    Recommandations pour les surfaces tactiles au sol pour personnes aveugles ou malvoyantes : rapport intermédiaire

    Le décret 99-756 sur l’accessibilité de la voirie aux personnes handicapées impose les bandes d’éveil de vigilance aux abaissés de trottoir. La norme existante NF P98-351 est appliquée alors que les aménagements de trottoirs et de voirie ont évolué. Les ingénieurs des villes et les représentants d’associations ont exprimé le besoin de recommandations, en parallèle de la révision de la norme. Les points prioritaires sont la pose dans les trottoirs en arrondi, sur les îlots de chaussée, le long des quais de tramways, le choix de surfaces pour les points d’arrêt de bus et pour l’aide à l’orientation ou à l’information.Une étape ayant été atteinte, le présent rapport intermédiaire présente les recommandations pour l’implantation des bandes d’éveil de vigilance, dont le relief est défini par la norme P98-351 pour alerter sur les situations de danger. Certaines de ces préconisations serviront à la révision de la norme qui sera engagée par le BNSR pour l’AFNOR début 2003. Les services techniques des villes pourront ainsi disposer d’éléments de réponse aux questions qu’ils se posent, dans le but d’améliorer la sécurité des déplacements piétons déficients visuels. Les recommandations concernent plus particulièrement l’implantation sur les abaissés de trottoirs en arrondi, au droit de traversées surélevées, sur les quais de transports guidés tels que tramways, sur les îlots-refuges et en haut des escaliers.Le travail de réflexion se poursuit sur l’aide à l’orientation et l’identification des points d’arrêt de bus,entre autres. Il permettra d’élaborer un guide de recommandations, en fin d’année 2003

    ESKIMO1 is a key gene involved in water economy as well as cold acclimation and salt tolerance

    <p>Abstract</p> <p>Background</p> <p>Drought is a major social and economic problem resulting in huge yield reduction in the field. Today's challenge is to develop plants with reduced water requirements and stable yields in fluctuating environmental conditions. <it>Arabidopsis thaliana </it>is an excellent model for identifying potential targets for plant breeding. Drought tolerance in the field was successfully conferred to crops by transferring genes from this model species. While involved in a plant genomics programme, which aims to identify new genes responsible for plant response to abiotic stress, we identified <it>ESKIMO1 </it>as a key gene involved in plant water economy as well as cold acclimation and salt tolerance.</p> <p>Results</p> <p>All <it>esk1 </it>mutants were more tolerant to freezing, after acclimation, than their wild type counterpart. <it>esk1 </it>mutants also showed increased tolerance to mild water deficit for all traits measured. The mutant's improved tolerance to reduced water supply may be explained by its lower transpiration rate and better water use efficiency (WUE), which was assessed by carbon isotope discrimination and gas exchange measurements. <it>esk1 </it>alleles were also shown to be more tolerant to salt stress.</p> <p>Transcriptomic analysis of one mutant line and its wild-type background was carried out. Under control watering conditions a number of genes were differentially expressed between the mutant and the wild type whereas under mild drought stress this list of genes was reduced. Among the genes that were differentially expressed between the wild type and mutant, two functional categories related to the response to stress or biotic and abiotic stimulus were over-represented. Under salt stress conditions, all gene functional categories were represented equally in both the mutant and wild type. Based on this transcriptome analysis we hypothesise that in control conditions the <it>esk1 </it>mutant behaves as if it was exposed to drought stress.</p> <p>Conclusion</p> <p>Overall our findings suggest that the <it>ESKIMO1 </it>gene plays a major role in plant response to water shortage and in whole plant water economy. Further experiments are being undertaken to elucidate the function of the ESKIMO1 protein and the way it modulates plant water uptake.</p

    A New Role for SAG12 Cysteine Protease in Roots of Arabidopsis thaliana

    Senescence associated gene (SAG) 12, which encodes a cysteine protease is considered to be important in nitrogen (N) allocation to Arabidopsis thaliana seeds. A decrease in the yield and N content of the seeds was observed in the Arabidopsis SAG12 knockout mutants (sag12) relative to the wild type (Col0) under limited nitrogen nutrition. However, leaf senescence was similar in both lines. To test whether SAG12 is involved in N remobilization from organs other than the leaves, we tested whether root N could be used in N mobilization to the seeds. Root architecture, N uptake capacity and 15N partitioning were compared in the wild type and sag12 under either high nitrogen (HN) or low nitrogen (LN) conditions. No differences in root architecture or root N uptake capacity were observed between the lines under HN or LN. However, under LN conditions, there was an accumulation of 15N in the sag12 roots compared to the wild type with lower allocation of 15N to the seeds. This was accompanied by an increase in root N protein contents and a significant decrease in root cysteine protease activity. SAG12 is expressed in the root stele of the plants at the reproductive stage, particularly under conditions of LN nutrition. Taken together, these results suggest a new role for SAG12. This cysteine protease plays a crucial role in root N remobilization that ensures seed filling and sustains yields when nitrogen availability is low

    Plants rely on “reserve” (stored) carbon (C) for growth and survival when newly synthesized C becomes limited. Besides a classic yet recalcitrant C reserve starch, fructans, a class of sucrose-derived soluble fructosyl-oligosaccharides, represent a major store of C in many temperate plant species including the economically important Asteraceae and Poaceae families (Hendry, 1993). Dicots typically accumulate inulin-type fructans as long-term storage (underground organs) whilst grasses and cereals accumulate fructans as short-term reserves in above-ground parts (Pollock and Cairns, 1991; Van Laere and Van den Ende, 2002). Unlike chloroplast-based water-insoluble starch, fructans are semi-soluble, possess flexible structures (Phelps, 1965; Valluru and Van den Ende, 2008), can be synthesized at low temperatures (Pollock and Cairns, 1991), and are degraded by a single type of fructan hydrolases, fructan exohydrolases (FEHs). Unlike starch that store in plastids, fructans store in vacuoles, which is physically less stressful to the active constituents of, and allows more C synthesis by, the photosynthetic cell, which may be different in dicots where fructans do not typically accumulate in green parts

    ESKIMO1 Disruption in Arabidopsis Alters Vascular Tissue and Impairs Water Transport

    Water economy in agricultural practices is an issue that is being addressed through studies aimed at understanding both plant water-use efficiency (WUE), i.e. biomass produced per water consumed, and responses to water shortage. In the model species Arabidopsis thaliana, the ESKIMO1 (ESK1) gene has been described as involved in freezing, cold and salt tolerance as well as in water economy: esk1 mutants have very low evapo-transpiration rates and high water-use efficiency. In order to establish ESK1 function, detailed characterization of esk1 mutants has been carried out. The stress hormone ABA (abscisic acid) was present at high levels in esk1 compared to wild type, nevertheless, the weak water loss of esk1 was independent of stomata closure through ABA biosynthesis, as combining mutant in this pathway with esk1 led to additive phenotypes. Measurement of root hydraulic conductivity suggests that the esk1 vegetative apparatus suffers water deficit due to a defect in water transport. ESK1 promoter-driven reporter gene expression was observed in xylem and fibers, the vascular tissue responsible for the transport of water and mineral nutrients from the soil to the shoots, via the roots. Moreover, in cross sections of hypocotyls, roots and stems, esk1 xylem vessels were collapsed. Finally, using Fourier-Transform Infrared (FTIR) spectroscopy, severe chemical modifications of xylem cell wall composition were highlighted in the esk1 mutants. Taken together our findings show that ESK1 is necessary for the production of functional xylem vessels, through its implication in the laying down of secondary cell wall components

    Interaction between sugar and abscisic acid signalling during early seedling development in Arabidopsis

    Sugars regulate important processes and affect the expression of many genes in plants. Characterization of Arabidopsis (Arabidopsis thaliana) mutants with altered sugar sensitivity revealed the function of abscisic acid (ABA) signalling in sugar responses. However, the exact interaction between sugar signalling and ABA is obscure. Therefore ABA deficient plants with constitutive ABI4 expression (aba2-1/35S::ABI4) were generated. Enhanced ABI4 expression did not rescue the glucose insensitive (gin) phenotype of aba2 seedlings indicating that other ABA regulated factors are essential as well. Interestingly, both glucose and ABA treatment of Arabidopsis seeds trigger a post-germination seedling developmental arrest. The glucose-arrested seedlings had a drought tolerant phenotype and showed glucose-induced expression of ABSCISIC ACID INSENSITIVE3 (ABI3), ABI5 and LATE EMBRYOGENESIS ABUNDANT (LEA) genes reminiscent of ABA signalling during early seedling development. ABI3 is a key regulator of the ABA-induced arrest and it is shown here that ABI3 functions in glucose signalling as well. Multiple abi3 alleles have a glucose insensitive (gin) phenotype comparable to that of other known gin mutants. Importantly, glucose-regulated gene expression is disturbed in the abi3 background. Moreover, abi3 was insensitive to sugars during germination and showed sugar insensitive (sis) and sucrose uncoupled (sun) phenotypes. Mutant analysis further identified the ABA response pathway genes ENHANCED RESPONSE TO ABA1 (ERA1) and ABI2 as intermediates in glucose signalling. Hence, three previously unidentified sugar signalling genes have been identified, showing that ABA and glucose signalling overlap to a larger extend than originally thought

    Crosstalks between Myo-Inositol Metabolism, Programmed Cell Death and Basal Immunity in Arabidopsis

    BACKGROUND: Although it is a crucial cellular process required for both normal development and to face stress conditions, the control of programmed cell death in plants is not fully understood. We previously reported the isolation of ATXR5 and ATXR6, two PCNA-binding proteins that could be involved in the regulation of cell cycle or cell death. A yeast two-hybrid screen using ATXR5 as bait captured AtIPS1, an enzyme which catalyses the committed step of myo-inositol (MI) biosynthesis. atips1 mutants form spontaneous lesions on leaves, raising the possibility that MI metabolism may play a role in the control of PCD in plants. In this work, we have characterised atips1 mutants to gain insight regarding the role of MI in PCD regulation. METHODOLOGY/PRINCIPAL FINDINGS: - lesion formation in atips1 mutants depends of light intensity, is due to PCD as evidenced by TUNEL labelling of nuclei, and is regulated by phytohormones such as salicylic acid - MI and galactinol are the only metabolites whose accumulation is significantly reduced in the mutant, and supplementation of the mutant with these compounds is sufficient to prevent PCD - the transcriptome profile of the mutant is extremely similar to that of lesion mimic mutants such as cpr5, or wild-type plants infected with pathogens. CONCLUSION/SIGNIFICANCE: Taken together, our results provide strong evidence for the role of MI or MI derivatives in the regulation of PCD. Interestingly, there are three isoforms of IPS in Arabidopsis, but AtIPS1 is the only one harbouring a nuclear localisation sequence, suggesting that nuclear pools of MI may play a specific role in PCD regulation and opening new research prospects regarding the role of MI in the prevention of tumorigenesis. Nevertheless, the significance of the interaction between AtIPS1 and ATXR5 remains to be established
