Stoichiometry and Change of the mRNA Closed-Loop Factors as Translating Ribosomes Transit from Initiation to Elongation
Protein synthesis is a highly efficient process and is under exacting control. Yet, the actual abundance of translation factors present in translating complexes and how these abundances change during the transit of a ribosome across an mRNA remains unknown. Using analytical ultracentrifugation with fluorescent detection we have determined the stoichiometry of the closed-loop translation factors for translating ribosomes. A variety of pools of translating polysomes and monosomes were identified, each containing different abundances of the closed-loop factors eIF4E, eIF4G, and PAB1 and that of the translational repressor, SBP1. We establish that closed-loop factors eIF4E/eIF4G dissociated both as ribosomes transited polyadenylated mRNA from initiation to elongation and as translation changed from the polysomal to monosomal state prior to cessation of translation. eIF4G was found to particularly dissociate from polyadenylated mRNA as polysomes moved to the monosomal state, suggesting an active role for translational repressors in this process. Consistent with this suggestion, translating complexes generally did not simultaneously contain eIF4E/eIF4G and SBP1, implying mutual exclusivity in such complexes. For substantially deadenylated mRNA, however, a second type of closed-loop structure was identified that contained just eIF4E and eIF4G. More than one eIF4G molecule per polysome appeared to be present in these complexes, supporting the importance of eIF4G interactions with the mRNA independent of PAB1. These latter closed-loop structures, which were particularly stable in polysomes, may be playing specific roles in both normal and disease states for specific mRNA that are deadenylated and/or lacking PAB1. These analyses establish a dynamic snapshot of molecular abundance changes during ribosomal transit across an mRNA in what are likely to be critical targets of regulation
Examining the Effects of a Well-Being Therapy Protocol on Sexual Minority Individuals’ Psychological Well-Being
Sexual minority individuals are at higher risk of mental health concerns than heterosexual individuals due to the impacts of minority stress. Previous counseling interventions for sexual minority individuals have focused on pathology and reducing negative symptoms. However, there is a need for more empirical counseling interventions that address these disparities from a positive psychology perspective. Utilizing a multiple baseline single case research design, the current study examined the impact of a wellbeing therapy protocol for improving the psychological well-being and decreasing depressive symptoms for three sexual minority individuals. There was no functional relationship observed for the Well-Being Therapy protocol on psychological well-being or depressive symptoms in the current study. Discussion of the findings and implications for future research are discussed
White-nose Syndrome at Mammoth Cave National Park: Actions Before and After Its Detection
Since it was identified in the United States in 2006, white-nose syndrome (WNS) in bats has become an important issue in the management of caves and bats at Mammoth Cave National Park (MACA). The threat of its arrival has led to more intense monitoring of bat populations, increased studies, and interventions with both the visiting public and researchers. The timeline of MACA’s WNS response is shown in Table 1
Observing Strategies for the NICI Campaign to Directly Image Extrasolar Planets
We discuss observing strategy for the Near Infrared Coronagraphic Imager
(NICI) on the 8-m Gemini South telescope. NICI combines a number of techniques
to attenuate starlight and suppress superspeckles: 1) coronagraphic imaging, 2)
dual channel imaging for Spectral Differential Imaging (SDI) and 3) operation
in a fixed Cassegrain rotator mode for Angular Differential Imaging (ADI). NICI
will be used both in service mode and for a dedicated 50 night planet search
campaign. While all of these techniques have been used individually in large
planet-finding surveys, this is the first time ADI and SDI will be used with a
coronagraph in a large survey. Thus, novel observing strategies are necessary

Partners in Water Quality Monitoring at Mammoth Cave National Park, Kentucky
Water resources are essential to landscape development and maintenance of the extraordinary ecosystem at Mammoth Cave National Park, Kentucky. The National Park Service has implemented many policies and management practices in an effort to maintain and improve the water quality in the park. As part of their resources management, the Park evaluates current hydrologic conditions, as well as, anticipates and responds to emerging issues. With regards to that goal, Mammoth Cave National Park Service partnered with Tennessee State University, the Mammoth Cave International Center for Science and Learning, and the U.S. Geological Survey on a series of water-related projects from 2007-2013. The objective of this paper is to highlight some of the findings and lessons learned from the past 6 years. Many of the results presented in this paper have been presented at other conferences or published in other reports. Collaborative projects included storm-water runoff from parking lots and roads, evaluating storm-water filters, and transport of chemicals in the caves. These projects purposefully engaged students to provide professional experience and educational outreach opportunities. Over 50 student presentations related to these monitoring activities have been made at regional and national conferences in the past 6 years, resulting in numerous awards and publications. Major funding or in-kind services were provided by the partnering agencies and institutions. Additional funding for supplies and student support was provided by the National Science Foundation (Opportunity for Enhancing Diversity in Geoscience, 2007-8; Undergraduate Research and Mentoring, 2009-13), and, the Department of Energy (Massey Chair – NNSA, 2007-13). Utility of the MMPI–2-RF (Restructured Form) Validity Scales in Detecting Malingering in a Criminal Forensic Setting: A Known-Groups Design
The current study examined the utility of the recently released Minnesota Multiphasic Personality Inventory—2 Restructured Form (MMPI–2-RF; Ben-Porath & Tellegen, 2008) validity scales to detect feigned psychopathology in a criminal forensic setting. We used a known-groups design with the Structured Interview of Reported Symptoms (SIRS; Rogers, Bagby, & Dickens, 1992) as the external
criterion to determine groups of probable malingering versus nonmalingering. A final sample of 125 criminal defendants, who were administered both the SIRS and the MMPI–2-RF during their evaluations, was examined. The results indicated that the two MMPI–2-RF validity scales specifically designed to detect overreported psychopathology, F-r and FP-r, best differentiated between the malingering and nonmalingering groups. These scales added incremental predictive utility to one another in this differentiation. Classification accuracy statistics substantiated the recommended cut scores in the MMPI–2-RF manual (Ben-Porath & Tellegen, 2008) in this forensic setting. Implications for these results in terms of
forensic assessment and detection of malingering are discussed.
