    Thermal degradation of citrus pectin in low-moisture environment - Influence of acidic and alkaline pre-treatment

    Pectin powder is degraded during storage and transport by demethoxylation and depolymerisation. The degradation mechanisms and especially the influence of pre-treatments on the degradation reactions are not completely understood. In this study, commercial citrus pectin was modified by either acidic or alkaline demethoxylation. The modified pectins, as well as the commercial pectin, were thermally degraded during four weeks of storage at 60 °C and 80% relative humidity. Demethoxylation and depolymerisation as well as colour alterations were examined during degradation, and the course of the reactions was monitored. It was found that the type of pre-treatment during modification determined the material properties and, thus, the water uptake of the modified pectin powders. The resulting water availability in the samples was crucial to the extent of demethoxylation and to the type and intensity of depolymerisation since some of these reactions competed for the water in the climate chamber. The pre-treatment also determined the content of neutral sugars and sodium ions of the modified pectins. High contents of these components limited the extent of degradation in different ways.     Skin sensitization in silico protocol

    The assessment of skin sensitization has evolved over the past few years to include in vitro assessments of key events along the adverse outcome pathway and opportunistically capitalize on the strengths of in silico methods to support a weight of evidence assessment without conducting a test in animals. While in silico methods vary greatly in their purpose and format; there is a need to standardize the underlying principles on which such models are developed and to make transparent the implications for the uncertainty in the overall assessment. In this contribution, the relationship of skin sensitization relevant effects, mechanisms, and endpoints are built into a hazard assessment framework. Based on the relevance of the mechanisms and effects as well as the strengths and limitations of the experimental systems used to identify them, rules and principles are defined for deriving skin sensitization in silico assessments. Further, the assignments of reliability and confidence scores that reflect the overall strength of the assessment are discussed. This skin sensitization protocol supports the implementation and acceptance of in silico approaches for the prediction of skin sensitization

Szukając odpowiedzi na pytanie, jaki kierunek rozwoju jest właściwy, trzeba uwzględnić różnorodność rzeczywistości miasta i konieczność podejścia kompleksowego. Założeniem jest to, że miasto jest wspólnotą polityczną, społeczną i gospodarczą, która powinna być przyjazna dla wszystkich użytkowników. Zgodnie z nim przedstawiono pokrótce koncepcje miasta kreatywnego, europejskiego i uczącego się oraz zbieżne z nimi założenia local governance i rewitalizacji. Na przykładzie miasta Bytomia podkreślono, że zawężanie się tylko do koncepcji miasta kreatywnego, bez horyzontalnego wykorzystania wszystkich czynników, może negatywnie wpłynąć na rozwój miasta.The article focuses on the problems of the city’s development. The question, which direction of development is appopriate, we need consider the diversity of the reality of the city and comprehensive approach to city problems and opportunities. The assumption is that the city is political, social and economic community and should be beneficial for all local actors. According to it, Authors present concepts: creative city, european city, learner city, revitalization and local governance. The article also describes the city of Bytom, emphasised that using only creative city concept, with non of all of the above, could adversely affect the development of the city

    The borderline range of toxicological methods : Quantification and implications for evaluating precision

    Test methods to assess the skin sensitization potential of a substance usually use threshold criteria to dichotomize continuous experimental read-outs into yes/no conclusions. The threshold criteria are prescribed in the respective OECD test guidelines and the conclusion is used for regulatory hazard assessment, i.e., classification and labelling of the substance. We can identify a borderline range (BR) around the classification threshold within which test results are inconclusive due to a test method's biological and technical variability. We quantified BRs in the prediction models of the non-animal test methods DPRA, LuSens and h-CLAT, and of the animal test LLNA, respectively. Depending on the size of the BR, we found that between 6% and 28% of the substances in the sets tested with these methods were considered borderline. When the results of individual non-animal test methods were combined into integrated testing strategies (ITS), borderline test results of individual tests also affected the overall assessment of the skin sensitization potential of the testing strategy. This was analyzed for the 2-out-of-3 ITS: Four out of 40 substances (10%) were considered borderline. Based on our findings we propose expanding the standard binary classification of substances into "positive"/"negative" or "hazardous"/"non-hazardous" by adding a "borderline" or "inconclusive" alert for cases where test results fall within the borderline range
