Detecting Imprinting and Maternal Effects Using Monte Carlo Expectation Maximization Algorithm
Numerous statistical methods have been developed to explore genomic
imprinting and maternal effects, which are causes of parent-of-origin patterns
in complex human diseases. However, most of them either only model one of these
two confounded epigenetic effects, or make strong yet unrealistic assumptions
about the population to avoid over-parameterization. A recent partial
likelihood method (LIME) can identify both epigenetic effects based on
case-control family data without those assumptions. Theoretical and empirical
studies have shown its validity and robustness. However, because LIME obtains
parameter estimation by maximizing partial likelihood, it is interesting to
compare its efficiency with full likelihood maximizer. To overcome the
difficulty in over-parameterization when using full likelihood, in this study
we propose a Monte Carlo Expectation Maximization (MCEM) method to detect
imprinting and maternal effects jointly. Those unknown mating type
probabilities, the nuisance parameters, can be considered as latent variables
in EM algorithm. Monte Carlo samples are used to numerically approximate the
expectation function that cannot be solved algebraically. Our simulation
results show that though this MCEM algorithm takes longer computational time,
and can give higher bias in some simulations compared to LIME, it can generally
detect both epigenetic effects with higher power and smaller standard error
which demonstrates that it can be a good complement of LIME method
Aplicação de medidas ambientais na Academia Militar-Sede: impacto económico
As Forças Armadas no geral, e o Exército em particular, têm procurado agir em conformidade com a política ambiental do Governo, cooperando em prol da preservação do ambiente e de um desenvolvimento sustentável. Neste âmbito, surge a tentativa de identificar qual a realidade ambiental existente na Academia Militar-Sede, instituição dedicada à formação dos futuros oficiais do Exército e da Guarda Nacional Republicana.
Deste modo, a investigação tem como objectivo avaliar o impacto económico consequente à adopção de determinadas medidas ambientais, de acordo com a realidade envolvente. O presente trabalho foi dividido em duas partes distintas: a primeira referindo-se ao enquadramento teórico e a segunda contendo os resultados obtidos através do trabalho de campo. Relativamente à primeira fase, foi realizada uma recolha de dados para a prossecução dos objectivos anteriormente referidos, que se consubstanciou essencialmente, numa análise documental decorrente de uma exaustiva pesquisa bibliográfica. Numa segunda fase, elaboraram-se entrevistas e inquéritos, e analisaram-se as propostas referentes à implementação de medidas de protecção ambiental, com o objectivo de avaliar a sua viabilidade económica.
De acordo com a análise dos dados recolhidos pelos supracitados métodos de investigação, concluiu-se que há actualmente, um fraco desempenho ambiental por parte da Academia Militar-Sede, razão pela qual se considera urgente a criação de uma política que defina claramente orientações no sentido de reverter a situação actual. Estas deverão incidir num consumo de energia e de água mais eficientes, na separação correcta de resíduos, nascompras públicas ambientalmente orientadas e na formação e sensibilização dos indivíduos que diariamente, frequentam o espaço da Academia Militar-Sede. No que respeita ao impacto económico subjacente a estas medidas, comprovou-se que elas estão intimamente ligadas à obtenção de novas fontes de receitas, devendo pois, ser encaradas como prioridades a definir.Abstract In general military forces, particularly the army, have sought to act in accordance with the environmental policy of the Government, working towards the preservation of the environment and for sustainable development. In this context, arise the attempt to identify the existing environmental reality in the Headquarters of the Military Academy, an institution dedicated to graduate future Army officers and National Guard offices. Thus, this research aims to assess the economic impact resultant of the adoption of environmental measures, according to the actual surroundings.
This work was divided into two distinct parts: the first refers to the theoretical framework and the second contains the results of the field work. For the first stage, was made a collection of data to achieve the objectives previously stated, that consisted essentially in a documentary analysis from an exhaustive literature research. In a second stage, interviews and questionnaires were prepared and the proposals were analyzed for the implementation of environmental protection measures in order to establish its economic viability.
According to the analysis of data collected by the aforementioned methods of investigation, it was concluded that there is currently a weak environmental performance by the Military Academy Headquarters, reason why it urges the establishment of a policy that sets out clear guidance on to reverse this situation. These should focus on consuming energy and water more efficiently, in the proper separation of waste, in green public procurement and training and in the awareness of individuals who daily attend the space of the Headquarters Military Academy Headquarters. Concerning the economic impact that underlies these measures, has been proved that they are closely related to tapping new sources of income, must therefore be seen as priorities to be defined
A Socioeconomic Well-Being Index
An annual well-being index constructed from thirteen socioeconomic factors is
proposed in order to dynamically measure the mood of the US citizenry.
Econometric models are fitted to the log-returns of the index in order to
quantify its tail risk and perform option pricing and risk budgeting. By
providing a statistically sound assessment of socioeconomic content, the index
is consistent with rational finance theory, enabling the construction and
valuation of insurance-type financial instruments to serve as contracts written
against it. Endogenously, the VXO volatility measure of the stock market
appears to be the greatest contributor to tail risk. Exogenously,
"stress-testing" the index against the politically important factors of trade
imbalance and legal immigration, quantify the systemic risk. For probability
levels in the range of 5% to 10%, values of trade below these thresholds are
associated with larger downward movements of the index than for immigration at
the same level. The main intent of the index is to provide early-warning for
negative changes in the mood of citizens, thus alerting policy makers and
private agents to potential future market downturns
Implementing Modifed Burg Algorithms in Multivariate Subset Autoregressive Modeling
The large number of parameters in subset vector autoregressive models often leads one to procure fast, simple, and efficient alternatives or precursors to maximum likelihood estimation. We present the solution of the multivariate subset Yule-Walker equations as one such alternative. In recent work, Brockwell, Dahlhaus, and Trindade (2002), show that the Yule-Walker estimators can actually be obtained as a special case of a general recursive Burg-type algorithm. We illustrate the structure of this Algorithm, and discuss its implementation in a high-level programming language. Applications of the Algorithm in univariate and bivariate modeling are showcased in examples. Univariate and bivariate versions of the Algorithm written in Fortran 90 are included in the appendix, and their use illustrated
O medo de falhar nos escalões jovens: O papel dos pais, a pressão competitiva/resultados e a sua influência no processo de formação desportiva
Neste relatório, é apresentado e analisado o processo de estágio realizado na equipa de iniciados A do Grupo Desportivo Rio de Mouro, Rinchoa e Mercês, na época desportiva 2017/2018, que esteve focado acima de tudo na componente da formação desportiva. Na área 1 (i.e., Gestão do processo de treino e competição), relacionada com o processo de treino, foi realizada uma planificação concetual, isto é, uma análise da época anterior e um planeamento centrado no modelo de jogo, numa periodização mais tática realizada essencialmente por microciclos, por permitir um melhor ajuste semana a semana de acordo com os problemas do jogo, tendo sido desenvolvido um microciclo padrão, para criar rotinas e facilitar a organização e gestão do processo de treino neste contexto condicionado em diversos aspetos. Na área 2 (i.e., Inovação e investigação), foi realizado um estudo de investigação para averiguar a existência do medo de falhar no plantel, pois é claramente um dos grandes problemas que surge na formação dos jovens atletas, devendo ser diagnosticado e compreendida a sua origem, para que o treinador e sua equipa técnica possam agir no sentido de o combater. Na área 3 (i.e., Relação com a comunidade), foi realizado um evento/reunião com os pais/encarregados de educação, visto que estes agentes desportivos têm uma tremenda influência no processo de formação dos jovens, que lhes é despercebida, tendo também estes agentes desportivos grandes responsabilidades no medo de falhar demonstrado pelas crianças e jovens, e por isso, devem também eles ser formados sobre o seu envolvimento no processo de formação desportiva dos seus filhos.In this report, it is presented and analyzed the internship process executed with the initiates A team of Grupo Desportivo Rio de Mouro, Rinchoa and Mercês, in the 2017/2018 sports season, which was focused above all on the component of sports formation. In area 1 (i.e., Management of the training and competition process), related to the training process, a conceptual planning was carried out, that is, an analysis of the previous season and a planning centered on the game model, in a more tactical periodization performed essentially by microcycles, for allowing a better adjustment week to week according to the problems of the game, having developed a standard microcycle, to create routines and facilitate the organization and management of the training process in this context, conditioned in several aspects. In area 2 (i.e., Innovation and research), a research study was carried out to ascertain the existence of fear of failure in the squad, as it is clearly one of the major problems that arise in the training of young athletes, and its origin must be diagnosed and understood, so that the coach and his technical team can act to combat it. In area 3 (i.e., Relation with the community), an event / meeting was held with parents/guardians, as these sports agents have a tremendous influence on the training process of young people, that is unnoticed by them, and these sports agents also have great responsibilities in the fear of failure shown by children and young people, and therefore, they too must be educated about their involvement in their children's sports training proces
Medidas Provisórias e pertinência temática : a importância do controle do “contrabando” legislativo como garantia da higidez da produção normativa infraconstitucional

Evidencia-se a forte dependência presidencial em relação às medidas provisórias e a pouca efetividade dos mecanismos de controle a elas associados, o que permite que diversas práticas abusivas sejam cometidas, tanto pelo Executivo quanto pelo Legislativo. Nesse passo, o trabalho examina uma dessas práticas, o “contrabando” legislativo. Trata-se de uma prática perniciosa que utiliza as medidas provisórias como carona para viabilizar, em flagrante desrespeito ao devido processo legislativo, a aprovação de matérias de interesse dos parlamentares, comprometendo a legitimidade e a higidez da produção legislativa infraconstitucional.In a multidisciplinary approach, this study puts into perspective the institute of provisional measure. Highlights the strong presidential dependence on provisional measures and the little effectiveness of control mechanisms associated with them, which allows many abuses are committed by both the Executive and the Legislature. In this step, the paper examines one such practice, the legislative smuggling. It is a pernicious practice, which affronts due process of law, and uses the provisional measures as lift to facilitate the approval of matters of interest to parliamentarians, compromising the legitimacy and healthiness of legislative production
Contribution of the adenine nucleotide carrier, porin, and sphingolipid metabolism to mitochondria membrane permeabilization in Saccharomyces cerevisiae
Tese de doutoramento do Programa Doutoral em Biologia Molecular e Ambiental (especialidade em Biologia Celular e Saúde)A decisive event in the cell‟s life-or-death decision is the mitochondrial outer
membrane permeabilization (MOMP). The biochemical events responsible for MOMP,
considered by many investigators the “point-of-no-return” in cell death, are not entirely
defined. Two major and very distinct mechanisms have been implicated in the control
of MOMP: i) the action of Bcl-2 family proteins, which can directly engage the outer
mitochondria membrane (OMM) and induce the opening of pores; and ii) the
mitochondrial permeability transition pore (PTP), an inner membrane unselective
channel that induces mitochondria swelling upon long term openings, and eventual
rupture of the OMM.
The growing interest in cell death biology, fostered by the relevant contributions of
yeast to the understanding of basic biological processes, brought the unicellular
eukaryote Saccharomyces cerevisiae into the scene. Yeast cells lack some of the major
regulators of apoptosis, such as Bcl-2 family proteins (though one related protein was
recently identified), but still possess homologues of mitochondrially enclosed proapoptotic
factors, as well as orthologues of the molecular components generally
ascribed to PTP, including the ADP/ATP carrier (AAC) and Porin (Por1). These
particular features of S. cerevisiae, along with the availability of genetic and molecular
tools and easy manipulation, provided an excellent opportunity to study Bcl-2 family
members in a “controlled” environment, or the contribution of the PTP and its
components to cell death.
In this work, the particular contribution of AAC‟s thiol goups, its oxidation, Por1,
and of a possible interaction between both proteins to cell death, using the wellestablished
model of acetic acid-induced yeast death, was explored. It was observed that
the cysteine residues of the Aac2p, when this AAC isoform was individually expressed,
do not contribute to the mechanisms of acetic acid-induced death. This finding indicates
that the oxidative modifications of Aac2p may not be implicated in the acetic acidinduced
cell death program, at least not those involving thiol crosslinking. This idea is
supported by the apparent absence of a particular Aac2p oxidation pattern.
Nevertheless, the AAC was previously demonstrated to be required for acetic acidinduced
cytochrome c release, and its absence promoted the survival of yeast cells.
Deletion of Por1, on the other hand, decreased the viability of yeast cells treated with
acetic acid. It was hypothesized that the two proteins could share the same pathway in the regulation of cell death. To test this, cytochrome c release was evaluated in
mitochondria isolated from Δaac1/2/3, Δpor1 and Δaac1/2/3Δpor1 cells following
acetic acid treatment. The data obtained suggest that absence of Por1 does not affect
cytcochrome c release, hence MOMP, during acetic acid induced-death, but it may be
important for its regulation. When both the AAC and Por1 were absent, yeast
mitochondria could still release cytochrome c, raising the possibility of an AACindependent
mechanism. Furthermore, we found both proteins have distinct effects that
regulate the cellular response to different stresses. Particularly, absence of the AACs
somehow contributed to increased cell wall resistance and osmotic stress resistance.
Finally, S. cerevisiae was explored as a model to study mechanistic aspects relative
to the function of Bcl-2 family proteins. Particularly, we assessed the role of
sphingolipids, using cells lacking sphingolipid metabolism enzymes, in the action of the
human pro-apoptotic regulator Bax. We found that absence of Isc1p, an inositol
phospholipase C that degrades complex sphingolipids into ceramides in yeast, favored
the viability of yeast cells expressing an active form of Bax. It was further revealed that
this effect is not associated with changes in the action of Bax; rather, it might be related
with the cellular consequences of Bax-action. Indeed, a parallel with the effect of Uth1p
absence in yeast cells expressing Bax suggests that the absence of Isc1p could affect the
selective degradation of mitochondria by mitophagy, and thus produce a different cell
death response.
This work provides new insights into the physiological events underlying the
contribution of mitochondrial proteins, previously associated with cell death responses,
and sphingolipid metabolism to cell death induced by acetic acid and Bax, respectively.
Once again, the yeast S. cerevisiae proved to be an excellent model in the particular
field of cell life and death research.A permeabilização da membrana mitocondrial externa (MOMP) é considerado o
evento decisivo para a sobrevivência celular. Os eventos bioquímicos subjacentes a este
processo, considerado o “ponto sem retorno” na morte celular, não estão ainda
completamente elucidados. Dois dos principais mecanismos apontados como
responsáveis pelo controlo deste processo consistem na acção das proteínas da família
Bcl-2, que podem interactuar directamente com a membrana mitocondrial externa
induzindo a abertura de poros; e o poro de permeabilidade transitória (PTP), um canal
não selectivo que ocorre na membrana mitocondrial interna e que, quando aberto por
longos períodos, induz a tumefacção mitocondrial podendo culminar na eventual
ruptura da membrana mitocondrial externa.
O crescente interesse no estudo da morte celular, potenciado pelas contribuições da
levedura na elucidação de processos celulares fundamentais, despoletou a utilização da
levedura enquanto modelo para a investigação da morte celular. Os principais
reguladores da apoptose, nomeadamente as proteínas da família Bcl-2 (à excepção de
um ortólogo recentemente identificado) não estão presentes na levedura. No entanto,
este organismo possui ortólogos de vários factores apoptóticos, assim como dos
constituintes moleculares do PTP, incluindo o transportador de ADP/ATP (AAC) e a
porina de membrana mitocondrial externa (Por1p). Estas características da levedura S.
cerevisiae, bem como a disponibilidade de ferramentas genéticas e moleculares aliadas
a uma fácil manipulação, fazem deste sistema uma excelente ferramenta para o estudo
da expressão de proteínas da família Bcl-2 num ambiente "controlado", ou do contributo
do PTP e dos seus componentes para a morte celular.
Neste trabalho é estudada a contribuição dos grupos tiol da proteína AAC, e da sua
oxidação assim como do papel das próprias proteínas AAC e Por1p, e uma possível
interacção entre ambas, para a morte celular induzida por ácido acético. Nas condições
testadas observou-se que os resíduos de cisteína do Aac2p, expresso como a única
isoforma do AAC, não contribuem para os mecanismos de morte induzida por ácido
acético. Este resultado sugere que o programa de morte celular induzido por este
composto não depende de modificações oxidativas do Aac2p, pelo menos dependentes
da oxidação de grupos tiol. Esta afirmação é suportada por aparente ausência oxidação
do Aac2p. No entanto, o AAC já foi previamente descrito como elemento essencial para
a libertação de citocromo c da mitocôndria durante a morte celular induzida por ácido acético. Para além deste efeito, a deleção do AAC promove igualmente a sobrevivência
das células de levedura. A ausência do gene POR1, por outro lado, diminui a viabilidade
deste mutante quando exposto ao tratamento com ácido acético.
Em relação a uma possível interação do AAC e da porina Por, foi sugerido que as
duas proteínas poderiam partilhar a mesma via na regulação do processo de morte
celular em levedura. Para testar esta hipótese, a libertação de citocromo c foi avaliada
em mitocôndrias isoladas dos mutantes Δaac1/2/3, Δpor1 e Δaac1/2/3Δpor1 após o
tratamento com ácido acético. Os dados obtidos sugerem que a ausência da proteína
Por1 não bloqueia a libertação de citocromo c, e consequentemente o MOMP, durante a
morte induzida por ácido acético mas pode ser importante para a sua regulação. Na
ausência dos genes AACs e POR1 foi possível observar libertação de citocromo c, o que
aponta para a existência de um mecanismo alternativo de regulação do MOMP nestas
condições. Verificou-se também que estas proteínas têm respostas distintas quando
sujeitas a diferentes stresses celulares. Em particular, a ausência dos AAC contribui, de
alguma forma, para o aumento da resistência ao stress osmótico e da resistência da
parede celular.
Por fim, a levedura S. cerevisiae foi usada como um modelo para estudo das
proteínas da família Bcl-2. Especificamente, avaliou-se o papel dos esfingolípidos, na
acção da proteína pro-apoptótica de mamíferos Bax usando estirpes mutadas no
metabolismo dos esfingolípidos. Foi observado que a ausência da proteína Isc1p, uma
inositol fosfolipase C que degrada esfingolípidos complexos em ceramidas, favorece a
viabilidade de células que expressam uma forma activa da Bax. Mostrou-se ainda que
este efeito parece não estar associado a uma alteração na acção da Bax, podendo, pelo
contrário, estar relacionado com os defeitos subsequentes à acção desta proteína. Assim,
e paralelamente ao que está descrito para ausência da proteína Uth1, a remoção do Isc1p
poderá estar a influenciar a degradação selectiva de mitocôndrias através de um
processo de mitofagia, conduzindo a um processo diferente na execução da morte
celular.
Com este trabalho foi possível aprofundar os conhecimentos sobre os mecanismos
fisiológicos subjacentes à contribuição de proteínas mitocondriais, anteriormente
associadas a processos de morte celular, e do metabolismo de esfingolípidos na morte
celular induzida por ácido acético e Bax, respectivamente. Mais uma vez, a levedura S.
cerevisiae mostrou-se um excelente modelo para compreensão dos processos de morte

A importância da interseccionalidade no movimento sufragista feminino na obra de Angela Davis
O seguinte trabalho tem o objetivo de ensaiar acerca da importância dainterseccionalidade entre gênero, raça e classe no movimento sufragista feminino na obra de Angela Davis; a autora faz, em Mulheres, Raça e Classe, uma análise a partir da história da escravidão e da Guerra Civil nos Estados Unidos, mostrando os problemas da supremacia, tanto dos brancos que escravizaram o povo negro, quanto da burguesia que se aproveitava da classe operaria com o surgimento das indústrias, quanto dos homens em relação às mulheres. Analisaremos como Davis mostra a importância de se vê as lutas como uma “união” e de se vê como tudo está relacionado; a importância da interseccionalidade entre gênero, raçae classe, mostrando de forma dialética como é necessário enxergar essas lutas como um conjunto e não como uma hierarquia. Esse trabalho também visa compreender o caráter de análise ética da visão da autora acerca das construções dos discursos e das ações de alguns dos “personagens” principais do ponto de vista de construção das bases do movimento sufragista feminino; por fim, uma análise do ponto de vista ontológico do significado de “Ser - Mulher” e de como a construção de caráter dos discursos de Soujouner Truth e de Elizabeth Cady Stanton nos dão pontos de vistas diferentes dessa construção, enquanto um discurso é pautado na “conveniência”, de modo que fica ambíguo, o outro nos leva a refletir e a questionar de que mulher que se está falando, permitindo então, uma análise mais fiel da filosofia da Davis, englobando questões de gênero, raça e classe de maneira interseccional no mesmo discurso.
