    Inhibiting CDK4/6 in pancreatic ductal adenocarcinoma via microRNA-21

    Pancreatic ductal adenocarcinoma (PDAC) is one of the most aggressive malignancies, with a 5-year survival rate of 5–10 %. The high mortality rate is due to the asymptomatic progression of clinical features in metastatic stages of the disease, which renders standard therapeutic options futile. PDAC is characterised by alterations in several genes that drive carcinogenesis and limit therapeutic response. The two most common genetic aberrations in PDAC are the mutational activation of KRAS and loss of the tumour suppressor CDK inhibitor 2A (CDKN2A), which culminate the activation of the cyclin-dependent kinase 4 and 6 (CDK4/6), that promote G1 cell cycle progression. Therapeutic strategies focusing on the CDK4/6 inhibitors such as palbociclib (PD-0332991) may potentially improve outcomes in this malignancy. MicroRNAs (miRs/miRNAs) are small endogenous non-coding RNA molecules associated with cellular proliferation, invasion, apoptosis, and cell cycle. Primarily, miR-21 promotes cell proliferation and a higher proportion of PDAC cells in the S phase, while knockdown of miR-21 has been linked to cell cycle arrest at the G2/M phase and inhibition of cell proliferation. In this study, using a CRISPR/Cas9 loss-of-function screen, we individually silenced the expression of miR-21 in two PDAC cell lines and in combination with PD-0332991 treatment, we examined the synergetic mechanisms of CDK4/6 inhibitors and miR-21 knockouts (KOs) on cell survival and death. This combination reduced cell proliferation, cell viability, increased apoptosis and G1 arrest in vitro. We further analysed the mitochondrial respiration and glycolysis of PDAC cells; then assessed the protein content of these cells and revealed numerous Kyoto Encyclopedia of Genes and Genomes (KEGG) pathways associated with PD-0332991 treatment and miR-21 knocking out. Our results demonstrate that combined targeting of CDK4/6 and silencing of miR-21 represents a novel therapeutic strategy in PDAC

    Histopathologic and immunohistochemical investigations of dental abscess formed in maxillofacial area

    Background: An abscess is a pocket of pus that forms around the root of an infected tooth. In this study, we aimed to investigate the extracellular matrix proteases ADAMTS1, ADAMTS4, osteonectin, and osteopontin expressions in abscess fluid cells in jaws after implantation and prosthesis operation. Materials and methods: In this clinical study, abscess fluids belonging to 17 patients who applied to the Department of Oral and Maxillofacial Surgery were examined histopathologically and immunohistochemically. In the histopathological examination of the abscess fluid, separation of chromatin bridges in the nuclei of neutrophil cells, pyknosis and apoptotic changes in the nucleus, degenerative change in the cytoplasm, and occasional vacuolar structures were observed. Results: The positive reaction of ADAMTS1 was observed in fibroblast cells, plasma cells, and macrophage cells. The positive reaction of ADAMTS4 was observed in fibroblast cells, osteoclast cells, and some apoptotic leukocyte cells. Osteopontin expression in osteoclastic cells and polymorphonuclear cells was defined as positive. Osteonectin expression was positive in polymorphonuclear leukocytes and hypertrophic fibroblast cells. Conclusions: ADAMTS1 and ADAMTS4 may induce bone destruction with its distinctive property in alveolar bone resorption, which promotes the activation of osteoclasts, which can accelerate the destruction of the extracellular matrix in the acute phase. Furthermore, osteoclastic activity increased with the increase of osteonectin and osteopontin protein expression due to inflammation in the abscess cases

    Application of Ion Implantation for Synthesis of Copper Nanoparticles in a Zinc Oxide Matrix for Obtaining New Nonlinear Optical Materials

    We have obtained a layered composite material by implantation of single crystal zinc oxide (ZnO) substrates with 160-keV Cu+ ions to a dose of 10(16) or 10(17) cm(-2). The composite was studied by linear optical absorption spectroscopy; the nonlinear optical characteristics were determined by means of Z-scanning at a laser radiation wavelength of 532 nm. The appearance of the optical plasmon resonance bands in the spectra indicated that ion implantation to the higher dose provides for the formation of copper nanoparticles in a subsurface layer of ZnO. The new nonlinear optical material comprising metal nanoparticles in a ZnO matrix exhibits the phenomenon of self-defocusing and possesses a high nonlinear absorption coefficient (beta = 2.07 x 10(-3) cm/W).     Using the Communication in Science Inquiry Project professional development model to facilitate learning middle school genetics concepts

    This study describes the effect of embedding content in the Communication in Inquiry Science Project professional development model for science and language arts teachers. The model uses four components of successful professional development (content focus, active learning, extended duration, participation by teams of teachers from the same school or grade level) and instructional strategies for inquiry, academic language development, written and oral discourse, and learning principles as components of science activities. Teachers were given a pre/ post-institute genetics assessment. There was a statistically significant increase in scores for the entire sample and a statistically significant difference between science and language arts pre and post scores, with science teachers scoring higher in both cases

    Peptidylarginine Deiminase Isozyme-Specific PAD2, PAD3 and PAD4 Inhibitors Differentially Modulate Extracellular Vesicle Signatures and Cell Invasion in Two Glioblastoma Multiforme Cell Lines

    Glioblastoma multiforme (GBM) is an aggressive adult brain tumour with poor prognosis. Roles for peptidylarginine deiminases (PADs) in GBM have recently been highlighted. Here, two GBM cell lines were treated with PAD2, PAD3 and PAD4 isozyme-specific inhibitors. Effects were assessed on extracellular vesicle (EV) signatures, including EV-microRNA cargo (miR21, miR126 and miR210), and on changes in cellular protein expression relevant for mitochondrial housekeeping (prohibitin (PHB)) and cancer progression (stromal interaction molecule 1 (STIM-1) and moesin), as well as assessing cell invasion. Overall, GBM cell-line specific differences for the three PAD isozyme-specific inhibitors were observed on modulation of EV-signatures, PHB, STIM-1 and moesin protein levels, as well as on cell invasion. The PAD3 inhibitor was most effective in modulating EVs to anti-oncogenic signatures (reduced miR21 and miR210, and elevated miR126), to reduce cell invasion and to modulate protein expression of pro-GBM proteins in LN229 cells, while the PAD2 and PAD4 inhibitors were more effective in LN18 cells. Furthermore, Kyoto Encyclopedia of Genes and Genomes (KEGG) pathways for deiminated proteins relating to cancer, metabolism and inflammation differed between the two GBM cell lines. Our findings highlight roles for the different PAD isozymes in the heterogeneity of GBM tumours and the potential for tailored PAD-isozyme specific treatment

    Performance and optimization of network-coded cooperative diversity systems

    In this paper, we study network–coded cooperative diversity (NCCD) systems comprising multiple sources, one relay, and one destination, where the relay detects the packets received from all sources and performs Galois field (GF) network coding over GF(2m) before forwarding a single packet to the destination. Assuming independent Rayleigh fading for all links of the network, we derive simple and accurate closed–formapproximations for the asymptotic symbol and bit error rates of NCCD systems. The derived error rate expressions are valid for arbitrary numbers of sources, arbitrary modulation schemes, and arbitrary constellation mappings and provide significantinsight into the impact of various system and channel parameters on performance.     Tracking the timing of Neotethyan oceanic slab break-off: Geochronology and geochemistry of the quartz diorite porphyries, NE Turkey

    The initiation of the break-off of the northern branch of the Neotethyan oceanic lithosphere is an important but poorly understood event in the geology of the Sakarya Zone (SZ) in northeastern Turkey. Although it is well-known that Latest Cretaceous intrusives (~70 Ma) and early Eocene adakitic magmatic rocks are present in the eastern SZ, the outcrops of the early Eocene non-adakitic rocks are very limited, and their tectono-magmatic evolution has not been studied. We describe a small outcrop of non-adakitic quartz diorite porphyry in the Kov area of the Gümüşhane region in northeastern Turkey. The genesis of these porphyries is significant in evaluating the syn- to post-collision-related magmatism. The LA-ICP-MS zircon U-Pb dating revealed that the Kov quartz diorite porphyries (KQDP) formed at ca. 50 Ma, coeval with adakitic rocks, and ~20 Myr later than the slab roll-back-related intrusive rocks. The KQDPs are calc-alkaline in composition and enriched in large ion lithophile elements (LILEs) and light rare earth elements (LREEs) and depleted in high field strength elements (HFSEs; e.g., Nb, Ta, Ti), with significant negative anomalies of Nb, Ta, and Ti but positive anomalies of Th, U, and Pb. Isotopic compositions of the samples show limited range of variation and slight enrichment of 87Sr/86Sr(t) (0.70489 to 0.70555), εNd(t) (−1.4 to −1.2) with TDM of 1.11 to 1.61 Ga. Pb isotopic ratios of the samples point to an enriched mantle source. They were likely crystallized from the melt that originated from an EM2-type spinel-facies subcontinental lithospheric mantle (SCLM), followed by the fractionation with insignificant crustal assimilation. The SCLM was metasomatically enriched, and the metasomatic agent was likely H2O-rich fluids rather than sediments released from subducting oceanic crust during the Late Cretaceous closure of the Neotethyan oceanic lithosphere. In conjunction with the geological background and previous data, we propose that the generation of the KQDPs resulted from a slab break-off event that caused ascending or infiltration of hot asthenosphere, triggering mantle melting. Such sporadic occurrences of the KQDPs, with coeval adakitic rocks in the SZ, are likely associated with the onset of extensional tectonics due to the earlier stage of slab break-off along the region during the early Eocene period.publishe
