    Towards an Agile Biodigital Architecture: Supporting a Dynamic Evolutionary and Developmental View of Architecture

    Architecture and biology are fields of high complexity. Generative design approaches provide access to continuously increasing complexity in design. Some of these methods are based on biological principles but usually do not communicate the conceptual base necessary to appropriately reflect the input from biology into architecture. To address this, we propose a model for analysis and design of architecture based on a multistaged integrated design process that extends the common morphological process in digital morphogenesis with a typology-based ontological model. Biomimetics, an emerging field to strategically search for information transfer from biology to technological application, will assist in delivering a frame of reference and methodology for establishing valid analogies between the different realms as well as integration of the biological concept into a larger framework of analogy to biological processes. As the biomimetic translation of process and systems information promises more radical innovation, this chapter focuses on the dynamic perspectives provided by biological development and evolution to model the complexity of architecture. The proposed process was used to inform five parallel workshops to explore dynamic biological concepts in design. The potential of the process to investigate biomimetic processes in architecture is then discussed, and future work is outlined

    VR in Nursing Facilities - A randomized controlled multicenter pilot study analyzing the changes in the state of mind of seniors in nursing facilities through the viewing of 360° videos

    Context: VR as an application to enhance well-being is sparsely researched in the elderly population. The aim of the pilot study was to analyze the effect of 360° videos of different categories on the state of mind of seniors in nursing facilities. Furthermore, for the implementation in everyday life, the usability of the system and the experience for seniors should be evaluated. Methods: The VR experience was used as a supplement to existing care services in three facilities on eight subjects. Mood state was assessed using the Questionnaire for the Assessment of Happiness before and after the intervention. Demographic data and technology acceptance were collected beforehand. After the intervention, subjects were interviewed about confounding factors and side effects, and nursing home staff were interviewed about the usability of the system and the organizational concept of implementation. Results: There was a positive effect on state of mind. Gender and spatial mobility turned out to be influencing factors. Categories containing people, animals and action achieved the highest increases in the state of mind. Interest in using technical devices correlated negatively with the change in mood state. None of the subjects found the VR goggles distracting or reported motion sickness. Very good usability was indicated by the employees. Conclusion: A very high willingness to use this technology was found among nursing staff and residents. The tendencies of the positive effect of 360° videos on the state of mind, as well as differentiation based on the mentioned characteristics gender and spatial mobility, should be verified by a larger sample to empirically validate the use of this technology to increase the quality of life

    Assessment of Global Ocean Biogeochemistry Models for Ocean Carbon Sink Estimates in RECCAP2 and Recommendations for Future Studies

    The ocean is a major carbon sink and takes up 25%–30% of the anthropogenically emitted CO2. A state-of-the-art method to quantify this sink are global ocean biogeochemistry models (GOBMs), but their simulated CO2 uptake differs between models and is systematically lower than estimates based on statistical methods using surface ocean pCO2 and interior ocean measurements. Here, we provide an in-depth evaluation of ocean carbon sink estimates from 1980 to 2018 from a GOBM ensemble. As sources of inter-model differences and ensemble-mean biases our study identifies (a) the model setup, such as the length of the spin-up, the starting date of the simulation, and carbon fluxes from rivers and into sediments, (b) the simulated ocean circulation, such as Atlantic Meridional Overturning Circulation and Southern Ocean mode and intermediate water formation, and (c) the simulated oceanic buffer capacity. Our analysis suggests that a late starting date and biases in the ocean circulation cause a too low anthropogenic CO2 uptake across the GOBM ensemble. Surface ocean biogeochemistry biases might also cause simulated anthropogenic fluxes to be too low, but the current setup prevents a robust assessment. For simulations of the ocean carbon sink, we recommend in the short-term to (a) start simulations at a common date before the industrialization and the associated atmospheric CO2 increase, (b) conduct a sufficiently long spin-up such that the GOBMs reach steady-state, and (c) provide key metrics for circulation, biogeochemistry, and the land-ocean interface. In the long-term, we recommend improving the representation of these metrics in the GOBMs

    Sialic Acid-Binding Immunoglobulin-like Lectin G Promotes Atherosclerosis and Liver Inflammation by Suppressing the Protective Functions of B-1 Cells.

    Atherosclerosis is initiated and sustained by hypercholesterolemia, which results in the generation of oxidized LDL (OxLDL) and other metabolic byproducts that trigger inflammation. Specific immune responses have been shown to modulate the inflammatory response during atherogenesis. The sialic acid-binding immunoglobulin-like lectin G (Siglec-G) is a negative regulator of the functions of several immune cells, including myeloid cells and B-1 cells. Here, we show that deficiency of Siglec-G in atherosclerosis-prone mice inhibits plaque formation and diet-induced hepatic inflammation. We further demonstrate that selective deficiency of Siglec-G in B cells alone is sufficient to mediate these effects. Levels of B-1 cell-derived natural IgM with specificity for OxLDL were significantly increased in the plasma and peritoneal cavity of Siglec-G-deficient mice. Consistent with the neutralizing functions of OxLDL-specific IgM, Siglec-G-deficient mice were protected from OxLDL-induced sterile inflammation. Thus, Siglec-G promotes atherosclerosis and hepatic inflammation by suppressing protective anti-inflammatory effector functions of B cells

    PowerCube: Design and Development of a 100 W Origami-Inspired Deployable Solar Array for NanoSatellites

    The rapid growth of the capabilities of small satellites have sparked the need for high-power deployable solar arrays. PowerCube addresses this need by proposing a unique solution that can generate up to 100W from a 1U stowed volume. At the core of this design is an innovative origami-inspired architecture, combined with advanced dual-matrix composite materials, to achieve excellent packaging efficiency and self-deployment capabilities. This paper provides an overview of the design of the system and presents the key analyses and breadboarding activities that supported its development. The next milestones in the ESA-funded PowerCube project are discussed, focusing on its qualification campaign. The paper is concluded by an overview of the PowerSat IOD mission, which will demonstrate a high-power 3U satellite, powered by the PowerCube solar array

    Relationship between volume and outcome for congenital diaphragmatic hernia: a systematic review protocol

    Background: Congenital diaphragmatic hernia is a rare and life-threatening anomaly that occurs during fetal development and results in an incomplete or incorrect formation of the diaphragm. Surgical therapy of the diaphragm should be performed after clinical stabilization of the neonate. Higher hospital or surgeon volume has previously been found to be associated with better clinical outcomes for different especially high-risk, low-volume procedures. Therefore, we aim to examine the relationship between hospital or surgeon volume and outcomes for congenital diaphragmatic hernia. Methods: This systematic review protocol has been designed according to the Preferred Reporting Items for Systematic Reviews and Meta-Analysis Protocol. We will perform a systematic literature search in MEDLINE, Embase, CINAHL and Biosis Previews without applying any limitations. In addition, we will search for relevant conference abstracts. We will screen titles and abstracts of retrieved studies, obtain potentially relevant full texts, and assess the eligibility of those full texts against our inclusion criteria. We will include comparative studies analyzing the relationship between hospital or surgeon volume and clinical outcomes. We will systematically assess risk of bias of included studies and extract data on the study design, patient characteristics, case-mix adjustments, statistical methods, hospital and surgeon volume, and outcomes into standardized tables. Title and abstract screening, full-text screening, critical appraisal, and data extraction of results will be conducted by two reviewers independently. Other data will be extracted by one reviewer and checked for accuracy by a second one. Any disagreements will be resolved by discussion. We will not perform a meta-analysis as we expect included studies to be clinically and methodologically very diverse. We will synthesize findings from primary studies in a structured narrative way and using GRADE. Discussion: Given the lack of a comprehensive summary of findings on the relationship between hospital or surgeon volume and outcomes for congenital diaphragmatic hernia, this systematic review will put things right. Results can be used to inform decision makers or clinicians and to adapt medical care. Systematic review registration: PROSPERO (CRD42018090231

    Low levels of IgM antibodies recognizing oxidation-specific epitopes are associated with human non-alcoholic fatty liver disease

    Background: Lipid oxidation of membrane phospholipids is accompanied by the formation of oxidation-specific epitopes (OSE). These epitopes are recognized by specific antibodies and represent danger-associated molecular patterns that are generated during chronic inflammatory processes. In a murine model for hepatic inflammation during non-alcoholic fatty liver disease (NAFLD), increased antibody levels targeting OSE were found to be protective. Here, our aim was to determine an association between OSE-specific antibody titers and NAFLD in humans. Methods: IgM and IgG levels with specificity for various OSE were assessed in the plasma of patients with NAFLD (n = 71) and healthy controls (n = 68). Antibody titers were comprehensively analyzed in patients with NAFLD after classification by histological evaluation of liver biopsies. Statistical analysis was performed to determine significant correlations and odds ratios. To study the specificity for NAFLD, plasma antibody titers were measured in patients with hepatitis C (n = 40) and inflammatory bowel disease (n = 62). Results: IgM titers against OSE were lower in patients with NAFLD compared to controls. Further biopsy-based classification of patients with NAFLD did not show any difference in IgM levels. Plasma IgM titers towards the P1 mimotope demonstrated an inverse correlation with markers for obesity, systemic inflammation, and liver damage. In contrast, hepatitis C and increased disease activity during inflammatory bowel disease was not associated with reduced IgM titers. Conclusions: Our data highlight the importance of immune recognition of OSE by IgM antibodies in the pathophysiology of NAFLD

    Global, neuronal or ÎČ cell-specific deletion of inceptor improves glucose homeostasis in male mice with diet-induced obesity

    Insulin resistance is an early complication of diet-induced obesity (DIO)1, potentially leading to hyperglycaemia and hyperinsulinaemia, accompanied by adaptive beta cell hypertrophy and development of type 2 diabetes2. Insulin not only signals via the insulin receptor (INSR), but also promotes beta cell survival, growth and function via the insulin-like growth factor 1 receptor (IGF1R)3-6. We recently identified the insulin inhibitory receptor (inceptor) as the key mediator of IGF1R and INSR desensitization7. But, although beta cell-specific loss of inceptor improves beta cell function in lean mice7, it warrants clarification whether inceptor signal inhibition also improves glycaemia under conditions of obesity. We assessed the glucometabolic effects of targeted inceptor deletion in either the brain or the pancreatic beta cells under conditions of DIO in male mice. In the present study, we show that global and neuronal deletion of inceptor, as well as its adult-onset deletion in the beta cells, improves glucose homeostasis by enhancing beta cell health and function. Moreover, we demonstrate that inceptor-mediated improvement in glucose control does not depend on inceptor function in agouti-related protein-expressing or pro-opiomelanocortin neurons. Our data demonstrate that inceptor inhibition improves glucose homeostasis in mice with DIO, hence corroborating that inceptor is a crucial regulator of INSR and IGF1R signalling. In male mice with diet-induced obesity, deletion of insulin inhibitory receptor (inceptor) in the whole body, in the brain and in pancreatic beta cells improves glucose homeostasis, underlining a role of inceptor in regulating glucose homeostasis in the brain and pancreas
