    Monitoring synaptic transmission in primary neuronal cultures using local extracellular stimulation

    Various techniques have been applied for the functional analysis of synaptic transmission in Cultured neurons. Here, we describe a method of studying synaptic transmission in neurons cultured at high-density from different brain regions such as the cortex, striatum and spinal cord. We use postsynaptic whole-cell recordings to monitor synaptic Currents triggered by presynaptic action potentials that are induced by brief stimulations with a nearby extracellular bipolar electrode. Pharmacologically isolated excitatory or inhibitory postsynaptic currents can be reliably induced, with amplitudes, synaptic charge transfers, and short-term plasticity properties that are reproducible from culture to culture. We show that the size and kinetics of pharmacologically isolated inhibitory postsynaptic Currents triggered by single action potentials or stimulus trains depend on the Ca2+ concentration, temperature and stimulation frequency. This method can be applied to study synaptic transmission in wildtype neurons infected with lentiviruses encoding various components of presynaptic release machinery, or in neurons from genetically modified mice, for example neurons carrying floxed genes in which gene expression can be acutely ablated by expression of Cre recombinase. The preparation described in this paper should be useful for analysis of synaptic transmission in inter-neuronal synapses formed by different types of neurons. (c) 2006 Elsevier B.V.     Analysis of all-optically tunable functionalities in sub-wavelength periodic structures by the Fourier modal method

    We propose the nonlinear Fourier Modal Method (FMM) [J. Opt. Soc. Am. B 31, 2371 (2014)] as a convenient and versatile numerical tool for the design and analysis of grating based next generation all-optical devices. Here, we include several numerical examples where the FMM is used to simulate all-optically tunable functionalities in sub-wavelength periodic structures. At first, we numerically investigate a 1-D periodic nonlinear binary grating with amorphous TiO2. We plot the diffraction efficiency in the transmitted orders against the structure depth for normally incident plane wave. Change in diffraction efficiencies for different incident field amplitudes are evident from the plots. We verify the accuracy of our implementation by comparing our results with the results obtained with the nonlinear Split Field-Finite Difference Time Domain (SF-FDTD) method. Next we repeat the same experiment with vertically standing amorphous Titanium dioxide (TiO2) nanowire arrays grown on top of quartz which are periodic in two mutually perpendicular directions and examine the efficiencies in the direct transmitted light for different incident field amplitudes. Our third example includes analysis of a form birefringent linear grating with Kerr medium. With FMM we demonstrate that the birefringence of such a structure can be tuned by all-optical means. As a final example, we design a narrow band Guided Mode Resonance Filter (GMRF). Numerical experiments based on the nonlinear FMM reveal that the spectral tunability of such a filter can be obtained by all-optical means.This work is partially supported by the Academy of Finland (contract 285880)

    Polarization conversion by dielectric subwavelength gratings in conical mounting

    Subwavelength dielectric gratings are examined in total-internal-reflection configuration. It is demonstrated experimentally that such elements, fabricated in TiO2, can perform full polarization conversion from incident TE to TM with nearly 100% efficiency. The dependence of the polarization conversion on the angle of incidence is analyzed. Rigorous diffraction theory is used to cross check the experimental results

    Reported prevalence of health conditions that affect drivers

    Drivers' health and fitness are essential factors in order to predict performance outcomes and to promote road safety. It is a demonstrated fact that drivers with (physical and mental) health impairments or illnesses have more probabilities of being involved in traffic crashes and suffering (or causing) several injuries. The general objective of this study was to examine the prevalence of adverse health conditions that may affect Spanish drivers and its perceived impact on driving fitness. It was used a sample of 1200 Spanish drivers (666 men and 534 women). The only selection criteria were to be in possession of any type of driving license for vehicles other than motorcycles and to drive frequently. As a first result, 42% of the participants considered that sometimes they were not in good health conditions to drive and 1.8% considered it frequently. Furthermore, fatigue, alcohol, negative emotions, drowsiness, headaches, respiratory disease, and fever the most prevalent health conditions impairing driver's fitness. Statistical analysis revealed gender differences in reported health and fitness between male and female drivers. The main cause reported to avoid driving task was to suffer illnesses related to physical health (77.6% of drivers). Based on this study, it can be stated that the Administration should work together with the primary health care system in order to improve drivers' awareness on the risk of driving while suffering symptoms of any common physical or mental illness

    Examining the Relationship Between Genetic Counselors’ Attitudes Toward Deaf People and the Genetic Counseling Session

    Given the medical and cultural perspectives on deafness it is important to determine if genetic counselors’ attitudes toward deaf people can affect counseling sessions for deafness genes. One hundred fifty-eight genetic counselors recruited through the National Society of Genetic Counselors Listserv completed an online survey assessing attitudes toward deaf people and scenario-specific comfort levels discussing and offering genetic testing for deafness. Respondents with deaf/Deaf friends or who work in prenatal or pediatric settings had more positive attitudes toward deaf people than those without deaf/Deaf friends or those working in ‘other’ settings. More positive attitudes toward deaf people correlated with higher comfort level talking about genetic testing for the two scenarios involving culturally Deaf clients; and correlated with higher comfort level offering genetic testing to culturally Deaf clients wishing to have a deaf child. Attitudes and comfort level were not correlated in the scenarios involving hearing or non-culturally deaf clients. These results suggest that genetic counselors’ attitudes could affect information provision and the decision making process of culturally Deaf clients. Cultural sensitivity workshops in genetic counseling training programs that incorporate personal interactions with culturally Deaf individuals are recommended. Additional suggestions for fostering personal interactions are provided

    Spatial chemical distance based on atomic property fields

    Similarity of compound chemical structures often leads to close pharmacological profiles, including binding to the same protein targets. The opposite, however, is not always true, as distinct chemical scaffolds can exhibit similar pharmacology as well. Therefore, relying on chemical similarity to known binders in search for novel chemicals targeting the same protein artificially narrows down the results and makes lead hopping impossible. In this study we attempt to design a compound similarity/distance measure that better captures structural aspects of their pharmacology and molecular interactions. The measure is based on our recently published method for compound spatial alignment with atomic property fields as a generalized 3D pharmacophoric potential. We optimized contributions of different atomic properties for better discrimination of compound pairs with the same pharmacology from those with different pharmacology using Partial Least Squares regression. Our proposed similarity measure was then tested for its ability to discriminate pharmacologically similar pairs from decoys on a large diverse dataset of 115 protein–ligand complexes. Compared to 2D Tanimoto and Shape Tanimoto approaches, our new approach led to improvement in the area under the receiver operating characteristic curve values in 66 and 58% of domains respectively. The improvement was particularly high for the previously problematic cases (weak performance of the 2D Tanimoto and Shape Tanimoto measures) with original AUC values below 0.8. In fact for these cases we obtained improvement in 86% of domains compare to 2D Tanimoto measure and 85% compare to Shape Tanimoto measure.
