Manifestations of magnetic vortices in equation of state of Yang-Mills plasma
The vacuum of Yang-Mills theory contains singular stringlike objects
identified with center (magnetic) vortices. Percolation of magnetic vortices is
known to be responsible for the color confinement in the low-temperature phase
of the theory. In our work we study properties of the vortices at finite
temperature using lattice simulations of SU(2) gauge theory. We show that
magnetic vortices provide a numerically large contribution to thermodynamic
quantities of the gluon plasma in Yang-Mills theory. In particular, we observe
that in the deconfinement phase at temperatures T_c < T < 3 T_c the magnetic
component of the gluon plasma produces a negative (ghostlike) contribution to
the anomaly of the energy-momentum tensor. In the confinement phase the vortex
contribution is positive. The thermodynamical significance of the magnetic
objects allows us to suggest that the quark-gluon plasma may contain a
developed network of magnetic flux tubes. The existence of the vortex network
may lead to observable effects in the quark-gluon plasma because the
chromomagnetic field of the vortices should scatter and drag quarks.Comment: 16 pages, 14 figures, RevTeX 4.0; revision: minor changes, references

Random Numbers Certified by Bell's Theorem
Randomness is a fundamental feature in nature and a valuable resource for
applications ranging from cryptography and gambling to numerical simulation of
physical and biological systems. Random numbers, however, are difficult to
characterize mathematically, and their generation must rely on an unpredictable
physical process. Inaccuracies in the theoretical modelling of such processes
or failures of the devices, possibly due to adversarial attacks, limit the
reliability of random number generators in ways that are difficult to control
and detect. Here, inspired by earlier work on nonlocality based and device
independent quantum information processing, we show that the nonlocal
correlations of entangled quantum particles can be used to certify the presence
of genuine randomness. It is thereby possible to design of a new type of
cryptographically secure random number generator which does not require any
assumption on the internal working of the devices. This strong form of
randomness generation is impossible classically and possible in quantum systems
only if certified by a Bell inequality violation. We carry out a
proof-of-concept demonstration of this proposal in a system of two entangled
atoms separated by approximately 1 meter. The observed Bell inequality
violation, featuring near-perfect detection efficiency, guarantees that 42 new
random numbers are generated with 99% confidence. Our results lay the
groundwork for future device-independent quantum information experiments and
for addressing fundamental issues raised by the intrinsic randomness of quantum
theory.Comment: 10 pages, 3 figures, 16 page appendix. Version as close as possible

Non-parametric inference of impurity transport coefficients in the ASDEX Upgrade tokamak
We present a non-parametric inference of impurity transport coefficients by using charge exchange recombination spectroscopy measurements of Ne X, Ne VIII, O VIII, and C VI lines. Due to their close atomic numbers, neon, oxygen and carbon impurity ions are assumed to have the same diffusion coefficient D and convection velocity v. Unlike conventional techniques that modulate or perturb the impurity contents, we employ a quasi-stationary plasma with static impurity profiles. Since the ratio of v to D only describes the equilibrated profile of the sum of all impurity charge states, steady-state measurements can still decouple D and v if different charge states are simultaneously observed. We have formulated a non-parametric analysis framework based on the Bayesian probability theory and conducted transport coefficient measurements for a Type III ELMy H-mode plasma at ASDEX Upgrade. The charge exchange reactions with the background neutrals, which are known to affect the impurity charge state balance, are taken into account by introducing additional free parameters. While D at the pedestal is close to the neoclassical level ( < 1 m s-2), a large diffusion coefficient and a strong outward convection are inferred right inside the pedestal top. This work has been carried out within the framework of the EUROfusion Consortium and has received funding from the Euratom research and training programme 2014–2018 and 2019–2020 under Grant Agreement No. 633053
Electronic structure of the MO oxides (M=Mg, Ca, Ti, V) in the GW approximation
The quasiparticle band structures of nonmagnetic monoxides, MO (M=Mg, Ca, Ti,
and V), are calculated by the GW approximation. The band gap and the width of
occupied oxygen 2p states in insulating MgO and CaO agree with experimental
observation. In metallic TiO and VO, conduction bands originated from metal 3d
states become narrower. Then the partial densities of transition metal e_g and
t_2g states show an enhanced dip between the two. The effects of static
screening and dynamical correlation are discussed in detail in comparison with
the results of the Hartree-Fock approximation and the static Coulomb hole plus
screened exchange approximation. The d-d Coulomb interaction is shown to be
very much reduced by on-site and off-site d-electron screening in TiO and VO.
The dielectric function and the energy loss spectrum are also presented and
discussed in detail.Comment: 10 pages, 5 figure
Identification of a novel biomarker candidate, a 4.8-kDa peptide fragment from a neurosecretory protein VGF precursor, by proteomic analysis of cerebrospinal fluid from children with acute encephalopathy using SELDI-TOF-MS

Abstract

Background

Acute encephalopathy includes rapid deterioration and has a poor prognosis. Early intervention is essential to prevent progression of the disease and subsequent neurologic complications. However, in the acute period, true encephalopathy cannot easily be differentiated from febrile seizures, especially febrile seizures of the complex type. Thus, an early diagnostic marker has been sought in order to enable early intervention. The purpose of this study was to identify a novel marker candidate protein differentially expressed in the cerebrospinal fluid (CSF) of children with encephalopathy using proteomic analysis.

Methods

For detection of biomarkers, CSF samples were obtained from 13 children with acute encephalopathy and 42 children with febrile seizure. Mass spectral data were generated by surface-enhanced laser desorption/ionization time-of-flight mass spectrometry (SELDI-TOF MS) technology, which is currently applied in many fields of biological and medical sciences. Diagnosis was made by at least two pediatric neurologists based on the clinical findings and routine examinations. All specimens were collected for diagnostic tests and the remaining portion of the specimens were used for the SELDI-TOF MS investigations.

Results

In experiment 1, CSF from patients with febrile seizures (n = 28), patients with encephalopathy (n = 8) (including influenza encephalopathy (n = 3), encephalopathy due to rotavirus (n = 1), human herpes virus 6 (n = 1)) were used for the SELDI analysis. In experiment 2, SELDI analysis was performed on CSF from a second set of febrile seizure patients (n = 14) and encephalopathy patients (n = 5). We found that the peak with an m/z of 4810 contributed the most to the separation of the two groups. After purification and identification of the 4.8-kDa protein, a 4.8-kDa proteolytic peptide fragment from the neurosecretory protein VGF precursor (VGF4.8) was identified as a novel biomarker for encephalopathy.

Conclusions

Expression of VGF4.8 has been reported to be decreased in pathologically degenerative changes such as Alzheimer's disease, amyotrophic lateral sclerosis (ALS), frontotemporal dementia, and encephalopathy. Thus, the VGF4.8 peptide might be a novel marker for degenerative brain conditions.</p
A practical device for pinpoint delivery of molecules into multiple neurons in culture
We have developed a device for pinpoint delivery of chemicals, proteins, and nucleic acids into cultured cells. The principle underlying the technique is the flow of molecules from the culture medium into cells through a rupture in the plasma membrane made by a needle puncture. DNA transfection is achieved by stabbing the needle tip into the nucleus. The CellBee device can be attached to any inverted microscope, and molecular delivery can be coupled with conventional live cell imaging. Because the position of the needle relative to the targeted cultured cells is computer-controlled, efficient delivery of molecules such as rhodamine into as many as 100 HeLa cells can be completed in 10 min. Moreover, specific target cells within a single dish can be transfected with multiple DNA constructs by simple changes of culture medium containing different plasmids. In addition, the nano-sized needle tip enables gentle molecular delivery, minimizing cell damage. This method permits DNA transfection into specific hippocampal neurons without disturbing neuronal circuitry established in culture
Boron site preference in ternary Ta and Nb boron suicides
X-ray single crystal (XSC) and neutron powder diffraction data (NPD) were used to elucidate boron site preference for five ternary phases. Ta3Si1-xBx (x=0.112(4)) crystallizes with the Ti3P-type (space group P4(2)/n) with B-atoms sharing the 8g site with Si atoms. Ta5Si3-x (x=0.03(1); Cr5B3- type) crystallizes with space group 14/mcm, exhibiting a small amount of vacancies on the 4 alpha site. Both, Ta-5(Si1-xBx)(3), X=0.568(3), and Nb-5(Si1-xBx)(3), x=0.59(2), are part of solid solutions of M5Si3 with Cr5B3-type into the ternary M-Si-B systems (M=Nb or Ta) with B replacing Si on the 8h site. The D8(8)-phase in the Nb-Si-B system crystallizes with the Ti5Ga4-type revealing the formula Nb5Si3B1-x (x=0.292(3)) with B partially filling the voids in the 2b site of the Mn5Si3 parent type. Strategic and practical guidelines for successful structured illumination microscopy
Linear 2D- or 3D-structured illumination microscopy (SIM or3D-SIM, respectively) enables multicolor volumetric imaging of fixed and live specimens with subdiffraction resolution in all spatial dimensions. However, the reliance of SIM on algorithmic post-processing renders it particularly sensitive to artifacts that may reduce resolution, compromise data and its interpretations, and drain resources in terms of money and time spent. Here we present a protocol that allows users to generate high-quality SIM data while accounting and correcting for common artifacts. The protocol details preparation of calibration bead slides designed for SIM-based experiments, the acquisition of calibration data, the documentation of typically encountered SIM artifacts and corrective measures that should be taken to reduce them. It also includes a conceptual overview and checklist for experimental design and calibration decisions, and is applicable to any commercially available or custom platform. This protocol, plus accompanying guidelines, allows researchers from students to imaging professionals to create an optimal SIM imaging environment regardless of specimen type or structure of interest. The calibration sample preparation and system calibration protocol can be executed within 1-2 d
Human Microglia Transplanted in Rat Focal Ischemia Brain Induce Neuroprotection and Behavioral Improvement
BACKGROUND AND PURPOSE: Microglia are resident immunocompetent and phagocytic cells of central nervous system (CNS), which produce various cytokines and growth factors in response to injury and thereby regulate disease pathology. The purpose of this study is to investigate the effects of microglial transplantation on focal cerebral ischemia model in rat. METHODS: Transient middle cerebral artery occlusion (MCAO) in rats was induced by the intraluminal filament technique. HMO6 cells, human microglial cell line, were transplanted intravenously at 48 hours after MCAO. Functional tests were performed and the infarct volume was measured at 7 and 14 days after MCAO. Migration and cell survival of transplanted microglial cells and host glial reaction in the brain were studied by immunohistochemistry. Gene expression of neurotrophic factors, cytokines and chemokines in transplanted cells and host rat glial cells was determined by laser capture microdissection (LCM) and quantitative real time-PCR. RESULTS: HMO6 human microglial cells transplantation group demonstrated significant functional recovery compared with control group. At 7 and 14 days after MCAO, infarct volume was significantly reduced in the HMO group. In the HMO6 group, number of apoptotic cells was time-dependently reduced in the infarct core and penumbra. In addition, number of host rat microglia/macrophages and reactive astrocytes was significantly decreased at 7 and 14 days after MCAO in the penumbra. Gene expression of various neurotrophic factors (GDNF, BDNF, VEGF and BMP7) and anti-inflammatory cytokines (IL4 and IL5) was up-regulated in transplanted HMO6 cells of brain tissue compared with those in culture. The expression of GDNF and VEGF in astrocytes in penumbra was significantly up-regulated in the HMO6 group. CONCLUSIONS: Our results indicate that transplantation of HMO6 human microglial cells reduces ischemic deficits and apoptotic events in stroke animals. The results were mediated by modulation of gliosis and neuroinflammation, and neuroprotection provided by neurotrophic factors of endogenous and transplanted cells-origin
