    Work-life balance in a Japanese sample: a person-centered approach

    2020 Fall.Includes bibliographical references.Work-life balance (WLB) has been linked to one's job satisfaction, well-being, and quality of life. Despite its importance, WLB remains elusive to many working people. The present study used a person-centered methodology called Latent Profile Analysis (LPA) to answer three research questions: (1) Can people be categorized into meaningful exclusive and exhaustive latent groups of varying degree of WLB based on their experiences in work and life domains?; (2) To what extent do demographic variables predict membership in certain profile of WLB?; (3) Are identified profiles of WLB related to individuals' well-being? In a sample of over 700 middle-aged workers from Tokyo, Japan, I identified three distinct subgroups that qualitatively differed in their symptomology of balance. I referred to these as the Moderate WLB Profile that was family-oriented and partially engaged in their multiple life roles, the High WLB Profile that was fully engaged and efficient at managing their roles, and the Low WLB Profile that was partially engaged and inefficient at juggling among several life roles. Regarding demographics, age, gender, and marital status seemed to be important predictors of one's latent profile membership. Furthermore, the latent profile membership was predictive of one's well-being. In sum, the study results suggested that WLB is indeed critical to workers' well-being and hence, further efforts to boost balance are needed. One-size-fit-all policies of WLB may not work well for all employees. Understanding workers' circumstances is critical for more targeted interventions/policies to enhance balance

    Detection of Mg2+-dependent, coaxial stacking rearrangements in a bulged three-way DNA junction by single-molecule FRET

    Three-way helical junctions (3WJs) arise in genetic processing, and they have architectural and functional roles in structured nucleic acids. An internal bulge at the junction core allows the helical domains to become oriented into two possible, coaxially stacked conformers. Here, the helical stacking arrangements for a series of bulged, DNA 3WJs were examined using ensemble fluorescence resonance energy transfer (FRET) and single-molecule FRET (smFRET) approaches. The 3WJs varied according to the GC content and sequence of the junction core as well as the pyrimidine content of the internal bulge. Mg2+ titration experiments by ensemble FRET show that both stacking conformations have similar Mg2+ requirements for folding. Strikingly, smFRET experiments reveal that a specific junction sequence can populate both conformers and that this junction undergoes continual interconversion between the two stacked conformers. These findings will support the development of folding principles for the rational design of functional DNA nanostructures

    Petrographic Characteristics and Depositional Environment Evolution of Middle Miocene Sediments in the Thien Ung - Mang Cau Structure of Nam Con Son Basin

    This paper introduces the petrographic characteristics and depositional environment of Middle Miocene rocks of the Thien Ung - Mang Cau structure in the central area of Nam Con Son Basin based on the results of analyzing thin sections and structural characteristics of core samples. Middle Miocene sedimentary rocks in the studied area can be divided into three groups: (1) Group of terrigenous rocks comprising greywacke sandstone, arkosic sandstone, lithic-quartz sandstone, greywacke-lithic sandstone, oligomictic siltstone, and bitumenous claystone; (2) Group of carbonate rocks comprising dolomitic limestone and bituminous limestone; (3) Mixed group comprising calcareous sandstone, calcarinate sandstone, arenaceous limestone, calcareous claystone, calcareous silty claystone, dolomitic limestone containing silt, and bitumen. The depositional environment is expressed through petrographic characteristics and structure of the sedimentary rocks in core samples. The greywacke and arkosic sandstones are of medium grain size, poor sorting and roundness, and siliceous cement characterizing the alluvial and estuarine fan environment expressed by massive structure of core samples. The mixed calcareous limestone, arenaceous dolomitic limestone, and calcareous and bituminous clayey siltstone in the core samples are of turbulent flow structure characterizing shallow bay environment with the action of bottom currents. The dolomitic limestones are of relatively homogeneous, of microgranular and fine-granular texture, precipitated in a weakly reducing, semi-closed, and relatively calm bay environment

    Supramolecular aggregation in dithia-arsoles: chlorides, cations and N-centred paddlewheels

    The benzo-fused dithia-chloro-arsole derivative C6H4S2AsCl (1) is found to crystallise in the triclinic space group P[1 with combining macron] with 17 molecules in the asymmetric unit whereas the tolyl derivative, MeC6H3S2AsCl (2) is polymorphic with the α-phase crystallising in the monoclinic space group P21/c with a single molecule in the asymmetric unit and the β-phase adopting a triclinic structure with two molecules in the asymmetric unit. Reaction of these dithia-chloro-arsole derivatives with LiN(SiMe3)2 in a 3 : 1 mole ratio afforded the unique paddlewheel structure (MeC6H4S2As)3N (4)

    Reaction of the anion-deficient, cation-ordered perovskite phase Ba<sub>2</sub>YFeO<sub>5</sub> with 80 atm of oxygen pressure at 410 °C results in the formation of the Fe<sup>4+</sup> phase Ba<sub>2</sub>YFeO<sub>5.5</sub>. The topochemical insertion of oxide ions lifts the inversion symmetry of the centrosymmetric host phase, Ba<sub>2</sub>YFeO<sub>5</sub> (space group <i>P</i>2<sub>1</sub>/<i>n</i>), to yield a noncentrosymmetric (NCS) phase Ba<sub>2</sub>YFeO<sub>5.5</sub> (space group <i>Pb</i>2<sub>1</sub><i>m</i> (No. 26), <i>a</i> = 12.1320(2) Å, <i>b</i> = 6.0606(1) Å, <i>c</i> = 8.0956(1) Å, <i>V</i> = 595.257(2) Å<sup>3</sup>) confirmed by the observation of second-harmonic generation. Dielectric and PUND ferroelectric measurements, however, show no evidence for a switchable ferroelectric polarization, limiting the material to pyroelectric behavior. Magnetization and low-temperature neutron diffraction data indicate that Ba<sub>2</sub>YFeO<sub>5.5</sub> undergoes a magnetic transition at 20 K to adopt a state which exhibits a combination of ferromagnetic and antiferromagnetic order. The symmetry breaking from centrosymmetric to polar noncentrosymmetric, which occurs during the topochemical oxidation process is discussed on the basis of induced lattice strain and an electronic instability and represents a new strategy for the preparation of NCS materials that readily incorporate paramagnetic transition metal centers

    Structure-property-reactivity studies on dithiaphospholes

    The reaction of either toluene-3,4-dithiol or benzene dithiol with phosphorus(III) trihalides generates the corresponding benzo-fused 1,3,2-dithiaphospholes, RC6H3S2PX (R = Me (1), R = H (2); X = Cl, Br, I). The P-chloro-dithiaphospholes undergo: (a) halogen abstraction reactions with Lewis acids forming phosphenium cations; (b) substitution with LiHMDS base and; (c) reduction chemistry with sodium metal to generate the P–P σ-bonded dimer, (RC6H3S2P)2. Reduction catalysis of aldehydes with pinacolborane using dithiaphospholes is compared with their dioxaphosphole and diazaphosphole counterparts as precatalysts, revealing interesting differences in the reactivity of this series of compounds

    Pichia pastoris versus Saccharomyces cerevisiae:a case study on the recombinant production of human granulocyte-macrophage colony-stimulating factor

    BACKGROUND: Recombinant human granulocyte-macrophage colony-stimulating factor (rhGM-CSF) is a glycoprotein that has been approved by the FDA for the treatment of neutropenia and leukemia in combination with chemotherapies. Recombinant hGM-CSF is produced industrially using the baker's yeast, Saccharomyces cerevisiae, by large-scale fermentation. The methylotrophic yeast, Pichia pastoris, has emerged as an alternative host cell system due to its shorter and less immunogenic glycosylation pattern together with higher cell density growth and higher secreted protein yield than S. cerevisiae. In this study, we compared the pipeline from gene to recombinant protein in these two yeasts. RESULTS: Codon optimization in silico for both yeast species showed no difference in frequent codon usage. However, rhGM-CSF expressed from S. cerevisiae BY4742 showed a significant discrepancy in molecular weight from those of P. pastoris X33. Analysis showed purified rhGM-CSF species with molecular weights ranging from 30 to more than 60 kDa. Fed-batch fermentation over 72 h showed that rhGM-CSF was more highly secreted from P. pastoris than S. cerevisiae (285 and 64 mg total secreted protein/L, respectively). Ion exchange chromatography gave higher purity and recovery than hydrophobic interaction chromatography. Purified rhGM-CSF from P. pastoris was 327 times more potent than rhGM-CSF from S. cerevisiae in terms of proliferative stimulating capacity on the hGM-CSF-dependent cell line, TF-1. CONCLUSION: Our data support a view that the methylotrophic yeast P. pastoris is an effective recombinant host for heterologous rhGM-CSF production

    COVIDiSTRESS Global Survey dataset on psychological and behavioural consequences of the COVID-19 outbreak

    This N = 173,426 social science dataset was collected through the collaborative COVIDiSTRESS Global Survey – an open science effort to improve understanding of the human experiences of the 2020 COVID-19 pandemic between 30th March and 30th May, 2020. The dataset allows a cross-cultural study of psychological and behavioural responses to the Coronavirus pandemic and associated government measures like cancellation of public functions and stay at home orders implemented in many countries. The dataset contains demographic background variables as well as measures of Asian Disease Problem, perceived stress (PSS-10), availability of social provisions (SPS-10), trust in various authorities, trustin governmental measures to contain the virus (OECD trust), personality traits (BFF-15), information behaviours, agreement with the level of government intervention, and compliance with preventive measures, along with a rich pool of exploratory variables and written experiences. A global consortium from 39 countries and regions worked together to build and translate a survey with variables of shared interests, and recruited participants in 47 languages and dialects. Raw plus cleaned data and dynamic visualizations are available.</div

    Structure of general-population antibody titer distributions to influenza A virus.

    Seroepidemiological studies aim to understand population-level exposure and immunity to infectious diseases. Their results are normally presented as binary outcomes describing the presence or absence of pathogen-specific antibody, despite the fact that many assays measure continuous quantities. A population's natural distribution of antibody titers to an endemic infectious disease may include information on multiple serological states - naiveté, recent infection, non-recent infection, childhood infection - depending on the disease in question and the acquisition and waning patterns of immunity. In this study, we investigate 20,152 general-population serum samples from southern Vietnam collected between 2009 and 2013 from which we report antibody titers to the influenza virus HA1 protein using a continuous titer measurement from a protein microarray assay. We describe the distributions of antibody titers to subtypes 2009 H1N1 and H3N2. Using a model selection approach to fit mixture distributions, we show that 2009 H1N1 antibody titers fall into four titer subgroups and that H3N2 titers fall into three subgroups. For H1N1, our interpretation is that the two highest-titer subgroups correspond to recent and historical infection, which is consistent with 2009 pandemic attack rates. Similar interpretations are available for H3N2, but right-censoring of titers makes these interpretations difficult to validate
