    Microbial Contamination of Locally Produced Cheese and Determination of their Antimicrobial Potential in Nigeria

    The high consumptionrate of soft cheese and manner of cheese production in Nigeria prompted the need to determine the microbial quality and  antimicrobial properties of locally produced cheese in Nigeria. A total of 20 cheese samples were obtained from different points in 4 cities in southern Nigeria, 5 cheeses per city. They were investigated for some physico-chemical properties, isolation and microbial counts and determination of antimicrobial potential. There was no significant variation in the  composition of physic-chemical properties of cheese samples from various cities except for the acidity of cheese sample obtained from Ilorin. All the 20 samples (100%) yielded low level of lactic acid bacteria (LAB) with counts ~ 103. Escherichia coli or Klebsiella species were constantly isolated in all the cheese samples. Similarly, yeast and Aspergillus species were isolated either alone or in a mixed culture. The result showed increase in total bacteria count from the point of production to the hawkers.  Antimicrobial potential was not found in cheese against the  microorganisms used in the study. The study identified local cheese (‘wara’) as a high risk food in Nigeria due to the high rate of contamination since they are ready-to-eat food item and no antimicrobial property detected in the soft cheese.Key Words: Cheese; Bacteria; Fungi; Nigeria, Susceptibility Le taux de fromage à pâte molle et les modalités de production de  fromage au Nigeria à forte consommation a incité la nécessité de déterminer la qualité microbienne et propriétés antimicrobiennes de  fromage produit localement au Nigeria. Un total de 20 échantillons de fromage ont été obtenues à partir de différents points dans 4 villes au sud du Nigeria, 5 fromages par ville. Ils ont été étudiés pour certaines  propriétés physico-chimiques, l'isolement et les numérations microbiennes et détermination du potentiel antimicrobien. Il n'y avait aucune variation significative dans la composition des propriétés physico- chimiques des échantillons de fromage à partir de différentes villes à l'exception de l'acidité de l'échantillon obtenu à partir de fromage de la ville d’Ilorin. Tous les 20 échantillons (100%) ont donné un faible niveau de bactéries lactiques (LAB) avec environs 103 espèces. Escherichia coli ou Klebsiellaont été constamment isolés dans tous les échantillons de fromage. De même, des espèces de levures et d'Aspergillus ont été isolés soit seuls, soit dans une culture mixte. Le résultat a montré l’augmentation des bactéries totales compté du point de production aux colporteurs. Potentiel antimicrobien n'a pas été trouvé dans le fromage contre les micro-organismes utilisés dans l'étude. L'étude a identifié fromage local (' wara ‘) comme un aliment à haut risque au Nigeria en raison du taux élevé de contamination, car ils sont prêts à consommer l'aliment et aucune  propriété antimicrobienne détecté dans le fromage à pâte molle.Mots clés: Fromage; bactéries; champignons; Nigeria, sensibilit

    Serial Monodomy in the Gypsy Ant, Aphaenogaster araneoides: Does Nest Odor Reduction Influence Colony Relocation?

    Serial monodomy is the nesting behavior in which a colony of animals maintains multiple nests for its exclusive use, occupying one nest at a time. Among serially monodomous ants, the availability of unoccupied nests reduces the probability and costs of army ant attacks. It has been proposed that nest odors mediate serial monodomy in the gypsy ant, Aphaenogaster araneoides Emery (Hymenoptera: Formicidae), and that colonies avoid returning to previously occupied nests that harbor colony odors. To evaluate this hypothesis, the odors inside the nests of A. araneoides colonies were experimentally reduced through ventilation; the nest movement behaviors of treatment and control colonies were compared. Odor reduction was found to have increased the frequency of movements into and out of the treated nest, without a change in the total occupation duration in the treated nest. Nests with a more open architecture that permitted natural flow of air were reoccupied more quickly than nests with smaller nest entrances. In summary, the openness of the architecture of A. araneoides nests and the ventilation of air through nests alters the use of these nests. These findings further support the working hypothesis that nest-bound odors mediate the pattern of serial monodomy in A. araneoides

    Seroprevalence of HTLV -I/II amongst Blood Donors in Osogbo, Nigeria.

    Background: HTLV type I/II is a blood borne infection that can be transmitted via blood transfusion.Objective: To determine the seroprevalence of human T – lymphotropic virus among blood donors in Osogbo, Nigeria.Methods: Diagnosis of Human T. Lymphotropic virus antigen was carried out on 372 serum samples among blood donors who visited the blood bank/transfusion unit of Ladoke Akintola University of Technology  Teaching Hospital and Our Lady of Fatima Catholic Hospital, Osogbo between January and July 2008 using Enzyme linked immunosorbent  assay techniques (ELISA) as described by the manufacturer. Western  blotting was used to confirm the serum reactive samples from ELISA.Results: Out of 372 samples analyzed, 14 (3.6%) samples were found to be positive for HTLV-I/II (7 HTLV-I and 7 HTLV-II) while 358 (96.4%) samples were negative after confirmation with Western blotting. The seroprevalence of HTLV-I/II among the blood donors in Osogbo, Nigeriawas found to be 3.6%. This has major implication for the blood transfusion service in Nigeria.Conclusion: The study concluded that there is need for screening of blood donor for HTLV-I/II in order to rule out this transfusion related infection.Keywords: human T-lymphotropic virus, T-cell leukaemia,  myelopathy/tropical spasticparaparesis

    GATA2 regulates the erythropoietin receptor in t(12;21) ALL.

    The t(12;21) (p13;q22) chromosomal translocation resulting in the ETV6/RUNX1 fusion gene is the most frequent structural cytogenetic abnormality in children with acute lymphoblastic leukemia (ALL). The erythropoietin receptor (EPOR), usually associated with erythroid progenitor cells, is highly expressed in ETV6/RUNX1 positive cases compared to other B-lineage ALL subtypes. Gene expression analysis of a microarray database and direct quantitative analysis of patient samples revealed strong correlation between EPOR and GATA2 expression in ALL, and higher expression of GATA2 in t(12;21) patients. The mechanism of EPOR regulation was mainly investigated using two B-ALL cell lines: REH, which harbor and express the ETV6/RUNX1 fusion gene; and NALM-6, which do not. Expression of EPOR was increased in REH cells compared to NALM-6 cells. Moreover, of the six GATA family members only GATA2 was differentially expressed with substantially higher levels present in REH cells. GATA2 was shown to bind to the EPOR 5'-UTR in REH, but did not bind in NALM-6 cells. Overexpression of GATA2 led to an increase in EPOR expression in REH cells only, indicating that GATA2 regulates EPOR but is dependent on the cellular context. Both EPOR and GATA2 are hypomethylated and associated with increased mRNA expression in REH compared to NALM-6 cells. Decitabine treatment effectively reduced methylation of CpG sites in the GATA2 promoter leading to increased GATA2 expression in both cell lines. Although Decitabine also reduced an already low level of methylation of the EPOR in NALM-6 cells there was no increase in EPOR expression. Furthermore, EPOR and GATA2 are regulated post-transcriptionally by miR-362 and miR-650, respectively. Overall our data show that EPOR expression in t(12;21) B-ALL cells, is regulated by GATA2 and is mediated through epigenetic, transcriptional and post-transcriptional mechanisms, contingent upon the genetic subtype of the disease

    Presumed stromal graft rejection after automated lamellar therapeutic keratoplasty: case report

    PURPOSE: To describe the development of presumed immune-mediated stromal rejection after automated lamellar therapeutic keratoplasty (ALTK) and its reversal after initiation of intensive topical corticosteroid therapy. METHODS: Observational case report. RESULTS: Stromal edema localized in the graft developed 42 days after ALTK for Avellino corneal dystrophy in a 65-year-old man. After one week of intensive topical corticosteroids, complete reversal of graft edema occurred, with full recovery of visual function. CONCLUSION: The clinical appearance and response to therapy in this case supported the diagnosis of immune-mediated stromal rejection. Ophthalmologists should be aware that stromal rejection may occur in lamellar corneal grafts

    Interleukin-6 gene (IL-6): a possible role in brain morphology in the healthy adult brain

    Background: Cytokines such as interleukin 6 (IL-6) have been implicated in dual functions in neuropsychiatric disorders. Little is known about the genetic predisposition to neurodegenerative and neuroproliferative properties of cytokine genes. In this study the potential dual role of several IL-6 polymorphisms in brain morphology is investigated. Methodology: In a large sample of healthy individuals (N = 303), associations between genetic variants of IL-6 (rs1800795; rs1800796, rs2069833, rs2069840) and brain volume (gray matter volume) were analyzed using voxel-based morphometry (VBM). Selection of single nucleotide polymorphisms (SNPs) followed a tagging SNP approach (e.g., Stampa algorigthm), yielding a capture 97.08% of the variation in the IL-6 gene using four tagging SNPs. Principal findings/results: In a whole-brain analysis, the polymorphism rs1800795 (−174 C/G) showed a strong main effect of genotype (43 CC vs. 150 CG vs. 100 GG; x = 24, y = −10, z = −15; F(2,286) = 8.54, puncorrected = 0.0002; pAlphaSim-corrected = 0.002; cluster size k = 577) within the right hippocampus head. Homozygous carriers of the G-allele had significantly larger hippocampus gray matter volumes compared to heterozygous subjects. None of the other investigated SNPs showed a significant association with grey matter volume in whole-brain analyses. Conclusions/significance: These findings suggest a possible neuroprotective role of the G-allele of the SNP rs1800795 on hippocampal volumes.     A Comparison of Wood Density between Classical Cremonese and Modern Violins

    Classical violins created by Cremonese masters, such as Antonio Stradivari and Giuseppe Guarneri Del Gesu, have become the benchmark to which the sound of all violins are compared in terms of their abilities of expressiveness and projection. By general consensus, no luthier since that time has been able to replicate the sound quality of these classical instruments. The vibration and sound radiation characteristics of a violin are determined by an instrument's geometry and the material properties of the wood. New test methods allow the non-destructive examination of one of the key material properties, the wood density, at the growth ring level of detail. The densities of five classical and eight modern violins were compared, using computed tomography and specially developed image-processing software. No significant differences were found between the median densities of the modern and the antique violins, however the density difference between wood grains of early and late growth was significantly smaller in the classical Cremonese violins compared with modern violins, in both the top (Spruce) and back (Maple) plates (p = 0.028 and 0.008, respectively). The mean density differential (SE) of the top plates of the modern and classical violins was 274 (26.6) and 183 (11.7) gram/liter. For the back plates, the values were 128 (2.6) and 115 (2.0) gram/liter. These differences in density differentials may reflect similar changes in stiffness distributions, which could directly impact vibrational efficacy or indirectly modify sound radiation via altered damping characteristics. Either of these mechanisms may help explain the acoustical differences between the classical and modern violins

    Beneficial effects of physical activity in an HIV-infected woman with lipodystrophy: a case report

    <p>Abstract</p> <p>Introduction</p> <p>Lipodystrophy is common in patients infected with human immunodeficiency virus receiving highly active antiretroviral therapy, and presents with morphologic changes and metabolic alterations that are associated with depressive behavior and reduced quality of life. We examined the effects of exercise training on morphological changes, lipid profile and quality of life in a woman with human immunodeficiency virus presenting with lipodystrophy.</p> <p>Case presentation</p> <p>A 31-year-old Latin-American Caucasian woman infected with human immunodeficiency virus participated in a 12-week progressive resistance exercise training program with an aerobic component. Her weight, height, skinfold thickness, body circumferences, femur and humerus diameter, blood lipid profile, maximal oxygen uptake volume, exercise duration, strength and quality of life were assessed pre-exercise and post-exercise training. After 12 weeks, she exhibited reductions in her total subcutaneous fat (18.5%), central subcutaneous fat (21.0%), peripheral subcutaneous fat (10.7%), waist circumference (WC) (4.5%), triglycerides (9.9%), total cholesterol (12.0%) and low-density lipoprotein cholesterol (8.6%). She had increased body mass (4.6%), body mass index (4.37%), humerus and femur diameter (3.0% and 2.3%, respectively), high-density lipoprotein cholesterol (16.7%), maximal oxygen uptake volume (33.3%), exercise duration (37.5%) and strength (65.5%). Quality of life measures improved mainly for psychological and physical measures, independence and social relationships.</p> <p>Conclusions</p> <p>These findings suggest that supervised progressive resistance exercise training is a safe and effective treatment for evolving morphologic and metabolic disorders in adults infected with HIV receiving highly active antiretroviral therapy, and improves their quality of life.</p

    Physician reported perception in the treatment of high blood pressure does not correspond to practice

    <p>Abstract</p> <p>Background</p> <p>High blood pressure is a significant health problem world-wide. Physician factors play a significant role in the suboptimal control of hypertension in the United States. We sought to better understand primary care physician's opinions regarding use of hypertension guidelines, patient and physician related barriers to treatment and physician treatment decision making in the management of hypertension as part of a first step in developing research tools and interventions designed to address these issues.</p> <p>Methods</p> <p>An IRB approved survey pertaining to physician opinion regarding the treatment of hypertension. Items consisted of questions regarding: 1) knowledge of hypertension treatment guidelines; 2) barriers to hypertension control (physician vs. patient); and 3) self-estimation of physician treatment of hypertension. Descriptive Statistics were used to describe results.</p> <p>Results</p> <p>All physicians were board certified in family or general internal medicine (n = 28). Practices were located in urban (n = 12), suburban (n = 14) and inner city locations (n = 1). All physicians felt they did a good job of treating hypertension. Most physicians felt the biggest barrier to hypertension control was patient non-compliance. Half of physicians would fail to intensify treatment for hypertension when blood pressure was above recommended levels for all disease states studied (essential hypertension, heart disease, diabetes, and renal disease).</p> <p>Conclusion</p> <p>Physician ability to assess personal performance in the treatment of hypertension and physician opinion that patient noncompliance is the greatest barrier to optimal hypertension control is contradictory to reported practice behavior. Optimal blood pressure control requires increased physician understanding on the evaluation and management of blood pressure. These data provide crucial formative data to enhance the content validity of physician education efforts currently underway to improve the treatment of blood pressure in the primary care setting.</p
