    Efficiency of DNA mini-barcoding to assess mislabeling in commercial fish products in italy: An overview of the last decade

    The problem of fish traceability in processed products is still an important issue in food safety. Major attention is nowadays dedicated to consumer health and prevention of possible frauds regulated by national and international laws. For this reason, a technical approach is fun-damental in revealing mislabeling at different levels. In particular, the use of genetic markers has been standardized and DNA barcoding is considered the gold-standard strategy to examine and prevent species substitution. Considering the richness of available DNA databases, it is nowadays possible to rapidly reach a reliable taxonomy at the species level. Among different approaches, an innovative method based on DNA mini barcoding has recently been proposed at an international level. Starting from this evidence, we herein illustrate an investigation dealing with the evolution of this topic in Italy over the last decade. The molecular analysis of 71 commercial fish samples based on mini-COI sequencing with two different primer sets reached an amplification success rate of 87.3 and 97.2%. The investigation revealed four major frauds (5.8%) and four minor ones (5.8%). Results highlighted a decrease in incorrect labeling in Italy from 32% to 11.6% over the last decade, although a recurrent involvement of “endangered” species sensu IUCN was still observed

    A Non-coded β2,2-Amino Acid with Isoxazoline Core Able to Stabilize Peptides Folding through an Unprecedented Hydrogen Bond

    Dedicated to Prof. Cesare Gennari on the occasion of his 70th birthday New peptidomimetics containing a beta(2,)(2)-isoxazoline amino acid, i.e. 5-(aminomethyl)-3-phenyl-4,5-dihydroisoxazole-5-carboxylic acid (Isox-beta(2,)(2)AA), were prepared and studied by NMR and theoretical calculation. Although similar amino acid derivatives have already been prepared via 1,3-dipolar cycloaddition reaction, neither experimental details nor characterization were found and they were never used for peptide synthesis. Both enantiomers were inserted in peptide sequences to verify their ability to induce a secondary structure. We found that an unexpected conformation is given by R-Isox-beta(2,)(2)AA, inducing the folding of short peptides thanks to an unprecedented H-bond involving C=N of the isoxazoline side chain of our beta(2,)(2)-AA

    Highly enantioselective “inherently chiral” electroactive materials based on a 2,2' -biindole atropisomeric scaffold

    Chiral oligothiophene monomers with C2 symmetry, based on 3,30 -bithiophene atropisomeric cores with high racemization barriers, have recently been shown to provide excellent chiral starting materials with high electroactivity for the easy preparation of enantiopure electroactive films endowed with powerful chirality manifestations. We now introduce an inherently chiral monomer based on a 2,20 -biindole core, as the prototype of a new inherently chiral monomer family, whose properties could be modulable through functionalization of the pyrrolic N atoms. By fast, regular electrooligomerization the new monomer yields inherently chiral films with high, reversible electroactivity and, above all, impressive enantioselectivity towards very different chiral probes, some of pharmaceutical interest, as generalscope electrode surfaces. Such results, while opening the way to a new, attractive inherently chiral selector class, nicely confirm the general validity of the inherent chirality strategy for chiral electrochemistry. Furthermore, the enantioselectivity of the new selectors not only holds with electroactive chiral probes, but also with circularly polarized light components as well as electron spins, resulting in good chiroptical and spin filter performances, which suggests fascinating correlations between the three contexts

    Il broncospasmo da eroina, di recente individuazione, è di raro riscontro probabilmente perché poco conosciuto o non adeguatamente indagato. L’eroina, principalmente attraverso un meccanismo di istamino-liberazione, induce ostruzione bronchiale che appare più grave negli asmatici cronici e negli atopici manifestandosi pochi minuti dopo l’inalazione della sostanza. Il narcotico può determinare: a) broncospasmo con insufficienza respiratoria che può richiedere cure intensive con ricorso alla ventilazione meccanica; b) depressione del centro bulbare del respiro; c) sintomatologia sistemica; d) sindrome da astinenza, che si può presentare poco dopo la risoluzione dell’episodio acuto. Un rapporto empatico con il paziente può agevolare il percorso diagnostico; infatti, la sua reticenza a svelare l’abitudine voluttuaria ritarda l’individuazione dell’esposizione alla sostanza. Ciò può comportare un trattamento terapeutico ritardato, con conseguente aggravamento della condizione clinica a possibile evoluzione infausta.Bronchospasm caused by inhaled heroin has recently been identified. The condition has been rarely identified and/or not sufficiently investigated. Through the reaction mediated by histamine, heroin causes a bronchial obstruction that seems more severe in chronic asthmatic and atopic individuals, immediately after inhalating the substance. Heroin can reveal: a) bronchospasm with respiratory failure that may require intensive care up to mechanical ventilation; b) depression of the bulbar center of breath; c) systemic symptoms; d) withdrawal syndrome that may be evident after an acute clinical event. Even though an empathic relationship between the patient and the doctor can facilitate the diagnostic process, a lack of early identification of exposure to the substance, due to the patient’s hesitation to disclose his/her behavior, may cause a delay in the treatment and a worsening of clinical conditions with unfavourable development

    Impact on Prehospital Delay of a Stroke Preparedness Campaign: A SW-RCT (Stepped-Wedge Cluster Randomized Controlled Trial)

    Background and Purpose—Public campaigns to increase stroke preparedness have been tested in different contexts, showing contradictory results. We evaluated the effectiveness of a stroke campaign, designed specifically for the Italian population in reducing prehospital delay. Methods—According to an SW-RCT (Stepped-Wedge Cluster Randomized Controlled Trial) design, the campaign was launched in 4 provinces in the northern part of the region Emilia Romagna at 3-month intervals in randomized sequence. The units of analysis were the patients admitted to hospital, with stroke and transient ischemic attack, over a time period of 15 months, beginning 3 months before the intervention was launched in the first province to allow for baseline data collection. The proportion of early arrivals (within 2 hours of symptom onset) was the primary outcome. Thrombolysis rate and some behavioral end points were the secondary outcomes. Data were analyzed using a fixed-effect model, adjusting for cluster and time trends. Results—We enrolled 1622 patients, 912 exposed and 710 nonexposed to the campaign. The proportion of early access was nonsignificantly lower in exposed patients (354 [38.8%] versus 315 [44.4%]; adjusted odds ratio, 0.81; 95% confidence interval, 0.60–1.08; P=0.15). As for secondary end points, an increase was found for stroke recognition, which approximated but did not reach statistical significance (P=0.07). Conclusions—Our campaign was not effective in reducing prehospital delay. Even if some limitations of the intervention, mainly in terms of duration, are taken into account, our study demonstrates that new communication strategies should be tested before large-scale implementation.     Brain-behaviour modes of covariation in healthy and clinically depressed young people.

    Understanding how variations in dimensions of psychometrics, IQ and demographics relate to changes in brain connectivity during the critical developmental period of adolescence and early adulthood is a major challenge. This has particular relevance for mental health disorders where a failure to understand these links might hinder the development of better diagnostic approaches and therapeutics. Here, we investigated this question in 306 adolescents and young adults (14-24 y, 25 clinically depressed) using a multivariate statistical framework, based on canonical correlation analysis (CCA). By linking individual functional brain connectivity profiles to self-report questionnaires, IQ and demographic data we identified two distinct modes of covariation. The first mode mapped onto an externalization/internalization axis and showed a strong association with sex. The second mode mapped onto a well-being/distress axis independent of sex. Interestingly, both modes showed an association with age. Crucially, the changes in functional brain connectivity associated with changes in these phenotypes showed marked developmental effects. The findings point to a role for the default mode, frontoparietal and limbic networks in psychopathology and depression.Wellcome Trus
