test it was observed that while the reduction of worms in the ground control was about 48%, the number of those that were in the ground with sawdust poultry manure or straw poultry manure decreased by 41% and 22% respectively. Finally, the effects on microorganisms showed that the enzymatic activities: dehydrogenase (DH) and phosphatase and basal respiration rate increased at the beginning of the test, and the differences were statistically significant compared with the values of the control group. During the test, all these parameters decreased (except DH activities) but they were always higher than in the ground control. This is why it is possible to deduce that the contribution of poultry manure caused an improvement in the conditions of fertilization and also for the soil

    The last decade has seen the expansion of trans identities that are gender queer, non-binary, androgynous, or multiply-sexed and gendered in Western Europe. These developments mark a shift from a uniformly gender-binaried system to one that encompasses some degree of gender pluralism, as reflected to an extent in policy changes in some European countries. However, gender binarism is still prevalent. This article uses the case of Norway to demonstrate a contrast between the citizenship statuses afforded to transsexual men and women, and the lack of citizenship rights that people with non-binary identities, and other gender-variant people who are not diagnosed as transsexual, face. The article addresses the historical role of the Norwegian state in perpetuating gender binaries, in key areas such as identity recognition. It then explores the ways in which Norwegian social policy is changing towards more trans-sensitive positions

    Parasitic manipulation of host behaviour: Baculovirus SeMNPV EGT facilitates tree-top disease in spodoptera exigua larvae by extending the time to death

One intriguing example concerns baculoviruses that induce hyperactivity and tree-top disease (i.e., climbing to elevated positions prior to death) in their caterpillar hosts. Little is known about the underlying mechanisms of such parasite-induced behavioural changes. Here, we studied the role of the ecdysteroid UDP-glucosyltransferase (egt) gene of Spodoptera exigua multiple nucleopolyhedrovirus (SeMNPV) in tree-top disease in S. exigua larvae. Larvae infected with a mutant virus lacking the egt gene exhibited a shorter time to death and died before the induction of tree-top disease. Moreover, deletion of either the open reading frame or the ATG start codon of the egt gene prevented tree-top disease, indicating that the EGT protein is involved in this process. We hypothesize that SeMNPV EGT facilitates tree-top disease in S. exigua larvae by prolonging the larval time to death.
