    Interactions of the drug amphotericin B with phospholipid membranes containing or not ergosterol: new insight into the role of ergosterol

    AbstractAmphotericin B (AmB) is an amphipathic polyene antibiotic which permeabilizes ergosterol-containing membranes, supposedly by formation of pores. In water, AmB forms chiral aggregates, modelled as stacks of planar dimers in which the joined polyene chains in each dimer turn round, from one dimer to the following in these stacks, by forming a helical array. Studies of the binding of AmB with L-dipalmitoylphosphatidylcholine (L-DPPC) and L-dilauroylphosphatidylcholine (L-DLPC) bilayers disclose the main following results. (1) An inversion of the helicity of the L-DPPC-bound AmB aggregates, when the L-DPPC bilayers are in the gel phase, is inferred from the evolution of the circular dichroism spectra of AmB+L-DPPC mixtures. (2) An AmB-induced gel-to-subgel transformation of L-DPPC bilayers, in the previous mixtures, is revealed by a differential scanning calorimetry study. (3) The role played by ergosterol in the location of phospholipid-bound AmB aggregates with respect to a phospholipid bilayer is directly demonstrated from atomic force microscopy observations of mica-supported AmB+L-DLPC mixtures, in the presence or absence of ergosterol. While in the absence of ergosterol AmB aggregates remained at the surface of the bilayer, in the presence of ergosterol they appeared embedded within this bilayer and became hollow-centered. As such an embedding in the hydrophobic core of a bilayer requires a rearrangement of the aggregates with respect to their architecture in water, this rearrangement is held responsible for the hollowing of aggregates. The hollow-centered sublayer-embedded AmB aggregates are thought to be the precursors of the formation of AmB pores

The ultimate goal of WP5.1 is to provide guidance on best practice restoration and management to the practitioners in River Basin Management. Therefore, a series of analyses was undertaken, each of which used a part of the WP5.1 database in order to track two major pathways of biological response: 1) the response of riverine biota to environmental pressures (degradation) and 2) the response of biota to the reduction of these impacts (restoration). This report attempts to provide empirical evidence on the environment-biota relationships for both pathways

    The role of emotion regulation in childhood obesity : implications for prevention and treatment

    Stress and negative emotions pose a major threat to public health, by increasing the risk of obesity. Since the management process for emotions (emotion regulation; ER) is developed in childhood, we present a novel conceptual framework model for the role of ER in the prevention and treatment of childhood obesity. A narrative review of the literature by electronic database search (MEDLINE, Web of Knowledge and Scopus) was conducted of observational and interventional/experimental literature on ER and obesity and the underlying concepts. We also present an overview of ER intervention techniques. Our model indicates that childhood ER is a link between stress and obesity. Stress along with ineffective ER leads to abnormal cortisol patterns, emotional eating, sedentary lifestyle, reduction of physical activity, and sleep problems. Simultaneously, a healthy lifestyle could show benefits on ER and in developing adaptive ER strategies. In the development of obesity and ER, parents also play a role. By contrast, effective ER skills decrease obesity-related unhealthy behaviour and enhance protective factors, which boost health. The literature contains some observational studies of children but very few intervention studies, most of which are pilot or on-going studies. In conclusion, encouraging effective ER could be a useful new approach for combating and treating childhood obesity. Future ER intervention studies are needed to confirm the validity of this model in children

    The Influence of Boron (B), Tin (Sn), Copper (Cu), and Manganese (Mn) on the Microstructure of Spheroidal Graphite Irons

    Most spheroidal graphite irons (SGIs) have a matrix consisting of ferrite, pearlite, or a mix of the two. To achieve the desired matrix composition, pearlite promoters such as Mn, Cu, or Sn, are added to the molten metal. Among these elements, Sn is the most potent pearlite promoter. However, each has a different impact on the solidification, graphite precipitation, eutectoid transformation, and ultimately the final structure of the material. Research has shown that B promotes ferrite in fully pearlitic grades where Cu and Mn were used to promote pearlite. The present work investigates the effect of B in SGI with additions of Sn, Cu, and Mn, and the effects of varying amounts of the different pearlite promoters on the matrix composition. The results show that Mn alone at levels of approximately 0.9 wt% is not enough to promote a fully pearlitic matrix, while 0.5 wt% Cu combined with 0.67 wt% Mn is sufficient. Likewise, a fully pearlitic microstructure can be obtained by alloying with 0.06 wt% Sn and 0.67 wt% Mn. B was found to promote ferrite in fully pearlitic SGI alloyed with Sn or Cu. However, in the absence of those elements, B promoted pearlite when alloyed with just Mn. Graphite protrusions were observed on the graphite nodule surface only for B-added alloys with Sn and Cu. In these cases, it is believed B promotes ferrite by changing the growth mechanism of graphite after solidification from spherical to lamellar. However, a different graphite morphology is observed when B is added with just Mn. Thermal analysis data is in agreement with the microstructural observations regarding the ferrite promoting effect of B

    NEW RECORDS FOR \u3ci\u3eACERIA ANTHOCOPTES\u3c/i\u3e (ACARI: ERIOPHYIDAE) OCCURRING ON CANADA THISTLE IN COLORADO, NEBRASKA, AND WYOMING, U.S.A.

    Canada thistle [Cirsium arvense (L.) Scop.] growing in eastern Colorado, Wyoming and western Nebraska were surveyed for the presence and distribution of Aceria anthocoptes (Nal.). Of the 34 sites surveyed in 2004, mites were abundant at 42%, present in lesser numbers at 52%, and not present at 6% of the sites. In 2005, two new sites were added and ten sites sampled in 2004 were revisited. Of these 12 sites, mites were abundant at 17%, present to a lesser extent at 58%, and not present at 25% of the sites. The results demonstrate that Canada thistle growing in this region commonly harbor A. anthocoptes. How long A. anthocoptes has been present in this region is unknown, however, anecdotal evidence demonstrating a dramatic decline in the population of Canada thistle at one Colorado site from 2000 to 2007 suggests that the mite may have been present since 2002
