    Identifizierung der zytosolischen Kinase CSK als Bindungspartner von VE-Cadherin:Auswirkung dieser Assoziation auf das Zellwachstum

    Das endothelspezifische AdhĂ€sionsprotein VE-Cadherin und die damit assoziierten Proteine sind an der Bildung und der Regulation endothelialer Zell-Zellkontakte beteiligt. Mit Hilfe des "yeast two hybrid"-Systems konnte die "C-terminal-Src-Kinase"(CSK) als zytosolischer Bindungspartner von VE-Cadherin identifiziert werden. CSK ist ein negativer Regulator der Src-Kinasen und wird ĂŒber die BindungsdomĂ€nen an die Orte der Src-AktivitĂ€ten rekrutiert. Die Assoziation mit VE-Cadherin erfolgt ĂŒber die CSK-SH2-DomĂ€ne und erfordert die Phosphorylierung des VE-Cadherin-Tyrosins 685. FĂŒr eine erhöhte Cadherin-Phosphorylierung ist die Bindung der isolierten SH2-DomĂ€ne ausreichend, eine aktive Phosphorylierung des Cadherins durch die Kinase CSK findet nicht statt. In Zellstudien mit CHO-Zellen, die induzierbares CSK zusammen mit VE-Cadherin(wt) oder VE-Cadherin(Y685F) exprimieren, konnte nachgewiesen werden, dass die Assoziation der beiden Proteine die Zellproliferation signifikant reduziert

    Compact Frontend-Electronics and Bidirectional 3.3 Gbps Optical Datalink for Fast Proportional Chamber Readout

    The 9600 channels of the multi-wire proportional chamber of the H1 experiment at HERA have to be read out within 96 ns and made available to the trigger system. The tight spatial conditions at the rear end flange require a compact bidirectional readout electronics with minimal power consumption and dead material. A solution using 40 identical optical link modules, each transferring the trigger information with a physical rate of 4 x 832 Mbps via optical fibers, has been developed and commisioned. The analog pulses from the chamber can be monitored and the synchronization to the global HERA clock signal is ensured.Comment: 13 pages, 10 figure

    PANIC: the new panoramic NIR camera for Calar Alto

    PANIC is a wide-field NIR camera, which is currently under development for the Calar Alto observatory (CAHA) in Spain. It uses a mosaic of four Hawaii-2RG detectors and covers the spectral range from 0.8-2.5 micron(z to K-band). The field-of-view is 30x30 arcmin. This instrument can be used at the 2.2m telescope (0.45arcsec/pixel, 0.5x0.5 degree FOV) and at the 3.5m telescope (0.23arcsec/pixel, 0.25x0.25 degree FOV). The operating temperature is about 77K, achieved by liquid Nitrogen cooling. The cryogenic optics has three flat folding mirrors with diameters up to 282 mm and nine lenses with diameters between 130 mm and 255 mm. A compact filter unit can carry up to 19 filters distributed over four filter wheels. Narrow band (1%) filters can be used. The instrument has a diameter of 1.1 m and it is about 1 m long. The weight limit of 400 kg at the 2.2m telescope requires a light-weight cryostat design.     Personal identity (de)formation among lifestyle travellers: A double-edged sword?

    This article explores the personal identity work of lifestyle travellers – individuals for whom extended leisure travel is a preferred lifestyle that they return to repeatedly. Qualitative findings from in-depth semi-structured interviews with lifestyle travellers in northern India and southern Thailand are interpreted in light of theories on identity formation in late modernity that position identity as problematic. It is suggested that extended leisure travel can provide exposure to varied cultural praxes that may contribute to a sense of social saturation. Whilst a minority of the respondents embraced a saturation of personal identity in the subjective formation of a cosmopolitan cultural identity, several of the respondents were paradoxically left with more identity questions than answers as the result of their travels

    Enhanced Radiation Hardness and Faster Front Ends for the Beetle Readout Chip

    This paper summarizes the recent progress in the development of the 128 channel pipelined readout chip Beetle, which is intended for the silicon vertex detector, the inner tracker, the pile-up veto trigger and the RICH detectors of LHCb. Deficiencies found in the front end of the Beetle Version 1.0 and 1.1 chips resulted in the submissions of BeetleFE 1.1 and BeetleFE 1.2, while BeetleSR 1.0 implements test circuits to provide future Beetle chips with logic circuits hardened against single event upset (SEU). Section I. motivates the development of new front ends for the Beetle chip, and section II. summarizes their concepts and construction. Section III. reports preliminary results from the BeetleFE 1.1 and BeetleFE 1.2 chips, while section IV. describes the BeetleSR 1.0 chip. An outlook on future test and development of the Beetle chip is given in section V

    Trans synaptic assemblies link synaptic vesicles and neuroreceptors

    Synaptic transmission is characterized by fast, tightly coupled processes and complex signaling pathways that require a precise protein organization, such as the previously reported nanodomain colocalization of pre and postsynaptic proteins. Here, we used cryo electron tomography to visualize synaptic complexes together with their native environment comprising interacting proteins and lipids on a 2 to 4 nm scale. Using template free detection and classification, we showed that tripartite trans synaptic assemblies subcolumns link synaptic vesicles to postsynaptic receptors and established that a particular displacement between directly interacting complexes characterizes subcolumns. Furthermore, we obtained de novo average structures of ionotropic glutamate receptors in their physiological composition, embedded in plasma membrane. These data support the hypothesis that synaptic function is carried by precisely organized trans synaptic units. It provides a framework for further exploration of synaptic and other large molecular assemblies that link different cells or cellular regions and may require weak or transient interactions to exert their functio

    GRAVITY: getting to the event horizon of Sgr A*

    We present the second-generation VLTI instrument GRAVITY, which currently is in the preliminary design phase. GRAVITY is specifically designed to observe highly relativistic motions of matter close to the event horizon of Sgr A*, the massive black hole at center of the Milky Way. We have identified the key design features needed to achieve this goal and present the resulting instrument concept. It includes an integrated optics, 4-telescope, dual feed beam combiner operated in a cryogenic vessel; near infrared wavefront sensing adaptive optics; fringe tracking on secondary sources within the field of view of the VLTI and a novel metrology concept. Simulations show that the planned design matches the scientific needs; in particular that 10 microarcsecond astrometry is feasible for a source with a magnitude of K=15 like Sgr A*, given the availability of suitable phase reference sources.Comment: 13 pages, 11 figures, to appear in the conference proceedings of SPIE Astronomical Instrumentation, 23-28 June 2008, Marseille, Franc
