    In-peptide synthesis of di-oxazolidinone and dehydroamino acid-oxazolidinone motifs as \u3b2-turn inducers

    Small and easy-to-do mimetics of \u3b2-turns are of great interest to interfere with protein-protein recognition events mediated by \u3b2-turn recognition motifs. We propose a straightforward procedure for constraining the conformation of tetrapeptides lacking a pre-formed scaffold. According to the stereochemistry array, N-Ts tetrapeptides including Thr or PhSer (phenylserine) at the positions 2 or 3 gave rise in a single step to the sequences Oxd 2-Oxd3 or \u394Abu2-Oxd3 (Oxd, oxazolidin-2-one; \u394Abu, 2,3-dehydro-2-aminobutyric). These pseudo-Pro residues displayed highly constrained, and \u3c7 dihedral angles, and induced clear \u3b2-turns or inverse turns of type I or II, as determined by extensive spectroscopic and computational analyses. \ua9 The Royal Society of Chemistry 2013

    Autonomic function in amnestic and non-amnestic mild cognitive impairment : spectral heart rate variability analysis provides evidence for a brain–heart axis

    Mild cognitive impairment (MCI) is a heterogeneous syndrome with two main clinical subtypes, amnestic (aMCI) and non-amnestic (naMCI). The analysis of heart rate variability (HRV) is a tool to assess autonomic function. Cognitive and autonomic processes are linked via the central autonomic network. Autonomic dysfunction entails several adverse outcomes. However, very few studies have investigated autonomic function in MCI and none have considered MCI subtypes or the relationship of HRV indices with different cognitive domains and structural brain damage. We assessed autonomic function during an active orthostatic challenge in 253 oupatients aged\u2009 65\u200965, [n\u2009=\u200982 aMCI, n\u2009=\u200993 naMCI, n\u2009=\u200978 cognitively normal (CN), neuropsychologically tested] with power spectral analysis of HRV. We used visual rating scales to grade cerebrovascular burden and hippocampal/insular atrophy (HA/IA) on neuroimaging. Only aMCI showed a blunted response to orthostasis. Postural changes in normalised low frequency (LF) power and in the LF to high frequency ratio correlated with a memory test (positively) and HA/IA (negatively) in aMCI, and with attention/executive function tests (negatively) and cerebrovascular burden (positively) in naMCI. These results substantiate the view that the ANS is differentially impaired in aMCI and naMCI, consistently with the neuroanatomic substrate of Alzheimer's and small-vessel subcortical ischaemic disease

    Allergic anaphylactic risk in farming activities: A systematic review

    Allergic disorders in the agriculture sector are very common among farm workers, causing many injuries and occupational diseases every year. Agricultural employees are exposed to multiple conditions and various allergenic substances, which could be related to onset of anaphylactic reactions. This systematic review highlights the main clinical manifestation, the allergens that are mostly involved and the main activities that are usually involved. This research includes articles published on the major databases (PubMed, Cochrane Library, Scopus), using a combination of keywords. The online search yielded 489 references; after selection, by the authors, 36 articles (nine reviews and 27 original articles) were analyzed. From this analysis, the main clinical problems that were diagnosed in this category were respiratory (ranging from rhinitis to asthma) and dermatological (eczema, dermatitis, hives) in nature, with a wide symptomatology (from a simple local reaction to anaphylaxis). The main activities associated with these allergic conditions are harvesting or cultivation of fruit and cereals, beekeepers and people working in greenhouses. Finally, in addition to the allergens already known, new ones have emerged, including triticale, wine, spider and biological dust. For these reasons, in the agricultural sector, research needs to be amplified, considering new sectors, new technologies and new products, and ensuring a system of prevention to reduce this risk.peer-reviewe

    ARBORIZAÇÃO URBANA E SUSTENTABILIDADE: UMA PROPOSTA PARA O CENTRO EXPERIMENTAL DE TECNOLOGIAS HABITACIONAIS SUSTENTÁVEIS (CETHS)

    O Centro Experimental de Tecnologias Habitacionais Sustentáveis (CETHS) propôs um assentamento urbano experimental, destinado à população de baixa renda, localizado na área metropolitana de Porto Alegre. O projeto especifica novas tecnologias vinculadas a habitações sustentáveis, incorporando, por um lado, os preceitos do desenvolvimento sustentável e, por outro, a idéia de habitação como extensão do habitat do homem, considerando também o seu contexto social. Desta forma, integra também uma proposta de paisagismo, procurando agregar o maior número de funções possíveis à vegetação nesse espaço. Para tanto, foi elaborada uma pesquisa com o objetivo de produzir informações capazes de orientar especificamente o projeto de arborização urbana do loteamento. A seleção das espécies recomendadas partiu da listagem de espécies apresentada pelo Plano Diretor de Arborização Urbana de Porto Alegre e de cruzamento de informações de revisões bibliográficas, onde chegou-se à indicação de 72 espécies arbóreas, pertencentes a 29 famílias botânicas, todas pertencentes à região. Espera-se, portanto, que este trabalho possa, servir de subsídio e de incentivo para a comunidade que irá habitar o CETHS, no sentido de que as pessoas efetivamente conheçam, respeitem e usufruam das potencialidades das espécies que irão fazer parte do entorno de suas habitações

    Cannabinoid receptor agonist WIN 55,212-2 inhibits rat cortical dialysate gamma-aminobutyric acid levels

    The effects of the cannabinoid receptor agonist WIN 55,212-2 (0.1-5 mg/kg i.p.) on endogenous extracellular gamma-aminobutyric acid (GABA) levels in the cerebral cortex of the awake rat was investigated by using microdialysis. WIN 55,212-2 (1 and 5 mg/kg i.p.) was associated with a concentration-dependent decrease in dialysate GABA levels (-16% +/- 4% and -26% +/- 4% of basal values, respectively). The WIN 55,212-2 (5 mg/kg i.p.) induced-inhibition was counteracted by a dose (0.1 mg/kg i.p.) of the CB(1) receptor antagonist SR141716A, which by itself was without effect on cortical GABA levels. These findings suggest that cannabinoids decrease cortical GABA levels in vivo, an action that might underlie some of the cognitive and behavioral effects of acute exposure to marijuana
