Procuramos compreender alguns dos mecanismos que permitiram a um professor de Física de uma escola pública da cidade de Belo Horizonte, Brasil, construir práticas inovadoras em meio a um grupo de professores fortemente resistentes a inovações curriculares. Com base no referencial teórico de René Kaës, pudemos concluir que elementos da formação inicial, ancorados pela implantação de uma política educacional potencialmente emancipadora, puderam se constituir em intermediários que sustentaram os impasses vividos pelo professor naquela escola: continuar ocupando o lugar a ele assinalado pelo grupo de professores resistentes ou pautar sua prática na aprendizagem dos alunos. Ao enfrentar esse impasse o professor pôde valorizar seu processo de formação inicial na medida em que houve a criação de um espaço transicional entre-dois lugares: formação e profissão

Dominant Role of PI3K p110α over p110β in Insulin and β-Adrenergic Receptor Signalling

Attribution of specific roles to the two ubiquitously expressed PI 3-kinase (PI3K) isoforms p110α and p110β in biological functions they have been implicated, such as in insulin signalling, has been challenging. While p110α has been demonstrated to be the principal isoform activated downstream of the insulin receptor, several studies have provided evidence for a role of p110β. Here we have used isoform-selective inhibitors to estimate the relative contribution of each of these isoforms in insulin signalling in adipocytes, which are a cell type with essential roles in regulation of metabolism at the systemic level. Consistent with previous genetic and pharmacological studies, we found that p110α is the principal isoform activated downstream of the insulin receptor under physiological conditions. p110α interaction with Ras enhanced the strength of p110α activation by insulin. However, this interaction did not account for the selectivity for p110α over p110β in insulin signalling. We also demonstrate that p110α is the principal isoform activated downstream of the β-adrenergic receptor (β-AR), another important signalling pathway in metabolic regulation, through a mechanism involving activation of the cAMP effector molecule EPAC1. This study offers further insights in the role of PI3K isoforms in the regulation of energy metabolism with implications for the therapeutic application of selective inhibitors of these isoforms

Anatomy and tectonic significance of WNW-ESE and NE-SW lineaments at a transpressive plate boundary (Nubia-Iberia)

Recent mapping of the Gulf of Cadiz seafloor permitted to identify major tectonic lineaments: the SWIM lineaments (Zitellini et al., 2009) and Cadiz lineament, striking WNW-ESE and NE-SW, respectively. Multibeam swath bathymetry and interpretation of multi-channel seismic data indicate that these features can be interpreted to correspond to the seafloor morphological expression of active dextral strike-slip faults. Based on the interpreted data and recently published GPS plate kinematic velocity vectors of Nubia with respect to Iberia and the Alboran block (e.g. Fernandes et al.; 2003 Stich et al., 2006) we propose that the SWIM Faults are related to the general NW-SE convergence of Nubia with respect to Iberia, and the Cadiz fault is related to the westward movement of the Gibraltar orogenic arc

Encontros e desencontros da formação em serviço

Neste texto analisamos as tensões entre docentes de uma escola pública – em suas tentativas de mudanças no currículo por meio da construção de Projetos de Trabalho – e os gestores responsáveis pela formação em serviço, com seus aportes conceituais. Os encontros de formação foram marcados por mudanças na política educacional da cidade de Belo Horizonte, Brasil. Para compreensão das transformações que ocorreram nas produções curriculares dos professores utilizamos conceitos retirados do referencial teórico de René Kaës. Nosso objetivo é obter indicações que possibilitem compreender melhor a formação em serviço, tendo a escola como espaço privilegiado de formação. Os dilemas enfrentados, se por um lado mostram as dificuldades inerentes à construção de projetos coletivos, por outro levam ao reconhecimento, pela comunidade escolar, dos atores envolvidos neste processo

Monomorphic subtelomeric DNA in the filamentous fungus, Metarhizium anisopliae, contains a RecQ helicase-like gene.

In most filamentous fungi, telomere-associated sequences (TASs) are polymorphic, and the presence of restriction fragment length polymorphisms (RFLPs) may permit the number of chromosome ends to be estimated from the number of telomeric bands obtained by restriction digestion. Here, we describe strains of Metarhizium, Gliocladium and Paecilomyces species in which only one or a few telomeric bands of unequal intensity are detectable by Southern hybridization, indicating that interchromosomal TAS exchange occurs. We also studied an anomalous strain of Metarhizium anisopliae, which produces polymorphic telomeric bands larger than 8 kb upon digestion of genomic DNA with XhoI. In this case, the first XhoI site in from the chromosome end must lie beyond the presumed monomorphic region. Cloned telomeres from this strain comprise 18?26 TTAGGG repeats, followed at the internal end of the telomere tract by five repeats of the telomere-like sequence TAAACGCTGG. An 8.1-kb TAS clone also contains a gene for a RecQ-like helicase, designated TAH1, suggesting that this TAS is analogous to the Y elements in yeast and the subtelomeric helicase ORFs of Ustilago maydis (UTASRecQ) and Magnaporthe grisea (TLH1). The TAS in the anomalous strain of M. anisopliae, however, appears distinct from these in that it is found at most telomeres and its predicted protein product possesses a significantly longer N-terminal region in comparison to the M. grisea and U. maydis helicases. Hybridization analyses showed that TAH1 homologues are present in all other anomalous M. anisopliae strains studied, as well as in some other polymorphic strains, where the recQ-like gene also appears to be telomere-associated.Published online: 2 June 2005
