    Differential response of two common human papillomavirus 16 E6 variants to small interfering RNA

    High-risk human papillomaviruses (HPVs), such as HPV16, are implicated in causing virtually all cases of cervical cancer. Molecular therapies capable of targeting HPV16 E6 oncoprotein expression can selectively eliminate infected cells without the unwanted side effects in healthy cells seen with traditional treatments. An efficient approach is the use of synthetic small interfering RNA (siRNA) to target and degrade E6 mRNA. Two common variants of the HPV16 E6 gene, European Prototype (EP E6) and Asian-American (AA E6), differ due to six single nucleotide polymorphisms (SNPs) and AA E6 has been shown to have a greater oncogenic potential than EP E6. In silico modeling was used to demonstrate that these AA E6 SNPs cause its mRNA to have a less compacted structure than that of EP E6. However, transfection of an E6 siRNA containing no base pair mismatches with either variant demonstrated that these structural changes did not result in AA E6 having a significantly different amount of E6 knockdown than EP E6, as determined using RT-qPCR. Interestingly, equivalent knockdown of E6 mRNA in both variants yielded differing responses in the restoration of downstream cellular process abrogated by E6. hTERT mRNA levels were equally restored to those of PHFKs in AA E6. There was a greater elevation of IFN B mRNA expression in EP compared to AA E6. AA E6 IFN K mRNA and p53 protein levels were elevated more following treatment with a lower dose of E6 siRNA than those of EP E6. These data add to the growing body of evidence that the AA E6 protein may have unique properties or disrupt cellular processes through different mechanisms compared to EP E6. These findings also have translational implications (e.g. different doses of E6 siRNA may be required to achieve the same therapeutic effect, depending on the variant involved)

    P-type gallium nitride semiconductor development and characterization for LEDs and other devices

    Migration Enhanced Afterglow (AIEAglow) is proposed as a fabrication technology to produce high indium content indium gallium nitride (IriGaN) LEDs operating in the green wavelengths. This research presents results for the first confirmed p-type gallium nitride films grown in a AIEAglow system for these applications. Films were fabricated at a variety of low temperatures, illustrating the technology’s capability to grow p-GaN at indium preserving conditions for high indium content InGaN LEDs. Films were resistive and exhibited a degree of codoping that stemmed from background oxygen concentration present during growth, rendering Hall effect measurements unsuitable. Electroluminescent spectra exposed three different emission sources: that from the bandgap, that from magnesium to nitrogen vacancy transitions, and the well known yellow defect. Current- Voltage characterization of films grown on top of an ii-GaN template determined the Fermi level inside the p-layer, yielding a net hole concentration of 9.9 x 10[superscript 16] cm [superscript -3] in one film. p-GaN semiconductors were investigated using Scanning Electron Microscopy (SEM), Atomic Force Alicroscopy (AFM), transmission spectroscopy and X-Ray Diffraction (XRD). It was found that growth rate decreased as the growth temperature increased from 545°C to 635°C indicating the desorption of gallium and nitrogen from the lattice at low growth pressures. Conditions used in the fabrication of films were also conducive to producing nanowires and nanostructures, which were shown to correlate with a decrease in luminosity; a worsening of the crystal structure, and is indicative of metal rich material. Attempts were made at optimizing the molecular flow rates of gallium, nitrogen and magnesium to yield better materials. A single heterojuriction GaN-IiiGaN LED was fabricated using the material studied in this thesis. It was shown that a small degree of codoping reduced the resistivity of commercially acquired p-GaN by two orders of magnitude, down to 0.038 [symbol]. cm. This p-GaN was placed below an IriGaN active layer, removing the need for magnesium doping at low temperatures. The low series resistance along the p-GaN material allowed large currents to be applied without much overheating. The high iridium content IiiGaN layer produced yellow light with high electrical conversion efficiency not usually characteristic of nitride or phosphide semiconductor LEDs

    Multi-Step Biomass Fractionation of Grape Seeds from Pomace, a Zero-Waste Approach

    Grape seeds are the wineries' main by-products, and their disposal causes ecological and environmental problems. In this study seeds from the pomace waste of autochthonous grape varieties from Lebanon, Obeidi (white variety) and Asswad Karech (red variety) were used for a multi-step biomass fractionation. For the first step, a lipid extraction was performed, and the obtained yield was 12.33% (w/w) for Obeidi and 13.04% (w/w) for Asswad Karech. For the second step, polyphenols' recovery from the defatted seeds was carried out, resulting in 12.0% (w/w) for Obeidi and 6.6% (w/w) for Asswad Karech, with Obeidi's extract having the highest total phenolic content (333.1 ± 1.6 mg GAE/g dry matter) and antioxidant activity (662.17 ± 0.01 µg/mL of Trolox equivalent). In the third step, the defatted and dephenolized seeds were subsequently extracted under alkaline conditions and the proteins were isoelectric precipitated. The recovered protein extract was 3.90% (w/w) for Obeidi and 4.11% (w/w) for Asswad Karech seeds, with Asswad Karech's extract having the highest protein content (64 ± 0.2 mg protein/g dry matter). The remaining exhausted residue can be valorized in cosmetic scrubs formulations as a replacement for plastic microbeads. The designed zero-waste approach multi-step biomass fractionation has the potential to improve the valorization of the side products (grape seeds) of these two Lebanese autochthonous grape varieties

    Começando por nos situarmos epistemologicamente face à multiplicidade de significados e práticas curriculares associadas à Educação Artística, focamo-nos na prática de Educadores de Infância e Professores do 1o Ciclo do Ensino Básico, colocando-nos na perspetiva de discutir modelos e estratégias que garantam, quer nas práticas quotidianas de Jardins e Escolas, quer na formação de professores: i) a efetiva integração curricular (empreendimento interdisciplinar) das expressões artísticas (Artes Visuais, Música, Teatro e Dança); ii) a capacitação dos docentes dos pontos de vista epistemológico e atitudinal, técnico e didático- pedagógico. Mobilizamos dados e reflexões da experiência e avaliação resultantes do desenvolvimento, desde 2015, de um projeto de intervenção (I-A) em Educação Artística num Agrupamento de Escolas do concelho de Abrantes. Propomos algumas reflexões com base nas quais seja possível melhor compreender e tomar decisões curriculares em Educação Artística.Starting by epistemologically framing our perspective on the multiplicity of meanings and curricular practices associated with Artistic Education, we then focus on the practice of Early Childhood Educators and Primary Teachers.
