    Testing the alkenone D/H ratio as a paleo indicator of sea surface salinity in a coastal ocean margin (Mozambique Channel)

    Reconstructing past ocean salinity is important for assessing paleoceanographic change and therefore past climatic dynamics. Commonly, sea water salinity reconstruction is based on planktonic foraminifera oxygen isotope values combined with sea surface temperature reconstruction. However, the approach relies on multiple proxies, resulting in rather large uncertainty and, consequently, relatively low accuracy of salinity estimates. An alternative tool for past ocean salinity reconstruction is the hydrogen isotope composition of long chain (C37) alkenones (dDalkenone). Here, we have applied dDalkenone to a 39 ka sedimentary record from the Eastern South African continental shelf in the Mozambique Channel, close to the Zambezi River mouth. Despite changes in global seawater dD related to glacial – interglacial ice volume effects, no clear changes were observed in the dDalkenone record throughout the entire 39 ka. The BIT index record from the same core, which provides information on relative contributions of soil organic matter (OM) vs. marine input, indicates high soil OM input during the glacial and low input during the Holocene. This suggests a more pronounced freshwater influence at the core location during the glacial, resulting in alkenones depleted in D during that time, thereby explaining the lack of a clear glacial-interglacial alkenone dD shift. The correlation between the BIT index and dDalkenone during the glacial period suggests that increased continental runoff potentially changed the growth conditions of the alkenone-producing haptophytes, promoting coastal haptophyte species with generally more enriched dDalkenone values. We therefore suggest that the application of dDalkenone for reconstructing past salinity in coastal settings may be complicated by changes in the alkenone-producing haptophyte community

    Acylsucrose-Producing Tomato Plants Forces Bemisia tabaci to Shift Its Preferred Settling and Feeding Site

    [Background] The whitefly Bemisia tabaci (Genn.) causes dramatic damage to plants by transmitting yield-limiting virus diseases. Previous studies proved that the tomato breeding line ABL 14-8 was resistant to B. tabaci, the vector of tomato yellow leaf curl disease (TYLCD). This resistance is based on the presence of type IV glandular trichomes and acylsucrose production. These trichomes deter settling and probing of B. tabaci in ABL 14-8, which reduces primary and secondary spread of TYLCD.[Methodology/Principal Findings] Whitefly settlement preference was evaluated on the adaxial and abaxial leaf surfaces of nearly-isogenic tomato lines with and without B. tabaci-resistance traits, 'ABL 14-8 and Moneymaker' respectively, under non-choice and free-choice conditions. In addition, the Electrical Penetration Graph technique was used to study probing and feeding activities of B. tabaci on the adaxial and abaxial leaf surfaces of the same genotypes. B. tabaci preferred to settle on the abaxial than on the adaxial surface of 'Moneymaker' leaves, whereas no such preference was observed on ABL 14-8 tomato plants at the ten-leaf growth stage. Furthermore, B. tabaci preferred to feed on the abaxial than on the adaxial leaf surface of 'Moneymarker' susceptible tomato plants as shown by a higher number of sustained phloem feeding ingestion events and a shorter time to reach the phloem.     Proteomic Insights into the Hidden World of Phloem Sap Feeding

    The physical interface between a phloem-feeding insect and its host plant is a single cell buried deep within the plant tissue. As such, the molecular interactions between these notorious agricultural pests and the crop plants upon which they feed are diffi cult to study. ‘Omic’ technologies have proved crucial in revealing some of the fascinating detail of the molecular interplay between these partners. Here we review the role of proteomics in identifying putative components of the secreted saliva of phloem-feeding insects, particularly aphids, and discuss the limited knowledge concerning the function of these proteins

    A reference time scale for Site U1385 (Shackleton Site) on the SW Iberian Margin

    Weproduced a composite depth scale and chronology for Site U1385 on the SWIberianMargin. Using log(Ca/Ti)measured by core scanning XRF at 1-cm resolution in all holes, a composite section was constructed to166.5 meter composite depth (mcd) that corrects for stretching and squeezing in each core. Oxygen isotopesof benthic foraminifera were correlated to a stacked d18O reference signal (LR04) to produce an oxygen isotopestratigraphy and age model.Variations in sediment color contain very strong precession signals at Site U1385, and the amplitude modulationof these cycles provides a powerful tool for developing an orbitally-tuned agemodel.We tuned the U1385 recordby correlating peaks in L* to the local summer insolation maxima at 37°N. The benthic d18O record of Site U1385,when placed on the tuned agemodel, generally agrees with other time scaleswithin their respective chronologicuncertainties.The age model is transferred to down-core data to produce a continuous time series of log(Ca/Ti) that reflectrelative changes of biogenic carbonate and detrital sediment. Biogenic carbonate increases during interglacialand interstadial climate states and decreases during glacial and stadial periods. Much of the variance in thelog(Ca/Ti) is explained by a linear combination of orbital frequencies (precession, tilt and eccentricity), whereasthe residual signal reflects suborbital climate variability. The strong correlation between suborbital log(Ca/Ti)variability and Greenland temperature over the last glacial cycle at Site U1385 suggests that this signal can beused as a proxy for millennial-scale climate variability over the past 1.5 Ma.Millennial climate variability, as expressed by log(Ca/Ti) at Site U1385, was a persistent feature of glacial climatesover the past 1.5Ma, including glacial periods of the early Pleistocene (‘41-kyrworld’)when boundary conditionsdiffered significantly from those of the late Pleistocene (‘100-kyr world’). Suborbital variability was suppressedduring interglacial stages and enhanced during glacial periods, especially when benthic d18O surpassed ~3.3–3.5‰.     The unexpectedly short Holocene Humid Period in Northern Arabia

    The early to middle Holocene Humid Period led to a greening of today's arid Saharo-Arabian desert belt. While this phase is well defined in North Africa and the Southern Arabian Peninsula, robust evidence from Northern Arabia is lacking. Here we fill this gap with unprecedented annually to sub-decadally resolved proxy data from Tayma, the only known varved lake sediments in Northern Arabia. Based on stable isotopes, micro-facies analyses and varve and radiocarbon dating, we distinguish five phases of lake development and show that the wet phase in Northern Arabia from 8800-7900 years BP is considerably shorter than the commonly defined Holocene Humid Period (similar to 11,000-5500 years BP). Moreover, we find a two century-long peak humidity at times when a centennial-scale dry anomaly around 8200 years BP interrupted the Holocene Humid Period in adjacent regions. The short humid phase possibly favoured Neolithic migrations into Northern Arabia representing a strong human response to environmental changes

    Brown Planthopper (N. lugens Stal) Feeding Behaviour on Rice Germplasm as an Indicator of Resistance

    BACKGROUND: The brown planthopper (BPH) Nilaparvata lugens (Stal) is a serious pest of rice in Asia. Development of novel control strategies can be facilitated by comparison of BPH feeding behaviour on varieties exhibiting natural genetic variation, and then elucidation of the underlying mechanisms of resistance. METHODOLOGY/PRINCIPAL FINDINGS: BPH feeding behaviour was compared on 12 rice varieties over a 12 h period using the electrical penetration graph (EPG) and honeydew clocks. Seven feeding behaviours (waveforms) were identified and could be classified into two phases. The first phase involved patterns of sieve element location including non penetration (NP), pathway (N1+N2+N3), xylem (N5) [21] and two new feeding waveforms, derailed stylet mechanics (N6) and cell penetration (N7). The second feeding phase consisted of salivation into the sieve element (N4-a) and sieve element sap ingestion (N4-b). Production of honeydew drops correlated with N4-b waveform patterns providing independent confirmation of this feeding behaviour. CONCLUSIONS/SIGNIFICANCE: Overall variation in feeding behaviour was highly correlated with previously published field resistance or susceptibility of the different rice varieties: BPH produced lower numbers of honeydew drops and had a shorter period of phloem feeding on resistant rice varieties, but there was no significant difference in the time to the first salivation (N4-b). These qualitative differences in behaviour suggest that resistance is caused by differences in sustained phloem ingestion, not by phloem location. Cluster analysis of the feeding and honeydew data split the 12 rice varieties into three groups: susceptible, moderately resistant and highly resistant. The screening methods that we have described uncover novel aspects of the resistance mechanism (or mechanisms) of rice to BPH and will in combination with molecular approaches allow identification and development of new control strategies
