    Breaking the silence of the 500-year-old smiling garden of everlasting flowers: The En Tibi book herbarium

    We reveal the enigmatic origin of one of the earliest surviving botanical collections. The 16th-century Italian En Tibi herbarium is a large, luxurious book with c. 500 dried plants, made in the Renaissance scholarly circles that developed botany as a distinct discipline. Its Latin inscription, translated as “Here for you a smiling garden of everlasting flowers”, suggests that this herbarium was a gift for a patron of the emerging botanical science. We follow an integrative approach that includes a botanical similarity estimation of the En Tibi with contemporary herbaria (Aldrovandi, Cesalpino, “Cibo”, Merini, Estense) and analysis of the book’s watermark, paper, binding, handwriting, Latin inscription and the morphology and DNA of hairs mounted under specimens. Rejecting the previous origin hypothesis (Ferrara, 1542–1544), we show that the En Tibi was made in Bologna around 1558. We attribute the En Tibi herbarium to Francesco Petrollini, a neglected 16th-century botanist, to whom also belongs, as clarified herein, the controversial “Erbario Cibo” kept in Rome. The En Tibi was probably a work on commission for Petrollini, who provided the plant material for the book. Other people were apparently involved in the compilation and offering of this precious gift to a yet unknown person, possibly the Habsburg Emperor Ferdinand I. The En Tibi herbarium is a Renaissance masterpiece of art and science, representing the quest for truth in herbal medicine and botany. Our multidisciplinary approach can serve as a guideline for deciphering other anonymous herbaria, kept safely “hidden” in treasure rooms of universities, libraries and museums

    De bijdrage van toevoeging van cacao aan tabak aan rookverslaving

    In this report the effect of these compounds on the addiction to cigarette smoking was assessed, using currently available information in the literature on psychoactive compounds of cocoa. The investigated psychoactive cocoa compounds were theobromine, caffeine, serotonin, histamine, tryptophan, tryptamine, tyramine, phenylethylamine, octopamine and anandamide. The general conclusion is that the level of these compounds in added cocoa in cigarettes is not sufficient to increase the addiction to cigarette smoking.In dit rapport wordt de mogelijke bijdrage van cacao aan rookverslaving beschreven. Cacao wordt aan tabak toegevoegd om de smaak te verbeteren. Daarnaast bevat cacao tal van psychoactieve stoffen die mogelijk bijdragen aan rookverslaving Dit literatuuronderzoek beschrijft de blootstelling, farmacologie, farmacokinetiek, toxicologie, interacties en verslavende eigenschappen van de tien meest bekende stoffen in cacao. De onderzochte stoffen zijn theobromine, caffeine, serotonine, histamine, tryptofaan, tryptamine, tyramine, fenylethylamine, octopamine en anandamide. Deze stoffen komen ook via dranken en voedsel het lichaam binnen of worden door het lichaam zelf aangemaakt. Dit rapport laat zien dat de aan roken gerelateerde blootstelling aan de psychoactieve stoffen uit cacao gering is ten opzichte van de inname via voeding en dranken en/of de lichaamseigen productie van deze stoffen. Een systemisch effect lijkt derhalve onwaarschijnlijk ook al omdat lichaamseigen stoffen snel worden afgebroken. Daarnaast kunnen deze stoffen, omdat ze geinhaleerd worden, een direct effect op de luchtwegen hebben. Daarmee zou de opname van nicotine beinvloed kunnen worden. De nicotine opname zou bijvoorbeeld kunnen toenemen via luchtwegverwijding door theobromine en cafe6ne of kunnen afnemen door luchtwegvernauwing door histamine. Dit rapport laat zien dat de aan roken gerelateerde blootstelling aan deze stoffen waarschijnlijk te gering is voor een direct effect op de luchtwegen. Verder dient te worden opgemerkt dat de hoeveelheid tryptamine, tyramine en fenylethylamine die via cacao wordt toegevoegd verwaarloosbaar is ten opzichte van de hoeveelheid die in tabak zelf aanwezig is. Tot slot is aandacht besteed aan de verbrandingsproducten van cacao. Amine verbindingen als serotonin, tryptofaan, tyramine, tryptamine en fenylethylamine vormen tijdens het roken stoffen die het enzym mono amine oxidase (MAO) remmen. MAO-remmers hebben een anti-depressieve werking en kunnen op die manier bijdragen aan rookverslaving. De conclusie van dit literatuur onderzoek is dat de afzonderlijke psychoactieve stoffen in tabak als gevolg van toevoeging van cacao niet direct bijdragen aan rookverslaving. De verbrandingsproducten van cacao doen dit, via remming van het enzym mono amine oxidase, mogelijk wel. Ook de smaak van cacao wordt geassocieerd met verslaving. De literatuur biedt geen inzicht in het effect op gezondheid en verslaving van het inhaleren van de combinatie van de 10 onderzochte stoffen uit cacao

    Linking forest cover, soil erosion and mire hydrology to late-Holocene human activity and climate in NW Spain

Because of the wide range of factors involved, understanding these complex links requires long-term multi-proxy approaches and research on the best proxies to focus. A peat core from NW Spain (Cruz do Bocelo mire), spanning the last ~3000 years, has been studied at high resolution by physical (density and loss on ignition (LOI)), geochemical (elemental composition) and palynological (pollen and non-pollen palynomorphs) analyses. Proxies related to mineral matter fluxes from the catchment (lithogenic tracers, Glomus and Entorrhiza), rainfall (Bromine), mire hydrology (HdV-18), human pressure (Cerealia-type, nitrophilous taxa and coprophilous fungi) and forest cover (mesophilous tree taxa) were the most useful to reconstruct the evolution of the mire and its catchment. Forest clearance for farming was one of the main drivers of environmental change from at least the local Iron Age (~2685 cal. yr BP) onwards. The most intense phase of deforestation occurred during Roman and Germanic times and the late Middle Ages. During these phases, the entire catchment was affected, resulting in enhanced soil erosion and severe hydrological modifications of the mire. Climate, especially rainfall, may have also accelerated these processes during wetter periods. However, it is noteworthy that the hydrology of the mire seems to have been insensitive to rainfall variations when mesophilous forest dominated. Abrupt changes were only detected once intense forest clearance commenced during the Iron Age/Roman transition (~2190 cal. yr BP) phase, which represented a tipping point in catchment's ability to buffer impacts.     The diverse nature of island isolation and its effect on land bridge insular faunas

    Aim: Isolation is a key factor in island biology. It is usually defined as the distance to the geographically nearest mainland, but many other definitions exist. We explored how testing different isolation indices affects the inference of impacts of isolation on faunal characteristics. We focused on land bridge islands and compared the relationships of many spatial and temporal (i.e., through time) isolation indices with community‐, population‐ and individual‐level characteristics (species richness, population density and body size, respectively). Location: Aegean Sea islands, Greece. Time period: Current. Taxon: Many animal taxa. Methods: We estimated 21 isolation indices for 205 islands and recorded species richness data for 15 taxa (invertebrates and vertebrates). We obtained body size data for seven lizard species and population density data for three. We explored how well indices predict each characteristic, in each taxon, by conducting a series of ordinary least squares regressions (controlling for island area when needed) and a meta‐analysis. Results: Isolation was significantly (and negatively) associated with species richness in 10 of 15 taxa. It was significantly (and positively) associated with body size in only one of seven species and was not associated with population density. The effect of isolation on species richness was much weaker than that of island area, regardless of the index tested. Spatial indices generally out‐performed temporal indices, and indices directly related to the mainland out‐performed those related mainly to neighbouring islands. No index was universally superior to others, including the distance to the geographically nearest mainland. Main conclusions: The choice of index can alter our perception of the impacts of isolation on biological patterns. The nearly automatic, ubiquitous use of distance to the geographically nearest mainland misrepresents the complexity of the effects of isolation. We recommend the simultaneous testing of several indices that represent different aspects of isolation, in order to produce more constructive and thorough investigations and avoid imprecise inference

    Effects of bumetanide on neurodevelopmental impairments in patients with tuberous sclerosis complex: An open-label pilot study

    Background: Tuberous sclerosis complex (TSC) is an autosomal dominant disease that affects multiple organs including the brain. TSC is strongly associated with broad neurodevelopmental disorders, including autism spectrum disorder symptomatology. Preclinical TSC studies have indicated altered neuronal chloride homeostasis affecting the polarity of γ-aminobutyric acid (GABA) ergic transmission as a potential treatment target. Bumetanide, a selective NKCC1 chloride importer antagonist, may attenuate depolarizing GABA action, and in that way reduce disease burden. In this open-label pilot study, we tested the effect of bumetanide on a variety of neurophysiological, cognitive, and behavioral measures in children with TSC. Methods: Participants were treated with bumetanide (2dd 0.5-1.0 mg) for 13 weeks in an open-label trial. The Aberrant Behavior Checklist-Irritability (ABC-I) subscale was chosen as the primary endpoint. Secondary endpoints included other behavioral questionnaires in addition to event-related potentials (ERP) and neuropsychological tests if tolerated. Additionally, the treatment effect on seizure frequency and quality of life was assessed. Endpoint data were collected at baseline, after 91 days of treatment and after a 28-day wash-out period. Results: Fifteen patients (8-21-years old) with TSC were included of which 13 patients completed the study. Treatment was well-tolerated with only expected adverse events due to the diuretic effects of bumetanide. Irritable behavior (ABC-I) showed significant improvement after treatment in 11 out of 13 patients (t(12) = 4.41, p =.001, d =.773). A favorable effect was also found for social behavior (Social Responsiveness Scale) (t(11) = 4.01, p =.002, d =.549) and hyperactive behavior (ABC-hyperactivity subscale) (t(12) = 3.65, p =.003, d =.686). Moreover, patients rated their own health-related quality of life higher after treatment. At baseline, TSC patients showed several atypical ERPs versus typically developing peers of which prepulse inhibition was significantly decreased in the TSC group. Neuropsychological measurements showed no change and bumetanide had no effect on seizure frequency. Limitations: The sample size and open-label design of this pilot study warrant caution when interpreting outcome measures. Conclusions: Bumetanide treatment is a potential treatment to alleviate the behavioral burden and quality of life associated with TSC. More elaborate trials are needed to determine the application and effect size of bumetanide for the TSC population.     Current Pyuria Cutoffs Promote Inappropriate Urinary Tract Infection Diagnosis in Older Women

    Background: Pre-existing lower urinary tract symptoms (LUTS), cognitive impairment, and the high prevalence of asymptomatic bacteriuria (ASB) complicate the diagnosis of urinary tract infection (UTI) in older women. The presence of pyuria remains the cornerstone of UTI diagnosis. However, >90% of ASB patients have pyuria, prompting unnecessary treatment. We quantified pyuria by automated microscopy and flowcytometry to determine the diagnostic accuracy for UTI and to derive pyuria thresholds for UTI in older women. Methods: Women ≥65 years with ≥2 new-onset LUTS and 1 uropathogen ≥104 colony-forming units (CFU)/mL were included in the UTI group. Controls were asymptomatic and classified as ASB (1 uropathogen ≥105 CFU/mL), negative culture, or mixed flora. Patients with an indwelling catheter or antimicrobial pretreatment were excluded. Leukocyte medians were compared and sensitivity–specificity pairs were derived from a receiver operating characteristic curve. Results: We included 164 participants. UTI patients had higher median urinary leukocytes compared with control patients (microscopy: 900 vs 26 leukocytes/µL; flowcytometry: 1575 vs 23 leukocytes/µL; P < .001). Area under the curve was 0.93 for both methods. At a cutoff of 264 leukocytes/µL, sensitivity and specificity of microscopy were 88% (positive and negative likelihood ratio: 7.2 and 0.1, respectively). The commonly used cutoff of 10 leukocytes/µL had a poor specificity (36%) and a sensitivity of 100%. Conclusions: The degree of pyuria can help to distinguish UTI in older women from ASB and asymptomatic controls with pyuria. Current pyuria cutoffs are too low and promote inappropriate UTI diagnosis in older women
