    Warp/Weft/Word: Inscriptive Materiality, Epistemological Violence, and the Inka Khipu

    Many competing theories of the Indigenous inscription practice known as the khipu have been offered, from L. Leland Locke’s long-standing postulation that khipus are accounting devices, to Walter Ong’s description of them as aide-mémoire, to Gary Urton’s more experimental theory that they constituted an early form of binary composition. Just as fraught is the history of the khipu, which were utilized by the Inka, intermediated by Spanish and Catholic authorities in their legal and religious systems, and, finally, banned and burned as seditious and sacrilegious. Contemporary khipus are primarily limited to those used by herders, but Chilean American poet-artist Cecilia Vicuña utilizes the khipu form to compose artworks such as Quipu in the Gutter, Skyscraper Quipu, Quipu That Remembers Nothing, among many others. This article argues that Vicuña utilizes the formal possibilities of khipu materiality, refusing to either limit her khipu-based works as simply mathematical or to romanticize their history. Instead, she intermediates the khipu form with elements of conceptual, visual, and performance art. In doing so, she participates in the colonial history of the khipu—just as Spanish authorities intermediated khipus with written Spanish in colonial courts and in khipu-based religious confessions. However, Vicuña in turn rejects the colonial violence imposed on the khipu in the same process, indicating that the possibilities of the form continue through the form’s intermediation and growth.     Low Temperature Opacities

    Previous computations of low temperature Rosseland and Planck mean opacities from Alexander & Ferguson (1994) are updated and expanded. The new computations include a more complete equation of state with more grain species and updated optical constants. Grains are now explicitly included in thermal equilibrium in the equation of state calculation, which allows for a much wider range of grain compositions to be accurately included than was previously the case. The inclusion of high temperature condensates such as Al2_2O3_3 and CaTiO3_3 significantly affects the total opacity over a narrow range of temperatures before the appearance of the first silicate grains. The new opacity tables are tabulated for temperatures ranging from 30000 K to 500 K with gas densities from 104^{-4} g cm3^{-3} to 1019^{-19} g cm3^{-3}. Comparisons with previous Rosseland mean opacity calculations are discussed. At high temperatures, the agreement with OPAL and Opacity Project is quite good. Comparisons at lower temperatures are more divergent as a result of differences in molecular and grain physics included in different calculations. The computation of Planck mean opacities performed with the opacity sampling method are shown to require a very large number of opacity sampling wavelength points; previously published results obtained with fewer wavelength points are shown to be significantly in error.     A common framework for collecting evidence of real-life outcomes of aphasia interventions: thoughts and applications

    Two related conceptual frameworks have been developed to guide outcome measurement. The Framework for Outcome Measurement (FROM) captures the integration of key generic concepts central to recent international thinking in the area of health and disability. The second, Living with Aphasia: Framework for Outcome Measurement (A-FROM), focuses directly on communication and is intended to guide thinking about outcome measurement in aphasia. User-friendly and accessible schematics represent potential outcome domains and highlight the centrality of quality of life. The frameworks and methods of development will be discussed. Guidelines for potential applications will be presented with examples from two A-FROM derived tools

    A Computational Simulation Study of Benzamidine Derivatives Binding to Arginine-Specific Gingipain (HRgpA) from Periodontopathogen Porphyromonas gingivalis

    We have shown that the binding free energy calculation from molecular dynamics can be adapted successfully to cysteine proteinases, such as arginine-specific gingipain (HRgpA) from Porphyromonas gingivalis. The binding free energy obtained is in good agreement with the available experimental data for eight benzamidine derivatives including urea and ether linker. The calculations showed that the electrostatic energies between HRgpA and inhibitors were important in determining the relative affinities of the inhibitors to the HRgpA, with an average binding free energy of about −5 kcal/mol. The average structures of the eight complexes suggest that benzamidine inhibitors interact with Asp387, His435, and Cys468 by hydrogen bonding and with Trp508 by hydrophilic interactions that are essential for the activities of benzamidine inhibitors. It can therefore be expected that the method provides a reliable tool for the investigation of new HRgpA inhibitors. This finding could significantly benefit the future design of HRgpA inhibitors

    Ovine Pulmonary Adenocarcinoma: A Unique Model to Improve Lung Cancer Research

    Lung cancer represents a major worldwide health concern; although advances in patient management have improved outcomes for some patients, overall 5-year survival rates are only around 15%. In vitro studies and mouse models are commonly used to study lung cancer and their use has increased the molecular understanding of the disease. Unfortunately, mouse models are poor predictors of clinical outcome and seldom mimic advanced stages of the human disease. Animal models that more accurately reflect human disease are required for progress to be made in improving treatment outcomes and prognosis. Similarities in pulmonary anatomy and physiology potentially make sheep better models for studying human lung function and disease. Ovine pulmonary adenocarcinoma (OPA) is a naturally occurring lung cancer that is caused by the jaagsiekte sheep retrovirus. The disease is endemic in many countries throughout the world and has several features in common with human lung adenocarcinomas, including histological classification and activation of common cellular signaling pathways. Here we discuss the in vivo and in vitro OPA models that are currently available and describe the advantages of using pre-clinical naturally occurring OPA cases as a translational animal model for human lung adenocarcinoma. The challenges and options for obtaining these OPA cases for research purposes, along with their use in developing novel techniques for the evaluation of chemotherapeutic agents or for monitoring the tumor microenvironment in response to treatment, are also discussed

    A Mendelian randomization study of circulating uric acid and type 2 diabetes

    We aimed to investigate the causal effect of circulating uric acid concentrations on type 2 diabetes risk. A Mendelian randomization study was performed using a genetic score with 24 uric acid associated loci. We used data of the EPIC-InterAct case-cohort study, comprising 24,265 individuals of European ancestry from eight European countries. During a mean (SD) follow-up of 10 (4) years, 10,576 verified incident type 2 diabetes cases were ascertained. Higher uric acid associated with higher diabetes risk following adjustment for confounders, with a HR of 1.20 (95%CI: 1.11,1.30) per 59.48 µmol/L (1 mg/dL) uric acid. The genetic score raised uric acid by 17 µmol/L (95%CI: 15,18) per SD increase, and explained 4% of uric acid variation. Using the genetic score to estimate the unconfounded effect found that a 59.48 µmol/L higher uric acid concentration did not have a causal effect on diabetes (HR 1.01, 95%CI: 0.87,1.16). Including data from DIAGRAM consortium, increasing our dataset to 41,508 diabetes cases, the summary OR estimate was 0.99 (95%CI: 0.92, 1.06). In conclusion, our study does not support a causal effect of circulating uric acid on diabetes risk. Uric acid lowering therapies may therefore not be beneficial in reducing diabetes risk

    Anti-cancer therapy is associated with long-term epigenomic changes in childhood cancer survivors

    Childhood cancer survivors (CCS) exhibit significantly increased chronic diseases and premature death. Abnormalities in DNA methylation are associated with development of chronic diseases and reduced life expectancy. We investigated the hypothesis that anti-cancer treatments are associated with long-term DNA methylation changes that could be key drivers of adverse late health effects. Genome-wide DNA methylation was assessed using MethylationEPIC arrays in paired samples (before/after therapy) from 32 childhood cancer patients. Separately, methylation was determined in 32 samples from different adult CCS (mean 22-years post-diagnosis) and compared with cancer-free controls (n = 284). Widespread DNA methylation changes were identified post-treatment in childhood cancer patients, including 146 differentially methylated regions (DMRs), which were consistently altered in the 32 post-treatment samples. Analysis of adult CCS identified matching methylation changes at 107/146 of the DMRs, suggesting potential long-term retention of post-therapy changes. Adult survivors also exhibited epigenetic age acceleration, independent of DMR methylation. Furthermore, altered methylation at the DUSP6 DMR was significantly associated with early mortality, suggesting altered methylation may be prognostic for some late adverse health effects in CCS. These novel methylation changes could serve as biomarkers for assessing normal cell toxicity in ongoing treatments and predicting long-term health outcomes in CCS. [Abstract copyright: © 2022.
