    Environmental epigenetics in zebrafish

    It is widely accepted that the epigenome can act as the link between environmental cues, both external and internal, to the organism and phenotype by converting the environmental stimuli to phenotypic responses through changes in gene transcription outcomes. Environmental stress endured by individual organisms can also enforce epigenetic variations in offspring that had never experienced it directly, which is termed transgenerational inheritance. To date, research in the environmental epigenetics discipline has used a wide range of both model and non-model organisms to elucidate the various epigenetic mechanisms underlying the adaptive response to environmental stimuli. In this review, we discuss the advantages of the zebrafish model for studying how environmental toxicant exposures affect the regulation of epigenetic processes, especially DNA methylation, which is the best-studied epigenetic mechanism. We include several very recent studies describing the state-of-the-art knowledge on this topic in zebrafish, together with key concepts in the function of DNA methylation during vertebrate embryogenesis

    Testicular endocrine activity is upregulated by D-Aspartic acid in the green frog Rana esculenta

    This study investigated the involvement of D-aspartic acid (D-Asp) in testicular steroidogenesis of the green frog Rana esculenta and its effect on stimulation of thumb pad morphology and glandular activity, a typical testosterone-dependent secondary sexual characteristic in this amphibian species. In the testis, D-Asp concentrations vary significantly during the reproductive cycle: they are low in pre- and post-reproductive periods, but reach peak levels in the reproductive period (140-236 nmol/g wet tissue). Moreover, the concentrations of D-Asp in the testis through the sexual cycle positively match the testosterone levels in the gonad and the plasma. The racemase activity evaluated during the cycle expresses its peak when D-Asp and testosterone levels are highest, that is, during the reproductive period, confirming the synthesis of D-Asp from L-Asp by an aspartate racemase. Short-term in vivo experiments consisting of a single injection of D-Asp (2.0 micro mol/g body weight) demonstrated that this amino acid accumulates significantly in the testis, and after 3 h its uptake is coupled with a testosterone increase in both testis and plasma. Moreover, within 18 h of amino acid administration, the D-Asp concentration in the testis decreased along with the testosterone titer to prestimulation levels. Other amino acids (L-Asp, D-Glu and L-Glu) used instead of D-Asp were ineffective, confirming that the significant increase in testicular testosterone was a specific feature of this amino acid. In long-term experiments, D-Asp had been administered chronically to frogs caught during the three phases of the reproductive cycle, inducing testosterone increase and 17beta-estradiol decrease in the gonad during the pre- and post-reproductive period, and vice versa during the reproductive period

    Diversification of spatiotemporal expression and copy number variation of the echinoid hbox12/pmar1/micro1 multigene family

    Changes occurring during evolution in the cis-regulatory landscapes of individual members of multigene families might impart diversification in their spatiotemporal expression and function. The archetypal member of the echinoid hbox12/pmar1/micro1 family is hbox12-a, a homeobox-containing gene expressed exclusively by dorsal blastomeres, where it governs the dorsal/ventral gene regulatory network during embryogenesis of the sea urchin Paracentrotus lividus. Here we describe the inventory of the hbox12/pmar1/micro1 genes in P. lividus, highlighting that gene copy number variation occurs across individual sea urchins of the same species. We show that the various hbox12/pmar1/micro1 genes group into three subfamilies according to their spatiotemporal expression, which ranges from broad transcription throughout development to transient expression in either the animal hemisphere or micromeres of the early embryo. Interestingly, the promoter regions of those genes showing comparable expression patterns are highly similar, while differing from those of the other subfamilies. Strikingly, phylogenetic analysis suggests that the hbox12/pmar1/micro1 genes are species-specific, exhibiting extensive divergence in their noncoding, but not in their coding, sequences across three distinct sea urchin species. In spite of this, two micromere-specific genes of P. lividus possess a TCF/LEF-binding motif in a similar position, and their transcription relies on Wnt/ f-catenin signaling, similar to the pmar1 and micro1 genes, which in other sea urchin species are involved in micromere specification. Altogether, our findings suggest that the hbox12/pmar1/micro1 gene family evolved rather rapidly, generating paralogs whose cis-regulatory sequences diverged following multiple rounds of duplication from a common ancestor

    Seasonal Fish Losses through Hauser Dam, Montana, 2007- 2008

    Management of fish populations in Hauser Reservoir, Montana, is hindered by undesirable and unpredictable downstream fish entrainment through Hauser Dam. We quantified fish entrainment through the dam using hydroacoustics at turbine intakes from July 2007 to November 2008 and over the spillway from 21 May to 18 July 2008. Species composition was characterized using multiple netting gears. Total estimated entrainment was 145,470 ± 6,204. Annual entrainment from summer to autumn was higher in 2007 (n = 79,031 ± 4378) than in 2008 (n = 52,513 ± 3,966). Spillway entrainment was 19 percent of annual entrainment in 2008 and was correlated with spillway discharge; turbine entrainment was not. Turbine entrainment increased from summer to autumn in both years, probably in response to autumn turnover and releases of hatchery rainbow trout (Oncorhynchus mykiss). Spill entrainment in 2008 resulted in similar entrainment between summer and autumn, but autumn turbine entrainment increased in 2008. About 60 percent of entrained fish were smaller than 220 mm. We applied species composition by size to the hydroacoustic data to identify fish species entrained, but many fish (n = 55,529 ± 4,397) could not be reliably assigned to species because concurrent net catches did not include individuals of similar size. Total entrainment of identified fish was made up of mostly rainbow trout (Oncorhynchus mykiss; 33.3%) and walleye (Stizostedion vitreum; 30.2%). Identification of patterns in spatial and temporal fish losses affords fishery managers the ability to make more informed decisions about operation of this dam

    Identification of GABA receptor genes and evidence of GABA signaling during embryogenesis of the sea urchin

    Gamma-aminobutyric acid (GABA) is the principal inhibitory neurotransmitter in the mammalian central nervous system and acts via ionotropic (GABAA-Rs) and metabotropic (GABAB-Rs) receptors. GABAA-Rs are Cl- selective hetero-pentameric channels assembled by combinations of 19 distinct gene products. Instead, GABAB-Rs are bi-subunit G-protein coupled receptors linked to K+ or Ca2+ channels. Dysfunctions of GABA-signaling (GS) cause psychotic disorders and correlate with epigenetic alterations, such as over-expression of DNA methyl transferase-1 which in turn imposes iper-methylation of GABA-regulated genes. The sea urchin embryo, which presents a rudimentary nervous system, offers a big opportunity to study the GS and its potential epigenetic implications in a simple eukaryote model. To this purpose, we performed a comprehensive in silico analysis of the sea urchin genome and identified a 450 Kb long cluster containing two genes encoding for the GABAB-R subunits, and two genes respectively encoding for a \u3b1/\u3b3/\u3b5-type and a \u3b2/\u3c1/\u3b4/\u3c4/\u3c0-type GABA B A-R subunit. From an evolutionary perspective, this result revealed a unique genomic organization of these genes in sea urchin. Next, to preliminarily evaluate the role of GS during development, Paracentrotus lividus embryos were cultured in the presence of GABA at concentrations ranging from 0.01 to 1.0 mM. Compared to controls, treated embryos showed aberrations in axial patterning, with a dose-dependent effect. In particular, at 48 hours post-fertilization control embryos were normal bilateral symmetric plutei whereas GABA-treated embryos displayed a radial organization with supranumerary spicules. Washout experiments allowed to determine that the period of sensitivity is restricted from the blastula to the gastrula stage. Altogether, these results suggest that dysregulation of GS affects the polarization of the ectoderm. Although preliminary, this study provide the first evidence of GS activity during development of echinoderms

    Optimization of graphene-based materials outperforming host epoxy matrices

    The degree of graphite exfoliation and edge-carboxylated layers can be controlled and balanced to design lightweight materials characterized by both low electrical percolation thresholds (EPT) and improved mechanical properties. So far, this challenging task has been undoubtedly very hard to achieve. The results presented in this paper highlight the effect of exfoliation degree and the role of edge-carboxylated graphite layers to give self-assembled structures embedded in the polymeric matrix. Graphene layers inside the matrix may serve as building blocks of complex systems that could outperform the host matrix. Improvements in electrical percolation and mechanical performance have been obtained by a synergic effect due to finely balancing the degree of exfoliation and the chemistry of graphene edges which favors the interfacial interaction between polymer and carbon layers. In particular, for epoxy-based resins including two partially exfoliated graphite samples, differing essentially in the content of carboxylated groups, the percolation threshold reduces from 3 wt% down to 0.3 wt%, as the carboxylated group content increases up to 10 wt%. Edge-carboxylated nanosheets also increase the nanofiller/epoxy matrix interaction, determining a relevant reinforcement in the elastic modulus

    On the nature of the near-UV extended light in Seyfert galaxies

    We study the nature of the extended near-UV emission in the inner kiloparsec of a sample of 15 Seyfert galaxies which have both near-UV (F330W) and narrow band [OIII] high resolution Hubble images. For the majority of the objects we find a very similar morphology in both bands. From the [OIII] images we construct synthetic images of the nebular continuum plus the emission line contribution expected through the F330W filter, which can be subtracted from the F330W images. We find that the emission of the ionised gas dominates the near-UV extended emission in half of the objects. A further broad band photometric study, in the bands F330W (U), F547M (V) and F160W (H), shows that the remaining emission is dominated by the underlying galactic bulge contribution. We also find a blue component whose nature is not clear in 4 out of 15 objects. This component may be attributed to scattered light from the AGN, to a young stellar population in unresolved star clusters, or to early-disrupted clusters. Star forming regions and/or bright off-nuclear star clusters are observed in 4/15 galaxies of the sample.Comment: 23 pages, 6 figures, 3 tables; accepted for publication in MNRA

    Nitrogen Experiments on a Supersonic Linear Cascade For ORC Applications

    A novel experiment has been conceived at Politecnico di Milano for the study of the flow within and downstream of supersonic cascades of Organic Rankine Cycle (ORC) turbines. This paper documents the first phase of the research, focused on the preliminary tests and studies performed by operating the facility with nitrogen as working fluid, to demonstrate the technical relevance of the experiment and the validity of the measurement system in a simplified thermodynamic condition. The set of measured data includes, beside the inlet total thermodynamic state, eight static pressure values obtained via taps manufactured on the test section rear end-wall, both within the bladed and semi-bladed region of the cascade, as well as a total pressure probe to retrieve the cascade performance. A double-passage Schlieren equipment was also employed to visualize the density gradients. Experiments show an outstanding repeatability, indicate a quasi -steady cascade operation during the blow-down process for all the pressure signal considered, and demonstrate a remarkable periodicity among two consecutive channels also in off-design conditions. Experimental data were also compared with CFD simulations, resulting in an excellent agreement for the pressure data acquired both within and downstream of the cascade

    Testing the specificity of predictors of reading, spelling and maths: a new model of the association among learning skills based on competence, performance and acquisition

    In a previous study (Zoccolotti et al., 2020) we examined reading, spelling, and maths skills in an unselected group of 129 Italian children attending fifth grade by testing various cognitive predictors; results showed a high degree of predictors’ selectivity for each of these three behaviors. In the present study, we focused on the specificity of the predictors by performing cross-analyses on the same dataset; i.e., we predicted spelling and maths skills based on reading predictors, reading based on maths predictors and so on. Results indicated that some predictors, such as the Orthographic Decision and the Arithmetic Facts tests, predicted reading, spelling and maths skills in similar ways, while others predicted different behaviors but only for a specific parameter, such as fluency but not accuracy (as in the case of RAN), and still others were specific for a single behavior (e.g., Visual-auditory Pseudo-word Matching test predicted only spelling skills). To interpret these results, we propose a novel model of learning skills separately considering factors in terms of competence, performance and acquisition (automatization). Reading, spelling and calculation skills would depend on the development of discrete and different abstract competences (accounting for the partial dissociations among learning disorders reported in the literature). By contrast, overlap among behaviors would be accounted for by defective acquisition in automatized responses to individual “instances”; this latter skill is item specific but domain independent. Finally, performance factors implied in task’s characteristics (such as time pressure) may contribute to the partial association among learning skills. It is proposed that this new model may provide a useful base for interpreting the diffuse presence of comorbidities among learning disorders
