Diacrisia obliqua is a polyphagous pest especially on oil seed crops. Adult female sex pheromone blend consists of five pheromone components, which include (3Z,6Z)-cis-9,10-epoxyl,3,6-henicosatriene and (3Z,6Z)-cis-9,10-epoxy3,6-henicosadiene. Synthesis of these enantiomers was achieved through alkylative epoxide rearrangement and stereoselective Wittig olefination reactions as key steps. The small RNA locus map for Chlamydomonas reinhardtii.
Small (s)RNAs play crucial roles in the regulation of gene expression and genome stability across eukaryotes where they direct epigenetic modifications, post-transcriptional gene silencing, and defense against both endogenous and exogenous viruses. It is known that Chlamydomonas reinhardtii, a well-studied unicellular green algae species, possesses sRNA-based mechanisms that are distinct from those of land plants. However, definition of sRNA loci and further systematic classification is not yet available for this or any other algae. Here, using data-driven machine learning approaches including Multiple Correspondence Analysis (MCA) and clustering, we have generated a comprehensively annotated and classified sRNA locus map for C. reinhardtii. This map shows some common characteristics with higher plants and animals, but it also reveals distinct features. These results are consistent with the idea that there was diversification in sRNA mechanisms after the evolutionary divergence of algae from higher plant lineages
Size control of Charge-Orbital Order in Half-Doped Manganite, LaCaMnO
Motivated by recent experimental results, we study the effect of size
reduction on half-doped manganite, LaCaMnO, using the
combination of density functional theory (DFT) and dynamical mean field theory
(DMFT). We find that upon size reduction, the charge-ordered antiferromagnetic
phase, observed in bulk, to be destabilized, giving rise to the stability of a
ferromagnetic metallic state. Our theoretical results, carried out on
defect-free nanocluster in isolation, establish the structural changes that
follow upon size reduction to be responsible for this. Our study further points
out the effect of size reduction to be distinctively different from application
of hydrostatic pressure. Interestingly, our DFT+DMFT study, additionally,
reports the correlation-driven stability of charge-orbitally ordered state in
bulk LaCaMnO, even in absence of long range magnetic order.Comment: 5 pages, 3 figures; to appear in Physical Review Letter
Cardiomyocyte ionic currents in intact young and aged murine Pgc-1β-/- atrial preparations.
INTRODUCTION: Recent studies reported that energetically deficient murine Pgc-1β-/- hearts replicate age-dependent atrial arrhythmic phenotypes associated with their corresponding clinical conditions, implicating action potential (AP) conduction slowing consequent upon reduced AP upstroke rates. MATERIALS AND METHODS: We tested a hypothesis implicating Na+ current alterations as a mechanism underlying these electrophysiological phenotypes. We applied loose patch-clamp techniques to intact young and aged, WT and Pgc-1β-/-, atrial cardiomyocyte preparations preserving their in vivo extracellular and intracellular conditions. RESULTS AND DISCUSSION: Depolarising steps activated typical voltage-dependent activating and inactivating inward (Na+) currents whose amplitude increased or decreased with the amplitudes of the activating, or preceding inactivating, steps. Maximum values of peak Na+ current were independently influenced by genotype but not age or interacting effects of genotype and age on two-way ANOVA. Neither genotype, nor age, whether independently or interactively, influenced voltages at half-maximal current, or steepness factors, for current activation and inactivation, or time constants for recovery from inactivation following repolarisation. In contrast, delayed outward (K+) currents showed similar activation and rectification properties through all experimental groups. These findings directly demonstrate and implicate reduced Na+ in contrast to unchanged K+ current, as a mechanism for slowed conduction causing atrial arrhythmogenicity in Pgc-1β-/- hearts
Oral tolerance to cancer can be abrogated by T regulatory cell inhibition
Oral administration of tumour cells induces an immune hypo-responsiveness known as oral tolerance. We have previously shown that oral tolerance to a cancer is tumour antigen specific, non-cross-reactive and confers a tumour growth advantage. We investigated the utilisation of regulatory T cell (Treg) depletion on oral tolerance to a cancer and its ability to control tumour growth. Balb/C mice were gavage fed homogenised tumour tissue – JBS fibrosarcoma (to induce oral tolerance to a cancer), or PBS as control. Growth of subcutaneous JBS tumours were measured; splenic tissue excised and flow cytometry used to quantify and compare systemic Tregs and T effector (Teff) cell populations. Prior to and/or following tumour feeding, mice were intraperitoneally administered anti-CD25, to inactivate systemic Tregs, or given isotype antibody as a control. Mice which were orally tolerised prior to subcutaneous tumour induction, displayed significantly higher systemic Treg levels (14% vs 6%) and faster tumour growth rates than controls (p<0.05). Complete regression of tumours were only seen after Treg inactivation and occurred in all groups - this was not inhibited by tumour feeding. The cure rates for Treg inactivation were 60% during tolerisation, 75% during tumour growth and 100% during inactivation for both tolerisation and tumour growth. Depletion of Tregs gave rise to an increased number of Teff cells. Treg depletion post-tolerisation and post-tumour induction led to the complete regression of all tumours on tumour bearing mice. Oral administration of tumour tissue, confers a tumour growth advantage and is accompanied by an increase in systemic Treg levels. The administration of anti-CD25 Ab decreased Treg numbers and caused an increase in Teffs. Most notably Treg cell inhibition overcame established oral tolerance with consequent tumor regression, especially relevant to foregut cancers where oral tolerance is likely to be induced by the shedding of tumour tissue into the gut
Distinct roles of Argonaute in the green alga Chlamydomonas reveal evolutionary conserved mode of miRNA-mediated gene expression
Abstract: The unicellular green alga Chlamydomonas reinhardtii is evolutionarily divergent from higher plants, but has a fully functional silencing machinery including microRNA (miRNA)-mediated translation repression and mRNA turnover. However, distinct from the metazoan machinery, repression of gene expression is primarily associated with target sites within coding sequences instead of 3′UTRs. This feature indicates that the miRNA-Argonaute (AGO) machinery is ancient and the primary function is for post transcriptional gene repression and intermediate between the mechanisms in the rest of the plant and animal kingdoms. Here, we characterize AGO2 and 3 in Chlamydomonas, and show that cytoplasmically enriched Cr-AGO3 is responsible for endogenous miRNA-mediated gene repression. Under steady state, mid-log phase conditions, Cr-AGO3 binds predominantly miR-C89, which we previously identified as the predominant miRNA with effects on both translation repression and mRNA turnover. In contrast, the paralogue Cr-AGO2 is nuclear enriched and exclusively binds to 21-nt siRNAs. Further analysis of the highly similar Cr-AGO2 and Cr-AGO 3 sequences (90% amino acid identity) revealed a glycine-arginine rich N-terminal extension of ~100 amino acids that, given previous work on unicellular protists, may associate AGO with the translation machinery. Phylogenetic analysis revealed that this glycine-arginine rich N-terminal extension is present outside the animal kingdom and is highly conserved, consistent with our previous proposal that miRNA-mediated CDS-targeting operates in this green alga

Chern-Simons Solitons, Toda Theories and the Chiral Model
The two-dimensional self-dual Chern--Simons equations are equivalent to the
conditions for static, zero-energy solutions of the -dimensional gauged
nonlinear Schr\"odinger equation with Chern--Simons matter-gauge dynamics. In
this paper we classify all finite charge solutions by first
transforming the self-dual Chern--Simons equations into the two-dimensional
chiral model (or harmonic map) equations, and then using the Uhlenbeck--Wood
classification of harmonic maps into the unitary groups. This construction also
leads to a new relationship between the Toda and chiral model
solutions
Existence of global strong solutions in critical spaces for barotropic viscous fluids
This paper is dedicated to the study of viscous compressible barotropic
fluids in dimension . We address the question of the global existence
of strong solutions for initial data close from a constant state having
critical Besov regularity. In a first time, this article show the recent
results of \cite{CD} and \cite{CMZ} with a new proof. Our result relies on a
new a priori estimate for the velocity, where we introduce a new structure to
\textit{kill} the coupling between the density and the velocity as in
\cite{H2}. We study so a new variable that we call effective velocity. In a
second time we improve the results of \cite{CD} and \cite{CMZ} by adding some
regularity on the initial data in particular is in . In this
case we obtain global strong solutions for a class of large initial data on the
density and the velocity which in particular improve the results of D. Hoff in
\cite{5H4}. We conclude by generalizing these results for general viscosity
coefficients
